약모밀(Houttuynia cordata) 뿌리에서 추출한 순차분획물의 항균활성

  • Antimicrobial activities, extraction yields and chemical compositions of the extracts from the powder of vacuum dried Houttuynia cordata root on various solvents were investigated. Antimicrobial activity was investigated by disc diffusion method against several microorganisms, nine species of Gram positive and thirteen Gram negative bacteria. Ethanol, ethylacetate and butanol extracts exhibited potent antimicrobial activities against B. megaterium C. xerosis, S. aureus, S. epidermidis and E.coli. Chloroform and aqueous extracts from H. cordata showed weak antimicrobial activity against the tested bacteria. Ethylacetate and butanol extracts exhibited strong antimicrobial activity against the tested microorganisms. The extraction yields were 15.7%, 3.7%, 0.13%, 0.5% and 5.9% in ethanol, chloroform, ethylacetate, butanol and aqueous fractions, respectively. To further elucidate the effective components, solvent extracts was analyzed by GC/MSD.

  • To investigate the function and effectiveness of natural antimicrobial agent(NAA), as freshness preserving material, extracted from medicinal herbs, their antimicrobial activities were tested against Bacillus cereus, Corynebacterium xerosis, Peudomonas syringae and Enterobacter aerogenes causing the postharvest decay of greenhouse produce. The ethanol extracts of Polygonum cuspidatum Sieb. et Zucc and Syzyrium aromaticum (L.) Merr. et Perry showed remarkable inhibitory effect on the growth of above-mentioned reference microorganisms when treated with mon than 500ppm. The antimicrobial activities of NAA were very stable in the wide spectrum of temperature (40$^{\circ}C$-180$^{\circ}C$) and pH(4-10).

  • Some surfactants known as biosurfactants, are produced biologically by yeast and bacteria from various substances. They are more effective and environmentally friendly than many other synthetic surfactants. Antimicrobial activities of sophorolipids produced by Candida bombiocola were investigated against various microorganisms. Minimal Inhibitory concentration (MIC) of sophorolipid against S. mutans, C. xerosis, and P. acnes were 0.005, 0.05, and 0.005%, respectively. The antimicrobiological activities were more effective than those of SLS and APG. Skin exfoliating and moisturizing effects of vehicles with the sophorolipids were tested. The skin turnover time of aqueous solution with 0.25% sophorolipids was similar to that of aqueous solution with 5.0% lactic acid. Higher moisturizing effects on skin showed as the concentration of sophorolipids increased. We suggest that the sophorolipids can be used for cosmetics as an antimicrobiological agent and an active materials of skin moisturizing and exfoliating.

  • The antimicrobial effect of mixed solutions of grapefruit seed extract(250ppm) and ascorbic acid(1%) on the spoilage microorganisms such as Staphylococcus epidermis, Escherichia coli, Pseudomonas syringae, Candida albicans and Corynebacterium xerosis isolated from the spoiled soybean sprouts were investigated. Cell wall and membrane were partially destroyed and the contents of the destroyed cell were exuded after treatment. Packages with 30 m cast polypropylene(CPP), 16 m polyolefin(RD 106) and 10 m high density polyethylene(HDPE) were applied for soybean sprouts dipped in mixed solutions respectively. Oxygen and carbon dioxide concentration inside packages were dependent on the kind of films during storage at 5oC. The antimicrobial activity of mixed solutions was maintained for 5 days at CPP package. Package with HDPE showed a severe browning than the others after 5 days. Ascorbic acid content of mixed solution treatment was higher than that of control for each package.

  • Antimicrobial activity and chemical composition of volatile flavor extracts from Agastache rugosa were investigated. The volatile flavor extracts were obtained from leaves and stems of Agastache rugosa by simultaneous distillation extraction (SDE) method. Antimicrobial activity was investigated by disc diffusion and broth dilution methods against several microorganisms of Bacillus cereus, bacillus megaterium, Bacillus subtilis, Corynebacterium xerosis, Staphylo coccus aureus, Staphylococcus epidermidis, Agrobacterium rhizogenes , Agrobacterium tumefaciences, Enterobacter cloacae, Escherichia coli, Salmonella typhi, Vibrio parahaemolyticus, Candida utilis and Saccharomyces cerevisiae. Volatile flavor extractsfrom leaves have strong antimicrobial activity against C.utilis and S.cerevisiae. When 0.12% volatile flavor extracts from fresh leaves were included in the medium, lag phase of C. utilis was extended 6 hr and that of S.utilis and S.cerevisiae was extended 2hr. Further analyses were performed to elucidatethe effective component of the extracts. The major component of volatile flavor was estragole, a phenolic compound. Minor components were determined to be terpenes , alcohols, acids , esters, ketones and aldethydes.

  • Sophorolipid is a biological surfactant of the glycolipid structure produced by Candida bombicola, which generally exists as a mixture of acidic and lactonic forms. In this study, we investigated physico-chemical properties, antibacterial activities, and cytotoxicity of the sophorolipid containing more than 96% of the lactonic form, produced by the gene regulation of production strains and application of a metabolic engineering technique. The lactonic sophorolipid showed a weak acidity in the range of pH 3.2~4.6 when diluted in water at the concentrations from 1 to 0.001 wt%. The $pK_a$ value of the lactonic sophorolipid was estimated to be around 4.3 from the acid-base titration curve. The critical micelle concentration (CMC) of the lactonic sophorolipid was $10^{-2}wt%$, at which the surface tension of aqueous solution was reduced to 36 mN/m. The lactonic sophorolipid showed the minimum inhibitory concentrations (MIC) of $1{\times}10^{-3}$ and $5{\times}10^{-3}g/mL$ against Propionibacterium acnes and Corynebacterium xerosis, respectively. The MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide] assay showed that cytotoxicity of the lactonic sophorolipid was ten times lower than that of triclosan.

Chemical Characterization and Antibacterial Effect of Volatile Flavor Concentrate from Houttyunia cordata Thunb (어성초의 화학적 특성과 휘발성 향기성분 추출물의 항균효과)

  • For developing natural antibacterial agents from Houttuynia cordata Thunb., antibacterial effects of volatile flavor component using various bacterial sp. were tested. Extraction from Houttuynia cordata Thunb. by using SDE (Simultaneous steam Distillation-Extraction) showed strong antibacterial activities against Vibrio and Bacillus genus, such as Vibrio. cholerae, V. parahaemolyticus, V. vulnificus, Bacillus. cereus, and B. subtilis. Then chemical compositions of leaf and stem were analyzed. The contents of crude protein, lipid, and ash in stem were less than those of leaf, but fiber contents were higher than those of leaf. Among the amino acids, aspartic acid, glutamic acid, glycine, and arginine were higher than those of other amino acids. Linolenic acid, linoleic acid, oleic acid, and palmitic acid were major fatty acids. Major minerals of Houttuynia cordata Thunb. were potassium, calcium, phosphorus, magnesium, iron, zinc, and copper. Especially, in the case of potassium, it was highest.

Compound K Activates Hyaluronan Synthase 2 in transformed human Keratinocytes and Fibroblasts and Increases hyaluronan in hairless mouse skin

  • Ginsenosides, the major active ingredients of ginseng, show a variety of biomedical efficacies such as anti-aging, anti oxidation and anti-inflammatory activities. To understand the effects of compound K (20-O-D-glucopyranosyl-20 (S)-protopanaxadiol), one of the major metabolite of ginsenosides on the skin, we assessed the expression level of ∼ 100 transcripts in compound K-treated HaCaT cells using cDNA microarray analysis. Compound K treatment induced differential expression of 21 genes, which have been reported to be involved in the organization of ECM structure as well as defense responses in human skin cells. One of the most interesting findings is 2-fold increase in hyaluronan synthase2 (HAS2) gene expression by compound K. We found that change in expression of HAS2 gene represents a specific response of HaCaT cells to compound K because hyaluronan synthase 1, 3 was not changed by treatment with compound K. We also demonstrated that the compound K effectively induced hyaluronan synthesis in human skin cells and hairless mouse skin. The human clinical study indicates that topical application of compound K-containing oil-in-water emulsion showed improvement of xerosis, wrinkle and fine lines in the aged skin. We concluded that compound K has anti-aging effects by the induction of HAS2 gene expression and following hyaluronan synthase.

Anti-microbial Activity of Various Herbal Extracts on Six Types of Bacteria Related to Skin Diseases and Effects of Saussurea lappa Exteract on Inflammatory Mechanism (數種의 한약이 피부질환과 관련된 균주 6종에 미치는 항균력 및 木香이 염증기전에 미치는 영향)

  • This study was carried out to investigate the anti-microbial of various herbal extracts on six types of bacteria related to skin diseases and effects of Saussurea lappa extract on dermatitis and acne. The results are followed ; 1. In the experiment on P. acnes using paper disk methods, Eugenia caryophyllata extract made 19mm clear zone, Saussurea lappa extract and Morus alba extract made 13mm and MIC of Morus alba extract and Eugenia caryophyllata extract was below 0.01$\%$. 2. In the experiment on S. aureus, extract of Eugenia caryophyllata, Poncirus trifoliata and Rubus coreanus made 9-10mm clear zone. 3. In the experiment on C. xerosis, Sesamum indicum extract made 16mm clear zone and MIC of that was 0.3$\%$. 4. In the experiment on C. albicans, extract of Cinnamomum cassia, Eugenia caryophyllata and Asparagus cochinchinensis made 9-10mm clear zone. 5. In the experiment on P. ovale, Cinnamomum cassia extract made 25mm clear one, MIC of that was 0.05$\%$. 6. In the experiment on T. mentagrophytes, Cinnamomum cassia extract made 26mm clear zone, extract of Eugenia caryophyllata, Rhizoma kaempferiae, Piper longum, Saussurea lappa and Zingiber officinale made 18-22mm clear zone, MIC of all extracts was below 0.02$\%$. 7. Saussurea lappa extract inhibited 85$\%$ and 28$\%$ of NO production at 50㎍/㎖ and 5 ㎍/㎖ 8. Saussurea lappa extract inhibited activity of COX Ⅱ over 50$\%$ at 10㎍/㎖ 9. Saussurea lappa extract had not radical scavenging activity and Saussurea lappa extract didn't inhibit manifestation of IL-1${\beta}$, IL-6 and TNF-${\alpha}$, and activity of 50- reductase I.

  • Paeonia lactiflora Pall. was extracted with water and the Paeonia lactiflora Pall. extract (PLE) was tested for antimicrobial activities against Corynebacterium xerosis, Candida albicans, Listeria monocytogenes and Pseudomonas syringae. PLE showed pronounced antimicrobial effects at concentrations at or above 50 ${\mu}g$/mL. The activities were stable at $100^{\circ}C$ and over the pH range of 3-11. PLE may serve as a natural antimicrobial agent in food preservation. It is suggested that hydrophillic components in the extract synergistically perturb microbial membrane functions.
