Effect of Electroacupuncture at SP-6 with Different Durations on Minimum Alveolar Concentration and the Cardiovascular System under Isoflurane Anesthesia in Dogs (개에서 Isoflurane 마취시 SP-6 혈위의 전침자극시간이 최소폐포농도 및 심맥관계에 미치는 영향)

  • The effects of electroacupuncture (EA) at SP-6 with different durations on the minimum alveolar concentration (MAC) and on the cardiovascular system were evaluated in dogs under isoflurane anesthesia. Eight healthy male beagles were randomly assigned to four study groups (n = 5/group) with washout period of 7 days for recovery and anesthetic withdrawal between experiments. Four study groups were control, nonacupoint electrical stimulation (NA), EA for 30 minutes (SP-6) and continuous EA for 70 or 90 minutes (SP-6C). For the nonacupoint electrical stimulation group, needles were inserted into the nonacupoint at the muscle bellies of left triceps brachii and right quadriceps femoris. MAC and cardiovascular parameters were determined after EA at SP-6 acupoint and at nonacupoint. Thirty minutes of EA and continuous EA until re-determination of MAC at SP-6 acupoint lowered the MAC of isoflurane by 21.3$\pm$8.0% and 16.1$\pm$4.6%, respectively (p<0.05). The decrements in MAC values were not significantly different between two EA groups. However, electrical stimulation of nonacupoint did not induce a significant change in MAC. In SP-6 and SP-6C groups, significant changes in cardiovascular parameters were not observed. These results indicate that EA at SP-6 have an advantage in isoflurane anesthesia in terms of reducing the requirement for anesthetics and minimizing cardiovascular side effects. EA for 30 minutes at maximum might be the sufficient time to produce acupuncture analgesia.

Isolation of an Agarase-producing Persicobacter sp. DH-3 and Characterization of its β-agarase (Agarase를 생산하는 Persicobacter sp. DH-3의 분리 및 β-agarase의 특성)

  • The purpose of this study was to isolate a new marine agarase-producing bacterium. Agarase can hydrolyze agar and agarose to produce agarooligosaccharides or neoagarooligosaccharides, which possess many physiological functions. Strain DH-3 was isolated from seawater collected from the coast of Yeosu at Jeollanam province, Korea. A 16S rDNA sequence analysis showed this strain to be Persicobacter sp. DH-3. Extracellular agarase was prepared from culture media of Persicobacter sp. DH-3 and used for characterization. Relative activities at 20, 30, 40, 50, 60, and $70^{\circ}C$ were 50, 55, 70, 100, 90, and 50%, respectively. Relative activities at pH 5, 6, 7, and 8 were 75, 100, 90, and 75%, respectively. The enzyme showed maximum activity at $50^{\circ}C$ in a 20 mM Tris-HCl buffer at pH 6. This enzyme could be useful, as agar is in liquid state at $50^{\circ}C$. Agarase activities were maintained at 80% or more for 2 hr at 20, 30, and $40^{\circ}C$. Thin layer chromatography analysis suggested that Persicobacter sp. DH-3 produced extracellular ${\beta}$-agarases as it hydrolyzed agarose to produce neoagarohexaose and neoagarotetraose. In addition, zymogram analysis confirmed that Persicobacter sp. DH-3 produces at least three agar-degrading enzymes with molecular weights of 45, 70, and 140 kDa. Therefore, it is expected that agarases from Persicobacter sp. DH-3 could be used to produce functional neoagarooligosaccharides.

Molecular Cloning and Analysis of Phosphate Specific Transport (pst) Operon from Serratia marcescens KCTC 2172 (Serratia marcescens KCTC 2172로부터 pst operon의 클로닝 및 해석)

  • A recombinant plasmid, pDH3, was obtained from the genomic library of Serattia marcescens KCTC 2172, and several recombinant subclones constructed from pDH3. The nucleotide sequence of a 5,137 bp segment, pPH4, was determined and three open reading frames were detected. The three ORFs encoded the phosphate specific transport (pst) operon, which was pstC, pstA, and pstB, with the same direction of transcription. Comparison of the pst operon of S. marcescens with that of other organisms revealed that the genes for pstS and phoU were missing. A potential CRP bonding site and pho box sequence was found in the upstream of the putative promoter at the regulatory region. Analysis of the nucleotide sequence showed that homology in amino acid sequences between the PstC protein and Yersinia sp., Vibrio sp., and Pseudomonas sp. were 49, 37 and 33%, respectively. The PstA protein and Yersinia sp., Vibrio sp., and Pseudomonas sp. showed homologies of 64, 51, and 47%, respectively. PstB protein and Methanocaldococcus sp., E. coli, and Mycoplasma sp. showed homologies of 60, 50, and 48%, respectively. The pst genes could be expressed in vivo and positively regulated by cAMP-CRP. The E. coli strain harboring plasmid pPH7, with pst genes, increased with the transport of phosphate.

Descriptions of Four New Species of Actinolaimoidea (Dorylaimida:Nematoda) from Korea (한국산 Actinolaimoidea상과의 (Dorylaimida:Nematoda) 4신종 기재)

  • Four new species of predatory nematodes belonging to the Actinolaimoidea, Dorylaimida are described. Neoactinolaimus gosungensis n. sp. is 2.2-2.4 mm long, b = 4.3-4.9; c = 10.7-11.2; species 60-62 ${\mu}{\textrm}{m}$ long and is distinguishable by low lip region, asymmetrical odontostyle and its wider lumen. Egtitus arcuatus n. sp. is 1.5-1.8 mm long, b = 3.6-4.2; c = 14-15 and is characterized by having dorsally arcuate odontostyle and presence of cardiac glands at base of oesophagus. Paractinolaimus tuberculantus n. sp. is 1.9-2.5 mm long, b = 3.3-4.0, c = 7.2-8.8, spicules 66-69 ${\mu}{\textrm}{m}$ long and is unique in having coiled body posture and a tubercle at the base of oesophagus. Carcharolaimus koriensis n. sp. is 1.7-1.9 mm long, b = 3.6-4.5, c = 65-76 and is characterized by having very thick body cuticle and larger odontostyle aperture.

  • Lytic activities of the egg white lysozyme from Korea-native Ogol fowl against the alkalophilic and thermophilic Bacillus sp. TA-11 were investigated and compared. Lytic activity of the Ogol fowl lysozyme for Bacillus sp. TA-11 was the highest for the cell of post-logarithm phase and optimum concentration of the lysozyme was 0.25%, Optimum reaction pH and temperature were 4.5 and 35$^{\circ}C$, respectively. Lytic activity of egg white lysozyme from fowl for Bacillus sp. TA-11 was the highest for the cell of stationary phase and optimum concentration of the lysozyme was 0.5%. Optimum reaction pH and temperature were 5.5 and 4$0^{\circ}C$, respectively.

  • Four species of Actinosporeans, Aurantiactinomyxon sp. A, B, C and Neoctinomyxon sp. D were released from Oligochaete, Branchiura sowerbyi captured from three reserviors, where intestinal giant-cystic disease in carp had occured from June to September, 1994. All part of the intestinal epithelial tissue from the gullet to the anus of B. sowerbyi were infected by Actino-sporeans, and many mature Actinosporean were seen more easily at the posterior parts of the body. Just before releasing, mature Actinosporean sporozoites were divied into each individual from the intestinal epithelial tissue of Oligochaete, while immature ones had 6 spores ($20{\times}25{\mu}m$ in size) per each in the oocyst ($60{\times}65{\mu}m$ in size). A total of 1, 762 of B. sowerbyi were investigated in three reservoirs, 86 individuals (4.88%) of them were infected; 0. 74% (13 ind.) of Aurantiactinomyxon sp. A, 2. 27% (40 ind.) of Aurantiactinomyxon sp. B, 1. 59% (28 ind.) of Aurantiactinomyxon sp. C, and 0. 28% (5 ind.) of Neoactinomyxon sp. D. At the room temperature of 22.6-$30.7^{\circ}C$, number of extrusion dates of Actinosporeans from B. sowerbyi for 32 days are 1 day (23.3% of total, 1 time) or 5 days (11.7%. 5 times), and the majority was finished within 15 days, however, 6.7% of total were released for 32 days.

Comparative Quantitative Study of Surfactant Protein C mRNA by Filter Hybridization and Solution Hybridization in Rats (Filter Hybridization과 Solution Hybridization 방법에 의한 백서 Surfactant Protein C mRNA 정량측정의 비교)

  • Background : Surfactant protein C(SP-C) is a hydrophobic 5,000 dalton molecule. SP-C has the primary roles in accelerating surface spreading of a surfactant phospholipid. The filter hybridization and solution hybridization assays are both rapid and sensitive and can be used to measure the RNAs complementary to any cloned DNA sequence. Methods : The authors measured the SP-C mRNA levels quantitatively using solution hybridization and filter hybridization assays to obtain a standard curve equation to quantify the mRNA of unknown samples comparatively. Results : 1. The minimum level of the specimens by solution hybridization was 3 pg for SP-C mRNA. 2. The standard curve equation of the solution hybridization assay between the counts per minute(Y) and the SP-C mRNA transcript input(X) was Y=6.46 X+244. The correlation coefficient was 0.99. 3. The minimum detection level of specimens by filter hybridization was 0.1 ng for SP-C mRNA. 4. The standard curve equation of the filter hybridization assay between the counts per minute(Y) and SP-C mRNA transcript input(X) is Y=2541.6 X+252.7. The correlation coefficient was 0.99. Conclusions : A comparison of CPM/filter in the linear range allowed an accurate and reproducible estimation of the SP-C mRNA copy number. Filter hybridization and solution hybridization assays are both rapid and sensitive and can be used to measure the RNAs complementary to any cloned DNA sequence. It is ideally suited to situations where accurate quantitation of multiple samples is required.

A coding technique for the smoothly perfect 8-connected contours in the spatial domain (공간 영역에서의 평활한 완전 8방향 윤곽선 부호화 기법)

  • In this paper, we introudce the notion of the smoothly perfect 8-conntected (SP8C) contour and describe a coding technique for the SP8C contours. Based on the simplificationusing the majority filter proposed by Gu, SP8C contours are restricted on the contour lattice from the segmented image. By noting thating SP8C contours are restricted to travel in only 3 different directions along the contours, we attempt to encode the SP8C contours using the following two techniques. The one is the neighbouring direction segmentcoding, while the other is to employ the notion of the entropy endoding. Computer simulations reveal that the contours can be efficiently encoded by the two technique.

Isolation of Bacillus sp. SW29-2 and Its Antifungal Activity against Colletotrichum coccodes (Bacillus sp. SW29-2의 분리 및 Colletotrichum coccodes에 대한 항진균 활성)

  • Antifungal bacterium against Colletotrichum coccodes causing black dot disease of potatoes and anthracnose of tomatoes was isolated from sewage sludge. The isolate showed a 99% sequence homology of partial 16S rRNA of Bacillus methylotrophicus CBMB205 and Bacillus amyloliquefaciens subsp. plantarum FZB42. The isolate was identified as Bacillus sp. SW29-2, using the neighbor-joining phylogenetic tree, BlastN sequence analysis, and morphological and cultural characteristics. Bacillus sp. SW29-2 is an aerobic, Gram-positive, endospore-forming bacterium, of which the morphological and physiological characteristics were the same as those of type strain B. lichniformis CBMB205, except for the cell growth of over 4% NaCl. The cell growth of the temperature and the initial pH of the medium was shown at $18-47^{\circ}C$ (opt. ca. $38^{\circ}C$) and 3-9 (opt. ca. 6.0), respectively. The inhibition size (diameter) of Bacillus sp. SW29-2 against four strains of C. coccodes ranged from 23 to 29 mm. Also, the isolate showed antifungal activity against penicillium rot-causing Penicillium expansum in apples. Thus far, any report on the antifungal activity of Baciilus spp. against C. coccodes has not been found. These results suggest that the Bacillus sp. SW29-2 isolate could be used as a possible biocontrol agent against C. coccodes, and further applied to other plant pathogenic fungi.

A Study on Material Degradation Evaluation of 9Cr1MoVNb Steel by Micromechanics Test Method (미소역학 시험기법에 의한 9Cr1MoVNb강의 열화도 평가)

  • The Micromechanics test is new test method which uses comparatively smaller specimen than that required in conventional material tests. There are several methods, such as small-specimen creep test, the continuous indentation test, and small punch(SP) test. Among them, the small punch(SP) test method has been applied to many evaluation fields, such as a ductile-brittle transition temperature, stress corrosion cracking, hydrogen embrittlement, and fracture properties of advanced materials like FGM or MMC. In this study, the small punch(SP) test is performed to evaluate the mechanical properties at high/low temperature from $-196^{\circ}C$ to $650^{\circ}C$ and the material degradation for virgin and aged materials of 9Cr1MoVNb steel which has been recently developed. The ${\Delta}P/{\Delta}{\delta}$ parameter defined a slope in plastic membrane stretching region of SP load-displacement curve decreases according to the increase of specimen temperature, and that of aged materials is higher than the virgin material in all test temperatures. And the material degradation degrees of aged materials with $630^{\circ}C$ -500hrs and $630^{\circ}C$ -1000hrs are $36^{\circ}C$ and $38^{\circ}C$ respectively. These behaviors are good consistent with the results of hardness($H_v$) and maximum displacement(${\delta}_{max}$).
