Stability of Ethanolic Extract from Fermented Cirsium setidens Nakai by Bioconversion during Different Storing Conditions (저장조건에 따른 생물전환 발효고려엉겅퀴 주정추출물의 안정성 조사)

  • This study investigated the stability of ethanolic extract from fermented Cirsium setidens Nakai (FCSN) in order to develop functional materials during different storage conditions. We evaluated pectolinarin and pectolinarigenin contents, total phenol content and antioxidant activity (DPPH radical scavenging activity and FRAP assay) of ethanolic extract from FCSN obtained by bioconverison at various temperatures (4, 25 and $50^{\circ}C$) and pHs (4.0, 7.0 and 10.0). Our results show that the pectolinarin, pectolinarigenin, and total phenol contents in ethanolic extract from FCSN were decreased during the storage periods. Moreover, the DPPH radical scavenging activity did not significantly change at $4^{\circ}C$ and $25^{\circ}C$. Pectolinarin and pectolinarigenin contents, total phenol content and DPPH radical scavenging activity of ethanolic extract from FCSN at acidic pH (pH 4.0) and neutral pH (pH 7.0) were higher than those at the alkaline pH range. These results indicate that the optimum storage condition of the ethanolic extract from FCSN was $4^{\circ}C$ and pH 4.0~7.0 range.

  • The purpose of this study was to evaluate anti-obesity activity of Cirsium setidens Nakai test material (CNTM) in 3T3-L1 adipocytes and obese C57BL/6J mice fed with a high-fat diet using various obesity-related in vitro experiments. During adipocyte differentiation, CNTM significantly inhibited lipid accumulation and ROS production compared to controls. To evaluate whether CNTM could exert glycerol release effects on mature 3T3-L1 adipocytes, we treated cells with various concentrations of CNTM for 1 h. Treatment of mature adipocytes with $160-320{\mu}g/mL$ of CNTM increased the release of glycerol, but not in a significant dose-dependent manner. Anti-adipogenic and anti-lipogenic effects of CNTM seemed to be mediated by the inhibition of $PPAR{\gamma}$ and $C/EBP{\alpha}$. Moreover, CNTM stimulated fatty acid oxidation in an AMPK-dependent manner. CNTM-treated groups of C57BL/6J mice showed reduced body weights and adipose tissue weight with improving serum lipid profiles and adiponectin protein expression in obese C57BL/6J mice fed with a high-fat diet. These results suggest that CNTM might have anti-obesity effect on adipogenesis and lipid metabolism in vitro and in vivo. This presents the possibility of developing a treatment for obesity using nontoxic natural resources.

Scopoletin from Cirsium setidens Increases Melanin Synthesis via CREB Phosphorylation in B16F10 Cells

  • In this study, we isolated scopoletin from Cirsium setidens Nakai (Compositae) and tested its effects on melanogenesis. Scopoletin was not toxic to cells at concentrations less than $50{\mu}M$ and increased melanin synthesis in a dose-dependent manner. As melanin synthesis increased, scopoletin stimulated the total tyrosinase activity, the rate-limiting enzyme of melanogenesis. In a cell-free system, however, scopoletin did not increase tyrosinase activity, indicating that scopoletin is not a direct activator of tyrosinase. Furthermore, Western blot analysis showed that scopoletin stimulated the production of microphthalmia-associated transcription factor (MITF) and tyrosinase expression via cAMP response element-binding protein (CREB) phosphorylation in a dose-dependent manner. Based on these results, preclinical and clinical studies are needed to assess the use of scopoletin for the treatment of vitiligo.

The Specific Plant Species and Naturalized Plants in the Area of Naejangsan National Park, Korea (내장산국립공원 일대의 특정식물과 귀화식물)

  • Endangered species designated by The Ministry of Environment in region of Naejangsan National Park were found Lycoris chinensis var. sinuolata K.H.Tae & S.T.Ko, Iris koreana Nakai, Cymbidium macrorrhizum Lindl and Vexillabium yakushimensis (Yamam.) F.Maek.(4 taxa). The floristic special plants were recorded to a total of 159 taxa; that is, class I species (91 taxa) were containing as Chloranthus fortunei (A.Gray) Solms, Vicia anguste-pinnata Nakai and Euscaphis japonica (Thunb.) Kanitz, etc, class II species (17 taxa) were Pseudostellaria coreana (Nakai) Ohwi, Dryopteris expansa (C.Presl) Fraser-Jenkins et Jermy and Gymnocarpium dryopteris (L.) Newman, etc, class III species (32 taxa) were containing; Cirsium setidens (Dunn) Nakai, Parasenecio pseudotaimingasa (Nakai) B.U.Oh and Saussurea eriophylla Nakai, etc, class IV species (9 taxa) were Deutzia paniculata Nakai, Carex ligulata Nees and Thuja orientalis L., etc, and class V species (10 taxon) were Orobanche filicicola Nakai, Lycoris chinensis var. sinuolata K.H.Tae & S.T.Ko and Lycoris sanguinea var. koreana (Nakai) T.Koyama, etc. Endemic species of korea were identified 40 taxa such as Broussonetia kazinoki var. humilis Uyeki, Pseudostellaria coreana (Nakai) Ohwi and Silene seoulensis Nakai, etc. Plant species designated as Natural Monument were 2 species that Daphniphyllum macropodum forest(No. 91) and Torreya nucifera forest(No. 153) receive protection. The naturalized plants were identified 30 taxa; Fallopia dumetorum (L.) Holub, Persicaria orientalis (L.) Spach and Rumex crispus L., etc. Among them the ecosystem disturbance wild plants was not discovered.

Chromosome Number Evolution in Cirsium Mill. and Carddus L. (Asteraceae)

  • Chromosome numbers and karyotypes in flowering plants have been considered to be prominent features in taxonomic and evolutionary context. Despite the increasing numbers of cytological studies in Asteraceae, karyotype analysis of Cirsium Mill. and Carddus L. in Korean population have not been performed carefully. In this study, the chromosome numbers and karyotype analysis of all eight species of the genus Cirsium Mill. and one species of Carddus L. were analyzed. While the chromosome number in Carduus crispus L. was diploid (2n = 2x = 18 or 18+2Bs) with x = 9 as the base chromosome number, all seven species of Cirsium were diploid with x = 17 except for Cirsium lineare (Thunb.) Sch. Bip. (x = 14). The chromosome number in C. pendulum Fisch. ex DC. presented 2n = 2x = 34 from two populations and C. lineare exhibited 2n = 2x = 28 from one population. Aneuploidy was occasionally found in C. japonicum Fisch. ex DC. var. spinossinum Kitam. (2n = 2x = 34, 35, 36), C. rhinoceros (H. $L{\acute{e}}v.$ & Vaniot) Nakai (2n = 2x = 32, 34), C. setidens (Dunn) Nakai (2n = 2x = 30, 31, 32) and C. vlassovianum Fisch. ex DC. (2n = 2x = 31, 32). While Cirsium japonicum Fisch. ex DC. var. japonicum possessed several B-chromosomes (2n = 2x = 34, 35, 36), polyploidy was only encountered in Cirsium nipponicum (Maxim.) Makino. (2n = 4x = 68) from two populations in Ulleung Island. The present cytological data might be contributed to the taxonomic and evolutionary studies in the genus Cirsium.

Combined Treatment of Aqueous Chlorine Dioxide, Organic Acid, and Blanching for Microbial Decontamination of Wild Vegetables after Harvest (수확 후 산채류의 미생물 제어를 위한 이산화염소수와 유기산 및 Blanching 병합 처리)

  • To improve the microbiological safety of wild vegetables after harvest, Aster scaber and Cirsium setidens Nakai were treated with combinations of 50 ppm aqueous chlorine dioxide ($ClO_2$)/0.5% citric acid or fumaric acid, and 50 ppm $ClO_2$/0.5% fumaric acid/blanching at $90^{\circ}C$ for 2 min. Combined treatment of 50 ppm $ClO_2$ and 0.5% citric acid reduced populations of total aerobic bacteria, yeast, and molds in Aster scaber and Cirsium setidens Nakai by 2.80~3.64 and 2.02~2.67 log CFU/g, respectively, compared to those of the control. Combined treatment of 50 ppm $ClO_2$ and 0.5% fumaric acid reduced total aerobic bacteria, yeast and molds populations by 3.62~3.82 and 2.47~3.02 log CFU/g, respectively. Based on the results, combined treatment of $ClO_2$ and fumaric acid was more effective in controlling microorganisms in the wild vegetables than either $ClO_2$ or citric acid. In addition, combined treatment of $ClO_2$/fumaric acid/blanching reduced the populations of total aerobic bacteria by 4.59~5.12 log CFU/g, and populations of yeast and molds were not detected by treatment. These results suggest that combined treatment of $ClO_2$/fumaric acid/blanching is the most effective method for improving microbiological safety of wild vegetables after harvest.
