건강 Ethyl acetate 분획의 선충에 대한 수명연장 효과 (Longevity Property of Ethyl acetate Fraction of Dried Zingiber officinale in Caenorhabditis elegans)

  • This study was performed to evaluate the lifespan extending effects of the dried ginger (Zingiber officnale Roscoe, Zingiberaceae). Ethanol extract of the dried ginger was successively partitioned as methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions, and lifespan-extending effects of the fractions were investigated using Caenorhabditis elegans model system. Ethyl acetate-soluble fraction showed the most potent longevity property. Furthermore, ethyl acetate-soluble fraction elevated thermal stress tolerance, and increased expression of stress resistance protein. Further studies were performed to investigate several aging-related factors such as reproduction, food intake, growth and movement of C. elegans. The results revealed that there were no significant changes in aging-related factors including reproduction, food intake and growth, however, ethyl acetate-soluble fraction treatment led to an up-regulation of locomotory ability of aged worms, suggesting ethyl acetate-soluble fraction affected healthspan as well as lifespan of nematode.

생강, 울금, 강황 추출물의 항산화 효과, AChE 억제활성 및 GABA 함량 (Effects of Ethanol Extracts from Zingiber officinale Rosc., Curcuma longa L., and Curcuma aromatica Salisb. on Acetylcholinesterase and Antioxidant Activities as well as GABA Contents)

  • 생강과 식물의 인지기능 관련 활성을 검토하고 종류와 산지에 따른 차이를 비교하기 위하여, 시중에서 많이 사용되고 고품질로 알려져 있는 국산 생강과 울금, 미얀마산 울금과 인도산 강황의 항산화 효과, GABA 함량, 활성성분 분석, 그리고 AChE 억제 활성을 측정하였다. 미얀마산 울금 에탄올추출물에서 총 폴리페놀(98 ${\mu}g/mg$), curcumin(97.1 ${\mu}g/mg$), GABA 함량(1.31 ${\mu}g/mg$)이 가장 높았으며, DPPH, ABTS 라디칼 소거능 및 AChE 저해능도 비교적 높게 나타났다. 한편, 인도산 강황 추출물도 미얀마산 울금과 비슷한 항산화 효과와 AChE 저해 효과가 나타났으나, curcumin (12.2 ${\mu}g/mg$)과 GABA(0.04 ${\mu}g/mg$) 함량은 매우 낮게 나타났다. 국산 생강과 울금 추출물의 경우 총 폴리페놀 함량은 비슷하였으나(39.7~42.5 ${\mu}g/mg$) 6-gingerol, 6-shogaol 등의 진저롤류를 함유하고 있는 생강 추출물에서 울금보다 높은 라디칼 소거능, 특히 강한 ABTS 라디칼 소거능을 나타내었으며, AChE 저해능은 두 추출물 모두 매우 낮은 것으로 나타났다. 국내산 2가지 추출물의 GABA 함량(약 1.11 ${\mu}g/mg$)은 미얀마산 울금과 비슷한 정도로 높게 검출되어 라디칼 소거능과 AChE 억제능은 GABA 함량과는 상관성이 없었다. 따라서 본 실험에 사용한 4가지 생강과 식물 추출물의 AChE 억제능은 총 폴리페놀 함량과 밀접한 관계가 있으며, curcumin 함량, 라디칼 소거능 및 GABA 함량과는 직접적인 상관관계가 없는 것으로 나타났다. 이상의 결과와 같이, 미얀마산 울금은 나머지 추출물과 비교하여 높은 항산화 활성과 curcumin 및 GABA 함량, 그리고 비교적 높은 AChE 저해활성을 갖는 강력한 인지능 개선 소재로 활용될 수 있을것으로 사료된다.

생강(Zingiber officinale Roscoe) 다대기의 저장 중 품질개선을 위한 첨가물 효과 (Effects of Additives on the Quality Improvement of Minced Ginger (Zingiber officinale Roscoe) During Storage)

  • 본 연구에서는 식초와 주정 및 아스코르빈산의 첨가량을 달리하여 제조한 생강다대기 (MG)를 $30^{\circ}C$에서 저장하면서 첨가물이 저장 중 품질특성에 미치는 영향을 살펴보았다. 생강다대기의 저장 중 색도의 경우 T-I 처리구 색도값이 다른 첨가구와 비교하여 높은 값을 보여주었고, 총 색차인 ${\Delta}E$는 T-III 처리구에서 저장 28일 경과 후에도 색도 변화가 가장 적은 것으로 나타났다. 생강다대기의 일반세균수를 측정한 결과, 저장 초기 무첨가구(T-0)에서 일반세균수가 $5.37{\times}10^7\;CFU/g$ 이었으나, MG-처리구의 초기 일반세균수가 T-0와 비교하여 4 log 이상 감소하는 경향을 보였다. 이는 저장기간 중 미생물의 증식없이 초기 일반세균수를 유지하는 경향을 보여 첨가물의 미생물 증식억제 효과를 확인할 수 있었다. 생강다대기의 관능평가를 실시한 결과, T-0와 MG-처리구간의 향기에 대한 강도 평가에 있어서 유의적인 차이를 보이지 않았다(p < 0.05). 전자코 분석장치에 의한 생강다대기의 향기성분 분석 결과, 초기치 생강다대기 향은 T-0에 비하여 T-I, T-II, T-III 처리구의 향기가 좀 더 강하게 인지되었으나, 저장 중 향기성분의 변화는 첨가물의 종류나 첨가량에 관계없이 저장온도 $30^{\circ}C$에서도 안정적으로 유지되는 것으로 나타났다.

토굴저장 생강의 부패에 관여하는 미생물의 동정 및 병원성 (Identification and Pathogenicity of Microorganisms Associated with Seed-Rhizome Rot of Gingers in Underground Storage Caves)

  • Microorganisms associated with seed-rhizome rot of gingers preserved in three underground storage caves were identified with respect to rot types. Rot patterns were grouped into 4 different types : yellow soft rot, brown rot, localized ring rot, and water-soaked rot. Water-soaked rot was highest in frequency with 40% and ring rot the least with 14%. Causal pathogens differed with rot type, yellow soft rot by Erwinia carotovora and Pseudomonas aeruginosa, brown rot by Fusarium solani and Pseudomonas aeruginosa, localized ring rot by F. solani, and water-soaked rot by Pythium spinosum and P. ultimum. Pythium myriotylum, the causal pathogen of ginger rhizome rot which occurs severely in fields was rarely detected from storage seed-rhizomes suggesting its minor involvement with storage rot. Pathogenic Pythium isolates were frequently obtained from both rhizome surface and inner tissues of rotten rhizomes. Detection frequency of Pythium isolates in inner tissues decreased as increasing distance from rhizome surface. In wound-inoculation tests, above pathogens caused a varying degree of rot on healthy rhizomes at 15$^{\circ}C$, 2$0^{\circ}C$ and 3$0^{\circ}C$ with increasing severity at higher temperatures.

  • The inclusion of crude ginger rhizome (zingiber officinale) extract at 0.5 to 1.0% (v/w) in the marination of marginally acceptable lean beef improved meat tenderness by 20-30% in the absence of 2% salt and by 35-45% in the presence of 2% salt. Ginger extract also retarded the development of rancidity and increased shelf-life of precooked lean beef two-fold in saran-wrap (no vacuum) storage at $4^{\circ}C$. Ginger rhizome improves the palatability and acceptability of lean beef from carcasses of marginal quality. It is particularly beneficial for the preparation of pre-cooked ready-to-eat beef products that are not vacuum-packaged.

화장품중 생약성분의 분석(I) 육제중의 당귀 및 천궁추출액의 함량분석 (Analysis of Medicinal Plants in Cosmetics(I) Determiantion of Korean Angelica Extract and Cnidium Rhizome Extract in bath Essence)

  • Angelica gigas extract and Cnidium of ficinale extract in Bath essence were determined by high performance liquid chromatography with a UV detector by use of Bondapak C-18 column. Decursin and Ligustilide were used as indicator ingredient for analysis of Angelica gigas extract and Cnidium of ficinale extract. Each indicator ingredient was isolated from each plant and purified using silver-ni-trate treated silica column. Their structure were conformed with IR, NMR and HPLC. The recoveries of Angelica gigas extract and Cnidium officinale extract added to Bath essence sample were 96.5-109.5% and 112-115%.

생강뿌리썩음병균 Pythium myriotyrum의 병원성 및 균학적 특성 (Pathogenicity and Mycological Characteristics of Pythium myriotylum Causing Rhizome Rot of Ginger)

  • 병든 생강으로부터 분리한 6개 Pythium 균주들은 여러 가지 형태적, 생리적 특성에서 Pythium myriotylum Drechsler로 동정되었다. 이 균들은 생강 지하부의 싹, 근경, 뿌리, 땅가줄기 뿐만 아니라 지상부 잎, 줄기 전부분에 걸쳐 병원성이 강하였다. 이 균들은 오이, 수박 등 10개 작물 혹은 잡초의 어린 묘를 부패시켰으며 보리, 옥수수 등 공시한 3개 작물 유묘의 지상부 생장이나 뿌리 생장을 현저히 억제하였다. 이 균들의 생육 최고, 최적, 최저온도는 각각 39~45$^{\circ}C$, 33~37$^{\circ}C$, 5~7$^{\circ}C$였으며 생육최적 pH는 6~7이었다. 이 균들의 균사생장은 V-8 juice 배지에서 가장 빨랐으나 기중균사량은 PDA와 옥수수전즙배지에서 가장 많았다. 유주자낭과 장난기는 V-8 juice 배지에서 가장 잘 형성되었으며 유주자낭 형성 최적온도는 균주에 따라 20~35$^{\circ}C$, 장난기 형성은 15$^{\circ}C$의 저온에서 가장 양호하였다

Effects of Natural Products on the Induction of NAD(P)H: Quinone Reductase in Hepa 1c1c7 Cells for the Development of Cancer Chemopreventive Agents

  • NAD(P)H:quinone reductase (QR) is one of the protective phase II enzymes against toxicity that accomplishes the capacity of detoxification by modulating the effects of mutagens and carcinogens. The detoxification mechanism is that quinone reductase promotes the 2-electron reduction of quinones to hydroquinones which are less reactive. This study is to search new inducers of quinone reductase from natural products, which can be used as cancer chemopreventive agents. Plant extracts were evaluated by using quinone reductase generating system With Hepa 1c1c7 murine hepatoma cell lines for enzyme inducing properties and crystal violet staining method for the measurement of cytotoxicity provoked. We have tested approximately 106 kinds of natural products after partition into n-hexane, ethyl acetate and aqueous layers from 100% methanol extracts of natural products. The ethyl acetate fractions of Vitex rotundifolia $(fruits,\;2FC:\;12.7\;{\mu}g/ml)$, Cnidium officinale $(aerial\;parts,\;2FC:\;10.5\;{\mu}g/ml)$, Chrysanthemum sinese $(flowers,\;2FC:\;17.4{\mu}g/ml)$ and the hexane fractions of Angelica gigas $(roots,\;2FC:\;13.2\;{\mu}g/ml)$, Smilax china $(roots,\;2FC:\;l1.9\;{\mu}g/ml)$, Sophora flavescens $(roots,\;2FC:\;16.3\;{\mu}g/ml)$ revealed the significant induction of quinone reductase in a murine hepatic Hepa 1c1c7 cell culture system.

Efficacy of Soil Amendment with Medicinal Plant Materials for the Control of Root-knot Nematode (Meloidogyne incognita) in Tomato

  • Soil amendments with oriental herbal medicines such as fruit of Anethum graveolens, flower buds of Syzygium aromaticum, rhizome of Cnidium officinale, rhizome of Coptis chinensis, root bark of Paeonia suffructicosa, stem bark of phellodendron amurense, and stem bark of Cinnamomum cassia at the rate of 0.2% (weight by volume of soil) significantly reduced Meloidogyne incognita infection (root gall formation) of tomato seedlings compared with the control. The most effective treatments were root bark of p. suffructicosa and stem bark of C. cassia as they gave minimum numbers of galls on tomato roots (4.7% and 8.9%, respectively, relative to control) as compared to other treatments. Another study with root bark of p. suffructicosa and C. cassia at different application doses also showed consistent results in reducing gall number. The control efficacy decreased as the application doses were lower-ed, indicating their dose-dependent control activities. These treatments significantly enhanced aboveground plant growths (total masses).

In vitro Effects of Plant Extracts, and Phytohormones on Mycelial Growth of Anthracnose Fungi

  • Water extracts of six plants, such as Allium sativum, A. cepa, Zingiber officinale, Platycodon grandiflorum, Oenanthe javanica, and Capsella brusapastoris, were tested in vitro for inhibitory activity against mycelial growth of anthracnose fungi, Colletotrichum gloeosporioides, C. dematium, and C. coccodes. Among the plant extracts, an Allium sativum extract has good inhibitory effects in all the fungi. Four phytohormones namely, IAA(indole-3-acetic acid), NAA(a-Naphthyl acetic acid), 2,4-D(2,4-Dichloro phenoxy acetic acid) and BAP(Benzyl adenine purine) were used to find out the role over mycelial growth of these fungi. All the concentrations of BAP have good inhibitory effect against mycelial growth of these fungi than that of other tested plant hormones.
