Codonopsis lanceolata has been used as an herbal medicine for several lung inflammatory diseases, such as asthma, tonsillitis, and pharyngitis. Previously, we showed the neuroprotective effect of steamed and fermented C. lanceolata (SFC) in vitro and in vivo. In the current study, the treatment of HT22 cells with SFC decreased glutamate-induced cell death, suggesting that SFC protected HT22 cells from glutamate-induced cytotoxicity. Based on these, we sought to elucidate the mechanisms of the neuroprotective effect of SFC by measuring the oxidative stress parameters and the expression of Bax and caspase-3 in HT22 cells. SFC reduced contents of ROS, $Ca^{2+}$ and NO. Moreover, SFC restored contents of glutathione and glutathione reductase as well as inhibited Bax and caspase-3 activity in HT22 cells. These results indicate that steamed and fermented C. lanceolata (SFC) extract protected HT22 cells by anti-oxidative effect and inhibition of the expression of Bax and caspase-3.
This study was conducted to evaluate the cytotoxicity and ${\alpha}-Amylase$, ${\alpha}-Glucosidase$, pancreatic lipase inhibition in vitro by different solvent fractions from the roots of Codonopsis lanceolata. The values of $IC_{50}$ against Calu-6 cell showed a high effect in n-hexane fraction ($10.13{\mu}g/mL$) whereas DW fraction exhibited the weakest inhibition on cell viability, having an $IC_{50}$ value of over $1,000{\mu}g/mL$. The values of $IC_{50}$ against HCT-116 cell showed the highest activity in n-BuOH fraction ($102.01{\mu}g/mL$), followed by n-hexane fraction ($145.85{\mu}g/mL$), methylene chloride fraction ($332.02{\mu}g/mL$), ethyl acetate fraction ($462.93{\mu}g/mL$) and DW fracion ($>1,000{\mu}g/mL$). ${\alpha}-Amylase$ inhibitory activity in methylene chloride fraction and ethyl acetate fraction was found to have a higher inhibitory effect with 24.5% and 25.6% than the other fractions. The highest ${\alpha}-Glucosidase$ inhibitory activity was observed from the ethyl acetate fraction extract, while the extract of DW fraction showed the lowest level of inhibitory activity at given experiment concentration. The pancreatic lipase inhibitory activity of C. lanceolata was found to have a higher the effect in ethyl acetate fraction. Inhibition of lipase activity of the ethyl acetate fraction and n-hexane fraction showed a relatively high, while the extract of DW fraction showed the lowest level at given experiment concentration. These results suggested that the roots of C. lanceolata may assist in the potential biological activity on carbohydrate, lipid Inhibitory activity and anticancer activity.
Background: This study was conducted to investigate the effects of fertilization with the application of mixed organic matter after cultivating green manure crops on the growth and saponin content of Codonopsis lanceolata Trautv. Methods and Results: Five type of cultivation of green manure crops were done: hairy vetch as a single seedling crop (HV), hairy vetch and rye as a mixture of seedling (HV/R), additional fertilization with mixed organic matter after cultivation of a seedling mixture (HV/R/MO), chemical fertilizer (CF), and a non-treated control (NT). The total nitrogen content was the highest in the HV/R/MO treatment, followed by the HV, CF, HV/R and NT treatments, respectively. Nitrogen fixation was higher in the single seedling treatment with hairy vetch (HV) than in the treatment with a mixture of hairy vetch and rye seedling (HV/R). Moreover, the growth of C. lanceolata was greatly increased by the application of additional fertilizer after treatment with the mixture of hairy vetch and rye seedlings. The HV/R/MO treatment produced the highest total lancemaside content among the treatments, followed by HV, HV/R, CF, and NT, respectively. Conclusions: It was verified that C. lanceolata experienced superior growth with the application of mixed organic matter as additional fertilizer after the cultivation of green manure crops, and the content of its major saponins, lancemasides were also increased by this treatment.
Calmodulin, a $Ca^{2+}$-binding protein, has no enzyme activity. It combines with $Ca^{2+}$ and makes variable proteins to an active form. Calmodulin 2 is a ubiquitous protein in plants. To investigate the defense mechanism against various stresses, a clone encoding a calmodulin 2 protein was isolated from a cDNA library prepared from taproot mRNAs of Codonopsis lanceolata. The cDNA, designated CICAM2, is 719 nucleotides long and has an open reading frame of 450 bp with a deduced amino acid sequence of 149 residues. The deduced amino acid sequence of CICAM2 showed a high similarity with calmodulins of P. x hybrida (P27163) 97%, N. tabacum (BAB61908) 97%, S. tuberosum (AAA74405) 96%, Z. mays (CAA74307) 92%, C. richardii (AF510075) 93%, M. truncatula (AAM81203) 91%, and G. max (P62163) 91%. A cyclophilin 1 cDNA clone(GenBank accession no.CF924191) was isolated from the taproot of C. lanceolata and designed as C1CyP1. Determination of the nucleotide sequence of C1CyPl identified an open reading frame of 525bp, which shared high homologies with cyclophilins that were previously reported in other organisms. The C1CyP1 amino acid sequence possesses 7 amino acid residue stretch(KSGKPLH) that is characteristic of plant cytosolic dehydrins. Currently available amino acid residues of plant cyclophilins were compared to examine their phylogenetic relationship to C1CyP1. In the phylogenetic analysis, based on the aligned sequences, C1CyP1 showed high homology with arabidopsis ROC2 and rice CyP1. The transcript that corresponded to C1CyP1 was abundant in callus, but only basal level of transcript was detected in stem, leaf and root. For the study in the defense mechanism against various stresses, we report expression patterns of this gene by quantative RT-PCR.
This study was executed to evaluate the phenolic content, DPPH radical scavenging rate, and the cytotoxic effect in human cancer cell, 3T3-L1 cell from C. lanceolata extracts at various ethanol concentration. Total polyphenol and flavonoid content of the C. lanceolata at various ethanol concentration showed the high amount in 70%, 100% ethanol extract. The DPPH radical scavenging activity progressively increased in a dose-dependent manner, and showed the highest in 100% ethanol extract. The cytotoxic effect against human cancer cell of the C. lanceolata was higher in 50% and 70% ethanol extracts. In particular, the cytotoxic effect in MCF-7 cell was relatively higher than in other cells. The $IC_{50}$ (concentration causing 50% cell death) value showed the highest on MCF-7 cell ($538.39{\mu}g/m{\ell}$ in 70% ethanol extract, and exhibited significant activity against Hela cell ($637.87{\mu}g/m{\ell}$, Calu-6 cell ($728.64{\mu}g/m{\ell}$. The extract of 70% ethanol at $1,000{\mu}g/m{\ell}$ exhibited a pronounced cytotoxic effect on 3T3-L1 cell comparable to that of the other extracts, and reduced in a concentration-dependent manner.

Background: This study determined the effects of mulching, an environment friendly organic cultivation method, on antioxidant compound contents and growth in Codonopsis lanceolata, commonly known as Deodeok. Methods and Results: C. lanceolata was treated by mulching with several different methods (a non-woven fabric, biodegradable film, or rice husks) and also treated with hand weeding. A non-treatment plot was used as a control. The growth and levels of weed control in C. lanceolata were better in plants cultivated under mulching treatments (non-woven fabric, biodegradable film, and rice husks) than in those under non-mulching treatments (hand weeding and non-treatment). The contents of antioxidant compounds, such as total flavonoids, phenolics, and anthocyanins, were highest under the biodegradable film treatment, followed by the non-woven fabric treatment, rice husks treatment hand weeding, and non-treatment. There were identifiable differences in DPPH and ABTS activity in comparison to antioxidant compound content by solvent fractions. Mulching treatments resulted in higher DPPH scavenging activity in water and ethyl ether fractions and ABTS scavenging activity in n-butanol fractions than in other fractions, as opposed to hand weeding and non-treatment groups, although total activity of DPPH and ABTS did not increase with mulching treatments. Conclusions: Mulching C. lanceolata with biodegradable film and non-woven fabric is an effective method for improving plant growth and inhibiting the occurrence of weeds as well as for increasing antioxidant compound content and altering antioxidant activity.
A cDNA clone (GenBank accession no. CF924621) homologous to aluminum induced protein gene was isolated and characterized from Codonopsis lanceolata (ClAIP). The ClAIP is 906 nucleotides long and has an open reading frame of 711 bp with a deduced amino acid sequence of 236 residues. The ClAIP shows high homology to A. marina (84%), G. hirsutum(83%), V. radiata (83%), A. thaliana (80%), B. nap us (78%) and T. aestivum (68%). The deduced amino acid sequence of ClAIP also has homology to the N-terminal end of plant Asn synthetase. This region does not contain the active sites of the enzyme and the significance of this conservation is currently not clear. To investigate the expression of ClAIP against several heavy metal stresses, we treated the sliced tap root of C. lanceolata with various heavy metals. The expression of ClAIP was increased by 25 uM $Al_2$(SO$_3$)$_4$ in proportion to incubation time and also increased by 50 uM CdCl$_2$.
The immunostimulating activities of the hot-water extract from Codonopsis lanceolata were investigated. The proliferation of BSA-primed lymph node cells was enhanced between 2.8- to 11.2-fold compare to control, when cultured with 1 to $25\;{\mu}g/mL$ of C. lanceolata extract. It showed strong immunopotentiating activity than ginseng extract and as remarkable as Bifidobacterium adolescentis M101-4 known as a positive immunostimulator. The proliferation of splenocytes and Peyer's patch cells was enhanced between 4.2- to 13.8-fold and 3.1- to 6.9-fold, respectively, when cultured with 1 to 25 $25\;{\mu}g/mL$ of C. lanceolata extract. It enhanced the production of cytokines such as $TNF-{\alpha}$ and IL-6 in the culture of RAW 264.7 macrophage cells. In the culture of lipopolysaccharide-stimulated RAW 264.7 cells, production of cytokines was as compared to controls. In unstimulated RAW 264.7 cells, both $TNF-{\alpha}$ and IL-6 production were enhanced between 12.6- to 67.8-fold and 2.8- to 10.1-fold, respectively. The hot-water extract from C. lanceolata is expected to be a safe immunopotentiator to maintain the host immunity and develop a physiologically functional food.
