Genetic Analysis of Medicinal Plants in Adenophorae Radix Using DNA Barcode (DNA바코드를 활용한 사삼(沙蔘)의 종 감별)

  • 사삼(沙蔘, Adenophorae Radix)은 "대한민국약전외한약(생약)규격집(KHP)"에 잔대 Adenophora triphylla var. japonica Hara 또는 사삼(당잔대, A. stricta Miq.)의 뿌리로 수재되어 있으나, 형태학적으로 유사한 제니(모시대, A. remotiflorus Miquel), 층층잔대(윤엽사삼, A. tetraphylla (Thunb.) Fisch), 더덕 Codonopsis lanceolata (Sieb. et Zucc.)과 오 혼용 우려가 있어 이들을 구별하기 위한 종 감별법이 필요하다. 본 연구에서는 '사삼'과 오 혼용 우려가 있는 종들을 구별할 수 있는 유전자 마커 개발을 위하여 DNA 바코드로 활용되고 있는 유전자 부위를 분석하여 ITS (25%), atpB-rbcL (15%), atpF-atpH (14%), rpl16 (13%), trnL-F (10%), matK (9%), rpoC1 (7%)에서 변이율(percent of variable sites)을 확인하였다. 또한, 분석한 유전자 부위 중 종간 차이를 확인하기 용이한 matK 구간을 활용해 기원종인 잔대, 당잔대와 형태적으로 유사하여 오 혼용될 우려가 있는 층층잔대, 모시대 및 더덕을 감별 할 수 있는 유전자 마커를 개발하였다. 본 연구를 통해 얻어진 염기서열과 분자 마커는 '사삼'의 품질관리에 유용하게 활용 가능할 것으로 사료된다.

  • 제니(薺苨, Adenophorae Remotiflori Radix)는 "대한민국약전외한약(생약)규격집(KHP)"에 모시대(Adenophora remotiflorus Miquel)의 뿌리로 수재되어있으나, 형태학적으로 유사한 잔대(A. triphylla), 당잔대(A. stricta) 및 더덕(Codonopsis lanceolata)과 오 혼용 우려가 있어 이들을 구별하기 위한 정확하고 객관적인 종 감별법이 필요하다. 본 연구에서는 '제니'의 기원인 모시대와 오 혼용 우려가 있는 종들을 구별 할 수 있는 유전자 마커를 개발하기 위하여 Genbank에 등록된 ycf2 구간을 활요하여 모시대와 잔대, 당잔대를 구분 할 수 있는 INDEL (insertion/deletion) 마커를 개발하였다. 또한, 보다 정확한 종감별을 위해 DNA 바코드로 활용되고 있는 유전자 부위의 염기서열을 분석하여 ITS (25%), atpB-rbcL (15%), atpF-atpH (14%), rpl16 (13%), trnL-F (10%), matK (9%), rpoC1 (7%)에서 변이율(percent of variable sites)을 확인하였다. 향후, 본 연구에서 개발된 INDEL 마커와 더불어 추가적으로 개발을 진행 중인 분자 마커는 한약재 '제니'의 품질관리에 활용 가능할 것으로 사료된다.

Development of Revegetation Methods Using Fresh Woodchip from Construction Works (건설현장 발생재를 활용한 비탈면 녹화에 관한 연구)

  • This study was conducted to develop recycle revegetation methods for the restoration of the steep slopes by using fresh wood chip from construction sites. In general, the fresh wood chips can be used as soil media for the restoration works, because they can increase infiltration of rainfall and give enough porous to breathe and elongate for the root growth as well as economic value. The experiment was carried out to compare the effect of fresh wood chips from different mixing with soil, organic material and macromolecular compound which used for slope restoration works conducted by Hyunwoo green(Ltd.). The main results by monitoring for two years are summarized as follows; 1. The soil media made with low percentage of fresh wood chip covered quickly by herb plants. Especially, the soil mixture Type C (wood chip 20%) showed 80 percent ground coverage within two months after seeding. 2. The soil mixture type E (wood chip 40%) and type F (wood chip 50%) which contains more fresh wood chips than soil type C was under 30 percent ground coverage because wood plants are germinated well. If the restoration works aims at making forest, then the soil type E and F would be recommended than using soil type C. 3. Among the woody plants, Ailanthus alfissima, Pinus rigida, Pinus densiflora, and Albizzia julibrissin showed high percentage of germination rates and vigorous growth. In case of shrubs, Lespedeza cyrtobotria and Indigofera pseudo-tintoria scored high percentage of germination rates. 4. In native plants, Chrysanthemum indicum, Artemisia princeps, Lutos corniculatus and Imperata cylindrica showed high percentage of appearance. In case of introduced herbs, Coreopsis lanceolata, Coreopsis tinctoria and Oenothera oborata grew so vigorously. 5. The soil types which including fresh wood chips over 30-40 percentage showed the most diverse plant composition and the most effective germination rates and growth pattern with woody plants. 6. This works to develop recycle revegetation methods using fresh wood chips need more efforts for monitoring the exact effect of fresh wood chips as the soil media.

A Study of the Vegetation in the Deogyusan National Park - Focused on the deciduous forest at Namdeogyu area - (덕유산국립공원의 식생에 관한 연구 -남덕유지역 낙엽활엽수림을 중심으로-)

  • This study was carried out to classify vegetation structure on the deciduous forest at Namdeogyu area in Deogyusan National Park from May 2007 to September 2008 using the gradient analyses and phytosociological method. The vegetation was classified into Quercus mongolica community(Typical subcommunity, Rhododendron sehlippenbaehii-Q. mongolica subcommunity, Carex lanceolata-Q. mongolica subcommunity), Q. Serrata community(Typical subcommunity, Q. variabilis -Q. serrata subcommunity, Carpinus laxiflora - Q. serrata subcommunity), C. laxiflora community, Fraxinus mandshurica community and Corn us controversa community. Ecological characteristics such as species composition, layer structure, vegetation ratio, and the distribution of individual trees by DBH(diameter at breast height) were significantly different among communities. The order of important value of the forest community with DBH 2cm above plants was Q. mongolica(67.8), F. mandshurica(44.8), Q. serrata(27.5), Acer pseudo-sieboldianum(17.0), C. controversa(l5.9), F. sieboldiana(l1.5), R. schlippenbachii(11.0), C. cordata(11.0), Styrax obassia(10.9), Magnolia sieboldii(10.4) and C. laxiflora(9.8). Distribution of DBH of Q. mongolica had a formality distribution, suggesting a continuous domination of these species over the other species for the time being. In contrast, F. mandshurica appeared limited to the valley of the sheet and a higher frequency of young individuals, suggesting a continuous domination of these species the development of a climax forest terrain. Q. serrata had a higher frequency of young individuals and middle individuals, suggesting a continuous domination of these species over the other species for the time being in contrast. This study examined the correlation between each community and the environment according to DCCA ordination. The Q. mongolica community predominated in the highest elevation habitats which had many C.E.C and T-N. F. mandshurica community predominated in the highest elevation habitats which had many Moisture and EX-Cation. The Q. serrata community and C. laxiflora community mainly occurred in the low elevation habitats which had few C.E.C and T-N. The C. controversa community appeared on the park in the middle habitats.

Synecology of the Forest Vegetation of Yeongjongdo (영종도 삼림식생의 군락생태)

  • The vegetation in this study area was divided into 10 plant communities: Pinus thunbergii, P. densiflora, Quercus acutissima, Q. mongolica, Q. serratea, Q. variabilis, Q. aliena, Q. dentata, Carpinus coreana, Betula davurica and 3 afforestations; Pinus rigida, Robinia pseudo-acacia, Castanea crenata. The dominant species in each stratum as determined by R-NCD appeared to be Q. acutissima, P. densiflora, Platycarya strobilacea, Sorbus alnifolia and Q. aliena in the tree-2 layer, Rhododendron mucronulatum, P. strobilacea, Q. serrata, Q. aliena and Q. acutissima in the shrub layer, and Carex lanceolata, C. humilis, R. mucronulatum, Mischanthus sinensis var. purpurascens and Oplismenus undulatifolius in the herb layer. The distribution of diameter at breast height(DBH) showed the highest percentage (56%) in the range of 2∼10㎝ in the study area and it was investigated as being in the restoration phase after disturbance. The average of the age of trees was under 40-years-od. The pH of soil collected at each site appeared to be within the range between 4.53 and 5.10(the average of 4.73) in each community, organic matters appeared higher than the value in Q. mongolica, P. rigida, C. crenata communities compared to P. thunbergii, P. densiflora communities. And soil organic matters and total nitrogen were highly correlated with their contents.

Phytosociological Study of Weed Vegetation around the Climbing Paths on Mt. Chungyeong (경기도 축령산 등산로 주변 잡초 식생의 식물사회학적 연구)

  • Mountain Chungyeong, 879m in altitude, is located in the northeast of the middle area in Korea. Around Mt. Chungyeong, many Korean endemic and rare plants are populated, so it is considered a very important biogeographical area where the temperature zones of northern and southern plants are crossed. Because it is close to Seoul, a capital of Korea, it is a common mountain where many tourists visit frequently. Continuous tourist's visit may cause a bad influence on vegetation around the climbing paths. Therefor, weed community around the climbing paths on Mt. Chungyeong, where visitors exert a bad influence directly on its community by coming in and out, was surveyed phytosocialogically. Our surveys have been accomplished from August, 2001 to September, 2002. weed communities formed around the climbing paths on Mt. Chungyeong were divided into several patterns and analysed. They have been divided into 5 communities and 5 subcommunities. Community A: Plantago asiatica community, A-a: Erigeron annuus subcommunity, A-b: Carex. lanceolata subcommunity, B: Pseudostellaria palibiniana community, B-a: Carex siderosticta subcommunity, B-b: Galium trachyspermum subcommunity, C: Pueraria thunbergiana community, D: Lespedeza maximowiczii community, E: Rubus crataegifolius community, F: Oplismenus undulatifolius community, The flora surveyed in these communities was constituted of 47 families, 101 genera, 17 varieties, and 149 species. Wild plants such as Plantago asiatica, Erigeron annuus, Erigeron strigosus, Pueraria thunbergiana, Lespedeza maximowiezii, Rubus crataegifolius, Artemisia princeps var. orientalis, Artemisia japonica and Lysimachia clethroides were mostly light loving plants and higher resistant plants against the stamping pressure. Our result from the ranking all surveyed areas by the Bray-Curtis ordination method was very similar to the results from phytosocialogical table analysis.

Effects of the Feeding Platycodon grandiflorum and Codonopsis Ianceolata on the Fatty Acid Composition of Serum and Liver in Rats (도라지 및 더덕 첨가식이가 흰쥐의 혈청 및 간장의 지방산 조성에 미치는 영향)

  • This study was designed to observe the effects of the feeding Platycodon grandiflorum, Codonopsis Ianceolata, perilla oil and safflower oil on the fatty acid composition in serum and liver of dietary hypercholesterolemic rats. Experimental groups mixed with 5% cellulose +10% lard (group 1, control group), 2% cholestyraemine +10% lard (group 2), 5% C. ianceolata+ 10% perilla oil (group 3), 5% P. grandiflorum+10% perilla oil (group 4), 5% C. ianceolata+10% safflower oil (group 5) and 5% P. grandiflorum+10% safflower oil (group 6) were administered to the male rats of the Sprague Dawley for 3 weeks. In the fatty arid compositions of serum total fatty arid, phospholipid, triglyceride and cholesteryl ester, the linoleic acid content was most in the PUFA and it was major fatty acid. Particularly, the other components except the phospholipid fraction in serum lipids were influenced by the fatty acid composition of the test lipids from the fact that linoleic acid content was remarkably more in the groups 5 and 6. In the fatty arid compositions of liver total fatty acid, phospholipid, triglyceride and cholesteryl ester, the linoleic acid content was more in the PUFA and so it was major fatty acid. Particularly, the arachidonic and eicosapentaenoic acid contents of phospholipid fraction in liver lipids were rather more. from the above research, fatty arid composition of serum and liver lipid components were influenced by the fatty acid composition of the test lipids from the fact that linoleic acid takes up the highest percentage.

Fish Community Characteristics and Distribution Aspect of Four Endangered Species in the Byekgye Stream, Korea (벽계천의 어류군집 특성 및 멸종위기 4종의 서식양상)

  • This study conducted a survey to investigate the characteristics of fish communities and the inhabiting status of endangered species in the Byekgye Stream, Korea from April to September 2020. A total of 3,415 fish of 9 families and 31 species were collected from 7 survey stations during the survey period. The dominant species was Zacco koreanus (relative abundance of 31.2%), and the subdominant species was Z. platypus (15.0%), followed by Pungtungia herzi (11.7%), Acheilognathus yamatsutae (5.4%), A. lanceolata intermedia (4.8%), Rhinogobius brunneus (4.4%), and Pseudopungtungia tenuicorpa (4.3%). Among the fish species collected, 19 (61.3%) were identified as Korean endemic species, and two cold-water fish species sensitive to climate change (Rhynchocypris kumgangensis and Cottus koreanus) were collected. Four species were designated as class II endangered wildlife by the Ministry of Environment: A. signifer, P. tenuicorpa, Rhodeus pseudosericeus, and C. koreanus. A. signifer and P. tenuicorpa mainly inhabited the mid to lower streams, R. pseudosericeus in the midstream, and R. pseudosericeus in the upstream. P. tenuicorpa inhabited in large numbers, and estimating the age by total length-frequency distribution in July, the total length of the 26-35 mm group was estimated as 0 years old, the 54-75 mm group as 1 year old, 82-97 mm group as 2 years old, 104-109 mm group as 3 years or older. The cluster analysis showed that the dominance index decreased from upstream to downstream, but the diversity, evenness, and richness index increased. The water quality of Byekgye Stream was evaluated as good overall since the river health (fish assessment index, FAI) using fish was evaluated as excellent (5 stations) and good (2 stations). Byekgye Stream has relatively well-preserved habitats, but conservation measures are required as habitats are disturbed by river repair work in some parts of the midstream and downstream areas where many endangered species inhabit.

  • Forest fires often destory forests that have taken years to grow in a few minutes. Forest fire therefore, is an important problem in forest management and have caused heavy losses to the nations economy. In order to resolve this problem many investigations have been made in many countries. However, ecological studies on the forest after accidental fire have not yet been made in Korea. In order to conduct such a study, a burned area on Mt. Samak which is located at Dukduwon-ri, Seo-myon, Chunsung-gun, Kangwon-do, was chosen as experimental plot in 1967. The remaining seeds were collected from the burned area, and investigations on their germination rate and their productivity were made comparing to those of the seeds of undemaged area, and following results were obtained. 1. The number of seed collected from the control plots were 740 while it was 537 from the test plots (Table 3, 4). It was considered that this difference between burned and unburned area was mainly due to the fact that some of the seeds had been burnt by the fire, and the unfavorable environmental conditions in the burned area was also considered to be a reason. In the germination rate in the control plots showed 28.1% while it was 3.2% in the test plots. This difference was considered to be due to complete loss of viability of the seed by burning and high heat. 2. In the test plots, sixteen seeds of the Alnus japonica were collected and six of these seeds germinated (index number 100) which was the highest germination rate among the species of collected seeds. From these results, it was considered that a high temperature (above $150^{\circ}C$) caused reduction of the germination rate (Quadrat. 1.2). Seeds of Carex lanceolata var. Nana, were appeared much more in the higher plots than in the lower plots and it seemed to be due to the fact that the forest floor plants were much more abundant in the lower plots than in the higher plots which is covered with shrubbery. And some small seeds midght be able to avoid the effect of fire being burried in the soil or under the gravel. 3. With Pinus densiflora, 43 seeds were collected, and 11 of these germinated in the control plots. However in the test plots, 11 seeds were collected and no seed germinated. This shows that the Pinus densiflora was the weakest in resisting to heat among the observed species in this study. 4. Without exception the germination rate showed a higher index in the herbs than in the woody plants and it is believed that the herbs produced more seed than the wood plants because of the abundance of herbs colony.

  • The aim of this study was to characterize the forest vegetation and site of Tongdaesan(Odaesan National Park) for ecological forest management. The deciduous forests of Tongdaesan are the result of cool, temperate climate. Direction and slope morphology contribute to differences in species composition and vegetaion structure. Based on the BRAUN-BLANQUET-approach, 242 releves were recorded and subsequently classified. The vegetation could be divided into two forest community groups, seven forest communities, and several groups. South exposed, dry slopes were covered with forests of the Quercus mongolica-Acer pseudo-sieboldianum community group. Differential species included grasses and sedges, e.g., Calamagrostis langsdorfii, Carex humilis and C. lanceolata. Quercus mongolica-Acer pseudo-sieboldianum community group could be subdivided into Rhododendron mucronulatum var. ciliatum community, Lespedeza${\times}$tomentella community, Carex humilis-Maackia amurensis community, Quercus mongolica-Acer pseudosieboldianum typical community and Pimpinella brachycarpa community. Carex humilis-Maackia amurensis community was divided into Galium kinuta subcommunity and typical subcommunity. Pimpinella brachycarpa community was divided into Vicia venosa var. cuspidata subcommunity and typical subcommunity. On north exposed, more humid and moist sites, forests of the Dryopteris crassirhizoma-Cornus controversa community group were found. This group was subdivided into Acer tschonoskii var. rubripes-Acer tegmentosum community and Deutzia glabrata-Ulmus laciniata community. Deutzia glabrata-Ulmus laciniata community was divided into Acer tegmentosum subcommunity, Saxifraga punctata subcommunity and typical subcommunity.
