Effect of the supplementation of Coconopsis lanceolata extract on lipid metabolism amelioration in type 2 diabetes mouse model induced by high fat diet (울릉도 섬더덕 추출물의 급여가 제2형 당뇨 동물의 지질대사 개선에 미치는 영향)

  • The present study was designed to investigate the antihyperlipidemic effect of Coconopsis lanceolata extract in C57BL/6J mice. The mice were divided into four groups: normal diet group (ND), high fat diet group (HFD), positive control group with 0.05% metformin (PC), Coconopsis lanceolata extract group (UCL). After 5 weeks of feeding, average body weight of the UCL group mice was slightly decreased, while that of the HFD group significantly increased) Also, liver and adipose tissue weights in the UCL group significantly increased. The levels of trigliceride (TG) and total in the plasma of UCL-supplemented group were significantly lower than those of high fat diet group. On the other hand, HDL-cholesterol level was increased. Expression level of proteins related with adipogenesis such as SREBP-1c, ACC, and FAS in the liver of the UCL group mice was much lower comparing with the HFD group mice. In conclusion, the results showed that the Coconopsis lanceolata extract possesses significant antihyperlipidemic effects in C57BL/6J mice.

Neuroprotective effect of Coreopsis lanceolata extract against hydrogen-peroxide-induced oxidative stress in PC12 cells

  • The present study investigated the neuroprotective effects of Coreopsis lanceolate extract against hydrogen-peroxide (H2O2)-induced oxidative damage and cell death in pheochromocytoma 12 (PC12) cells. Reactive oxygen species (ROS), 2,2'-azinobis (3-ethylbebzothiazoloine-6-sulfonic acid) diammonium salt, and 1,1-diphenyl-2-picrrylhydrazyl radical scavenging activities, as well as the expression levels of proteins associated with oxidative damage and cell death were investigated. According to the results, C. lanceolate extract exhibited inhibitory activity against intracellular ROS generation and cell-damaging effects induced by hydroxyl radicals in a dose-dependent manner. Total phenolic and flavonoid contents were 22.3 mg·g-1 gallic acid equivalent and 16.2 mg·g-1 catechin equivalent, respectively. Additionally, a high-performance liquid chromatography (HPLC) assay based on the internal standard method used to detect phenolic compounds. The phenolic compounds identified in C. lanceolata extract contained (+)-catechin hydrate (5.0 ± 0.0 mg·g-1), ferulic acid (1.6 ± 0.0 mg·g-1), chlorogenic acid (1.5 ± 0.0 mg·g-1), caffeic acid (1.2 ± 0.0 mg·g-1), naringin (0.9 ± 0.0 mg·g-1), and p-coumaric acid (0.5 ± 0.0 mg·g-1). C. lanceolata extract attenuated pro-apoptotic Bax expression levels and enhanced the expression levels of anti-apoptotic Bcl-2, caspase-3, and caspase-9 proteins. Therefore, C. lanceolata is a potential source of materials with neuroprotective properties against neurodegenerative disorders, such as Alzheimer's and Parkinson's diseases.

Effects of Codonopsis lanceolata Extracts on bFGF-induced Angiogenesis in vitro (더덕추출물에 의한 bFGF-유도 시험관내 혈관신생의 억제)

  • In this study, we examined the effects of the methanolic extract(CL-ex) of Codonopsis lanceolata on the angiogenesis stimulated with basic fibroblast growth factor(bFGF) in vitro, using porcine pulmonary arterial endothelial cells(PPAECs). In addition, we investigated the endothelial functions involved in angiogenesis, such as proliferation, migration and secretion of matrix metalloproteinases(MMPs), using human umbilical vein endothelial cells(HUVECS). CL-ex inhibited FGF-induced sprout formation in vitro at concentrations of 0.1-100 ug/ml. Although CL-ex did not affect the proliferation of endothelial cells, CL-ex strongly inhibited the FGF-induced migration of HUVECS at concentrations of 0.1-1 ug/ml; the degree of inhibition of endothelial cells by C-ex was 49.4% at 0.1 ug/ml and 71.9 % at 1.0 ug/ml. Moreover, CL-ex inhibited the secretion of MMPs from HUVECS stimulated with FGF. Therefore, the inhibitory effect of CL-ex on angiogenesis in vitro could be explained by the inhibition of endothelial cell migration. From these results, we suggest that Codonopsis lanceolata is a useful herb for the development of therapeutics or preventive food factors for angiogenesis related diseases, such as tumors.

Effects of Fermented Milk with Hot Water Extract from Acanthopanax senticosus and Codonopsis lanceolata on the Immune Status of Mouse (가시오가피와 더덕 추출물을 첨가한 발효유가 마우스의 면역기능에 미치는 영향)

  • We investigated the immunomodulatory actions of water extract from Acanthopanax senticosus in male ICR mice. The mice were treated with fermented milk containing three added doses of freeze dried extract: 3 mg/kg (A), 9 mg/kg (B), and 27 mg/kg (C) of body weight with Acanthopanax senticosus: Codonopsis lanceolata (8:2) for 7 and 10 weeks, respectively. Organ weights, plaque-forming cell tests, agglutination tests, IgG tests, differential white cell counts, and histological tests were performed at the 7th and 10th weeks of dietary treatment. There were no significant differences in body weight and organ weight. The spleen indices of group B at 7 weeks and group C at 10 weeks were significantly higher than those of the control group (p<0.05). For the plaque-forming cell test, groups B and C at 7 weeks, and group C at 10 weeks, showed significant increases over the control group (p<0.05). The agglutination test decreased with an extended experimental period. Groups A, B, and C at 7 weeks, and groups B and C at 10 weeks, had greater antibody responses to sheep red blood cells (SRBC) than the control group. The IgG antibody production of group C at 7 weeks and groups B and C at 10 weeks were significantly higher than the control group (p<0.05). In groups B and C, lymphocyte percentage was higher than the control group, and their spleen and thymus tissues showed active immune reactions.

Neuroprotective effect of Deodeok (Codonopsis lanceolata) bud extracts in H2O2-stimulated SH-SY5Y cells (더덕순 에탄올 추출물의 신경세포 보호 효과)

  • Purpose: Deodeok (Codonopsis lanceolata) is generally used in conventional medicines and is considered to have remedial properties to cure several diseases. However, application of the C. lanceolata bud as a novel food ingredient has not been fully explored. Hydrogen peroxide (H2O2) is associated with the production of oxidative damage that results in mutagenesis, carcinogenesis, and cell death. This study examines the neuroprotective effect of C. lanceolate bud extracts (CLBE) on H2O2-stimulated apoptosis in SH-SY5Y cells. Methods: C. lanceolata bud of length 10 to 15 cm was collected and extracted using 70% ethanol. Cytotoxicity was evaluated by the EZ-cytox reagent, measurement of lactic dehydrogenase (LDH) release and reactive oxygen species (ROS). The morphological changes of the nuclei were determined using the Hoechst 33258 dye. Enzyme activities were analyzed using the caspase activity assay kit. Related protein expressions were quantified by the Western blot immunoassay in H2O2-stimulated SH-SY5Y cells. Results: Cell viability, LDH release and ROS generation, demonstrated neuroprotective effects of CLBE in H2O2-stimulated SH-SY5Y cells. The occurrence of apoptosis in H2O2-stimulated cells was confirmed by caspase activity, which was increased in H2O2-stimulated SH-SY5Y cells compared to the unexposed group. Pretreatment of CLBE was observed to inhibit the H2O2-stimulated apoptosis. In addition, exposure to CLBE resulted in increased expression of the Bcl-2 (B cell lymphoma 2) protein and decreased expression of the Bax (Bcl2 associated X) protein. Conclusion: This study shows that exposure to CLBE alleviates the H2O2-stimulated neuronal damage in SH-SY5Y cells. Our results indicate the potential application of CLBE in neurodegenerative disease therapy or prevention.

  • To evaluate the nutritive value, 13 Wild vegetables which commonly used for side dish were analyzed. Their Moisture are lower than that of other vegetables. Crude Protein content of thess Wild vegetables is more than 2% , which is higher than that of other vegetables and these tendencies were also shown in the content of Crude Ash. The content of Crude Fat and of Crude Fiber are similar in the Wild vegetables, but Crude Fiber of C. lanceolata TRAUTV is three times higher than that of other Wild vegetables. In all Wild vegetables, the K content of Mineral is similar, but that of C. lanceolata TRAUTV is very low. The content of Ca and P are quite different among Wild vegetables. The content of Mg is also similar each other, about 16mg%, but that of P. agwilinum var. latiusculum UNDERW, A. elata SEEM, A. altissima SWINGLE, root of A. triphylla (Thunb.)A.DC., H.Thunbergii BAK. is 8.99, 7.69, 4.30, 4.13 and 1.92mg% respectively. Total Amino acids were contained in order of A. elata; 3072.4mg%>C. lanceolata TRAUTV; 2709.4mg%>P. agwilinum var. latiusculum UNDERW, A. altissimB SWTNGLE, A. Japonica KOIDZ, S.palmata MAX. : about 2350mg%>L.Jaluensis KOM., P.Caponicus MAX., leaf of A. triphylla(Thunb.)A.DC., C.Japonicum var. ussuriense, H.plantaginea ASCHERS; about 1400mg%>H.Thunbergii BAK., A.scaber THUNB, root of A. triphylla(Thunb.) A.DC. ; about 600mg%, Asp.A.A. and Glu.A.A. content, 46% of Total A.A. is higher than other Amino acids and the content of Arg. A.A. in C.lanceolata TRAUTV which is higher than that of other Amino acids is 6 times highly than that of other Wild vegetables. A. altissima SWINGLE, H.plantaginea ASCHERS, A.Japonica KOIDZ contain Vitamin C a little avobe, 45.9mg%, 36.8mg%, 30.2mg% respectively. Glucoslde content of L.Jaluensis KOM., C.lanceolata TRAUTV which are 4.1%, 3.5%, were highest of all ana that of P.Caponicus MAX., P.agwilinum var. latiusculum UNDERW, 2.2%, 2.0% is in the second, which seems to be the cause of bitter taste.

Swimming Performance Evaluation of Four Freshwater Fish Species from the South Korea (국내에 서식하는 담수어류 4종에 대한 유영능력 평가)

  • Swimming performance of fish is an important factor in the survival of fish. Also, swimming performance of fish is used in the form of habitat, or as a condition to consider when creating a fish ladder. However in Korea, researches in swimming performance of Korean freshwater fish were scarce and inadequate in some part, thus fish ladders were installed without considering their swimming performance. Therefore, in this study, we measured swimming performance of 4 Korean freshwater fish species to consider importance of swimming performance test. The fish used in this study were Carassius auratus, Zacco koreanus, Gnathopogon strigatus, Acheilognathus lanceolata intermedia species which was collected during October to November, 2018 at Geum River, and measurement for swimming speed of each fish was done by using $Loligo^{(R)}$ System, swim tunnel respirometer in January to February of 2019. The average value of the burst critical swimming speed ($U_{crit}$) for each species was $0.8{\pm}0.04m\;s^{-1}$ for C. auratus, $0.77{\pm}0.04m\;s^{-1}$ for Z. koreanus, $0.95{\pm}0.04m\;s^{-1}$ for G. strigatus, $0.73{\pm}0.03m\;s^{-1}$ for A. lanceolata intermedia and the average value of prolonged critical swimming speed was $0.54m\;s^{-1}$ for C. auratus, $0.67m\;s^{-1}$ for Z. koreanus, $0.7m\;s^{-1}$ for G. strigatus, $0.54m\;s^{-1}$ for A. lanceolata intermedia. Since the fish used in this experiment were collected from a small part of the water system in Korea and there were only 4 species, they were not enough to represent the species that inhabit the entire Korean water system. It will be necessary to continue evaluating the swimming performance of other freshwater species in Korea.

Effectof natural type ABA foliar application on growth, yield of Codonopsis lancelata (天然型 ABA의 葉面散布가 더덕의 生長, 收量에 미치는 影響)

  • In order to improve cultivation technuque of C. lanceolata, natural ABA was treated with foliar application periodically during differenctitation of node. The higher is concentration and the earier its foliar application was, the shoter plant height was. Especially, when $10mg;.L{-1}$ of ABA was treated at differentiated stage of 3rd node, plant height was inhibited to 60% of control. But leaf length, leaf width, and number of branches have no significant differnence in comparison wiht control. The fresh weight of subterranean part was similar to control independent of treat-time in the case of $10mg;.L{-1}$. When 1,5 and $10mg;.L{-1}$ of ABA were treated at initial differentiated node stage, plant height inhibited to 20~30% of control, but subterranean part was similar to control. All treatement showed slight antitumor activity by the P388 cytotoxic screening test.

  • Codonopside lc is an active natural compound isolated from the roots of Codonopsis lanceolata (Campanulaceae), a Korean medicinal herb. In the present study, we investigated the in vitro effect of Codonopside 1c on the proliferation and induction of apoptosis in HL-60 human promyelocytic cells. When HL-60 cells were treated with Codonopside 1c, evidence of apoptotic features, including DNA fragmentation, formation of DNA ladder in agarose gel elecrophoresis and increse of annexin V binding, were obtained.
