설사자돈으로부터 병원성대장균, 캠필로박터속균 및 살모넬라속균의 분리동정 (Isolation of Enteropathogenic Escherichia coli, Thermophilic Campylobacter and Salmonellae from Scouring Piglets)

  • This study was conducted to isolate etiological agents from the 103 scouring piglets in Gyeongnam area and also carried out antimicrobial drug susceptibility test and epidemiogical served. The incidence of scouring piglet was most prevalent as 81.6% in the age of 2 to 4 weeks after birth, while the rate was less than 10% in the age of 5 to 6 weeks and under 1 week after birch. When compared the isolation frequency of the each etiological agent, enteropathogenic E. coli was most prevalent as 46.6%, thermophilic Campylobacter 26.2% and Salmonellae was 8.7% in order. In the OK serotyping for 117 isolates of enteropathogenic E. coli, type 0141 : K85 (20.5%), 0157:K88ac(14.5%), 0138:K81 and 0149:K91 (13.3%) were encountered most frequently. In the biotyping for 27 isolates of thermophilic Campylobacter, most strains of C. jejuni were belong to type I (50.0%) and II (25.0%), and most strains of C. coli were belong to biotype I (78.9%). In the serotyping for 9 strains of Salmonellae, 3 strains were grouped as D, 2 strains as C. and each 1 strain was group B and E. The other 2 strains were untypable. The 117 isolates of enteropathogenic E. coli were resistant more than 90% to erythromycin, penicillin, tetracycline and streptomycin, wherease about 90% of the isolates were sensitive to kanamycin and gentamicin. In the case of Salmonellae, all of the isolates were resistant to penicillin, but about 89% of the isolates were sensitive to gentamicin and colistin. All of C. jejuni and C. coli isolates were resistant to cephalothin, but more than 89% of C.jejuni and C. coli were sensitive to kanamycin and gentamicin.

백년초 추출물에 의한 신선절단 사과의 저장 중 E. coli O157:H7과 Listeria monocytogenes의 생육저해 (Effects of Opuntia ficus indica Extracts on Inactivation of Escherichia coli O157:H7 and Listeria monocytogenes on Fresh-cut Apples)

  • 신선절단 사과에 E. coli O157:H7과 L. monocytogenes를 인위적으로 접종한 후 백년초 추출물 처리에 따른 저장 중 미생물 수 변화를 조사하였다. 신선절단 사과에 접종된 E. coli O157:H7과 L. monocytogenes의 초기 균수가 5~6 log CFU/g이 되게 하였고, 사용한 백년초 추출물은 25, 50, 100, 200 mg/mL이었으며 4, $21^{\circ}C$에서 8일간 저장하였다. E. coli O157:H7은 백년초 추출물 50 mg/mL 이상 처리구에서 유의적으로 E. coli O157:H7 생육저해 효과가 나타났음을 확인할 수 있었으며, 온도별로는 $21^{\circ}C$ 저장 시에 생육 억제효과가 더 큰 것으로 판단되었다. L. monocytogenes는 E. coli O157:H7에 비교하여 매우 민감하게 반응하였으며 저장 온도별 차이는 크지 않았다. 특히 50 mg/mL 이상 처리구에서 2일째부터 4, $21^{\circ}C$ 저장 시 모두 L. monocytogenes가 검출한계 이하로 관찰되어 생육억제에 매우 효과적임을 확인할 수 있었다. 그리고 관능평가에서는 100~200 mg/mL 이상의 백년초 추출물 처리가 신선절단 사과의 품질을 잘 유지하는 것으로 나타났다. 따라서 본 연구결과, 백년초 추출물 처리는 신선 절단 사과의 저장, 유통 중 오염될 수 있는 E. coli O157:H7과 L. monocytogenes의 생육억제에 매우 효과적인 천연물이며, 향후 미생물학적 안전성 확보와 신선식품의 품질유지 향상에 효과적인 천연물 항균제로서 개발 가능하리라 판단된다.

Escherichia coli O157:H7의 살균을 위한 감마선과 가열처리의 효과 (Effects of Gamma-Ray and Heat Treatment on Sterilization of Escherichia coli O157:H7)

  • 본 연구는 Escherichia coli O157:H7 균주를 살균하고자 감마선 단독 및 가열과의 병용처리 효과를 조사하였다. E. coli O157:H7 균주의 가열 단독처리시 D값은 $50^{\circ}C$에서 129.2분, $55^{\circ}C$에서 27.1분, $60^{\circ}C$에서 2.4분으로 나타났다. E. coli O157:H7 균주는 0.03 M cysteine, 1% sodium citrate 및 5% sucrose가 첨가된 배지에서는 열에 대한 저항성이 증가하였고 1%, meat extract, 1% casein 및 1% casamino acid가 첨가된 배지에서는 감소하였다. 감마선 단독처리시는 $D_{10}$ 값과 불활성화 계수가 0.116 kGy와 $17{\sim}25$ 이였으나 가열과의 병용처리시는 약 0.07 kGy와 $25{\sim}41$로 나타났다. 이로서 가열과 방사선의 병용처리는 E. coli O157:H7 균주의 방사선 감수성을 현저하게 상승시켜 본 균주의 살균에 효과적이었다.

은/셀라이트 복합체의 항균특성에 관한 연구 (Study of Antimicrobial Properties for Silver/Celite Composites)

  • 본 연구에서는 은 나노 입자와 탄산은(Silver Carbonate)을 이용하여 제조된 은나노/셀라이트(SN/C)와 탄산은/셀라이트(SC/C) 복합체의 대장균에 대한 항균특성을 조사하였다. SN/C와 SC/C 복합체의 특성은 주사전자현미경, X-선 회절법, 그리고 원자흡수분광법에 의해 알아보았다. SN/C와 SC/C 복합체의 대장균에 대한 최소 억제농도는 0.541과 0.344 ppm이었고 완전 사멸농도는 1.427과 1.623 ppm으로 나타났다. 상기의 결과로 SN/C와 SC/C 복합체의 대장균에 대한 항균효과를 확인할 수 있었다.

농산물 접촉 표면 재질에 따른 Escherichia coli O157:H7의 생존 및 상추로의 교차오염도 조사 (Survival and Cross-contamination of Escherichia coli O157:H7 on Various Agricultural Product-Contact Surfaces)

  • 본 연구는 polyvinyl chroride (PVC)와 stainless steel 표면에서 E. coli O157:H7의 부착, 바이오필름형성, 온 습도에 따른 생존율을 조사하고, E. coli O157:H7이 오염된 작업대 표면에서 상추로의 교차오염도 조사를 통하여 E. coli O157:H7에 오염된 작업대가 상추의 미생물학적 안전성에 미치는 영향을 평가하고자 수행하였다. 작업대 표면 재질인 PVC와 stainless steel에 E. coli O157:H7 배양액을 1시간 노출시켰을 때, PVC의 경우, stainless steel 보다 10배 정도 부착력이 높았으나, 6시간째는 두 재질간의 차이가 없었다. 또한, E. coli O157:H7의 바이오필름 형성은 TSB > 10% 상추추출액 > 1% 상추추출액 > phosphate buffer 순이었으며, 재질별로는 PVC에서 바이오필름형성능이 높은 것으로 나타났다. 온도 $20^{\circ}C$, $30^{\circ}C$, 습도 43%, 69%, 100%에서 각각 노출 시켰을 때, E. coli O157:H7은 온도 $30^{\circ}C$, 습도 43%, 69% 조건에서 12시간 내에 약 5.0 log CFU/coupon이 감소한데 반해, 상대습도 100%에서는 농도의 큰 변화 없이 5일 이상 생존이 지속되었다. 또한, 유기물이 작업대 표면에 존재 시, E. coli O157:H7의 감소 속도가 느렸다. E. coli O157:H7에 오염된 작업대에 상추를 접촉시키고 상추의 오염도를 조사한 결과, 상추 표면에 수분이 존재할 때 오염된 작업대 표면에서 E. coli O157:H7의 이동수준이 상추에 수분이 없을 때 보다 10배 이상 증가하는 것으로 나타났다. 따라서 E. coli O157:H7에 오염된 작업대는 상추의 안전성에 직접적인 영향을 미칠 수 있어 작업 후에 세척, 소독을 통하여 위생적으로 관리하는 것이 필요하다.

Cloning and Expression of pcbC and pcbD Genes Responsible for 2,3-Dihydroxybiphenyl Degradation from Pseudomonas sp. P20

  • Pseudomonas sp. P20 was shown to be capable of degrading biphenyl and 4-chlorobiphenyl (4CB) to produce the corresponding benzoic acids wnich were not further degraded. But the potential of the strain for biodegradation of 4CB was shown to be excellent. The pcbA, B, C and D genes responsible for the aromatic ring-cleavage of biphenyl and 4CB degradation were cloned from the chromosomal DNA of the strain. In this study, the pebC and D genes specifying degradation of 2, 3-dihydroxybiphenyl (2, 3-DHBP) produced from biphenyl by the pebAB-encoded enzymes were cloned by using pBluescript SK(+) as a vector. From the pCK102 (9.3 kb) containing pebC and D genes, pCK1022 inserted with a EcoRI-HindIII DNA fragment (4.1 kb) carrying pebC and D and a pCK1092 inserted with EcoRI-XbaI fragment (1.95 kb) carrying pebC were constructed. The expression of pcbC and D' in E. coli CK102 and pebC in E. coli CK1092 was examined by gas chromatography and UV-vis spectrophotometry. 2.3-dihydroxybiphenyl was readily degraded to produce meta-cleavage product (MCP) by E. coli CK102 after incubation for 10 min, and then only benzoic acid(BA) was detected in the 24-h old culture. The MCP was detected in E. coli CK1022 containing pebC and 0 genes (by the resting cells assay) for up to 3 h after incubation and then diminished completely in 8 h, whereas the MCP accumulated in the E. coli CK1092 culture even after 6 h of incubation. The 2, 3-DHBP dioxygenases (product of pebC gene) produced by E. coli CK1, CK102, CK1023, and CK1092 strains were measured by native PAGE analysis to be about 250 kDa in molecular weight, which were about same as those of Pseudomonas sp. DJ-12, P. pseudoa1caligenes KF707, and P. putida OU83.

Effect of Temperature on Survival of Escherichia coli O157:H7 and Listeria monocytogenes in Livestock Manure Compost

  • Animal manure compost is a commonly used fertilizer in organic vegetable and fruit production in Korea. However, livestock manure compost produced from animal feces can contain a lot of the non-pathogenic and pathogenic bacteria. Of particular concern are bacteria causing human food-borne illness such as Escherichia coli O157:H7 and Listeria monocytogenes. The objective of this study was to investigate effect of temperature on survival of E. coli O157:H7 and L. monocytogenes in livestock manure compost. Commercial livestock manure compost (manure 60%, sawdust 40%) was inoculated with E. coli O157:H7 and L. monocytogenes. Compost was incubated at four different temperatures (10, 25, 35, and $55^{\circ}C$) for 20 weeks. Samples were taken every week during incubation depending on the given conditions. E. coli O157:H7 persisted for up to 1 day in livestock manure compost at $55^{\circ}C$, over 140 days at $10^{\circ}C$, 140 days at $25^{\circ}C$, and 120 days at $35^{\circ}C$, respectively. L. monocytogenes persisted for up to 1 day in livestock manure compost at $55^{\circ}C$ and 140 days at $10^{\circ}C$, 70 days at $25^{\circ}C$, and 40 days at $35^{\circ}C$, respectively. The results indicated that E. coli O157:H7 and L. monocytogenes persisted longer under low temperature condition. E. coli O157:H7 survived longer than L. monocytogenes at three different temperatures (10, 25, and $35^{\circ}C$). The results are being used to develop guidelines on the management of manure to reduce the risks of E. coli O157:H7 and L. monocytogenes transmission to foods produced in the presence of animal waste.

수생조류에서 분리한 대장균의 항균제 내성 및 Tetracycline 내성인자의 분포 (Antimicrobial resistance and distribution of tetracycline resistance determinants in Escherichia coli isolated from aquatic birds)

  • One hundred and sixty nine Escherichia (E.) coli strains isolated from fecal samples of aquatic birds in Geumho river basin and Dalseong park were tested by agar dilution method to determine their susceptibility patterns to 14 antimicrobial agents. The distribution of tetracycline resistance determinants (tetA, tetB, tetC, tetD and tetE) were also examined by PCR in 76 tetracycline-resistant ($TC^r$) E. coli isolates. The high resistance was observed in tetracycline, cephalothin and ampicillin (45.0~36.7%). Resistance of E. coli isolates derived from Dalseong park to tetracycline, cephalothin, ampicillin and streptomycin (65.7~44.8%) were significantly higher than those isolated from Geumho river basin (31.4~14.7%). About seventy percent (70.4%) of the strains isolated were resistant to one or more drugs tested. Thirty (39.5%) of 76 $TC^r$ E. coli isolates which were resistant to one or more drugs transferred all or a part of their resistance patterns to the recipient strain of E.coli J53 by conjugation. All of $TC^r$ E. coli isolates contained at least one or more of 5 tet genes examined. The most common genes found in these isolates were tetA (60.6%) and followed by tetB (7.9%) and tetC (1.3%). However, tetD and tetE were not found in any of the isolates tested. Twenty one (27.6%) of $TC^r$ E. coli isolates had two determinants, tetA/tetB (20 strains), tetA/tetC (1 strain). And two strains (2.6%) contained three determinants (tetA/tetB/tetC).

Constitutive Expression of Lipase on the Cell Surface of Escherichia coli using OmpC Anchoring Motif

  • We have developed a constitutive display system of the Pseudomonas fluorescens SIK W1 TliA lipase on the cell surface of Escherichia coli using E. coli outer membrane protein C (OmpC) as an anchoring motif, which is an economical compared to induced system. For the constitutive expression of truncated OmpC-TliA fusion proteins, gntT104 promoter was employed. Cell growth was not affected by over expression of fusion protein during entire culture time, suggesting cell lysis was not a problem. The localization of truncated OmpC-TliA fusion protein on the cell surface was confirmed by immunofluorescence microscopy and measuring whole cell lipase activity. Constitutively displayed lipase was very stable, retaining activity enantioselectivity throughout the five repeated reactions. These results suggest that OmpC from E. coli be a useful anchoring motif for displaying enzymes on the cell surface without any inducers, and this stable surface display system can be employed for a broad range of biotechnological applications.

과량 생산된 대장균 chitin 분해효소의 정제 및 특성 조사 (Purification and Characterization of the Overproduced E. coli Endochitinase)

  • 실험실 배양 조건에서는 발현되지 않는 대장균 K-12의 endochitinase 유전자(yheR)를 PCR로 증폭하여 pET28c와 pQE9벡터에 각각 클로닝 하였다. yheB유전자를 가진 pET28c와 pQE9 벡터를 함유한 대장균에서 생산된 endochitinase는 생장배지 내로 일부 분리되었다. 과량 생산된 endochitinase를 His-affinity 크로마토그래피와 DE-52 크로마토그래피로 부분 정제하였으며 SDS-PAGE에 의한 단백질의 분자량은 약 97,000 이었다. 정제된 효소의 최적 pH는 6이었으며 최적 온도는 $40^{\circ}C$이었다.
