1999년 여름동안 동해안의 7개 석호와, 내륙의 5개 인공호를 대상으로 autotrophic picoplankton (APP)의풍부도와 총 식물플랑크톤 생물량에 대한 기여도를 평가하였다. 석호에서는 부영양화가 심한 호수에서 주로 나타나는 phycocyanin-rich APP가 우점한 반면 인공호에서는 주로 phycoerythrin-rich APP가 우점하였다. 동해안 석호에서 APP의 세포밀도와 생물량은 각각 $3.6{\times}10^3{\sim} 5.0{\times}10^6\;cells/ml$와 $1.0{\sim}1,385.0\;{\mu}gC/L$의 범위를 나타냈고 인공호에서는 각각 $3.8{\times}10^4{\sim}3.6{\times}10^5\;cells/ml$와 $15.3{\sim}128.2\;{\mu}gC/L$의 범위로 동해안석호에서 더 높은 APP의 풍부도를 나타냈다. 특히 경포호에서는 3회조사 모두 $10^6\;cells/ml$이상의 높은 세포밀도를 보였는데 이는 세계적으로 보고된 가장 높은 밀도 수준이다. 소양호에서 APP의 수직분포를 조사한 결과 수온약층에서 최대 세포밀도를 나타냈는데 이는 APP가 낮은 광도에서도 성장이 가능하며 심층의 높은 영양염류를 이용할 수 있었기 때문으로 사료된다. APP세포밀도는TN/TP비와 음의 상관관계,TP와는 양의 상관관계를 나타내어 APP의 풍부도가 호수의 부영양화와 함께 증가하는 경향을 나타내었다. 총 식물플랑크톤 생물량에 대한 APP의 기여도는 $0.1{\sim}85.0%$로 호수간의 큰 차이를 보였다. 본 연구결과는 지금까지 식물플랑크톤 연구에서 소외되어져 왔던 APP가 호수 생태계의 일차생산자로써 중요한 부분을 차지하고 있음을 보여주고 있다.
Arthrospira platensis (A. platensis) is a microscopic and filamentous cyanobacterium that derives its name from the spiral or helical nature of its filaments. In this study, we induced mutants of A. platensis through NMU treatment and selected two strains by level of ipid contents. We named mutant '1-9', '2-5', and they were cultivated in the same way with the wild type. During 12 days cultivation, cell growth, dry cell weight, pigment content, and lipid content were measured for characteristics of mutants. As a result, pigment and lipid content of mutants were increased about 3.6, 1.8 times compared with wild type, respectively. It was shown that total flavonoid and polyphenol contents of mutants were increased about 1.5 times compared with wild type. And radical scavenging effect of mutants were increased about 10% compared with wild type.
Riceyeotgangjung (RYG) with added spirulina was prepared with citron juice for masking of odor. Oil temperature for puffing was found to be $220^{\circ}C$ for frying dry rice based upon the results of expansion rate and color. RYGs with added spirulina (1, 2, and 3%) were prepared under the above conditions, and their physicochemical and sensory properties were examined under storage at $60^{\circ}C$ for 15 days. Moisture content of RYG decreased proportionally with the spirulina content and storage time. The hardness levels of the freshly made RYGs with spirulina were higher than that of control. During storage, the hardness of the control gradually increased, but those of the RYGs with spirulina gradually decreased, although no significant differences existed between the spirulina containing RYGs. Lightness and redness values in the Hunter color system decreased according to the spirulina content and storage time. Yellowness value also increased with the spirulina content and storage time. Phycocyanin and total phenolic content increased with spirulina content, but decreased with the storage period. Sensory evaluation results showed that RYG with 2% added spirulina received the highest scores for color, taste, overall acceptability, and intention of purchase. As a result, the optimal addition amount of spirulina to RYG is recommended to be 2%.
사상체 토착 남세균을 경상남도 합천군 합천호의 수화시료로부터 무균적으로 분리하였으며, 형태적·분자적 동정 결과 림노트릭스 속에 속하는 것으로 밝혀졌다. 따라서, 본 남세균 균주는 림노트릭스 속 KNUA012 균주로 명명하였으며, 분리균주의 최적생장 온도는 섭씨 25도였다. 지질성분 분석 결과, 에스테르 교환반응을 거치지 않고 직접 연료로 사용할 수 있는 펜타데칸(C15H32)과 헵타데칸(C17H36)과 같은 알칸들이 본 균주에 의해 광독립 영양적으로 생합성 된다는 것이 밝혀졌다. 또한 알칸 생합성에 관여하는 유전자들이 본 남세균 내에 존재하는 것을 발견하였다. 일반적인 미세조류 바이오디젤 구성성분으로 알려진 미리스트올레산(C14:1), 팔미트산(C16:0) 및 팔미톨레산(C16:1) 역시 KNUA012 균주에 의해 주요 지방산 성분으로서 생산되는 것으로 확인되었다. 근사분석 결과 KNUA012 균주의 휘발성물질 함량은 86.0%였으며, 원소분석 결과 고위발열량은 19.8 MJ kg−1으로 나타났다. 또한, 본 분리균주는 고부가가치 항산화물질로 알려져 있는 피코시아닌을 광독립영양적으로 21.4 mg g−1의 농도로 생산할 수 있는 것을 확인하였다. 따라서, 본 연구결과는 KNUA012 균주가 미세조류 기반 바이오연료와 바이오매스 원료의 경제적인 생산에 있어 이상적인 자원이 될 수 있음을 제시하였다.

Intact phycobilisomes from a wild-type red Chondrus crispus and its vegetatively derived green mutant were isolated by centrifugation through a discontinuous sucrose density gradient. Pigment composition was subsequently characterized by spectrophotometry. Vegetative thalli of the two strains grown together for six months in the laboratory resulted in different pigment profiles. Two pigmented phycobilisome bands appeared in the sucrose gradient of the wild-type alga, a purple coloured one, and a pink one, whereas only a single blue band appeared in the gradient of the green mutant. Spectrophotometric and fluorescence analyses identified the phycobiliprotein composition of the purple band as the typical phycoerythrin-phycocyanin-allophycocyanin complement in the wild-type, but there was no detectable phycoerythrin present in the blue band of the green mutant. Sodium dodecyl sulphate, preparative polyacrylamide gel electrophoresis analysis confirmed the presence of allophycocyanin subunits in all extracts, but firm evidence of an R-phycoerythrin linker polypeptide in the blue band was missing. These results highlight the ability of C. crispus to adapt to a phycoerythrin deficiency by adjusting light harvesting pigment ratios.
The cyanobacterium Anabaena doliolum Ind1 isolated from a coal mining site was tested for removal of cadmium at optimum pH 7.0 and temperature $25^{\circ}C$. The organism recorded high percentage of metal removal (92-69%) within seven days of exposure to 0.5-2.0 ppm cadmium. Biosorption onto the cell surface was the primary mode of metal removal. Fourier transform infrared spectroscopy (FTIR) established hydroxyl, amides, carboxyl, sulphate and carbonyl groups to be the major functional groups on the cell surface involved in cadmium binding. Cellular ultrastructure and a range of vital physiological processes (i.e., photosynthetic pigments, respiration, photosynthesis, heterocyst frequency and nitrogenase activity) remained unaffected upon 0.5 ppm treatment; higher concentrations of cadmium exerted visible adverse effects. Amongst the five photosynthetic pigments tested, phycocyanin was the most targeted pigment (inhibition was 15-89%). Both respiration and photosynthetic activities were inhibited by cadmium with more severe effect seen on respiration. 2.0 ppm cadmium exposure also had drastic negative effect on nitrogenase activity (87% decreased).
A strain of the cyanobacterium Arthrospira was isolated from Lake Chahannaoer in northern China and was characterized according to microscopic morphology, photosynthetic oxygen-evolving activity, growth rate, and nutritional profile. Compared with thermophilic Arthrospira species occurring naturally in tropical and subtropical lakes, this isolate is mesophilic and grows optimally at ${\sim}20^{\circ}C$. The total protein, fatty acid, phycocyanin, carotenoid, and chlorophyll a contents were 67.6, 6.1, 4.32, 0.29, and 0.76 grams per 100 grams of dry weight, respectively. The strain is rich in polyunsaturated fatty acids (PUFAs). An essential omega-3 fatty acid, docosahexaenoic acid (DHA), was detected, and ${\gamma}$-linolenic acid (GLA) and DHA accounted for 28.3% of the total fatty acid content. These features of this newly isolated strain make it potentially useful in commercial mass culture in local areas or as a biofuel feedstock. It is also an alternative resource for studying the metabolic PUFA pathways and mechanisms of cold stress tolerance in cyanobacteria.
Quality characteristics and antioxidant activity of spirulina with added tofu were investigated in the presence of different coagulants ($CaSO_4$, $MgCl_2$, Glucono delta lactone (GDL), vinegar). The yield of the spirulina tofu was the highest when coagulated with GDL. The pH of spirulina tofu with $MgCl_2$ was the highest compared to those of other coagulants. The tumidity of spirulina tofu was the highest with $MgCl_2$. In the color analysis of spirulina tofu, the L (lightness) value was the highest with $CaSO_4$, whereas a (redness) value was the highest with $MgCl_2$. C-phycocyanin contents were the highest with the $CaSO_4$. Phycoerythrin of spirulina tofu for vinegar was 0.194 mg/mL, and GDL content was 0.214 mg/mL. Additionally, the antioxidant activity of spirulina tofu was the highest with $MgCl_2$$IC_{50}$ values of the DPPH radical scavenging activity of spirulina tofu with $MgCl_2$ was 89.2 mg/g, GDL 93.5 mg/mL, $CaSO_4$ 116.7 mg/mL, and vinegar 118.0 mg/mL. The texture analyses (TPA) results showed that hardness was in the order of vinegar>$MgCl_2$>GDL>$CaSO_4$. For the overall acceptability of the preference test, the spirulina tofu that was made using $MgCl_2$ coagulant had the highest score. Based on these results, it is suggested that $MgCl_2$ is the proper coagulant for the preparation of spirulina with added tofu.
Pure lines were isolated from young gametophytic blades of pigmentation and morphological mutants in Porphyra tenera. Growth, blade-shape and photosynthetic pigment content of pure lines were compared with the wild type. Growth of blade length in the wild type (W, R-B), with round shape and brown color, was fastest at $5{\sim}10^{\circ}C$ and became slower as temperature increased. The blade-shape of the wild type changed from linear to round as temperature increased. The green type (R-G), with round shape and green color, showed slower growth, and the red type (R-R) 'with round shape and red color' showed faster growth than the wild type. The blade-shapes of the green and red types changed from elliptical or linear to round as temperature increased. The phycoerythrin (PE) / phycocyanin (PC) ratio of the green type was markedly lower and the PE/PC ratio of the red type was markedly higher than that of the wild type. The linear type (L-B), with liner shape and brown color, showed faster growth in blade length than the wild type at $10{\sim}20^{\circ}C$ and maintained its linear shape at $5{\sim}15^{\circ}C$. The content of photosynthetic pigments of the linear type was similar to that of the wild type. Each of the pure lines of pigmentation and morphological mutants that were isolated in the present study showed particular patterns in growth, blade-shape and photosynthetic pigment composition. Therefore, they are expected to be useful as new varieties by themselves and to be available for breeding and biotechnological studies.
마른 김의 배소(焙燒) 및 배소김의 저장중에 일어나는 색소(chlorophyll a, carotenoid 및 biliprotein). Trypsin Tndigestible Substrate(TIS) 및 dietary fiber 등 성분들의 변화를 측정하고 색소단백, TIS 및 소화율과의 관계를 고찰한 결과를 요약하면 다음과 같다 1. 마른김을 $50,\;70,\;90,\;100,\;120^{\circ}C$ 에서 각각 1시간 가열처리하였을 때 chlorophyll a와 carotenoid는 비교적 안정하여 $120^{\circ}C$ 에서도 $85\%$이상의 잔존량을 보였다. 그러나 biliprotein은 매우 불안정하여 $70^{\circ}C$에서 $60\%$$100\~120^{\circ}C$에서는 $20\~10\%$의 잔존량에 불과하였다. Chlorophyll a는 carotenoid보다 다소 안정하였고 biliprotein 색소중에서는 phycoerythrin보다 phycocyanin이 더 불안정하였다. 2. TIS는 열에 대하여 비교적 안정하였는데 $100^{\circ}C$ 이상에서 $10\%$감소하였고 소화율은 대조시료가 $80\%,\;100^{\circ}C$ 처리시료에서는 $85\%$로 증가하였다. TIS의 감소와 소화율의 증가는 색소 단백의 감소와 상관관계를 보였다. 3. 열처리중에 insoluble dietary fiber(IDF)가 다소 감소하는 경향을 보였으나 온도별 영향은 크지 않은 반면에 soluble dietary fiber(SDF)는 열처리 온도가 높을수록 다소 증가하는 경향을 보였다. 4. 배소김을 실온에서 20주간 저장하였을 때 chalorophyll a는 aw 0.65의 경우 약 $20\%$ aw 0.1에서는 $75\%$이상, carotenoid의 경우 aw 0.1일때 $40\%$, aw 0.65일 때 $2\%$ 정도와 잔존량을 보여 저장중 크게 감소하는 반면, biliprotein 색소는 안정하였다. 5. 저장중 TIS는 aw 0.65일 때 $15\%$ 정도가 감소하였고 같은 조건에서 소화율은 $92\%$로 증가하여 TIS와 소화율과의 관계를 나타내었고 SDF는 점차 증가하여 12주 경부터 상당한 변화를 보였다.
