Stability of Phycocyanin and Spectral Characteristic of Phycobilins from Spirulina platensis (Spirulina platensis가 생산하는 phycobilins의 spectral 특성 및 phycocyanin 색소의 안정성)

  • The stability of c-phycocyanin and spectral property of phycobilins obtained from Spirulina platensis cultured by helical tubular photobioreactor were determined. The c-phycocyanin with maximal absorption of 622 nm and allophycocyanin with maximal absorption of 652 nm fractions were isolated from phycobilins by Sephadex G-100 gel chromatoBraphy, The yield of partially purified c-phycocyanin was about $1.5{\%}$ to dried biomass. The stability of c-phycocyanin in the range of $pH 4{\~}9$ was high but c-phvcocvanin was unstable over pH 10. The c-phycocyanin was stable at temperatures below $40^{\circ}C$, and at light intensity below 15000 lux. And metal ions were not affect the stability of c-phycocyanin.

Content of Phycocyanins and Growth of Spirulina platensis with Culture Conditions (배양 조건에 따른 Spirulina piatensis의 성장 및 phycocyanin 함량 변화)

  • The growth of the microalgal Spiulina platensis in a batch photobioreactor had been studied to determine the influence of temperature, light intensity, and culture medium on the growth and c-phycocyanin content of the biomass. The most favorable conditions for high biomass and c-phycocyanin production were as follows: light intensity of 3500 lux, temperature of $35^{\circ}C,\;NaHCO_3\;of\;1.0{\%}$ for pH control, $0.2{\~}0.3{\%}\;Na_2CO_3$ for carbon source, and $0.2{\~}0.3{\%}\;NaCO_3$ for nitrogen source. The c-phycocyanin and chlorophyll content on most favorable condition were about $11{\%},\;1.0{\%}$, respectively.

Isolation of Chromoprotein and Its Amino Acids Composition in Korean Laver (한국산(韓國産) 김중의 색소단백질(色素蛋白質)의 분리(分離) 및 그 아미노산(酸) 조성(組成))

  • In this research chromoproteins, namely phycoerythrin and phycocyanin in Korean laver are isolated and purified and their characteristics and the contents of amino acid are investigated. Three kinds of laver, Yonpyungdo product, Jindo product, and Kohung product are used in the experiments. The results are as following: 1) 0.68g of phycoerythrin and 0.72g of phycocyanin are isolated from 50g of laver. 2) when observed by microscope (1,000 times amplification), the crystals of phycocrythrin have the shape of small needles and phycocyanin that of platelets. 3) Absorption spectra of purified phycoerythrin shows strong absorption peaks at 546, 526, and $496m{\mu}$,and phycocyanin shows strong absorption peaks at 610 and $546m{\mu}$. After the phycoerythrin and phycocyanin were kept at $4^{\circ}C$ for 24 and 48 hours. The peaks of phycoerythrin are slightly shifted and those of phycocyanin are decreased. 4) In amino acid analysis, sixteen kinds of amino acids are detected in phycoerythrin and phycocyanin. Amino acids found in the greatest amounts in phycoerythrin are glutamic acid, alanine, glycine and leucine. Essential amino acids are found to exist in phycocyanin in greater amounts than in phycoerythrin.

Differential Responses of Three Cyanobacteria to UV-B and Cd

  • Interactive effects of UV-B and Cd on growth, pigment, photosynthesis, and lipid peroxidation have been studied in Anabaena, Microcystis, and Nostoc; all the tested cyanobacteria showed differential sensitivity to different dosage of UV-B and Cd alone as well as in combination. Phycocyanin was severely affected by UV-B and Cd by all the strains; the degree of pigment bleaching was most pronounced in Anabaena followed by Microcystis and Nostoc. $UV-B_2+Cd_2$ produced nearly 83, 78, and 65% inhibition of phycocyanin in Anabaena, Microcystis, and Nostoc, respectively. The above treatment also significantly decreased the contents of Chl ${\alpha}$ and carotenoid. Presence of capsule in Microcystis protected the phycocyanin bleaching as compared to decapsulated cells. Laboratory-grown Microcystis revealed about 75 and 80% inhibition, following $UV-B_2+Cd_2$ treatment, respectively. in capsulated and decapsulated cells. Damage caused by Cd was more pronounced than UV-B. Inhibition of photosynthesis did occur in all the test strains, being maximum in Anabaena. PS II was the most sensitive component of the electron transport chain, showing 84, 80, and 70% inhibition in Anabaena, Microcystis, and Nostoc, respectively. As compared to control, significant lipid peroxidation (6.5-fold higher) was observed in Anabaena with $UV-B_2+Cd_2$, $^{14}C-uptake$ was more susceptible to Cd and Uv-B than oxygen-evolution. Approximately 84, 80, and 76% inhibition of $^{14}C-uptake$ was observed in Anabaena, Microcystis, and Nostoc, respectively. Similarly, $UV-B_2+Cd_2$ inhibited APT content of Anabaena by 87%. This ,study suggests that inhibition of carbon fixation was due to decreased ATP content of the test cyanobacteria by UV-B+Cd, where Anabaena was the most sensitive and Nostoc the most tolerant.

Studies on the Isolation of Phycobiliprotein from S. platensis (S. platensis로부터 phycobiliprotein의 분리에 관한 연구)

  • Isolation of phycobiliprotein from S. platensis was performed by using $30-60{\%}$ ammomium sulfate fractionation, Sephadex G-100 gel filtration and DEAE-Sephacel anionic exchange chromatography. Isolated phycobiliprotein was determined to be a c-phycocyanin with a maximum absorption wavelength at 620 nm. This phycobiliprotein consisted of $({\alpha}$ and $({\beta}$ subunit when analyzed through SDS-PAGE. The molecular weights of $({\alpha}$ and $({\beta}$ subunit were 14.5 kDa and 16 kDa, respectively. The native molecular weight of phycobiliprotein through gel filtration was about 100 kDa. These results show that the structure of phycobiliprotein from S. platensis might be aggregated form of $({\alpha}{\beta})_{3}-trimer$.

Pigment Stability of Lavers Porhyra tenera Kjellman during Processing and Storage (김의 가공 저장중에 있어서의 색소의 안정도)

  • The stability of three major pigments in lavers, namely chlorophylls, carotenoids, and phy-cobilins, during processing and storage was studied. The results of the pigment retention in fresh lavers during storage at low temperatures, of heat lability during heat treatment, and of the pigment loss during storage are summarized as follows: 1. The contents of chlorophyll a, xanthophyll (lutein+zeaxanthin), and carotene ($\alpha-+\beta-carotene$) averaged 1,525, 627, and $409{\mu}g/g$ of dry samples respectively The comparative composition of four carotenoids, lutein, $\beta-carotene$, zeaxanthin, and $\alpha-carotene$, was 35.9, 33.7, 12.2, and $5.8\%$ respectively. 2. In storage of fresh lavers at room ternperature ($20-25^{\circ}C$), 2 to $5^{\circ}C$, and $-15^{\circ}C$, the pigments generally were retained more at 2 to $5^{\circ}C$ than others while carotene only showed more retention at $-15^{\circ}C$ than at 2 to $5^{\circ}C$. Phycocyanin tended to decrease more rapidly than phyco-erythrin at low temperatures. 3. In regard to heat treatment of both dried and wet materials at 40, 60, 80, $100^{\circ}C$ respectively, the pigments were more stable at lower temperatures than at higher temperatures in both cases of materials. Carotenoids were retained more in wet materials than in the sun dried. Xanthophylls showed higher heat stability than carotenes. Phycoerythrin was considered more heat labile than phycocyanin, particularly at higher temperatures. 4. In storage of heat treated lavers for 50 days at room temperature, the retention of pigments appeared differently in accordance with the temperatures of treatment The materials treated at $60^{\circ}C$ showed better relention of pigments than those treated at the other temperatures. Degradation rate of xanthopllylls, carotenes, and phycobilins was decreased in the materials treated at higher temperatures, showing a tendency of great pigment retention even for a long period storage. Chlorophyll il was retained more in sun dried materials whereas xanthophyll and carotene were retained more in the wet. 5. In order to maintain a higher pigment stability during storage, the materials must be treated at $60^{\circ}C$ in a dryer with rapid ventilation until the moisture level reaches about $6.0\%$. In case of treatment at a higher temperature than $60^{\circ}C$, they must be treated for a short period of time to minimize pigment degradation by heat. 6. The lavers treated with the solutions of pH around 7.8 showed a higher retention of chlorophyll a. When treated with the solutions of algal extracts such as agar and alginic acid, uniform layer of laver sheets and better surface gloss of dried layers were obtained.

Quality Characteristics of Tofu Prepared with Spirulina (스피루리나 첨가 두부의 품질특성)

  • In this study, the quality characteristics of tofu prepared with added spirulina(0, 0.25, 0.5, 0.75, and 1%). As the concentration of spirulina increased, tofu yield decreased and the pH and turbidity increased. The Hunter L(lightness) and a(redness) values of tofu decreased as the amount of spirulina increased. Phycocyanin(C-phycocyanin, allophycocyanin and phycoerythrin) contents increased according to the spirulina amount. Moreover the antioxidant activity of tofu increased as the concentration of spirulina increased($IC_{50}$ values for control was 150.7 mg/g; 1% spirulina, 82.5 mg/g). Textural properties(TPA) for hardness, gumminess, and chewiness were higher in spirulina with added tofu, compared to those of control. Regarding the overall acceptability of sensory properties, spirulina with 0.5% added tofu had the highest scores among all treatments. From these results, it was suggested that the optimal amount of added spirulina was 0.5% for the preparation of high quality tofu.

Geosmin Concentration and Its Relation to Environmental Factors in Daechung Reservoir, Korea (대청호의 geosmin 농도와 환경요인과의 관계)

  • The biological and physicochemical factors and geosmin concentration were monitored in the Daechung Reservoir for 25 weeks from April to October in 1999. Geosmin was detected 5 times within a range of $1.16{\sim}5.28\;ng\;L^{-1}$, mostly in the late summer. The highest geosmin concentration was recorded on August 31, which also overlapped with the peaks of phycocyanin concentration, cyanobacterial density, and Anabaena spiroides density. A correlation analysis indicated that geosmin production was closely related with a high water temperature, pH, total dissolved phosphorus (TDP), and A. spiroides density. A water temperature of $27^{\circ}^C$, pH of 8.5, and TDP of $0.06\;mg\;L^{-1}$ were identified as the prerequisite environmental condition sand threshold values for geosmin production. Accordingly, under such conditions, an A. spiroides density above $10,000\;cells\;mL^{-1}$ indicates imminent geosmin occurrence.

Effect of Light with Different Wavelengths on Nostoc flagelliforme Cells in Liquid Culture

  • The effects of lights with different wavelengths on the growth and the yield of extracellular polysaccharides of Nostoc flagelliforme cells were investigated in a liquid cultivation. N. flagelliforme cells were cultured for 16 days in 500 ml conical flasks containing BG11 culture medium under $27{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ of light intensity and $25^{\circ}C$ on a rotary shaker (140 rpm). The chlorophyll a, phycocyanin, allophycocyanin, and phycoerythrin contents in N. flagelliforme cells under the lights of different wavelengths were also measured. It was found that the cell biomass and the yield of polysaccharide changed with different wavelengths of light. The biomass and the yield of extracellular polysaccharides under the red or violet light were higher than those under other light colors. Chlorophyll a, phycocyanin, and allophycocyanin are the main pigments in N. flagelliforme cells. The results showed that N. flagelliforme, like other cyanobacteria, has the ability of adjusting the contents and relative ratio of its pigments with the light quality. As a conclusion, N. flagelliforme cells favor red and violet lights and perform the complementary chromatic adaptation ability to acclimate to the changes of the light quality in the environment.
