Differential Diagnosis of Chemical-induced Hepatobiliary Toxicities Using a New Hepatobiliary Imaging Agent in Mice

  • We have synthesized $^{99m}$Tc-mercaptoacetyltriglycine (MAG3)-biocytin as a new imaging agent for hepatobiliary scintigraphy. The aim of this study was to evaluate the usefulness of $^{99m}$Tc-MAG3-biocytin scintigraphy in differentiating carbon tetrachloride ( $CCl_4$)-induced hepatotoxicity from $\alpha$-naphthylisothiocyanate (ANIT)-induced cholestasis in mice, which reflecting the differential diagnosis of neonatal jaundice caused by neonatal hepatitis from congenital biliary atresia in humans. Methods: Balb/c mice (female, 20 g, n=4-6) were pretreated with $CCl_4$(0.5 or $1.0m\ell$/kg) and ANIT ($150 or 300 m\ell$/kg) 18 h before scintigraphy. Biochemical and histopathological examinations showed a pattern of typical acute hepatitis (increase of transaminases and hepatocellular necnsis) in $CCl_4$-treated mice and cholestasis (increase of alkaline phosphatase and ${\gamma}$-glutamyltransferase, and biliary hyperplasia) in ANIT-treated mice, respectively, Mice were fasted at least 4 hr prior to the intravenous injection of $^{99m}$Tc-MAG3-biocytin (18.5 MBq/20$\mu\textrm{g}$) in 2% human serum albumin in saline. Scintigraphy was performed with a ${\gamma}$-camera equipped with a 1-mm diameter pin-hole collimator for 30 min and images were acquired every 15 s. We compared the values of physical parameters, such as peak liver/heart ratio ($${\gamma}$_{max}$) and peak ratio time ($t_{max}$) far $^{99m}$Tc-MAG3-biocytin scintigraphy. Results: Scintigraphic parameters of the $CCl_4$-pretreated (0.5 $m\ell$/kg) group showed a 81.9% decrease of r$_{max}$, and 42.2% decrease of $t_{max}$, whereas the ANIT-pretreated ( $150m\ell$/kg) group showed a 53% decrease of $r_{max}$, and 2.36-fold increase of $t_{max}$, (P<0.05). These results demonstrate that the decrease of $r_{max}$ and the shortening of $t_{max}$ are characteristic features for hepatotoxicity, in contrast to the increase of $t_{max}$ and decrease of $r_{max}$ for biliary hyperplasia. Conclusion: $^{99m}$Tc-MAG3-biocytin hepatobiliary scintigraphy can distinguish hepatitis from cholestasis in mice model and may be similarly useful in humans which differentiating the cause of neonatal jaundice in clinical study.cal study.cal study.cal study.

  • We are currently conducting studies on culturing and biocompatibility assessment of various cells such as neural stem cells and induced pluripotent stem cells(IPS cells) on carbon nanotube (CNT), on nerve regeneration electrodes, and on silicon wafers with a focus on developing nerve integrated CNT based bio devices for interfacing with living organisms, in order to develop brain-machine interfaces (BMI). In addition, we are carried out the chemical modification of carbon nanotube (mainly SWCNTs)-based bio-nanosensors by the plasma ion irradiation (plasma activation) method, and provide a characteristic evaluation of a bio-nanosensor using bovine serum albumin (BSA)/anti-BSA binding and oligonucleotide hybridization. On the other hand, the researches in the case of "novel plasma" have been widely conducted in the fields of chemistry, solid physics, and nanomaterial science. From the above-mentioned background, we are conducting basic experiments on direct irradiation of body tissues and cells using a micro-spot atmospheric pressure plasma source. The device is a coaxial structure having a tungsten wire installed inside a glass capillary, and a grounded ring electrode wrapped on the outside. The conditions of plasma generation are as follows: applied voltage: 5-9 kV, frequency: 1-3 kHz, helium (He) gas flow: 1-1.5 L/min, and plasma irradiation time: 1-300 sec. The experiment was conducted by preparing a culture medium containing mouse fibroblasts (NIH3T3) on a culture dish. A culture dish irradiated with plasma was introduced into a $CO_2$-incubator. The small animals used in the experiment involving plasma irradiation into living tissue were rat, rabbit, and pick and are deeply anesthetized with the gas anesthesia. According to the dependency of cell numbers against the plasma irradiation time, when only He gas was flowed, the growth of cells was inhibited as the floatation of cells caused by gas agitation inside the culture was promoted. On the other hand, there was no floatation of cells and healthy growth was observed when plasma was irradiated. Furthermore, in an experiment testing the effects of plasma irradiation on rats that were artificially given burn wounds, no evidence of electric shock injuries was found in the irradiated areas. In fact, the observed evidence of healing and improvements of the burn wounds suggested the presence of healing effects due to the growth factors in the tissues. Therefore, it appears that the interaction due to ion/radicalcollisions causes a substantial effect on the proliferation of growth factors such as epidermal growth factor (EGF), nerve growth factor (NGF), and transforming growth factor (TGF) that are present in the cells.

  • HWE 방법에 의해 CdSe 박막을 (100)방향 Si 기판 위에 성장시켰다. 증발원과 기판의 온도를 각각 $600^{\circ}C$, $430^{\circ}C$로하여 성장시킨 CdSe 박막의 이중 결정 X-선 요동곡선(DCRC)의 반폭치(FWHM)값이 380 arcsec로 가장 작았다. Van der Pauw 방법으로 Hall 효과를 측정하여 운반자농도의 In n 대 (1/T)에서 구한 활성화에너지는 0.19eV로 측정되었다. Hall 이동도의 온도 의존성은 30K에서 150K까지는 $T^{3/2}$에 따라 증가하여 불순물산란에 기인하고, 150K에서 293K까지는 $T^{-3/2}$에 따라 감소하여 격자산란에 기인한 것으로 고찰되었다. 광전도셀의 특성으로 spectral response, 최대 허용소비전력(MAPD), 광전류와 암전류(pc/dc)의 비 및 응답시간을 측정하였다. Cu 증기분위기에서 열처리한 광전도셀의 경우, 감도(${\gamma}$)는 0.99, pc/dc은 $1.39{\times}10^{7}$, 그리고 최대 허용소비전력(MAPD)은 335mW, 오름시간(rise time)은 10ms, 내림시간(decay time)은 9.5ms로 가장 좋은 광전도 특성을 얻었다.

방사선 입체조형치료용 다엽콜리메이터의 특성과 조직내 선량분포 측정 (Dose Distribution and Characterization for Radiation Fields of Multileaf Collimateor System)

  • 목적 : 다엽콜리메이터를 이용하여 부정형의 치료부위를 편리하고 정확히 차폐하며 삼차원 입체조형치료를 원활히 수행하기 위하여 다엽콜리메이터의 차폐특성을 계측하고 치료계획을 위한 최적방법을 확정한다. 대상 및 방법 : 차폐폭이 1cm이고 길이가 15cm인 52개의 금속차폐엽들이 양방향 각각 26개로 구성되어 컴퓨터계획에 의거 여러모양의 차폐면을 구사할 수 있는 다엽콜리메이터(Multileaf collimator)를 대상으로 방사선 치료에 요구되는 고에너지 방사선의 출력선량율, 차폐 및 투과효과, 반음영, 조직내 선량분포등을 전리함측정기와 열형광측정기 및 필름 선량측정방법을 이용하여 계측하였다. 다엽콜리메이터의 계단식 반음영 측정은 고체팬텀과 비디오밀도측정기를 이용하여 실효반음영(Effective penumbra)과 일반적반음영(General penumbra)으로 구분 평가하였다. 결과 : 다엽콜리메이터의 출력선량율은 표준차폐조리개에 비교하여 출력 선량율이 $1-2\%$ 증가 되었으며 치료오차 이내로 평가되었다. 다엽콜리메이터의 방사선투라 선량율은 6-10MV X선에서 평균 $2\%$로 표준차폐조리개보다 컸지만 혼합차폐벽돌보다 적었다. 원형으로 형성된 차폐엽 끝면의 조사선량은 약 $20\%$증가 되었으며 엽과 엽사이의 선량은 6mm 폭의 $5\%$증가로 심부치료에 영향이 없었다. 다엽콜리메이터의 실효반음영($20-80\%$)은 약 6mm로서 혼합차폐벽돌과 거의 같았으나 중심선에서 벗어날수록 $2-3\%$증가되었다. 다엽콜리메이터에 의한 계단식 모양의 반음영은 6-10MV x-선에서 차폐면 경사각이 45도일 때 약 11mm로 가장 컸으며 방사선속에 평행히 절단된 차폐벽돌보다 약 5mm 더 증가되었다. 결론 : 다엽콜리메이터는 1cm폭의 차폐엽들로 구성되어 계단식 차폐면에 대한 반음영이 다소 증가되고 있지만 조직내부의 다문조사 경우 반음영이 완화되므로 부정형 조사면 차폐에 적당하며 방사선 입체조형치료를 수행할 수 있는 가장 적당한 차폐기구로 생각된다.

제대혈 CD34+ 세포에서 유래된 지지세포의 분석 (Analysis of Stromal Cells Developed from Cord Blood CD34+ Cells)

  • 목적 : 제대혈의 조혈모세포 체외확장 시 조혈세포 증폭과 더불어 조혈미세환경의 변화가 일어난다. 이때 제대혈 $CD34^+$ 세포에서 유래되는 지지세포의 계열 분석조혈성장인자 분비능력을 알아보고 지지세포 증식 조건을 확립하여 효과적인 제대혈의 체외증폭을 제시하고자 하였다. 방법 : 제대혈부터 $CD34^+$ 세포를 분리하여 실험에 사용하였다. 무혈청배지에서 각종 조혈성장인자를 다양한 조합으로 첨가하여 배양하였고 증식정도는 현미경으로 관찰하여 배양용기를 점유한 면적 비율로 계산하였다. 세포외간질 단백의 효과를 분석하기 위하여 collagen S, fibronectin, laminin 및 poly-L-ly sine를 미리 coating한 용기에 배양하여 분석하였다. 제대혈 $CD34^+$ 세포를 조혈성장인자의 첨가 없이 3주간 액체배양하였다. 배양 시, 1주, 2주 및 3주에 상층액을 얻어 $-80^{\circ}C$에 보관하였다가 한꺼번에 IL-3, IL-6, GM-CSF, IL-$1{\beta}$ 및 TNF-$\alpha$등을 ELISA 방법으로 내부적으로 분비되는 량을 측정하였다. 분화된 지지세포의 계열을 분석하기 위해 E-selectin, VCAM-1, ICAM-1, PECAM-1, vWF, vimentin 및 CD 14 항체를 이용하여 면역화학염색 후 형광현미경으로 관찰하였다. 결과 : 제대혈 $CD34^+$ 세포 체외증폭시키는 과정에서 배양 4일에 지지세포가 출현하기 시작하여 7-10일이 지나면서 증식하기 시작하였고 14-2 1일 경에 서로 뭉치는 양상을 보여주었다. 제대혈 $CD34^+$ 세포 배양하면서 내부적으로 분비되는 GM-CSF, IL-6의 측정치는 시간이 지남에 따라 증가되었다. 제대혈 $CD34^+$ 세포 체외확장 시 지지세포의 증식 정도는 TPO+FL+SCF+LIF의 조합의 조혈성장인자가 첨가되었을 때 그리고 세포외간질 단백 성분 중 1% poly-L-lysine으로 처리한 경우 가장 효과적이었다. 결론 : 체외 증폭시 제대혈 $CD34^+$ 세포로부터 지지세포가 나타났으며 적절한 조혈성장인자의 첨가나 세포외간질 단백의 첨가에 의해 증폭될 수 있다.

기니피그 위 평활근의 자발적 수축과 전기적 활동에 대한 $Ca^{2+}$$Ca^{2+}$-길항제 영향 (Effect of $Ca^{2+}$ and $Ca^{2+}-antagonists$ on the Spontaneous Contractions and Electrical Activities of Guinea-pig Stomach)

  • The effects of external $Ca^{2+}$ and $Ca^{2+}-antagonists$ on the spontaneous contractions and electrical activities were investigated in guinea-pig stomach in order to clarify the mechanism for the generation of slow waves. Electrical responses of circular smooth muscle cells were recorded using glass capillary microelectrodes filled with 3 M KCl. All experiments were performed in tris-buffered Tyrode solution which was aerated with 100% $O_2$ and kept at $35^{\circ}C$. The results obtained were as follows: 1) The amplitude of spontaneous contractions was maximal at around 2-4 mM $Ca^{2+}$, whereas their frequency was inversely related with external $Ca^{2+}$ within the range of 0.5 to 16 mM $Ca^{2+}$. 2) Verapamil suppressed the amplitude of spontaneous contraction in a dose-dependent manner, while the frequency of spontaneous contractions was almost not changed over the whole concentration of verapamil $(0.01{\sim}5\;mg/l)$. 3) Manganese increased both the amplitude and the frequency of spontaneous contractions dose-dependently in low $Mn^{2+}$ (below 0.05 mM $Mn^{2+}$), while their amplitude and frequency were decreased in high $Mn^{2+}$ (above 0.1 mM $Mn^{2+}$). 4) The ampltude and maximum rate of rise of slow waves were incrased in high $Ca^{2+}$ solution. In $Ca^{2+}-free$ solution, the spontaneous contractions recorded simultaneously with slow waves ceased and tonic contraction ($Ca^{2+}-free$ contracture) was developed in parallel with membrane depolarization and the disappearance of slow waves. 5) Verapamil (1 mg/1) decreased the amplitude and maximum rate of rise of slow waves and it depolarized the membrane by about 6 mV, whereas the frequency of slow waves was not affected by verapamil. 6) Manganese showed different characteristic effects between low and high $Mn^{2+}$ on the slow waves: In low $Mn^{2+}$ (0.05 mM $Mn^{2+}$), the initial rapid increases and the subsequent gradual decreases in three parameters of slow waves (amplitude, rate of rise, and frequency of slow waves) till a new steady state were observed. However, in high $Mn^{2+}$ (0.5 mM $Mn^{2+}$) slow waves disappeared and membrane was depolarized. From the above results, the following conclusions could be made: 1) $Ca^{2+}$ is necessary for a generation of the slow waves, even though it is small amount. 2) Verapamil suppresses the spontaneous contractions of gastric antral strip by the decreases in amplitude and maximum rate of rise of slow waves, while this drug does not block the $Ca^{2+}-channel$ involved in the generation of slow waves. 3) Manganese has dual actions on the $Ca^{2+}-channels$; the $Ca^{2+}-channel$ involved in the generation of slow waves (or Na-Ca exchange system) or the channel for the generation of spike potentials are stimulated by a low concentration of $Mn^{2+}$, while both the $Ca^{2+}$. Channels are blocked by high concentration of $Mn^{2+}$.

  • POMS의 우수한 효용성을 국내에서도 활용하기 위해 K-POMS를 제작하여 그 신뢰도와 타당도를 점검하였고 만족할 만한 수준의 신뢰도와 타당도를 확보하였다. K-POMS의 요인 특성은 POMS와 비교하여 놀랄 만큼 유사한 특징을 보여준다. 단지 혼란-당황의 요소가 확실하지 않고 그 항목들이 여러 감정 요인에 나뉘어 부하되는 요인 분석 결과는 한국인의 정서와 그 표현이 영어권과 다를 가능성을 시사한다. 임상적으로 다양한 집단과 다양한 연령에 대한 자료를 보완하면 정신측정학적으로 훨씬 용이하게 사용할 수 있을 것이다. 나아가 이 연구에 나타난 제한점을 넘어서 한국인의 정서에 맞는 척도 항목을 개발하고 이 연구에서 나타난 '무기력과 불확실성' 요인이 일관성 있는 특성인지를 확인하는 작업이 필요할 것으로 생각한다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. To synthesize microspheres for biomedical application, "two stage shot growth method" was developed. The uniformity ratio of synthesized microspheres was always smaller than 1.05. And the surface charge density (or the number of ionizable functional groups) of the microspheres synthesized by "two stage shot growth method" was 6~13 times higher than that of the microspheres synthesized by conventional seeded batch copolymerization. As a previous step for biomedical application, adsorption experiments of bovine albumin on microspheres were carried out under various conditions. The maximum adsorbed amount was obtained in the neighborhood of pH 4.5. Isoelectric point of bovine albumin is pH 5.0, so experimental result shows that it shifted to acid area.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm.
