HPLC-FLD를 이용한 겨울철 PM2.5 중 아미노산 성분 분석 (Determination of Amino Acids on Wintertime PM2.5 using HPLC-FLD)

  • Ground-based measurements were conducted from January 6 to 12 of 2015 for understanding characteristics of nitrogen containing carbonaceous aerosols as 16 amino acids at the Mokpo National University, Korea. The detailed amino acid components such as Cystine ($(SCH_2CH(NH_2)CO_2H)_2$) and Methionine ($C_5H_{11}NO_2S$) and their sources were analyzed by High-Performance Liquid Chromatography with Fluorescence Detection (HPLC-FLD) for behavior of secondary products in particulate matter. In addition, organic carbon (OC) and elemental carbon (EC) based on the carbonaceous thermal distribution (CTD), which provides detailed carbon signature characteristics relative to analytical temperature, and water soluble organic carbon (WSOC) by total organic carbon (TOC) analyzer were used to understand the carbon compound behaviors. The backward trajectories were discussed for originations of carbonaceous aerosols as well. Different airmasses were classified with the amino acids and OC thermal signatures. The results can provide to understand the aging process influenced by the long-range transport from East Sea area.

비타민 C 첨가가-산란계의 생산성과 난각품질에 미치는 영향 (Effect of Vitamin C Supplementation on the Performance and Eggshell Quality of Layers)

  • 비타민 C 첨가가 산란계의 생산성과 난각의 품질에 미치는 영향을 측정하기 위하여 사양실험을 실시하였다. 71주령된 산란계(ISA-Brown) 800수를 공시하여 LG 비타민 C를 각각 0, 3, 10, 50, 100 ppm, flee ascorbic acid 100 ppm, Rovimix stay C-35 100 ppm, vitamin C 50 ppm + Zn chelate (100 ppm in Zn)를 첨가한 8처리구에 5반복, 반복당 20수씩 2수용 케이지에 수용하여 난괴법으로 배치하였다. 사양실험은 5월 중순에 시작하여 4주간 실시하였으며, 이 기간 중 평균온도는 23,8$^{\circ}C$(오후 5:00 기준)이었다. 비타민 C의 첨가시 대조구에 비해 산란을, 난중 및 난각 품질의 개선효과는 없었다. 비타민 C첨가구들 중 100 ppm의 LG 비타민 C 첨가구, Rovimix stay C-35 첨가구, LG 비타민 C 50 ppm+zn chelate 첨가구의 산란율이 대조구에 비해 낮았으며, 난중은 대조구와 LG 비타민 C 100 ppm 첨가구가 가장 높았다. 50, 100 ppm LG 비타민 C 첨가와 Rovimix stay C-35첨가는 연파란율을 증가시켰다. 난각두께는 LG 비타민 C 첨가구 모두 대조구와 차이가 없었다 이상의 결과를 종합해 볼 때 외기 온도가20-$26^{\circ}C$인 환경하에서는 산란계 사료에 3-100 ppd수준의 비타민 C첨가는 산란계의 생산성 개선에 유의한 영향을 주지 못하였다.

Genome Analysis and Optimization of Caproic Acid Production of Clostridium butyricum GD1-1 Isolated from the Pit Mud of Nongxiangxing Baijiu

  • Caproic acid is a precursor substance for the synthesis of ethyl caproate, the main flavor substance of nongxiangxing baijiu liquor. In this study, Clostridium butyricum GD1-1, a strain with high caproic acid concentration (3.86 g/l), was isolated from the storage pit mud of nongxiangxing baijiu for sequencing and analysis. The strain's genome was 3,840,048 bp in length with 4,050 open reading frames. In addition, virulence factor annotation analysis showed C. butyricum GD1-1 to be safe at the genetic level. However, the annotation results using the Kyoto Encyclopedia of Genes and Genomes Automatic Annotation Server predicted a deficiency in the strain's synthesis of alanine, methionine, and biotin. These results were confirmed by essential nutrient factor validation experiments. Furthermore, the optimized medium conditions for caproic acid concentration by strain GD1-1 were (g/l): glucose 30, NaCl 5, yeast extract 10, peptone 10, beef paste 10, sodium acetate 11, L-cysteine 0.6, biotin 0.004, starch 2, and 2.0% ethanol. The optimized fermentation conditions for caproic acid production by C. butyricum GD1-1 on a single-factor basis were: 5% inoculum volume, 35℃, pH 7, and 90% loading volume. Under optimal conditions, the caproic acid concentration of strain GD1-1 reached 5.42 g/l, which was 1.40 times higher than the initial concentration. C. butyricum GD1-1 could be further used in caproic acid production, NXXB pit mud strengthening and maintenance, and artificial pit mud preparation.

남대양산 크릴의 이용에 관한 연구 (STUDIES ON THE UTILIZATION OF ANTARCTIC KRILL 2. Processing of Paste Food, Protein Concentrate, Seasoned Dried Product, Powdered Seasoning, Meat Ball, and Snack)

  • 크릴을 원료로 하여 품에 대한 화학적 및 관능적 평가를 포함하는 일련의 실험을 하여 얻은 결과를 요약하면 다음과 같다. 1. krill paste의 제조에 있어서 krill juice는 $71\%$, krill scrap는 $29\%$의 수율을 나타내었으며, krill juice에서 분리한 krill paste는 $53\%$, krill broth는 $47\%$의 수율을 나타내었다. Krill paste의 아미노산 조성은 proline, glutamic acid, aspartic acid, lysine 및 leucine의 함량이 많았고, histidine, methionine, tyrosine, serine 및 threonine의 함량이 적었으며 필수아미노산에 있어서는 leucine 및 lysine의 함량이 높았고 methionine의 함량이 낮았다. 2. 크릴분말단백질의 제조에 있어 최적가공조건은 원료에 대하여 10배량의 $90\%$ isopropyl alcohol나 ethyl alcohol을 가하여 $80\%$에서 5분간 추출하는 조작을 5회 반복하는 것이었다. 추출용제로서는 ethyl alcohol 보다 isopropyl alcohol이 양호하였으며, 크릴분말단백질의 수율은 생동결크릴의 경우는 isopropyl alcohol 추출때가 $10.2\%$ ethyl alcohol 추출때는 $8.8\%$였고, 자숙동결크릴의 경우는 각각 $13.0\%$$11.8\%$였다. 크릴분말단백질의 아미노산조성은 생육질부의 아미노산조성과 거의 비슷하였다. 한편 크릴분말단백질을 첨가한 국수를 제조하여 관능검사를 한 결과, 원료 밀가루에 대하여 $3\%$까지 첨가 한 것이 국수 품질에 큰 영향을 주지않고 단백질을 강화할 수 있다고 판정하였다. 3. 크릴조미조림품의 품질은 전반적으로 볼 때 열풍건조크릴을 원료로 한 것이 가장 양호하였고, 다음이 일건크릴을 원료로 했을 때이며, 자숙냉결크릴을 원료로 한 것이 가장 못 하였다. 4. 크릴의 건조분말에 볶은참깨가루, 소금, 분말우육엑스, monosodium glutamate, 고추가루 및 후추가루를 각종 비율로 배합하여 분말조미료를 만들 었을 때의 품질은 색택에 있어서는 열풍건조크릴을 사용한 것이 양호하였고, 냄새는 일건크릴을 원료로 한 것이 양호하였으며, 열탕에 녹였을 때의 풍미는 sample B(크릴함량 $77\%$)가 가장 적당하였다. 5. 크릴 및 조기를 각종 비율로 배합하고 여기에 일정량의 밀가루, monosodium glutamate 및 소금을 첨가하여 krill meat ball을 만들어 그 색택, 맛 냄새, texture 및 젤리강도 등에 대하여 관능검사를한 결과, 크릴의 첨가량이 $25\%$까지의 제품이 양호하였으며, 생동결 및 자숙동결크릴을 원료로 했을 때의 품질차는 인정할 수 없었다. 6. 크릴, $\alpha$-전분, 고구마전분, 설탕, 소금, monosodium glutamate, glycine, 주석산칼리, 중탄산암모늄 및 중탄산소오다를 각종 비율로 배합하여 크릴${\cdot}$스낵${\cdot}$과자를 만들어 품질을 비교 검토한 결과 sample B(크림함량 $7.1\%$) 및 sample C(크릴함량 $10.8\%$)서 가장 양호하였다. 크릴${\cdot}$스낵${\cdot}$과자의 제조에 있어서 각종 전분의 가공적성을 시험한 결과 고구마전분이 가장 양호하였고 다음이 옥수수전분, 갈분의 순이었으며, 밀가루가 가장 못 하였다. 튀김방법은 소금튀김에 비하여 기름튀김이 좋은 결과를 나타내있고, 튀김온도는 $210\~215^{\circ}C$가 적당하였다.

Chemical Properties and Physiological Aacitivities of Stromata of Cordyceps militaris

  • The chemical properties and physiological activities of the freeze-dried stromata of Cordyceps militaris were examined. A proximate analysis exhibited that the stromata consisted mainly of crude carbohydrate (74.3%), crude protein (11.5%), and moisture content (8.7%), with a low content of crude ash (4.%) and fat (1.3%). The carbohydrate was mostly composed of glucose (88.6%). A large quantity of essential fatty acids, including linolenic acid (33.8%) was also observed. An analysis of the component amino acid showed a relatively high ratio of the essential amino acids, lysine (101.2 mg/g), methionine (62.7 mg/g), and acidic amino acids of glutamic aicd (57.5 mg/g) and aspartic acid (43.9 mg/g), whereas a low of tyrosine content (4.7 mg/g). An examination of the cordycepin content indicated tht the stromata and mycelium-embedded media (silkworm pupae) contained 0.2% and 0.5%, respectively. From the examination of the physiological activities, based on methanol extract (M), ethylacetate extract (EA), and hot-water extract (HW) fractions of the stromata, the hot-water extract (HW) fraction showed the most potent intestinal immune modulating activity, anticoagulant activity, and anticomplementary activity, whereas the ethylacetate extract (EA) fraction exhihited a radical scavenging activity. Therefore, the results from the present study indicate that the stromata of C. militaris contain various healthy chemical ingredients, and that especially boiled water extract of whole body would apprear to provide beneficial physiological activities.

한국인에서의 사이트린 결핍증의 경험 (Korean Experiences of Citrin Deficiency: Seven cases of citrin deficiency and nine major mutation screening in newborns in Korea)

  • Citrin deficiency resulting from mutations of SLC25A13is associated with two major clinical phenotypes; neonatal intrahepatic cholestasis caused by citrin deficiency (NICCD) and adult-onset type 2 citrullinemia (CTLN2). In Korea, 7 cases of citrin deficiency have been diagnosed based on biochemical and molecular findings. Four NICCD cases were identified by newborn screening using MS/MS or presenting symptoms like cholestatic jaundice. They are all males, presenting with conjugated hyperbilirubinemia, elevated liver enzymes, hypoalbuminemia, mild hyperammonemia, elevated citrullin, methionine and threonine. All of them have been spontaneously recovered from hepatic manifestation by the age of 6-8 months. Mutation analysis has been performed using their genomic & cDNAs obtained from skin fibroblasts. They turned out to be compound heterozygotes carrying each of 851del4, IVS11+1G>A, and IVS13+1G>A. Three CTLN2 patients were identified. Two adult male patients presented with a sudden loss of consciousness, seizure, vomiting, hyperammonemia and citrullinemia in their twenties. They carried an IVS13+1G>A, 851del4, and IVS11+1G>A mutant alleles. The other CTLN2 patient was 52 year old female patient, manifesting lethargy, altered consciousness, irritability and hyperammonemia. Similar clinical symptoms had recurred at the delivery of first and second babies in her past medical history. She was managed by hemodialysis and survived with neurological sequellae. Also, we screened the presence of 9 common mutations in 500 Korean newborns using dried blood spot of filter papers. Only a allele carried 854del4 mutation. In conclusion, the entire picture of citrin deficiency in Korea including incidence, genotype, clinical features and natural courses, is still vague at the present time.

Purification and Characterization of Glycerate Kinase From the Thermoacidophilic Archaeon Thermoplasma acidophilum: An Enzyme Belonging to the Second Glycerate Kinase Family

  • Thermoplasma acidophilum is a thermoacidophilic archaeon that grows optimally at $59^{\circ}C$ and pH 2. Along with another thermoacidophilic archaeon, Sulfolobus solfataricus, it is known to metabolize glucose by the non-phosphorylated Entner-Doudoroff (nED) pathway. In the course of these studies, the specific activities of glyceraldehyde dehydrogenase and glycerate kinase, two enzymes that are involved in the downstream part of the nED pathway, were found to be much higher in T. acidophilum than in S. solfataricus. To characterize glycerate kinase, the enzyme was purified to homogeneity from T. acidophilum cell extracts. The N-terminal sequence of the purified enzyme was in exact agreement with that of Ta0453m in the genome database, with the removal of the initiator methionine. Furthermore, the enzyme was a monomer with a molecular weight of 49kDa and followed Michaelis-Menten kinetics with $K_m$ values of 0.56 and 0.32mM for DL-glycerate and ATP, respectively. The enzyme also exhibited excellent thermal stability at $70^{\circ}C$. Of the seven sugars and four phosphate donors tested, only DL-glycerate and ATP were utilized by glycerate kinase as substrates. In addition, a coupled enzyme assay indicated that 2-phosphoglycerate was produced as a product. When divalent metal ions, such as $Mn^{2+},\;CO^{2+},\;Ni^{2+},\;Zn^{2+},\;Ca^{2+},\;and\;Sr^{2+}$, were substituted for $Mg^{2+}$ the enzyme activities were less than 10% of that obtained in the presence of $Mg^{2+}$. The amino acid sequence of T. acidophilum glycerate kinase showed no similarity with E. coli glycerate kinases, which belong to the first glycerate kinase family. This is the first report on the biochemical characterization of an enzyme which belongs to a member of the second glycerate kinase family.

Differentially Expressed Genes in Metastatic Advanced Egyptian Bladder Cancer

  • Background: Bladder cancer is one of the most common cancers worldwide. Gene expression profiling using microarray technologies improves the understanding of cancer biology. The aim of this study was to determine the gene expression profile in Egyptian bladder cancer patients. Materials and Methods: Samples from 29 human bladder cancers and adjacent non-neoplastic tissues were analyzed by cDNA microarray, with hierarchical clustering and multidimensional analysis. Results: Five hundred and sixteen genes were differentially expressed of which SOS1, HDAC2, PLXNC1, GTSE1, ULK2, IRS2, ABCA12, TOP3A, HES1, and SRP68 genes were involved in 33 different pathways. The most frequently detected genes were: SOS1 in 20 different pathways; HDAC2 in 5 different pathways; IRS2 in 3 different pathways. There were 388 down-regulated genes. PLCB2 was involved in 11 different pathways, MDM2 in 9 pathways, FZD4 in 5 pathways, p15 and FGF12 in 4 pathways, POLE2 in 3 pathways, and MCM4 and POLR2E in 2 pathways. Thirty genes showed significant differences between transitional cell cancer (TCC) and squamous cell cancer (SCC) samples. Unsupervised cluster analysis of DNA microarray data revealed a clear distinction between low and high grade tumors. In addition 26 genes showed significant differences between low and high tumor stages, including fragile histidine triad, Ras and sialyltransferase 8 (alpha) and 16 showed significant differences between low and high tumor grades, like methionine adenosyl transferase II, beta. Conclusions: The present study identified some genes, that can be used as molecular biomarkers or target genes in Egyptian bladder cancer patients.

Single Nucleotide Polymorphism of TBC1D1 Gene Association with Growth Traits and Serum Clinical-Chemical Traits in Chicken

  • TBC1D1 gene has known functional effects on body energy homeostasis and glucose uptake pathway in skeletal muscle tissue. This biological function is reported to have significant effects on traits of growth and meat quality in chicken. In this study, we focused on two single nucleotide polymorphisms (SNPs) (g.70179137A>G and g.70175861T>C) identified through SNP annotation information of Korean native chicken and previous literature for TBC1D1 in chicken. Association of SNPs in TBC1D1 with growth and serum clinical-chemical traits were evaluated. A total of 584 male and female birds from five Korean native chicken lines were used in the study. The SNP1 (g.70179137A>G) is located in intron 11 and SNP2 (g.70175861T>C) is a non-synonymous missense mutation in exon 10, responsible for the amino acid change from Methionine to Valine. The A allele of SNP1 and T allele of SNP2 had the highest allele frequencies. Both SNPs indicated moderate polymorphism information content values (0.25

한국산 낙화생의 가열 및 저장 중 품질의 이화학적인 변화 (I) (Changes of the Physicochemical Characteristics of Korean Peanuts with Roasting and Storaging(I))

  • This study was carried out to determine the proximate composition, amino acids and fatty acids contents and changes of physicochemical characteristics of each oil extracted from Spanish and Virginia type peanuts grown in Korea roasted at 110, 120, 130 and 14$0^{\circ}C$ for 2 minutes. 1. The moisture contents of raw Spanish and Virginia type peanuts were 6.5~6.8% respectively. The crude ash and reduced sugar contents of raw Spanish and Virginia type peanuts were 2.3% and 16.5% and the crude protein content was 27.0% in Spanish type peanuts and was aproximately 1% higher than in Virginia type peanut. The protein content was 25.7%~26.7% in Virginia type peanut roasted at 110, 120, 130 and 14$0^{\circ}C$. The crude fat content of Virginia type peanut was 46.0% which was aproximately 1% higher than that of Spanish type. But four kinds of oils content were 51.3%~51.8% in Spanish type peanut roasted at 110, 120, 130 and 14$0^{\circ}C$, which was about 2% higher than those of Virginia type. 2. Amino acids existed in peanut were glutamie acid, arginine, aspartic acid, leucine, glycine, phenylalanine, proline-lysine, tyrosine, valine and isoleucine, etc mainly. But methionine and threonine contents were very low. The content of glutamic acid was the highest in 71.6-81.7mg among amino acids. Glutamic acid content of Virginia type peanut was about 12% higher than that of Spanish type peanut. Total amino acid content was 441.8mg/g in Virginia type peanut and that was 16% higher than that of Spanish type peanut. The lysine content of Spanish and Virginia type peanuts roasted at 14$0^{\circ}C$ were 24% and 13%, these were lower than those of peanuts roasted at 11$0^{\circ}C$. 3. Main fatty acids of raw Spanish and Virginia type peanut oils were oleic(40.99-46.58%), linoleic(33.21-38.82%) and palmitic acid(9.72-11.58). Linoleic acid content of raw Virginia type peanuts was 5.6% higher than that of raw Spanish type peanut. And the oleic acid content of Spanish and Virginia type peanuts roasted at 11$0^{\circ}C$, 12$0^{\circ}C$, 13$0^{\circ}C$ and 14$0^{\circ}C$ was 50-53% and 41-43% respectively. Linoleic acid content of Spanish and Virginia type peanuts roasted at same temperatures as the former was about 28-31% and 37-38% respectively. That linoleic acid content of roasted peanuts was lower than that of raw peanuts. Linoleic acid content of raw and roasted Virginia type peanut, were higher than that of Spanish type peanuts. 4. Acid value and peroxide value of oils extracted from roasted Spanish and Virginia type peanuts were much higher than those of oils extracted from raw peanuts. The maximum AVs of oils extracted from Spanish and Virginia type roasted peanuts were samples roasted at 12$0^{\circ}C$and those AVs were 0.50 and 0.63 respectively. And the maximum POVs of oils extracted from Spanish and Virginia type roasted peanuts were samples roasted at 12$0^{\circ}C$ also and those POVs were 26.8 and 32.8 meq/kg. oil respectively. Acid value and peroxide value of oils extracted from roasted peanuts were increased with increasing the roasting temperatures from 11$0^{\circ}C$ to 12$0^{\circ}C$, then decreased, while TBA values were increased continuously with increasing the roasting temperatures.
