Desulfurization efficiency in photobioreactors dependent on the irradiation type of light sources (광생물반응기내 광조사 유형별 탈황 효율)

  • Removal rates of hydrogen sulfide were investigated to known effects of several light sources with external and internal irradiation on the desulfurization using C. thiosulfatophilum. In the case of internal illumination system, optical-fiber photobioreactor was applied to increase the light availability. Furthermore, sunlight was used as the main light energy in the daytime and metal-halide lamp was applied as an additional light energy at night. Light energy of 99% was saved by the application of the LED's array in comparison with the incandescent light source. $H_2S$ removal rates at 5,000 lux in a 4-L photobioreactor were shown as 0.040, 0.138, 0.136, and 0.134 (${\mu}mol$ $H_2S/min$)/(mg protein/L), respectively, in the following order of light sources, when several light sources such as fluorescent, energy-saving, incandescent, halogen lamp, and filtered light at 460 nm were applied. Removal rate per unit cell concentration with the internal light diffused optical-fibers increased about 1 six times as much as that with the external light sources. Removal rate per unit cell concentration, using sunlight in the daytime and a metal-halide lamp at night, was 0.41 less than 0.869 (${\mu}mol$ $H_2S/min$)/(mg protein/L) using a 400 W metal-halide lamp day and night, since the automatic sunlight collection system can transmit the light intensity as only 10% of that with a metal-halide lamp.

Constrained Spatiotemporal Independent Component Analysis and Its Application for fMRI Data Analysis

  • In general, Independent component analysis (ICA) is a statistical blind source separation technique, used either in spatial or temporal domain. The spatial or temporal ICAs are designed to extract maximally independent sources in respective domains. The underlying sources for spatiotemporal data (sequence of images) can not always be guaranteed to be independent, therefore spatial ICA extracts the maximally independent spatial sources, deteriorating the temporal sources and vice versa. For such data types, spatiotemporal ICA tries to create a balance by simultaneous optimization in both the domains. However, the spatiotemporal ICA suffers the problem of source ambiguity. Recently, constrained ICA (c-ICA) has been proposed which incorporates a priori information to extract the desired source. In this study, we have extended the c-ICA for better analysis of spatiotemporal data. The proposed algorithm, i.e., constrained spatiotemporal ICA (constrained st-ICA), tries to find the desired independent sources in spatial and temporal domains with no source ambiguity. The performance of the proposed algorithm is tested against the conventional spatial and temporal ICAs using simulated data. Furthermore, its performance for the real spatiotemporal data, functional magnetic resonance images (fMRI), is compared with the SPM (conventional fMRI data analysis tool). The functional maps obtained with the proposed algorithm reveal more activity as compared to SPM.

A Study on the Phased Cultural Product Design with Characteristics of Unlined Cheollik in Early 17th Century and Baby Typed Doll's Body (17세기 초 홑철릭 유물의 시대특성과 유아형 인형의 체형특성을 응용한 단계별 복식문화상품 디자인 연구)

  • This study will broaden the application of doll costume product with historical characteristics and the image of unlined cheollik in the $17^{th}$ century period of transition during the Joseon Dynasty. Historical design sources were extracted from old documents and precedent studies. Unlined cheollik of Shin Gyeong-yu, meritorious retainer, were selected as main reference-relic because of various fabric, preserved conditions and definite shape. 'Baby doll' was selected as main model because of consumer preferences and awareness. Design sources from unlined cheollik in the early $17^{th}$ century were about a 1:2 ratio of upper and under parts, removable separated doori-somae, knife-shaped collar (outside), projected square collar (inside), long pleats line, side slit, traditional flat fell seam sewing. Trapezoidal side line, round waistline, and thin fabric were applied in designs because of toddler-body of doll model. Three designs were produced in step 1: Cheollik A focusing on the historical remake (traditional type), Cheollik B with belt and side slit (crossover type), and Cheollik C with back- opening (modern type). In step 2, interview with fashion major student was conducted to increase the utility of designs. As a result, modern trend sources (frill, velcro, round armhole line, slope of sleeves, and floral pattern) were reflected in Cheollik B, C. Finally, three doll cheollik and cheollik-styled doll apron sample were produced. Various versions must be suggested in the study of doll costume products with traditional sources that balance historical characteristics and practicality to improve customer satisfaction.

Preparation of $MgO-ZrO_2$ Fibers by Sol-Gel Method and Their Characterization (졸-겔법에 의한 $MgO-ZrO_2$ 섬유의 제조와 특성)

  • From Zr(O-nC3H7)4-H2O-C2H5OH-HNO3 starting solutions, MgO-doped stabilized zirconia fibers with varying content of MgO (10~18 mol%) from different MgO sources were fabricated by sol-gel method. The MgO sources used are magnesium nitrate hexahydrate, magnesium acetate tetrahydrate, and magnesium ethylate. The phase transformation studies of a drawn MgO-ZrO2 fiber were carried out using X-ray diffraction, IR spectroscopy, and Raman spectroscopy. The microstructure, tensile strength, and microporosity of fibers were investigated using SEM, tensile strength test, and microporosimeter. Although various MgO sources such as magnesium nitrate, acetate, and ethylate were used, the crystallization behavior of MgO-ZrO2 fibers at different temperatures could be summarized as follows: CubiclongrightarrowMetastable TetragonallongrightarrowMonocliniclongrightarrowCoexistence of Monoclinic and CubiclongrightarrowCubic(trace of monoclinic). At 150$0^{\circ}C$, the phase transformation of MgO-ZrO2 fibers shows the following change depending on the amount of MgO[Mg(NO3)2.6H2O]: At 10 mol%, both monoclinic and cubic phase coexist, at 12 mol%, monoclinic phase decreases rapidly, and then at 14 mol%, only cubic phase remains. When the MgO-ZrO2 fibers containing 12 mol% magnesium nitrate were heated at 80$0^{\circ}C$ for 1hr, average tensile strength of fibers is 4.0 GPa at diameters of 20 to 30 ${\mu}{\textrm}{m}$. As the heat-treatment temperatures increase to 100$0^{\circ}C$ for 1 hr, tensile strength of MgO-ZrO2 fibers decreases rapidly to 0.7 GPa.

Serotype and Enzymatic Profile of Crypfococcus neoformans Isolates from Clinical and Environmental Sources in Korea (한국의 임상과 자연환경에서 분리된 Cryptococcus neoformans의 혈청형과 효소생성능)

  • Fifty eight Cryptococcus neoformans strains isolated from clinical and environmental sources in Korea were examined for their serotypes and extracellular enzyme activities. Among the 51 strains isolated from clinical sources, 48 strains were serotype A (94.1%), 2 strains were serotype B (3.92%), and 1 strain was serotype D (1.96%). All seven environmental strains isolated from pigeon excreta were identified as serotype A. All isolates of C. neoformtans were positive for the production of extracellular proteinase and phospholipase. In the API-ZYM system, all fifty eight isolates produced alkaine phosphatase, esterase C4, esterase lipase: C8, leucine arylamidase, acid phosphatase, naphthol-AS-BI-phosphohydrase, $\alpha$-glucosidase and $\beta$-glucosidase. Thirty nine isolates (67.2%) of C. neoformans produced N-acetyl-$\beta$-glucosidase. Two isolates, serotype B, and B only one serotype A produced $\beta$-glucuronidase. Analysis of enzymatic profiles to 21 enzymes revealed four biotypic patterns among the 58 strains. The enzymatic patterns of C. neoformans isolated from clinical and environmental sources represented a significant relationship with the serotypes.

Production of siderophore from L-glutamic acid as both carbon and nitrogen sole sources in Acinetobacter sp. B-W (글루탐산을 유일한 탄소원과 질소원으로 이용하는 Acinetobacter sp. B-W의 시드로포어 생산)

  • Catechol type siderophore different from 2, 3-dihydroxybenzoic acid (DHB) was produced from Acinetobacter sp. B-W grown in medium containing L-glutamic acid as both carbon and nitrogen sole sources at $28^{\circ}C$. Optimal concentration of glutamic acid for siderophore production was 3% and production of siderophore was decreased above 3% glutamic acid. In previous report, siderophore, 2, 3-DHB was produced from strain B-W grown in medium containing glucose as carbon source and glutamic acid as nitrogen source. Rf value of siderophore produced from strain B-W grown in medium glutamic acid as both carbon and nitrogen sole sources at $28^{\circ}C$ was 0.32, while 2, 3-DHB was 0.84 in butanol-acetic acid-water (12:3:5) as developing solvent. Antioxidative activity of 2, 3-DHB was not detected in that siderophore produced from glutamic acid. Catechol nature of siderophore was detected by Arnow test. Although in iron-limited media optimal cell growth was identified at $36^{\circ}C$, significant quantities of siderophore were produced only at $28^{\circ}C$. Biosynthesis of siderophore was strongly inhibited by growth at $36^{\circ}C$. Production of siderophore was completely inhibited by $10{\mu}M\;FeCl_3$.

Influence of Yeast Fermented Cassava Chip Protein (YEFECAP) and Roughage to Concentrate Ratio on Ruminal Fermentation and Microorganisms Using In vitro Gas Production Technique

  • The objective of this study was to determine the effects of protein sources and roughage (R) to concentrate (C) ratio on in vitro fermentation parameters using a gas production technique. The experimental design was a $2{\times}5$ factorial arrangement in a completely randomized design (CRD). Factor A was 2 levels of protein sources yeast fermented cassava chip protein (YEFECAP) and soybean meal (SBM) and factor B was 5 levels of roughage to concentrate (R:C) ratio at 80:20, 60:40, 40:60, 20:80, and 0:100, respectively. Rice straw was used as a roughage source. It was found that gas production from the insoluble fraction (b) of YEFECAP supplemented group was significantly higher (p<0.05) than those in SBM supplemented group. Moreover, the intercept value (a), gas production from the insoluble fraction (b), gas production rate constants for the insoluble fraction (c), potential extent of gas production (a+b) and cumulative gas production at 96 h were influenced (p<0.01) by R:C ratio. In addition, protein source had no effect (p>0.05) on ether in vitro digestibility of dry matter (IVDMD) and organic (IVOMD) while R:C ratio affected the IVDMD and IVOMD (p<0.01). Moreover, YEFECAP supplanted group showed a significantly increased (p<0.05) total VFA and $C_3$ while $C_2$, $C_2:C_3$ and $CH_4$ production were decreased when compared with SBM supplemented group. In addition, a decreasing R:C ratio had a significant effect (p<0.05) on increasing total VFA, $C_3$ and $NH_3$-N, but decreasing the $C_2$, $C_2:C_3$ and CH4 production (p<0.01). Furthermore, total bacteria, Fibrobacter succinogenes, Ruminococcus flavefaciens and Ruminococcus albus populations in YEFECAP supplemented group were significantly higher (p<0.05) than those in the SBM supplemented group while fungal zoospores, methanogens and protozoal population remained unchanged (p>0.05) as compared between the two sources of protein. Moreover, fungal zoospores and total bacteria population were significantly increased (p<0.01) while, F. succinogenes, R. flavefaciens, R. albus, methanogens and protozoal population were decreased (p<0.01) with decreasing R:C ratio. In conclusion, YEFECAP has a potential for use as a protein source for improving rumen fermentation efficiency in ruminants.

Effects of Dietary Protein Sources on Growth and Body Composition in Olive Flounder (Paralichthys olivaceus)

  • A 6-week feeding trial was conducted to investigate the effects of six different dietary animal protein sources on growth and body composition of olive flounder, Paralichthys olivaceus in recirculating system. White fish meal (WFM), flounder muscle (FLM), carp muscle (CM), blood meal (BM), squid liver powder (SLP) and casein (CA) were used as the main animal protein sources in the six experimental diets. Fish averaging $2.9\pm0.03g$ $(mean\pm SD)$ were distributed to each aquarium as a group of 15 fish and were fed one of the six experimental diets to each treatment of triplicate groups. After 6-week of the feeding trial, fish fed white fish meal (WFM) and flounder muscle (FLM) diets showed a significant higher weight gain $(WG\%)$ (P<0.05) than those of fish fed the CM, BM, SLP and CA diets. Fish fed BM diet showed the lowest WG among all the dietary treatments. Feed conversion ratio (FCR) and protein efficiency ratio (PER) showed the similar trend as WG. Hematocrit and hemoglobin were not affected by the dietary treatments. Fish fed the FLM and CM diets showed significant higher survival rate than those of fish fed BM diets, and there was no significant difference in survival of fish fed WFM, FLM, CM, SLP and CA diets. These results indicated that WFM and FLM are the best dietary protein sources tested in olive flounder.

A SOURCE EXTRACTION METHOD FOR THE AKARI MID-IR FAINT SOURCE CATALOGUE

  • We plan to produce a faint source catalogue from the AKARI mid-infrared (IR) all-sky diffuse maps. In the publicly-available AKARI mid-IR point source catalogue (PSC), sources were extracted from single-scan images, and confirmed by using the other scan images. By stacking multiple scan images, we can detect fainter sources which are not listed in the PSC. We optimized the source extraction process using a $6^{\circ}{\times}6^{\circ}C$ area around the star-forming region, Cepheus B. Then, we divided the all-sky data into three seasonal images, and checked the positions and the fluxes of the detected sources on the images. As a result, our new source extraction method works well; 90% of the sources are also identified in the WISE catalogue. In this method, we obtain the detection limit twice deeper than that of the PSC. The number of sources increases by a factor of 2, as compared with the PSC.

Estimation of In vitro Digestibility of Barley Straw by Using a Homogenized Rumen Fluid and Artificial Saliva Mixed with Nitrogen and Energy Sources

  • A $2{\times}2{\times}4$ factorial study was conducted to examine the possibility of improving estimates of in vitro digestibility, using untreated (UBS) and ammonia-treated (ABS) barley straw, through homogenization of rumen fluid (RF) and by additions of urea (U) and casein (C) as N sources and Xylose + Glucose (XG) as energy sources into artificial saliva. Digestibiltiy of ABS was significantly greater than that of UBS (p < 0.001). There was a significant decrease in digestibility when additions (U, UC, UCXG) were compared with the control (p < 0.001). A 2-way interaction between RF and straw type was significant (p < 0.05) for dry matter digestibility (DMD). Homogenization of RF increased DMD of ABS (p < 0.05) whereas it decreased DMD of UBS (p > 0.05). The study showed that addition of N alone or in combination with energy sources was not better than control, rather the reverse, digestion was inhibited by a combination of U and casein (UC). It was concluded that sufficient N and branched chain fatty acids were supplied in the inoculum from sheep fed high protein grass cubes to support the growth of cellulolytic microbes during in vitro incubation. Further studies are, however, required to explore the potential of homogenization in improving the in vitro method to estimate digestibility of cereal straws.
