본 연구의 목적은 생약복합제제인 SH21B의 adipogenesis 억제 효능에 대한 상세한 분자적 메커니즘을 3T3-L1 지방세포를 이용하여 밝히는 데 있다. 실험에 사용된 SH21B는 7가지 생약성 천연물질인, 황금, 행인, 마황, 석창포, 포황, 원지 및 하엽으로 이루어졌다. 최근, 본 연구진에 의해 3T3-L1을 이용한 in vitro 연구와 마우스를 이용한 in vivo 연구에서 SH21B의 adipogenesis 억제효능이 밝혀진 바 있다. 본 연구에서는 3T3-L1 지방세포가 분화될 때 작용하는 다양한 지방세포형성 조절자들의 유전자 발현이 SH21B에 의해 어떻게 변하는지 살펴보고자 하였다. 실시간중합효소반응(real time PCR) 기술을 이용하여 SH21B를 처리한 지방세포와 그렇지 않은 지방세포를 비교한 결과, 최종마커인 ADIPOQ와 SLC2A4의 유전자 발현이 SH21B에 의해 급격하게 감소함을 알 수 있었다. 최종마커의 발현을 유도하는 핵심전사인자인 $PPAR{\gamma}$와 C/$EBP{\alpha}$의 유전자 발현 역시 SH21B의 처리 시 유의하게 억제되었다. 좀 더 상세한 분자적 메커니즘을 규명하기 위해, 핵심전사인자의 상위에 위치한 다양한 조절자들의 유전자 발현을 분석하였다. 그 결과, 여러 지방세포형성 유도조절자 중, Krox20과 KLF15의 유전자 발현이 SH21B 처리에 의해 유의하게 감소된 반면, C/$EBP{\beta}$와 KLF5의 유전자 발현은 SH21B 처리에 영향을 받지 않았다. 그리고 지방세포형성 억제조절자인 KLF2와 CHOP의 유전자 발현은 SH21B 처리에 의해 유의하게 증가되었다. 이러한 결과들은 SH21B의 지방세포형성 억제효능이 지방세포의 분화에 작용하는 다양한 상위조절자 중 지방세포형성 유도조절자인 Krox20과 KLF15 그리고 지방세포형성 억제조절자인 KLF2와 CHOP 등의 유전자 발현이 변화되면서 일어나는 복합적인 반응의 결과임을 제시한다.

Chlorogenic acid is a phenolic compound found in Cudrania tricuspidata fruits. In the present study, the effect of chlorogenic acid on the inhibition of adipogenesis in 3T3-L1 adipocytes was investigated. Cells were stained with Oil red O reagent to detect lipid droplets in adipocytes. The 3T3-L1 cells were lysed and measured for intracellular triglyceride and adipokine by ELISA kit. The protein expression of adipogenesis-related gene was evaluated by Western blot analysis. Chlorogenic suppressed lipid droplet and intracellular triglyceride accumulation in a concentration manner and also decreased secretion of adipokines such as leptin and adiponectin, compared with fully differentiated adipocytes. Treatment of 3T3-L1 cells with chlorogenic acid reduced the protein levels of peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) and, CCAAT/enhancer binding proteins alpha ($C/EBP{\alpha}$). This indicates that chlrogenic acid was effective as an anti-obesity agent by repressing the differentiation of 3T3-L1 into adipocytes and inhibiting triglyceridef formation in adipocyte and that it exerts its role mainly through the significant down-regulation of $PPAR{\gamma}$ and $C/EBP{\alpha}$.
In this study, the effects of Ganoderma lucidum (GL) on fat metabolism were performed in 3T3-L1 adipocytes. The effects of GL on 3T3-L1 preadipocytes differentiation were also examined. Our results showed that GL decreased the TG content by ORO staining. To elucidate the mechanism of the effects of GL on lowering TG content in 3T3-L1 adipocytes, we examined whether GL modulate the expressions of transcription factors and adipokines related to control of energy expenditure process because adipokines regulate adipocyte mass and increased expenditure may consume much TG in adipocytes. As a result, the expression of C/$EBP{\beta}$, C/$EBP{\delta}$, C/$EBP{\alpha}$, and $PPAR{\gamma}$, genes, which induce the adipose differentiation and adipose-specific FAS, aP2, and adipsin genes, which compose fat formation were decreased. In addition, GL increased the expression of leptin, UCP2, adiponectin in 3T3-L1 adipocytes, resulting in energy homeostasis. In conclusion, GL could regulate transcript factor related to induction of adipose differentiation and control TG content by up-regulation of adipokines related to fat burn.
Up until today, the key to contouring has been resumed in these two alternatives, either limiting the adipocyte storing capacity by modulating lipogenesis, or by stimulating lipolysis to eliminate adipocyte lipid content. Another interesting way could be the regulation of adipocyte differentiation. In this work, we have evaluated the effect of a brown algal extract of Sphacelaria scoparia (SSE) on the differentiation of pre-adipocytes into adipocytes. A pre-adipocyte line (3T3-L 1) was used. The differentiation was evaluated by the measure of produced lipids thanks to red oil coloration and spectrophotometry, and also by the expression of adipocyte differentiation markers: enzymes such as fatty acid synthase (FAS) and stearoyl CoA desaturase (SCD), or membrane proteins such as glucose transporters (GLUT -4) and fatty acid transporters (FAT) expressed on the surface of human adipocytes. These genes are under control of two transcription factors: CAAT-enhancer binding protein (c/EBP alpha) and sterol response element binding protein (SREBP1). All these markers were analysed at different stages of differentiation by RT -PCR. Sphacelaria extract (SSE) inhibits pre-adipocytes differentiating into adipocytes following a dose-dependant relation, using a kinetics similar to retinoic acid. It decreases the expression of mRNA specific to FAS, FAT, GLUT -4, SCD1, c/EBP alpha and SREBP1. Moreover, SSE regulated on collagen 1 and collagen 4 expression. A stimulation of collagen 1 was also measured in human skin fibroblasts. Thus, SSE performs as a genuine differentiation inhibitor and not only as a lipogenesis inhibitor, and could be used in slimming products.
Previous our study demonstrated that ailanthoidol (3-deformylated 2-arylbenzo[b]furan), a neolignan from Zanthoxylum ailanthoides or Salvia miltiorrhiza Bunge, is a novel anti-inflammatory agent. In this investigation, we examined the anti-adipogenic effect of ailanthoidol. Our data showed that ailanthoidol suppressed lipid droplet formation and adipocyte differentiation in 3T3-L1 cells. Treatment of the 3T3-L1 adipocytes with ailanthoidol resulted in an attenuation of the releases of leptin and interleukin-6. The expression of peroxisome proliferator-activated receptor $(PPAR){\gamma}$ and CCAAT/enhancer-binding protein $(C/EBP){\alpha}$, the central transcriptional regulators of adipogenesis, was decreased by treatment with ailanthoidol. Additionally, ailanthoidol treatment increased the phosphorylation levels of 5' adenosine monophosphate-activated protein kinase. These results suggest that ailanthoidol effectively suppresses adipogenesis and that it exerts its role mainly through the significant down-regulation of $PPAR{\gamma}$ and $C/EBP{\alpha}$ expression. Our findings provide important insights into the mechanisms underlying the anti-adipogenic activity of ailanthoidol.
솔장다리(S. collina)는 건조한 대지에 널리 분포되어 있는 한해살이 식물로 한방에서는 고혈압의 치료에 사용되어왔으며 선행연구를 통해 솔장다리가 보유한 항산화 및 항암 활성을 밝힌바 있다. 본 연구에서는 솔장다리 에탄올 추출물(SCEE)의 항비만 활성을 췌장 lipase 효소 활성 억제능과 세포실험계를 이용하여 분석하였다. 그 결과 SCEE는 농도 의존적으로 lipase 효소 활성을 유의적으로 억제시켰으며, 3T3-L1 preadipocyte를 이용하여 지방세포 분화 및 지방생성, 생성된 지방의 분해에 미치는 영향을 분석한 결과 지방세포 분화, 세포 내 지방 축적, TG 함량 등을 독성 없이 농도의존적으로 억제하였고, 지방세포 내 중성지방을 유의적으로 분해시키는 것으로 나타났다. 이러한 솔장다리의 지방세포 분화 억제능은 핵심 작용 인자인 $C/EBP{\alpha}$, $C/EBP{\beta}$, 그리고 $PPAR{\gamma}$의 유전자 및 단백질 발현조절에서 기인함을 확인하였다. 이러한 결과는 솔장다리가 보유한 췌장 lipase 활성 저해능, 지방세포 분화 억제능, 지방세포 내 지방 분해능, 지방분화관련 인자 신호전달기전 조절을 통한 항비만 활성을 처음으로 밝혀낸 것이며 추후 계속적인 연구를 통해 활성 물질의 규명이 필요할 것으로 판단된다.
본 연구에서는 통초(Tetrapanax papyriferus)와 희렴(Siegesbeckia pubescens)의 항산화 및 항비만 활성을 DPPH radical 소거능과 세포실험계를 이용하여 분석하였다. 통초와 희렴의 DPPH radical 소거능의 50% 저해능($IC_{50}$)은 각각 65.23과 47.79 ${\mu}g/ml$로 나타났다. 또한 두 시료 모두 농도 의존적으로 lipase 효소 활성을 유의적으로 억제시켰으며, 3T3-L1 preadipocyte를 이용하여 지방세포 분화 및 지방생성에 미치는 영향을 분석한 결과 통초와 희렴 모두 지방 세포 분화, 지방 축적, TG 함량 등을 독성 없이 농도의존적으로 억제함을 보였다. 이러한 통초와 희렴의 지방세포분화억제능은 핵심 작용 인자인 $C/EBP{\alpha}$, $C/EBP{\beta}$, 그리고 $PPAR{\gamma}$의 유전자 및 단백질 발현조절에서 기인함을 확인하였다. 또한 통초와 희렴 간의 항비만 시너지 효과를 분석한 결과 각 시료의 단독 처리시보다 병용 처리시 더 높은 지방세포분화억제능을 보여 두 시료간에 시너지 효과를 보유함을 확인하였다. 이러한 결과는 통초 및 희렴의 항비만 활성 및 그 작용 기전을 밝힌 것이며 추후 계속적인 연구를 통해 활성 물질의 규명이 필요할 것으로 판단된다.
본 연구에서는 백두구(A. cardamomum L.) 메탄올 추출물(ACME)의 항산화 및 항비만 활성을 DPPH radical 소거능과 췌장 lipase 효소 활성 억제능, 그리고 세포실험계를 이용하여 분석하였다. 그 결과 ACME는 DPPH radical을 농도 의존적으로 소거하였으며 DPPH radical 소거능의 50% 저해농도($IC_{50}$)는 $25.15{\mu}g/ml$로 나타났다. 또한 ACME는 농도 의존적으로 lipase 효소 활성을 유의적으로 억제시켰으며, 3T3-L1 preadipocyte를 이용하여 지방세포 분화 및 지방생성, 생성된 지방의 분해에 미치는 영향을 분석한 결과 ACME는 지방세포 분화, 세포 내 지방 축적, TG 함량 등을 독성 없이 농도의존적으로 억제하였으며 지방세포 내 중성지방을 유의적으로 분해시키는 것으로 나타났다. 이러한 백두구의 지방세포 분화 억제능은 핵심 작용 인자인 $C/EBP{\alpha}$, $C/EBP{\beta}$, 그리고 $PPAR{\gamma}$의 유전자 및 단백질 발현조절에서 기인함을 확인하였다. 이러한 결과는 백두구가 보유한 항산화능과 췌장 lipase 활성 저해능, 지방세포 분화 억제능, 지방세포 내 지방 분해능을 통한 항비만 활성을 처음으로 밝혀낸 것이며 추후 계속적인 연구를 통해 활성 물질의 규명이 필요할 것으로 판단된다.
선행연구에서 Euptelea pleiosperma가 항산화능과 항염증 활성을 나타내는 유용한 소재임을 처음으로 밝혔다. 본 연구에서는 E. pleiosperma 에탄올 추출물(EPEE)의 항비만 활성을 췌장 리파아제 효소 활성 억제능 및 세포실험모델계를 이용하여 분석하였다. 먼저 EPEE는 농도 의존적으로 lipase 효소 활성을 유의적으로 억제시켰으며, 3T3-L1 preadipocyte에서 지방세포 분화, 세포 내 지방 축적, TG 함량 등을 독성 없이 농도의존적으로 억제하였으며 지방세포 내 중성지방을 유의적으로 분해시키는 것으로 나타났다. 이러한 EPEE의 지방세포 분화 억제능은 핵심 작용 인자인 $C/EBP{\alpha}$, $C/EBP{\beta}$, 그리고 $PPAR{\gamma}$의 유전자 및 단백질 발현조절에서 기인함을 확인하였다. 이러한 결과는 E. pleiosperma가 보유한 췌장 lipase 활성 저해능, 지방세포 분화 억제능, 지방세포 내 지방 분해능을 통한 항비만 활성을 처음으로 밝혀낸 것이며 추후 계속적인 연구를 통해 활성 물질의 규명이 필요할 것으로 판단된다.
FCEE를 3T3-L1 전지방세포의 분화과정 중에 처리한 후, FCEE가 지방의 축적에 미치는 영향을 확인하였다. FCEE($200{\mu}g/mL$)를 세포에 처리한 후 Oil Red O 염색법과 AdipoRed 형광염색법을 이용한 지방 축적과 세포내 중성지방 함량을 측정한 결과, 무처리한 지방세포와 비교하여 각각 10.2%, 13.7% 유의하게 감소하는 것을 확인하였다. 이러한 결과에 대한 기작을 확인하고자 포도당 유입량과 유리 글리세롤 방출량을 측정하였다. FCEE($200{\mu}g/mL$) 처리 시 지방세포와 비교하여 포도당 유입량이 36.6% 유의적으로 감소하였으며, 유리 글리세롤 방출량도 8일째에 유의적으로 감소하였다. 이는 지방세포의 분화과정 중에 FCEE의 처리로 분화가 억제되어 지방구의 형성이 억제되고 배양액으로 배출되는 유리 글리세롤의 총량이 감소한 것으로 판단된다. 이러한 결과가 지방세포 분화의 억제에 의한 것인지 판단하기 위해 지방 형성 관련인자의 mRNA 발현량을 측정하였다. FCEE($200{\mu}g/mL$)의 처리로 AMPK mRNA 발현량은 지방세포와 비교하여 3배 증가하였으며, SREBP-1c, $C/EBP{\alpha}$와 $PPAR{\gamma}$ mRNA 발현량은 각각 0.6, 0.6 및 0.8배 감소하는 것을 확인하였다. 이상의 결과들로부터 FCEE는 지방 합성을 억제하는 활성을 보유하고 있는 바, 향후 항비만 기능성 식품소재로 활용될 수 있을 것으로 사료된다.
