A study on the variation of physical properties of line heated classification DH32 thick plate steel (선상가열한 선급 DH32 후판 강재의 물성 변화에 관한 연구)

  • This study was performed to study the possibility of TMCP's application for side shell plating curved structures through mechanical property testing to understand if the rules and regulations of DNV and the quality standard of IACS after line heating for TMCP steel and normalizing DH32 material steel could be satisfied. Experimental results showed that TMCP's strength was measured to YS = 385 MPa, 18% higher than the required 315 MPa and TS = 525 MPa, complying with the required range of between 440 and 570 MPa. The minus 20 degree impact test for the Charpy V-Notch complied with the required standard and in addition the hardness test satisfied the requirement of 'Hv10 = 130 ~ 320' by reaching an average of T : 216 and L : 275 respectively.

Inhibitory Effects of Aralia cordata Thunb Extracts on Nitric Oxide Synthesis in RAW 264.7 Macrophage Cells (독활(Aralia cordata Thunb) 추출물의 Nitric Oxide Synthesis 저해효과)

  • Assessment was made of the effects of Aralia cordata Thunb (DH) on the cell proliferation, inducible nitric oxide synthase (iNOS) mRNA gene expression and nitric oxide (NO) production in RAW 264.7 macrophage cells. For the screening of anti-inflammatory activities, ethanolic extracts of 55 species of traditional herbal medicines were examined for inhibitory effects, and it was confirmed that DH possessed inhibitory effects on NO production. As a result, DH significantly decreased the production of NO and iNOS gene expression at a concentration of $250{\mu}g/mL$. The chloroformsoluble fractionates have the strongest No synthesis inhibitory effect. It is presumed that the inhibition of NO production in LPS-stimulated RAW 264.7 cells by DH components occurred via the modulation of iNOS and DH, and that the active compound from DH may be useful for therapeutic management of inflammatory-associate diseases.

Effect on Digestion Efficiency by Adding Microbial Agent in Mesophilic Two-stage Anaerobic Digester (중온2단혐기성소화조에 미생물제재 주입시 소화효율에 미치는 영향)

  • In the near future, the capacity of conventional anaerobic digester is thought to be insufficient because of the increase of the total solids from expansion of intercepting sewer, sewage quantity and direct input of night soil from near apartment districts. The objectives of this study was to investigate the improvement of digestion efficiency using microbial agent(Bio-dh). The system was a pilot-scale, two-staged, anaerobic sludge digestion system. The first-stage digester was heated and mixed. The agitation velocity of the first-stage digester was 120rpm. The second-stage digester was neither heated nor mixed. The Digestion temperature was kept at $35{\pm}1^{\circ}C$ The detention time of digester was 19 days. The dosage of sewage sludge and microbial agent were $0.65m^3/day$ and $0.5{\ell}/day$, respectively. The experiments was run for 25days. Three times a week, $COD_{Mn}$ and SS of effluent, TS, VS, and biogas production rate were measured. Temperature, pH, and alkalinity were measured daily. The results were as follows ; Without microbial agent, digestion efficiencies ranged 46.0%~50.9%(mean=48.6%), with microbial agent(Bio-dh), digestion efficiencies ranged 52.8%~57.3%(mean=54.2%). Consequently, microbial agent(Bio-dh) increased the sludge digestion efficiency about 12%. Also, Without microbial agent, the mean concentration of $COD_{Mn}$ and SS of second-stage digester effluent were 1,639mg/L, 4,888mg/L respectively. With microbial agent, the mean concentration of $COD_{Mn}$ and SS of second-stage digester effluent were 859mg/L, 2,405mg/L respectively. Consequently, microbial agent(Bio-dh) increased the removal efficiency of $COD_{Mn}$ and SS about 47.6% and 50.8%, respectively.

Development of Indole-3-Acetic Acid-Producing Escherichia coli by Functional Expression of IpdC, AspC, and Iad1

  • Biosynthesis of indole-3-acetic acid (IAA) via the indole-3-pyruvic acid pathway involves three kinds of enzymes; aminotransferase encoded by aspC, indole-3-pyruvic acid decarboxylase encoded by ipdC, and indole-3-acetic acid dehydrogenase encoded by iad1. The ipdC from Enterobacter cloacae ATCC 13047, aspC from Escherichia coli, and iad1 from Ustilago maydis were cloned and expressed under the control of the tac and sod promoters in E. coli. According to SDS-PAGE and enzyme activity, IpdC and Iad1 showed good expression under the control of $P_{tac}$, whereas AspC was efficiently expressed by $P_{sod}$ originating from Corynebacterium glutamicum. The activities of IpdC, AspC, and Iad1 from the crude extracts of recombinant E. coli Top 10 were 215.6, 5.7, and 272.1 nmol/min/mg-protein, respectively. The recombinant E. coli $DH5{\alpha}$ expressing IpdC, AspC, and Iad1 produced about 1.1 g/l of IAA and 0.13 g/l of tryptophol (TOL) after 48 h of cultivation in LB medium with 2 g/l tryptophan. To improve IAA production, a tnaA gene mediating indole formation from tryptophan was deleted. As a result, E. coli IAA68 with expression of the three genes produced 1.8 g/l of IAA, which is a 1.6-fold increase compared with wild-type $DH5{\alpha}$ harboring the same plasmids. Moreover, the complete conversion of tryptophan to IAA was achieved by E. coli IAA68. Finally, E. coli IAA68 produced 3.0 g/l of IAA after 24 h cultivation in LB medium supplemented with 4 g/l of tryptophan.

Enzymatic Hydrolysis Optimization of a Snow Crab Processing By-product (홍게 가공부산물의 효소적 단백질 가수분해 최적화)

  • The objectives of this study were to evaluate a protease suitable for the enzymatic hydrolysis of a snow crab processing by-product (SPB) and to optimize the hydrolysis conditions using response surface methodology (RSM). The SPB was hydrolyzed at $50^{\circ}C$ and pH 7.0-7.2 to obtain various degree of hydrolysis (DH) using Flavourzyme at an enzyme/substrate (E/S) ratio of 3.0%. The reaction progress curve exhibited an initial fast reaction rate followed by a slowing of the rate. The DH was increased to 30% at 90 min with a final DH 32 to 36%. A central composite experimental design having three independent variables (reaction temperature, reaction time, and E/S ratio) with five levels was used to optimize the enzymatic hydrolysis conditions. Based on the DH data, the optimum reaction conditions for the enzymatic hydrolysis of the SPB were a temperature of $51.8^{\circ}C$, reaction time of 4 hr 45 min, and an E/S ratio of 3.8%. It was demonstrated that the enzymatic hydrolysate of SPB could be used as a flavoring agent or a source of precursors for the production of reaction flavors.

Mild Heat Treatments for Quality Improvement of Fresh-cut Potatoes (열처리가 신선편이 감자 가공품의 품질에 미치는 영향)

  • Effect of pre- and post-mild heat treatment on quality of fresh-cut potatoes was investigated with the focus on surface color, firmness, phenolics, vitamin C and sensory characteristics. As the pre-heat treatment before cutting of potatoes, dried hot air (DH) and hot water (WH) treatment were conducted at $45^{\circ}C$ for 60 min, respectively. As a post-heat (CH) treatments, potato cubes were dipped into water of $55^{\circ}C$ for 45 seconds after cutting. The potato cubes were packed in 15 cm x 20 cm LDPE pouches $30^{\mu}m$ thick and then stored at $5{\pm}1^{\circ}C$. The respiration rate of fresh-cut potatoes was decreased by mild heat treatment Especially, the rate of cubes treated with DH was decreased by $20\%$ compared to that of the non-heat treated. L value of cubes treated with DH showed higher value than that of others. Firmness and free phenolics had no significant difference between the treatments. The fresh-cut potatoes prepared with non-heat treatment showed higher vitamin C content than that of the heat treated. In sensory visual color, CH treated cubes marked the best quality. Conclusively, mild heat treatment, especially CH treatment, showed positive effect on browning inhibition and quality enhancement of fresh-cut potatoes.

Studies on the Bleaching of Rice-Straw Chemical Pulp (II) -Two-Stages Bleaching- (볏짚화학펄프의 표백에 관한 연구(제2보) -2단 표백-)

  • This study was carried out to acquire basic data necessary for the use of rice-straw chemical pulp. It investigated the proper bleaching conditions when rice-straw chemical pulp(alkaline sulfite-${Na_2}{S_2}{O_4}$) was bleached using the various kinds of bleaching agents by the two-stages bleaching methods. And, physical properties of pulps bleached with eight kinds of two-stages bleaching methods were tested. The results of this study were as follow; 1. The first-stage bleaching was conducted under the proper conditions with chlorine(C). And then, the proper conditions related to the calcium chlorite(H), hydrogen peroxide(P) and sodium hydrosulfite(Y) bleaching as the second-stage bleaching were investigated. The proper conditions of CH stage were determined to be 0.3% concentration of calcium chlorite, $30^{\circ}C$ of reaction temperature and 20min. of reaction time. For CP stage, the proper conditions of concentration of hydrogen peroxide, reaction temperature and reaction time were 1.5%, $60^{\circ}C$ and 90min., respectively. And for CY stage, the proper conditions were 0.5% concentration of sodium hydrosulfite, $40^{\circ}C$ of reaction temperature and 90min. of reaction time. 2. The first-stage bleaching was conducted under the proper conditions with chlorine dioxide(D). And then, the proper conditions related to the calcium chlorite(H), hydrogen peroxide(P) and sodium hydrosulfite(Y) bleaching as the second-stage bleaching were investigated. The proper conditions of DH stage were determined to be 0.5% concentration of calcium chlorite, $25^{\circ}C$ of reaction temperature and 5min. of reaction time. For DP stage, the proper conditions of concentration of hydrogen peroxide, reaction temperature and reaction time were 1.0%, $70^{\circ}C$ and 90min., respectively. And for DY stage, the proper conditions were 0.3% concentration of sodium hydrosulfite, $50^{\circ}C$ of reaction temperature and 20min. of reaction time. 3. The first-stage bleaching was conducted under the proper conditions with calcium chlorite(H). kAnd then, the proper conditions related to the hydrogen peroxide(P) and sodium hydrosulfite(Y) bleaching as the second-stage bleaching were investigated. The proper conditions of HP stage were determined to be 0.3% concentration of hydrogen peroxide, $30^{\circ}C$ of reaction temperature and 60min. of reaction time. For HY stage, the proper conditions of concentration of sodium hydrosulfite, reaction temperature and reaction time were 2.5%, $60^{\circ}C$ and 30min., respectively. 4. When the rice-straw chemical pulps were bleached with eight kinds of two-stages bleaching methods in the proper conditions mentioned above, respectively, the final brightnesses after CH, CP, CY, DH, DP, DY, HP, and HY bleachings were 62.0, 74.3, 61.4, 58.9, 66.9, 62.9, 50.4 and 60.1, respectively. And strengthes of pulps bleached with DP and DH methods were comparatively higher than those of pulps bleached with other bleaching methods.

Cloning of the Endoglucanase Gene from Actinomyces sp. 40 in Escherichia coli and Some Properties of the Gene Products

  • The $\beta$-1,4-endoglucanase gene from Actinomyces sp. 40 was cloned into Escherichia coli DH5$\alpha$ with pUC19. Chromosomal DNA from Actinomyces sp. 40 was cleaved with the restriction enzyme Sau3AI and ligated into pUC19 for the transformation of Escherichia coli DH5$\alpha$. Positive clones of $\beta$-1,4-endoglucanase gene were detected as the clear zones on a medium supplemented with carboxymethylcellulose (CMC). This transformant possessed a single plasmid, designated pDS1, which contained the vector DNA and a 3.5 kilobase (kb) Sau3AI insertion fragment encoding endoglucanase. The size of the cloned fragment was reduced to 2.0 kb. The endoglucanase activity produced by the E. coli DH5$\alpha$ (pDS6) was higher than that of Actinomyces sp. 40 strain. The optimum pH and temperature of the cloned enzyme were pH 4.0$\sim$5.0 and 55$^{\circ}C$, respectively. The cloned enzyme was stable at 55$^{\circ}C$ or below and in buffer ranging from pH 4.0 to 7.0. The enzyme degraded CMC but did not degrade xylan, cellobiose, and methyl-umbelliferylcellobiopyranoside (MUC).

Carbon Monoxide Dehydrogenase in Cell Extracts of an Acinetobacter Isolate (Acinetobacter sp.1의 일산화탄소 산화효소의 특성)

  • Extracts of CO-autotrophically grown cells of Acinetobacter sp. 1 were shown to use thionin, methylene blue, or 2,6-dichlorophenol-indophenol, but not NAD, NADP, FAD, or FMN, as electron acceptors for the oxidation of CO under strictly anaerobic conditions. The CO dehydrogenase (CO-DH) in the thes bacterium was found to be an inducible enzyme. The enzyme activity was determined by an assay based on the CO-dependent reduction of thionin. Maximal reaction rates were found at pH 7.5 and $60^{\circ}C$, and the Arrhenius plot revealed an activation energy of 6.1 kcal/mol(25.5kJ/mol). THe $K_m$ m/ for CO was $154{\mu}M$. Known metalchelating agents tested had no effects on the CO-DH activity. No divalent cations tested affect the enzyme activity significantly escept $Cu^{2+}$ which suppressed the activity completely. The enzyme was inhibited by glucose and succinate. The same extracts catalyzed oxidation of hydrogen gas and formate with thionin as electron acceptor. The CO-DH of Acinetobacter sp. 1 was to have no immunological relationship with that of Pseudomonas carboxydohydrogena.

A Study on the Properties of TiN Films by Using Electron Beam Irradiation (전자-빔 조사를 이용한 TiN 박막의 물성변화에 관한 연구)

  • Titanium nitride (TiN) films were deposited on the polycarbonate substrate by using radio frequency (RF) magnetron sputtering without intentional substrate heating. After deposition, the films were bombarded with intense electron beam for 20 minutes. The intense electron irradiation impacts on the crystalline, hardness and surface roughness of the TiN films. The films irradiated with an electron beam of 300 eV show the small grains on the surface, while as deposited TiN films did not showany grains on the surface. Also the surface harness evaluated with micro indenter was increased up to 18 Gpa at electron energy of 900 eV after electron beam irradiation. In addition, surface root mean square (RMS) roughness of the films irradiated with intense electron beam affected strongly. The films irradiated by electron beam with 900 eV have the lowest roughness of 1.2 nm in this study.
