Targeting Acetate Kinase: Inhibitors as Potential Bacteriostatics

  • Despite the importance of acetate kinase in the metabolism of bacteria, limited structural studies have been carried out on this enzyme. In this study, a three-dimensional structure of the Escherichia coli acetate kinase was constructed by use of molecular modeling methods. In the next stage, by considering the structure of the catalytic intermediate, trifluoroethanol (TFE) and trifluoroethyl butyrate were proposed as potential inhibitors of the enzyme. The putative binding mode of these compounds was studied with the use of a docking program, which revealed that they can fit well into the enzyme. To study the role of these potential enzyme inhibitors in the metabolic pathway of E. coli, their effects on the growth of this bacterium were studied. The results showed that growth was considerably reduced in the presence of these inhibitors. Changes in the profile of the metabolic products were studied by proton nuclear magnetic resonance spectroscopy. Remarkable changes were observed in the quantity of acetate, but other products were less altered. In this study, inhibition of growth by the two inhibitors as reflected by a change in the metabolism of E. coli suggests the potential use of these compounds (particularly TFE) as bacteriostatic agents.

연속 혐기성 수소발효 공정에서 성공적인 start-up 방법 (Start-up Strategy for the Successful Operation of Continuous Fermentative Hydrogen Production)

  • The variations of performance and metabolites at an early stage were investigated for the successful start-up technology in continuous fermentative hydrogen production. Unsuccessful start-up was observed when the operation mode was changed from batch to continuous mode after the yield was reached to 0.5 mol $H_2$/mol $hexose_{added}$ by batch mode. $H_2$ production continued till 12 hours accompanied by butyrate production, but did not last with propionate production increase. It was suspected that the failure was due to the regrowth of propionic acid bacteria during batch mode which were inhibited by heat-shock but not completely killed. Thus, successful start-up was tried by early switchover from batch to continuous operation; continuous operation was started after the $H_2$ yield was reached to 0.2 mol $H_2$/mol $hexose_{added}$ by batch mode. Although $H_2$ production rate decreased at an early stage, stable $H_2$ yield of 0.8 mol $H_2$/mol $hexose_{added}$ was achieved after 10 days by lowering down propionate production. And it was also concluded that the reason for $H_2$ production decrease at an early stage was due to alcohol production by self detoxification mechanism against VFAs accumulation.

Influence of Dietary Butyrate on Growth Rate, Efficiency of Nutrient Utilization and Cost of Unit Gain in Murrah Buffalo (Bubalus bubalis) Male Calves

  • Eighteen Murrah male buffalo calves were allotted into three groups of six each. The calves in group I (control) were fed with whole milk, skim milk, calf starter and green maize fodder. The calves in group II (high butyric acid) and group III (low butyric acid) were fed with the same diet as control along with 24 ml and 12 ml of butyric acid/calf/day for 120 days, respectively for 120 days. Dry matter intake was higher in group II and III as compared with group I. Digestibility of dry matter, organic matter, crude protein, crude fibre, ether extract, neutral detergent fibre, acid detergent fibre, cellulose and hemicellulose was the highest in group II followed by group III and the control group. Body weight gain and conversion efficiency of dry matter, digestible crude protein and total digestible nutrients were better in group II. Cost of feed for per unit of live weight gain was the lowest in group II. It was concluded that dietary addition of butyric acid (24 ml/day) was economical and had positive effect on the performance of Murrah buffalo calves.

Preventive Effect of Ebelactone B, an Esterase Inhibitor on Rice Sheath Blight Caused by Rhizoctonia solani

  • Two types of Rhizoctonia solani esterases induced by cutin hydrolysate were partially purified by ammonium sulfate precipitation and gel filtration. The esterase I with hydrolyzing activity toward both ${\rho}-ni-trophenyl$ butyrate and ${\rho}-nitrophenyl$ palmitate and the esterase II with hydrolyzing activity toward only ${\rho}-ni-trophenyl$ butyrate were inhibited by ebelactone B, an esterase inhibitor produced by actinomycetes with $IC_{50}$ values of 0.01 and $0.09{\;}\mu\textrm{g}/l$, respectively. Spraying on rice seedling with ebelactone B at a concentration of $30{\;}\mu\textrm{g}/ml$ completely suppressed infection by R. solani. Ebelactone B could not protect the wounded rice seedling and did not show any inhibitory effect on the mycelial growth at a concentration of 1 mg/ml. These results indicate that ebelactone B, an esterase inhibitor protects rice plants from infection with R. solani by inhibition of penetration, not through fungitoxic or fungicidal effect.

Levanoligosaccharide(levanoctaose)의 장내미생물에 대한 생육효과 (Growth Effect of Levanoligosaccharide(Levanoctaose) on Intestinal Microflora)

  • Pseudomonas sp. K-52 유래의 levanase 에 의해 생산되는 levanoctaose 가 growth factor로서 각종 장내미생물에 미치는 영향을 조사하였다. In vitro 실험에서 0.5% levanoctaose를 탄소원으로 하여 glucose 일 경우와 상호 비교하여 분석한 결과 levanoctaose 는 Escherichia coli, Clostridium perfringens, Eubacterium limosum, Staphylococcus aureus 보다 Bifidobacterium adolescentis, Lactobacillius acidophilus, Bacteroid fragilis 등에 의해 효율적으로 이용되었다. 쥐를 이용한 in vivo 실험에서 탄소원의일부로 levanotaose를 제공한 경우, 장내 Bifidobacteria 수는 약 10배, butyrate 양은 2.3배, $\beta$-fructosidase 활성은 약 1.5배 증가하였다. 따라서 분리균주 Pseudomonas sp. K-2의 levanase로부터 생성되는 levanoctaose는 Bifidobacteria, Lactobacillus 와 같은 장내 유익균주에 선별적으로 이용되는 것이 확인되었다.

한우(韓牛) 말초혈액내(末梢血液內) rosette 형성세포(形成細胞)의 효소화학적(酵素化學的) 염색성(染色性) 비교(比較) (Comparison of enzyme cytochemical activities between rosetted cells in peripheral blood of Korean native cattle)

  • Peripheral blood mononuclear cells(PBMNC) of Korean native cattle rosetted with Korean goat erythrocytes(KGRBC) and blood monocytes were evaluated for four cytochemical reactions such as acid phosphatase(ACP), alkaline phosphatase-anti-boby(ALP-Ab), ${\alpha}$-naphthyl butyrate esterase(${\alpha}$-NBE) and peroxidase. The results obtained were as follows; In rosetted cells, the positivities of ACP in E AET-DeX, EA and EAC were 70.3%, 22.4% and 25.2%, those of ${\alpha}$-NB were 27.4%, 44.2% and 79.8%, and those of ALP-Ab were 9.5%, 88.3% and 91.5%, respectively. Whereas, the positivity for Peroxidase in monocytes was 100%. In non-rosetted (remained) cells, the positivities of ACP in E AET+DeX. EA and EAC were 41.4%, 57.2% and 61.9%, those of ${\alpha}$-NB were 38.6%, 16.5% and 18.9% and those of ALP-Ab were 98.2%, 5.3% and 6.3%, in order.

연속반응실험에서 수소생성에 대한 pH 영향 (Effect of pH on Continuous Hydrogen Fermentation)

  • 연속반응조에서의 수소생산에 대한 pH의 영향을 HRT 10시간으로 유지하고, pH 4.1부터 8.0까지의 범위에서 조사하였다. 실험조건에서의 생성된 수소가스 성분은 41-71% 범위로 발생되었다. $H_2$/$CO_2$ 비율은 pH 6.0 이상에서는 크게 변화가 없었으나, 대체적으로 pH가 증가함에 따라 $H_2$/$CO_2$ 비율도 증가하였다. 최대 수소생성수율은 pH 5.0에서 3.16$\ell$/g sucrose이었다. Acetate 생성은 pH 증가에 따라 증가하였으나, butyrate 생성은 pH 증가에 따라 감소하였다. 미생물량은 pH 증가에 따라 증가하였다.

Rhodobacter sphaeroides KD131에 의한 유기산 광합성 발효 최적화 (Optimization of Various Organic Acids on Photo-Fermentative Hydrogen Production using Rhodobacter sphaeroides KD131)

  • Photobiological $H_2$ production was compared using purple non-sulfur bacteria Rhodobacter sphaeroides KD131 in the medium containing various organic acids as the carbon source and electron doner under illumination of $110\;W/m^2$ using halogen lamp at $30^{\circ}C$. The organic acids used were 0~120 mM acetate, butyrate, lactate and malate. Initial pH 7.0 and cell concentration 1.0 at 660nm were increased to pH 8 and 4.4~5.1, respectively during 24hrs of photo-fermentation when lactate and malate were used. However, acetate and butyrate increased pH to 9 and cell concentration to 3.2~3.9 of malate at the same experimental conditions. Optimum ranges of organic acids concentration and carbon/nitrogen ratio were 30~60 mM and 10~20, respectively. When malate was used as the substrate, maximum $H_2$ production 1.1 ml $H_2$/ml broth, which is equivalent to 1.97 mol $H_2$/mol malate was observed.

미세조류를 이용한 중온 및 고온 혐기성 수소 발효 (Hydrogen Production from Microalgae in Anaerobic Mesophilic and Thermophilic Conditions)

  • This study was conducted to evaluate the characteristics of dark fermentative $H_2$ production from microalgae (Chlorella vulgaris) using batch reactors under mesophilic (25, $35^{\circ}C$) and thermophilic (45, $55^{\circ}C$) conditions. The $H_2$ yield and $H_2$ production rate increased with increasing temperature. The maximum $H_2$ yield and $H_2$ production rate were 56.77 mL $H_2/g$ dcw, 3.33 mL $H_2/g\;dcw{\cdot}h$ at $55^{\circ}C$, respectively. The activation energy calculated using Arrhenius equation was 36.24 kcal/mol, which was higher than that of dark $H_2$ fermentation of glucose by anaerobic mixed culture. Although the concentration of butyrate was maintained, the concentrations of lactate and acetate increased with increasing temperature. The $H_2$ yield was linearly proportional to acetate/ butyrate ratio.

음식물류 폐기물의 혐기성 소화 시 식종 방법에 따른 start-up 특성 평가 (Evaluation of Characterization During Start-up of Anaerobic Digestion Via Various Seeding Methods)

  • This study was performed to evaluate the characteristics of start-up of anaerobic digestion from food waste with different inoculum ratios. The hydrogen yield was similar with different inoculum ratios. The hydrogen production rate increased with increasing inoculum ratio. But the specific hydrogen production rate decreased with increasing inoculum ratio. Total volatile fatty acids composition analysis showed that butyrate and acetate were the prevalent products in all reactors, followed by lactate and propionate. The acetate was most prevalent product in reactors at $X_0/S_0=0.080$ and 0.159. But in reactors at $X_0/S_0=0.239$ and 0.318, butyrate accounted for greater than 50% of the total volatile fatty acids.
