Evaluation of Extractants for Bio-butanol Extraction Fermentation Using Organic Solvents and Ionic Liquids (유기용매와 이온성액체를 이용한 바이오 부탄올 추출발효 용매 선정 평가)

  • Oleyl alcohol, butyl butyrate, and two different ionic liquids were evaluated for the extraction of butanol from culture broth without toxic effect to cells. The tested solvents showed more than 50% extraction efficiency, and oleyl alcohol was chosen as the best extractant for butanol among the used extractants with a partition coefficient of 2.89. When oleyl alcohol was used as an extractant, more than 80% of butanol was extracted in the wide range of butanol concentrations (1-20 g/L) and pH values (pH 4-5.5). In extractive fermentation using oleyl alcohol only, there was 11% more butanol production and glucose consumption when compared to that without extractive fermentation, implicating a reduced inhibitory effect of butanol due to butanol removal to the oleyl alcohol phase. In addition, oleyl alcohol did not inhibit cell growth, while a mixture of oleyl alcohol and butyl butyrate with the volume ratio of 9:1~7:3 inhibited either butanol production or cell growth significantly due to the toxicity of butyl butyrate to cells. In conclusion, oleyl alcohol can be used as an efficient and non-toxic solvent for extractive fermentation for butanol production.

Effects of Bacterial Inoculants and Organic Acids on Silage Quality : Meta-analysis (미생물제제 및 유기산제제의 처리가 사일리지 품질에 미치는 영향 : 메타분석)

  • This study was conducted to estimate and compare the effects of bacterial inoculants and organic acids on silage quality. Silage pH, lactate, acetate, lactate:acetate ratio, propionate, butyrate, water-soluble carbohydrate, crude protein, ammonia-N, neutral detergent fiber and acid detergent fiber (ADF) were used as parameters for quality analysis and a meta-analysis technique was employed to determine the effect size. As a data pool for analysis, we examined 14 research papers. Bacterial inoculants were found to elevate pH, lactate, acetate, lactate:acetate ratio, propionate and ADF contents compared to the controls (p<0.01). In contrast bacterial inoclulants decreased butyrate, water-soluble carbohydrate, crude protein and ammonia-N contents (p<0.01). In the organic acid treatments, all parameters except ADF showed higher contents than the control (p<0.01). In the comparison of effect sizes between the two treatments, significant differences were detected in butyrate, water-soluble carbohydrate, crude protein and ammonia-N (p<0.05). It may be concluded that bacterial inoculants could improve silage quality in terms of the aforementioned four parameters compared with organic acid treatments.

Electrochemical Characteristics of the MFCs using the Ceramic Membrane as a Separator (세라믹막을 이용한 미생물연료전지의 전기화학적 특성 연구)

  • This study attempts to verify the applicability of ceramic membrane as a separator by comparing the power generation characteristics in single-chamber MFCs using ceramic membranes to those in the MFCs using nafion membrane. The generated power in MFCs by using acetate as a substrate was more stable than that by using formate, propionate and butyrate, respectively. It was shown that the generated power by using formate substrate in MFCs was unstable and a little higher than that by using acetate, and the power generated by using propionate and butyrate were lower than that by using acetate. In order to find out the Pt catalyst effect, it was compared the power generated in MFCs using Pt-coated carbon cloth as electrode to that power using normal carbon cloth. The power generated in MFCs using Pt-coated carbon cloth as electrode was 1.2 times higher than that using normal carbon cloth. The Pt-coated carbon cloth was about 5 times more expensive than normal carbon cloth. It is suggested that both power generation efficiency and cost together should be considered in selecting electrodes of MFCs. It was found that the ceramic membrane was superior to nafion membrane by comparing to the power generation characteristics obtained. It was shown that average voltage values were $523.67mV{\pm}49.41mV$ by using synthetic wastewater, in MFCs of ceramic membrane as a separator. While average voltage values were $424.09mV{\pm}79.95mV$ by using synthetic wastewater, in MFCs of nafion membrane as a separator. The organic removal efficiency, 41.7% by using ceramic membrane was a little bit higher than 40.8% by using nafion membrane. This research implies ceramic membrane can be a valid alternative to nafion membrane as a separator when considering the power generation and the efficiency of organics removal.

  • Allergic diseases have increased over the past several decade worldwide including developing countries. Allergic inflammatory responses are caused by Th (T helper)2 immune responses, triggered by allergen ingestion by antigen presenting cells such as dendritic cells (DCs). Intestinal microorganisms control the metabolism and physiological functions of the host, contribute to early immune system maturation during the early life, and homeostasis and epithelial integrity during life. Bifidobacteria have strain-specific immunostimulatory properties in the Th1/Th2 balance, inhibit TSLP (thymic stromal lymphopoietin) and IgE expression, and promote Flg (Filaggrin) and FoxP3 (Treg) expression to alleviate allergies. In addition, unmethylated CpG motif ODN (oligodeoxynucleotides) is recognized by TLR (toll-like receptors)9 of B cells and plasmacytoid dendritic cells (pDCs) to induce innate and adaptive immune responses, while the butyrate produced by Clostridium butyricum activates the GPR (G-protein coupled receptors)109a signaling pathway to induce the expression of anti-inflammatory gene of pDCs, and directly stimulates the proliferation of thymically derived regulatory T (tTreg) cells through the activation of GPR43 or inhibits the activity of HADC (histone deacetylase) to differentiate naive $CD4^+$ T cells into pTreg cells through the histone H3 acetylation of Foxp3 gene intronic enhancer.

Effect of Herbal Extracts on the Ruminal Dry Matter Digestibility, Volatile Fatty Acid Production and Growth Rate of Microbes in Vitro (한약재 추출물이 반추위 in vitro 건물소화율, 휘발성 지방산 생성 및 미생물 성장률에 미치는 영향)

  • This study was conducted to investigate the effect of herbal (Obtusifolia, Cinnamon, Chinese pepper, Licorice) extracts on the rumen fermentation in vitro. Comparing to the control, in vitro dry matter digestibility was significantly (P<0.05) decreased at zero hour in the Cinnamon and the Chinese pepper, and at three hour after supplementation in the Licorice. The ratio of volatile fatty acids were significant (P<0.05) differences at 3 hour after fermentation only, acetic acid was higher (P<0.05) in the control compare to the herbal extract treatments, but the ratios of butyrate, iso-butyrate, iso-valerate and valerate were lowest in the control. The growth rate of rumen microbes in vitro was significantly (P<0.05) higer in the herbal extract treatments excluding the Obtusifolia than the control during three hour fermentation, but was not significant difference among treatments in the other fermentation times. From above results, even though the extracts of Cinnamon, Chinese pepper and Licorice inclined to inhibit the activity of rumen microbes during early fermentation period, but did not affect on the growth rate of rumen microbes in vitro.

Application of lactic acid bacteria producing antifungal substance and carboxylesterase on whole crop rice silage with different dry matter

  • Objective: This study was conducted to investigate effects of antifungal substance and carboxylesterase-producing inoculant on fermentation indices and rumen degradation kinetics of whole crop rice (WCR) silage ensiled at different dry matter (DM) contents. Methods: Dual-purpose inoculants, Lactobacillus brevis 5M2 and Lactobacillus buchneri 6M1, confirmed both activities of antifungal and carboxylesterase in the previous study. The WCR at mature stage was chopped, and then wilted to obtain three different DM contents consisting of 35.4%, 43.6%, and 51.5%. All WCR forages were applied distilled water (CON) or mixed inoculants with 1:1 ratio at 1×105 colony forming unit/g (INO), and ensiled into 20 L mini silo (5 kg) in quadruplicates for 108 d. Results: The INO silages had lower lactate (p<0.001) and butyrate (p = 0.022) with higher acetate (p<0.001) and propionate (p<0.001) than those of CON silages. Ammonia-N (p<0.001), lactate (tendency; p = 0.068), acetate (p = 0.030), and butyrate (p<0.001) concentrations of INO silages decreased linearly with increasing DM content of WCR forage. The INO silages presented higher lactic acid bacteria (p<0.001) with lower molds (p<0.001) than those of CON silages. Yeasts (p = 0.042) and molds (p = 0.046) of WCR silages decreased linearly with increasing DM content of WCR forage. In the rumen, INO silages had higher the total degradable fraction (p<0.001), total volatile fatty acid (tendency; p = 0.097), and acetate (p = 0.007), but lower the fractional degradation rate (p = 0.011) and propionate (p<0.001) than those of CON silage. The total degradable fraction (p<0.001), total volatile fatty acid (p = 0.001), iso-butyrate (p = 0.036), and valerate (p = 0.008) decreased linearly with increasing DM content of WCR forage, while the lag phase (p<0.001) was increased linearly. Conclusion: This study concluded that application of dual-purpose inoculants on WCR silage confirmed antifungal and carboxylesterase activities by inhibiting mold and improving rumen digestibility, while increase of wilting times decreased organic acids production and rumen digestibility.

Effects of quercetin and coated sodium butyrate dietary supplementation in diquat-challenged pullets

  • Objective: This study was designed to investigate the hypothesis that dietary quercetin (QUE) and coated sodium butyrate (SB) supplementation alleviate oxidative stress in the small intestine of diquat (DIQ)-challenged pullets. Methods: A total of 200 13-week-old pullets were divided into four groups: the control group (CON), the DIQ group, the QUE group, and the coated SB group, and injected intraperitoneally with either saline (CON) or diquat (DIQ, QUE, and SB) to induce oxidative stress on day 0. Results: On the first day, the malondialdehyde and superoxide dismutase (SOD) concentrations in the SB group were significantly different from those in the DIQ and QUE groups (p<0.05), and dietary supplementation with SB increased serum glutathione peroxidase (GSH-PX) levels compared with the DIQ group (p<0.05). Quercetin and SB increased the levels of CLAUDIN-1 and zonula occludens-1 (ZO-1) in the jejunum. On the tenth day of treatment, QUE attenuated the decrease in GSH-PX levels compared to those of the CON group (p<0.05), while SB increased SOD, GSH-PX, and total antioxidant capacity levels compared to those of the DIQ group. Nuclear factor erythroid 2-related factor 2 (NRF2) and heme oxygenase-1 (HO-1) mRNA levels in the QUE and SB groups increased (p<0.05) and CLAUDIN-1 mRNA levels in the QUE and SB groups were upregulated compared to those in the DIQ group ileum tissue. Conclusion: Supplementation of QUE and SB demonstrated the ability to relieve oxidative stress in pullets post DIQ-injection with a time-dependent manner and QUE and SB may be potential antioxidant additives for relieving oxidative stress and protecting the intestinal barrier of pullets.

Characterization of Photosynthetic Bacteria in Swine Wastewater (양돈 계수에서 광합성 미생물의 특성)

  • Photosynthetic bacteria (PSB), strains IP-4 and IP-6-7 were isolated from nature, and were studied for swine wastewater treatment. It was confirmed that these PSB were very effective for the COD treatment. Growth of those PSB were increased to 4.5 fold in organic-acids (acetate, propionate and butyrate) added medium than cultivation in Lascelles basal medium and the amount of bacteriochlorophyll a were increased to 5 folds. The COD removal rate in swine wastewater using PSB, strains IP-4 and IP6-7 were obtained 91% and 85%, respectively.

  • Complexes of the type[Co(T)(L-pro)], where T is the quadridentate $N_2O_2$-type ligand, N,N'-dimethylethylenediamine-N,N'-diacetate or N,N'-dimethylethylenediamine-N,N'-di-$\alpha$-butyrate, have been prepared. The complexes were separated into the two stereoisomers, $\Delta$-s-cis-[Co(T)(L-pro)] and $\Lambda$-s-cis-[Co(T)(L-pro)]. They were characterized by their proton magnetic resonance, absorption and circular dichroism spectra, elemental analyses.
