작은소피참진드기에 대한 다양한 식물체 및 천궁유래물질의 살비 및 기피 효과 (Acaricidal and Repellent Activities of Cnidium officinale-derived Compounds Against Haemaphysalis longicornis (Acari: Ixodidae))

  • 충북 청주지역에서 채집된 작은소피참진드기(Haemaphysalis longicornis)에 대하여 다양한 식물체 메탄올 조추출물과 천궁유래 물질들을 이용하여 살비활성과 기피활성을 조사하였다. 총 24종의 식물추출물을 이용한 작은소피참진드기에 대한 살비활성을 조사한 결과, 천궁(Cnidium officinale)에서 93.3%의 살비율로 가장 높은 활성을 보였으며, 기피 효과의 경우 도라지, 미국자리공, 백목련, 어성초, 천궁, 칡잎 등에서 통계적으로 유의미한 차이를 보였다. 기피 및 살비활성이 가장 높은 천궁의 메탄올추출물을 용매 분획한 물질들과 천궁 유래물질인 butylidenephthalide의 활성을 비교한 결과, 헥산(hexane)층 처리 3일 후에 90%의 높은 살비효과를 보였으며 기피반응도 통계적으로 유의미한 차이를 보였다. 천궁유래물질인 butylidenephthalide은 처리 7일 후에 90%의 살비활성을 보였고 T-tube olfactometer 실험에서 모든 작은소피참진드기가 무처리구로 이동하여 기피활성이 있음을 알 수 있었다. 따라서 본 연구결과는 천궁 조추출물과 천궁유래물질인 butylidenephthalide를 이용하여 작은 소피참진드기의 방제 개발에 기초자료로 활용될 수 있을 것으로 생각된다.

자생종과 재배종 고본의 성분함량 비교 (Constituents Comparison of Components in Native and Cultivated Species of Angelica tenuissima Nakai)

  • The root of Angelica tenuissima Nakai (Umbelliferae) has been used in traditional medicines of Korea as a headache, common cold and a fever remedy. A. tenuissima contains ferulic acid and various compounds of essential oil group such as limonene, 3-butylidenephthalide, ${\gamma}$-terpinene, neocnidilide, ligustilide, senkyunolide and neocnidilide. This study carried out to compare the contents of ferulic acid, z-ligustilide and n-butylidenephthalide between native and cultivated species of A. tenuissima by HPLC. The average contents of ferulic acid, z-ligustilide and n-butylidenephthalide indicated that native species (9 samples) were 0.060%, 0.616%, 0.025% and cultivated species (15 samples) were 0.037%, 0.141%, 0.029%, respectively. All samples were collected from different places in Korea.

NFAT Transcription Factor Inhibitory Constituents from Cnidium officinale

  • Four hundred varieties of plant extracts were screened for inhibitory activity against the NFAT transcription factor which plays an important role in inducing immune response. Among them, the MeOH extract of Cnidium officinale showed potent activity, and the activity-guided separation yielded butylidenephthalide, senkyunolide A and falcarindiol as the active constituents. The $IC_{50}$ value of butylidenephthalide was $1.3{\times}10^{-4}\;M$ and was similar to that of senkyunolide A $(2.1{\times}10^{-4}\;M)$. Interestingly, falcarindiol showed higher activity $(IC_{50},\;2.6{\times}10^{-5}\;M)$ than the two phthalides.

  • In continuation of our studies on essential oils of Angelica genus(Umbelliferae), We report on the components of essential oils obtained from the root of Angelica tenuissima Nakai(藁子). Oils were obtained from the dried roots by steam distillation and fractionated by column chromatography. Each isolate or fraction was identified by GC, GC-MS and spectral analysis. Essential oils of the root of A. tenuissima(Gaoben) were found to contain $\alpha-pinene,\;camphene,\;\beta-pinene,\;myrcene,\;\alpha-phellandrene,\;\Delta-3-carene,\;p-cymene,\;limonene,\;\gamma-terpinene,\;terpinolene,\;4-vinylguauacol,\;\gamma-elemene$, one aromatic compound, three unidentified sesquiterpene alcohols, butylidenephthalide, senkyunolide and Z-ligustilide which was the most abundant compound comprising 75% of the whole oil. Also butylphthalide and hydroxybutylidenephalide were tentatively identified.

국내산 천궁의 향기 성분 (The Flavor Components of Korean Cnidium officinale Makino)

  • This study was performed to identify the flavor compnenets of Korean Cnidium officinale M.. The extraction of the flavor compounds of Cnidium officinale M. was accomplished by a simultaneous distillation and extraction method using a Likens and Nickerson's extraction apparatus. The concentrated extract was analyzed and identified by gas chromatography and GC-mass spectrometry. The change of the 림팩 contents in each samples was determined in terms of two different storage conditions. The flavor contents of sample at low temperature storage were higher than those at room temperature storage, and increased until storage period of 70 days. The major components of Cnidium officinale M. were terpene compounds including α-pinene, β -selinene, γ-selinene, and phthalides including butylidenephthalide, cnidilide, neocnidilide, senkyunolide A, ligustilide, which are believed to contribute to the characteristic flavor of the Cnidium officinale M.. Especially, great amount of cnidilide and ligustilide was shown in sample of low temperature storage.

천궁의 혈액순환과 항산화적 효과 (Effects of Ligusticum Chuanxiang on blood circulation and oxidative stress)

  • Ligusticum Chuanxiong and its components were reviewed in terms of pharmacodynamic mechanisms. Three components of about 40 chemical components in Chuanxiong tetramethylpyrazine, ferulinolol and Butylidenephthalide have mainly been studied for its pharmacodynamic mechanisms which are focused on the increase in blood flow and anti-oxidative stress. The mechanisms for the effects of Chuanxiong on the increase in blood flow can be summarized as four ways, 1) anti-coagulation 2) blocker of ${\beta}1$ adrenergic receptor 3) cellular control of Ca++ level 4) collagen synthesis. Chuanxiong also showed the effectiveness on free radical-induced injury. It seems that its effectiveness is also related to the mechanisms for the increase in blood flow.

당귀(Angelica sinensis)의 임상활용에 대한 효능 및 부작용연구 (Pharmacological action and Toxicity of Angelica sinensis)

  • Objectives : Angelica sinensis has been used extensively in Korean traditional medicine. Although thorough clinical trials are lacking, Various pharmacological actions for Angelica sinensis has been identified newly using animal models. In addition, it was reported that reactive intermediates, potentially causing toxic effects, was isolated from components in Angelica sinensis. In this article, it was purposed for explanation and introduction of new studies for Angelica sinensis. in terms of Pharmacological action and Toxicity. Methods : New studies for Angelica sinensis were reviewed and summarized in terms of pharmacological action and toxicity. Results and Conclusions : Angelica sinensis and its components including phthalide, phthalide dimers, polysaccharides, polyacetylenes, ligustilide와 butylidenephthalide as well as organic acid showed a variety of pharmacological actions on uterine, cardiovascular system and immune system. In addition, it was identified that three components of such as ligustilide, caffeic acid and safrole was biotransformed to reactive intermediates causing possibly toxicity.

참당귀, 중국당귀, 일당귀 및 그 구성 생화합물의 약리작용에 대한 고찰 (A Review of Pharmacological Effects of Angelica gigas, Angelica sinensis, Angelica acutiloba and their Bioactive Compounds)

  • Backgrounds: Danggui, one of the major herbs in Korean traditional medicine consists of three species: Angelica gigas of Korea, Angelica sinensis of China, and Angelica acutiloba of Japan. Despite its importance in Korean traditional medicine, recognition of the clinical differences between its three species is insufficient. Objectives: The purpose of the present review is to suggest evidence in using Angelica gigas, Angelica sinensis, and Angelica acutiloba distinctively in clinic, by comparing their pharmacological effects and bioactive compounds. Methods: We searched articles published from 2000 to 2009 in Pubmed, EMbase, and RISS. The search keywords were "Angelica gigas", "Angelica sinensis", "Angelica acutiloba", "dongquei", "toki", "Angelicae Radix", "Archangelica officinalis Hoffm.", "Garden Angelica", "Chinese angelica root", "tangkuei", and "danggui". 861 articles were searched. Among them, we selected 143 articles which met our inclusion criteria. Results: This review summarizes active constituents, experimental studies, clinical studies, pharmacokinetics, side effects and toxicity, drug interaction, and industrial use of Angelica gigas, Angelica sinensis, and Angelica acutiloba. Conclusions: While Angelica sinensis and Angelica acutiloba are relatively similar, Angelica gigas is quite different from the others in main active constituents and genetic form. The main experimental studies of Danggui are cardiovascular studies, central nervous system studies and anti-cancer activity. Even though there were cases in which the three species show similar pharmacological effects, the mechanism was not always shared. Therefore, distinguished use of Angelica gigas, Angelica sinensis and Angelica acutiloba is needed.

Ligustici Rhizoma(고본(藁本))와 Angelica tenuissimae Radix(한국고본(韓國藁本)뿌리)의 규격화(規格化) 및 유효성분(有效成分)의 진통효과(鎭痛效果) 비교(比較)에 관한 연구(硏究) (A Study on the Standardization of Ligustici Rhizoma and Angelicae Tenuissimae Radix including the Comparison for the Biological Activity on the Active Ingredients)

  • Major separation for the active ingredients and characterization of chemical properties in conjunction with screening test on animal were performed in order to analyze and standardize Ligustici Rhizoma or Angelicae Tenuissimae Radix as an important oriental herbal medicine for antiphlogistic or an important oriental herbal medicine for antiphlogistic or an anodyne. Furthermore the structure, composition and contents of ingredients for essential oil in Angelicae Tenuissimae Radix(Suckpo, Korea) were determined by means of Ge/MS followed by screening test on Z-ligustilide(82%) known as major ingredient as well as butylidenephthalide collected by HPLC with normal phase semiprep-column. The total active ingredient in Ligustici Rhizoma from China or Angelicae Tenuissimae harvested at Choonyang(Kyungnam, Korea), Jungsun(Kangwon, Korea), Suckpo(Kyungnam Korea), Youngchun(Kyungnam, Korea) have been determined showing higher abundant for three times on the product in Korea compared to that in China. In addition, the major component in Ahgelicae Tebyussunae Radux extract was found to be Z-ligustilide(70-80%) which is very different from that in Ligustici Rhizoma senkyunolide(39%) as major species. For screening test of Ligustici Rhizoma or Angelicae Tenuissimae Radix extracts toward the target animal, the efficiency has been shown the similarity on both extracts. Taking into account the level of ingredient, the total efficiency may be three times higher on Angelicae Tenuissimae Radix in Korea compared to Ligustici Rhizoma in China. As a result of present study, it is preferable to distinguish between Ligustici Rhizoma and Angelicae Tenuissimae Radix for better usage of oriental herbal medicine because of very different composition and abundant in spite of their similar screening effect.
