The complementary DNA (cDNA) of potato virus Y- vein necrosis strain (PVY-VN) replicase gene (Nlb) was transformed into tobacco (Nicotiana tabacum cv. Burley 21) plants. Out of 25 putative transformants regenerated, 3 were resistant to PVY-VN, one highly resistant plant with no symptom until seed harvest time and the other two with mild chlorotic spot symptoms at late stages after infection. No symptom was observed in the highly resistant plant, while mild vein necrotic symptoms were developed on suckers of the moderately resistant plants after seed harvest time, In the first generation (T1) via self fertilization, resistance to susceptibility frequency in transgenic plants from the highly resistant transformant was about 3 : 1, while it was lowered much (about 1:2 and 1:19) in T1 of the moderately resistant transformants. In the second generation (T2) of the highly resistant plant, resistance frequencies were similar to T1, but resistance levels varied greatly and appeared to be decreased. To investigate tile effects of growth characteristics on the yield, price per Kg, chemical contents and physical properties in Burley tobacco, ten varieties including Burley 21 were tested in this study. The results obtained are summarized as follows. 1 Forty to fifty days after transplanting, CCR (Crop Growth Rate) was the highest. RCR (Relative Growth Rate) increased up to 40 days, but decreased 50 days when maturation began. High- yielding varieties showed high CCR and RCR till 60 days. 2. Total alkaloid content of cured leaf increased about three times than that of topping stage, but the increased rates were some what different among varieties. 3. Leaf area, stalk diameter, stalk height and days to flower showed positive correlations to yield, whereas leaf thickness and weight per unit leaf area showed negative. 4 Varieties which are high in cured leaf weight ratio and weight per unit leaf thickeners showed relatively poor quality. 5 Nitrogen content was high in leafy and larger stalk diameter variety. 6. There are positive correlation between weight per unit leaf thickness and filling power. The time of combustion was positively correlated to leaf thickness and weight Per unit leaf. 7. It can be concluded that many characteristics are related to the yield, but not quality. It is, there fore, easy to Predict tile yield, but difficult to forecast the qualiffy.
This study was conducted to investigate the influence of the topping time and height on the nonvolatile organic and higher fatty acid contents and the correlation among acid contents of tobacco plants. Within the topping time, the flowering (normal) stage of topping was the lowest in total analyzed acid (TAA), total nonvolatile organic acid (TNOA) and magic acid contents of NC2326, however, the highest in those of Burley 21 on 30 days after budding. Total higher fatty acid (THFA) and linoleic acid contents tended to be low with normal stage of topping. The lower the topping height, the lower the TAA, THFA and linoleic acid contents in middle and upper leaves (or NC2326 and Burley 21 were observed. On the other hand, the lower the topping height, the lower the TNOA and malic acid contents in middle and upper leaves for Burley 21 and upper leaves for NC2326 were found. There was a significant positive correlation between magic acid and TNOA contents. Similarly, TAA content showed a significant positive correlation with TNOA, oxalic acid and citric acid contents. But there was a negative correlation between malic acid and THFA contents. There were highly positive correlations among oleic acid, linoleic acid and THFA contents, respectively.
The effects of shading - 90, 70, 50, 30, 10% on curing burley tobacco was investigated in pipe vinyl houses. The curing in sunlight under transparent materials resulted in an increase in temperature and a decrease in relative humidity. But amount of light on tobacco leaves were less than 10% sun light in all treatments. Shading was delayed curing period, but cured leaves under transparent shading materials were off color and brittle. Filling power and combustibility were deteriorated as decreased shading. But quality of cured leaves were non significant between treatments. Effects of shading were believed that resulted in a decrease in temperature and an increase in relative humidity than light.
This study was carried out to improve the analytical methods for determination of organic acids in tobacco leaf by HPLC. The samples for HPLC analysis were pre-treated by means of SPE. The calibration curve for each acid was linear and $R^2$ values ranged from 0.9990 to 1.0000. The limit of detection were obtained from the calibration curves and their values were between 1.39 to $4.87{\mu}g/mL$. Recovery rates of organic acids were between 88.6 % to 98.5 %. The concentrations of organic acids among the various tobacco leaves were compared to the concentration of organic acids, were in the order oriental, burley, flue-cured tobacco. In the case of flue-cured and oriental tobacco leaves, the order of concentration of organic acids was malic acid, oxalic acid, citric acid. But in the case of burley tobacco leaves, the order of concentration of organic acids was citric acid, oxalic acid, malic acid.
AFLP(Amplified Fragment Length Polymorphism) analysis was conducted to cultivars of tobacco, Nicotiana tabacum in order to select the cultivar-specific markers. AFLP results using 12 primer sets revealed genetic diversity among 12 field grown tobacco cultivars. Polymorphic fragments amplified by PCR was purified and cloned to identify their nucleotide sequences. From the sequences of them, 40 primer sets were designed to select cultivar-specific markers. When genomic DNA isolated from tobacco were used as PCR template, a set of primers, BrSF/BrSR showed Burley-specific band patterns. The results indicate that AFLP technique used in this experiments is useful for identifying tobacco cultivars in a rapid manner.
This study was conducted to investigate the effects of plant density and urea supplement on the yield, quality, value and total nitrogen, alkaloid contents of burley tobacco leaves. The results were as fellows; As urea supplement were increased from 0kg/10a to 50kg/10a, yield, total alkaloid, total nitrogen of cured leaf were significantly increased. The lower plant density levels also increased dry weight and leaf area per plant. Price of cured leaves were not influenced by plant density and amounts of urea supplemented. The effects of plant density and urea supplement on the number of leaves were not detected.
This experiment was carried out to fond out the proper transplanting time of burley tobacco since the transplanting dates In Jeonla Do varied from early April to late June. In this experiment , the first transplanting was lone on April 10 and continued until June, 20 with the total of 7 Plantings. The results of experiment from 1977 to 1978 showed no significant differences on value per kg (won /kg ) from April 10 to May 10 whether the plants were grown with mulching or non -mulching The yield and price per 10a were highest with May, 10 transplanting . The proper transplanting date obtained by regression analysis, however, was between May, 1 to May 5.
This experiment was carried out determine the effect of plowing depth and heavy fertilization on the yield and quality of Burley tobacco. In the experiment, three plowing depth (10, 20, 30cm) and three levels of compound fertilizer (138, 166, 193kg/10a) were employed with split plot design for three years (1976-1978). Yield and quality were increased with deeper plowing and heavier fertilization. Yield was increased by 5% at the plowing depth of 30cm and 13% at the fertilizing level, 193kg/10a. The value per kg was increased by 2% plowing depth of 30cm and at the fertilization of 193kg/10a, respectively.
This study was carried out to control the stalk curing period in Burley Tobacco, for its period was longer than 40 days. The 5 methods were treated to shorten the stalk curing period. The curing period was shortened for 1~3 days by the harvesting methods and the amount of predehydration, but it was shortened for 8 days by the hanging methods(sloping and horizontal curing)and the hanging density(90 plants/3.3m2) without the decrease of quality. In the ventilating condition of the curing house, the curing period could be controlled for 5 dyes with the quality like the conventional cured leaves by closing the curing house from the late curing stage. But when the curing house was not ventilated earlier, however the curing period was shortened but its quality was decreased.
