Effects of Feeding Bupleurum falcatum L. on Performance, Blood Component and Hormone Concentration in Broiler Breeder Hens (육용종계 사료에 시호(Bupleurum falcatum L.)의 첨가가 생산능력, 혈액의 성상 및 호르몬 분비에 미치는 영향)

  • An experiment was conducted to investigate the effects of feeding Bupleurum falcatum L.(BFL) on performance, blood component and hormonal secretion in broiler breeder hens. A total of 800 broiler breeder hens were allotted to four treatments(BFL ; 0, 0.5, 1.0, 2.0%) with four replications for eight weeks. Weight gain was not significantly different between the groups in growing period($20{\sim}24$ weeks), but egg production and egg mass of broiler breeder hens fed BFL were significantly(p<0.05) higher than those of the control groups in laying period($24{\sim}28$ weeks). Dietary BFL did not affect serum triglyceride concentration, whereas serum cholesterol concentration was significantly increased(p<0.05) in 2.0% BFL-fed groups. No statistical differences were found in serum glucose, Ca, P, Mg, total protein, albumin, GGT, AST, ALT and BUN concentrations. However, albumin/globulin ratio was statistically higher in BFL-fed groups compared to the control groups(p<0.05). Serum estrogen concentrations were significantly increased by feeding a diet containing 0.5% BFL, but progesterone were not influenced. Serum IGF-I of BFL groups was greater than that of control groups at 28 weeks(p<0.05). In conclusion, the results suggest that the dietary addition of BFL into broiler breeder diets improved egg production, serum estrogen and IGF-I concentration, and decreased serum cholesterol concentration and albumin/globulin ratio.

  • We examined the effects of various basal media and exogenous auxins on the growth and saikosaponin production in hairy root cultures of Bupleurum falcatum. Of the various media tested, 2RCM medium was superior for growth and saikosaponin content, in which saikosaponin a, c, and d was produced, however MS medium was resulted in extremely low level in saikosaponin production. By the treatment of 0.5 mg/L IBA the growth rate was increased by about 60% in 2RCM medium, whereas both IAA and IBA showed a potent inhibition effect in saikosaponin biosynthesis by the treatment of 0.01 mg/L~5mg/L.

  • Saikosaponin productivity was examined in a Bupleurum falcatum L. BFHR2 hairy root culture in response to changes in the sucrose content $(2{\sim}8%)$, nitrogen content $(0{\sim}250mM\;NH_4NO_3)$, phosphate content $(0{\sim}12mM\;NaH_2PO_4)$, and the potassium content $(0{\sim}87.2 mM\; KCl)$ of the culture media. We found that the conditions for maximal saikosaponin production differed from those for optimal root growth. Highest saikosaponin yield was achieved for 8% sucrose, 62mM $NH_4NO_3$ 1.2 mM $NaH_2PO_4$, and 0.5mM KCl.

Effects of Bupleurum falcatum Extract on the Survival of Cancered ICR Mouse and the Growth of Cancer Cells such as J774A.1 Cells and L1210 Cells (시호추출물의 ICR 발암생쥐의 생존율 및 J774A.1 세포와 L1210 세포의 증식에 미치는 영향)

  • The current investigation was carried out to find out the anticancer activity of the methanol extract from Buplerum falcatum against cancered ICR mouse and cancer cell lines such as J774A.1 and L1210 cells. Extract of Buplerum falcatum displayed the considerable augmentation(134%) of the survival of ICR mouse bearing Sarcoma 180 cancer. In addition, the cytotoxic effects of methanol extract of Buplerum falcatum against J774A.1 cells and L1210 cells were found to show $IC_{50}$ values of $57.3\;{\mu}g/ml$ and $54.6\;{\mu}g/ml$, respectively. In contrast to such cytotoxicity against cancer cells, the extract exerted only meagre toxicity against normal lymphocytes. The increased generation of $O_2^-$ and the considerably increased activities of super-oxide dismutase(SOD) and glutathione peroxidase(GPx) of both J774A.1 cells and L1210 cells in the presence of Buplerum falcatum extract implied that the observed cytotoxicities may have resulted from the detrimental effect of reactive oxygen species(ROS) evoked by Buplerum falcatum extract on the cancer cells.

Effect of Siho (Bupleurum falcatum L.) Extracts on Liver Function in $CCl_4$ Intoxicated rats (시호추출물이 간장기능 회복에 미치는 영향)

  • Effect of Siho (Bupleurum falcatum L.) extracts on liver function were investigated in CCl4 intoxicated rats. Plasma $\alpha$-fetoprotein and total protein concentration showed a tendency to decrease in Siho extracts groups. However plasma albumin concentration showed no significantly different in all treatment groups. Activities of plasma asparate aminotransferase and alanine aminotransferase in Siho extracts groups showed a lower values than control group. Alkan phosphatase and lactate dehydrogenase activities showed a tendency to decrease in Siho extracts groups. However ${\gamma}$-glutamyl transferase activity showed no significantly different in all treatment groups. Concentration of plasma triglyceride and total cholesterol showed no significantly different in all treatment groups.

Effects of Planting Date and Density on Growth Characteristics and Saikosaponins Content in Bupleurum falcatum L. (파종시기 및 재식밀도가 시호의 생육 및 Saikosaponin 함량에 미치는 영향)

  • This study was conducted to determine the optimum planting dates and density of one year old Bupleurum falcatum L. to improve its productivity and quality. Two cultivars of B. falcatum, originated from Jeongseon, Korea and Mishima, Japan were used. Some of the results obtained are as follows : Jeongseon cultivar showed less stem branches and shoot weight compared to Mishima. However, Jeongseon cultivar showed tall plant height, high root fresh and dry weight, and high levels of saikosaponin, but low saikosaponin content than that of Mishima. Both cultivars seeded on March 20 had long main root, big stem diameter, few stem branch, and high saikosaponin c content compared to those of late seeded one, April 30. Growth characteristics such as plant height, stem diameter, stem branch number, shoot weight, root diameter, root fresh and dry weight, and root branch number were increased in a low planting $density(30\;{\times}\;15cm)$, but the content of saikosaponin was not affected by planting density. Jeongseon and Mishima cultivars seeded on April 10 with $30\;{\times}\;15cm$ planting density and April 30 with $30\;{\times}\;10cm$ planting density contained the highest total saikosaponin levels, respectively. However, average root dry weight were not affected by planting time or density in both Bupleurum cultivars.

Embryogenesis and Regeneration from Anther Cultures of Bupleurum falcatum L. (시호의 약배양의 통한 배발생 및 재분화)

  • Effects of growth regulators(2,4-D, IAA, NAA, BA and kinetin) and chilling treatment on callus induction. embryogenesis and regeneration through anther cultures of B. falcatum L. were examined. Frequency of callus induction and embryogenesis was effectively increased by the treatment of chilling at 5$^{\circ}C$ for 10 days before anther inoculation. Optimal level of growth regulator for callus induction and embryogenesis from anther was 2,4-D 1.0 mg/L in Murashige and Skoog(MS) basal medium supplemented with 30 g/L sucrose, 8 g/L agar. Frequency of embryogenesis from anther derived callus was increased up to 48% or 45% by addition of IAA 0.1+ kinetin 1.0 mg/L of IAA 0.1+ BA 1.0 mg/L in MS medium, respectively, Optimal medium for obtaining green callus was MS basal supplemented with BA 1.0mg/L. Addition of auxins(IAA or NAA) inhibited the formation of green callus from anther derived callus.

Effect of Bupleurum falcatum on the immune system (시호 추출물이 면역계 세포의 활성에 미치는 영향)

  • The root of Bupleurum falcatum L.(BF) has been widely used in oriental medicine as a major camponent in many prescriptions for chronic hepatitis, renal disease, tuberculosis and some other infectious diseases. Many attempts have done to investigate the therapeutic effects of these principles. However, any kinds of screenig on immune regulatory- and antitumor- effects of BF has not been reported. The present study, therefore, was undertaken to investigate the BF-effects on cellular- and humoral-immune responses, phagocytic activities of macrophages, lymphokine- and Immunoglobulin(Ig)-production of lymphocytes, tumorigenesis of implanted sarcoma 180 cells and B16 melanoma cells, and proliferations of some tumor cell lines(Fsa II, 3LL and EL4). BF increased phagocytic activities of mouse peritoneal macrophages in a dose- and time-dependent fashion. Arthus reaction and antibody responses to SRBC were slightly enhanced but delayed hypersensitivity was depresed when BF was injected before- and after-SRBC sensitization. BF inhibited the proliferative responses of human tonsillar lymphocytes to PHA- and Con A-stimulation but slightly augmented the response of these cells to Staphylococcus aureus Cowan 1(SAC)-activation. Ig secretion of human mononuclear cells activated with SAC was slightly increased by BF. BF significantly augmented the SAC-induced IL 6 production of human mononuclear cells but not influenced Con Ainduced IL 2 secretion. NK cell activities of mouse splenocytes were somewhat increased when BF was pretreated and this responses were due to the increment of binding affinities of effector cells to target cells and of lytic activities of effector cells against target cells. In vitro BF significantly inhibited the proliferations of cancer cells such as Fsa II, 3LL and EL4 strains. BF decreased not only the frequency of tumor induction but also the tumor size per sarcoma 180 or B16 cell-implanted mouse. Taken together, these results indicate that BF is one of the potential immunomodulator, and suggest its possibility to be used as a desirable antitumor agent.
