The aim of this study was to compare two random regression models (RRM) fitted by fourth ($RRM_4$) and fifth-order Legendre polynomials ($RRM_5$) with a lactation model (LM) for evaluating Holstein cattle in Brazil. Two datasets with the same animals were prepared for this study. To apply test-day RRM and LMs, 262,426 test day records and 30,228 lactation records covering 305 days were prepared, respectively. The lowest values of Akaike's information criterion, Bayesian information criterion, and estimates of the maximum of the likelihood function (-2LogL) were for $RRM_4$. Heritability for 305-day milk yield (305MY) was 0.23 ($RRM_4$), 0.24 ($RRM_5$), and 0.21 (LM). Heritability, additive genetic and permanent environmental variances of test days on days in milk was from 0.16 to 0.27, from 3.76 to 6.88 and from 11.12 to 20.21, respectively. Additive genetic correlations between test days ranged from 0.20 to 0.99. Permanent environmental correlations between test days were between 0.07 and 0.99. Standard deviations of average estimated breeding values (EBVs) for 305MY from $RRM_4$ and $RRM_5$ were from 11% to 30% higher for bulls and around 28% higher for cows than that in LM. Rank correlations between RRM EBVs and LM EBVs were between 0.86 to 0.96 for bulls and 0.80 to 0.87 for cows. Average percentage of gain in reliability of EBVs for 305-day yield increased from 4% to 17% for bulls and from 23% to 24% for cows when reliability of EBVs from RRM models was compared to those from LM model. Random regression model fitted by fourth order Legendre polynomials is recommended for genetic evaluations of Brazilian Holstein cattle because of the higher reliability in the estimation of breeding values.

Genetic profiles of Iranian Holstein young bulls at the national artificial insemination station were determined on the basis of individual genotypes at 13 ISAG's recommended microsatellites, the most useful markers of choice for parentage identification. In the present study a total of 119 individuals were genotyped at 13 microsatellite loci and for possible parent-offspring combinations. A high level of genetic variation was evident within the investigated individuals as assessed from various genetic diversity measures. The mean number of observed alleles per microsatellite marker was 9.15 and the number of effective alleles as usual was less than the observed values (4.03). The average observed and expected heterozygosity values were 0.612 and 0.898, respectively. The mean polymorphic information content (PIC) value (0.694) further reflected a high level of genetic variability. The average exclusion of probability (PE) of the 13 markers was 0.520, ranging from 0.389 to 0.788. The combined exclusion of probability was 0.999, when 13 microsatellite loci were used for analysis in the individual identification system. Inbreeding was calculated as the difference between observed and expected heterozygosity. Observed homozygosity was less than expected which reflects inbreeding of -3.7% indicating that there are genetic differences between bull-sires and bull-dams used to produce young bulls. The results obtained from this study demonstrate that the microsatellite DNA markers used in the present DNA typing are useful and sufficient for individual identification and parentage verification without accurate pedigree information.
Recent favor on high quality food and concern on food safety have demonstrated the superiority of Hanwoo (Korean native cattle). In general, the price of cow is higher than those of steer and bull, causing cheating issues in the market. Hence, this study is to discriminate genders of Hanwoo with identification of factors which highly influence gender discrimination based on the big-size deboning data. Totally, there were 31 variables in the deboning data, and we divided into them two categories: data obtained before and after deboning. Discriminant function analysis was then applied into the data to determined the accuracy of gender discrimination in Hanwoo. The result showed that Hanwoo could be classified by gender with 99.2% of accuracy when using all 31 variables. In detail, it was possible to identify 93 of 94 bulls (98.9%), 96 of 96 cows (100%) and 74 of 75 steers (98.7%). The most significant variables was chuck, sirloin, armbone shin, plates, retail and cuts percentage, sequentially. With variables obtainable before deboning, accuracies of classification were 91.5% for bulls, 92.7% for cows, and 89.3% for steers. The most significant variables was water, cold carcass weight and back-fat thickness. The discrimination accuracy was higher with data obtainable after deboning: bulls (98.9%), cows (99.0%) and steers (98.7%). In this case, chuck, sirloin and armbone shin were the factors determined the classification ability. This study showed that Hanwoo can be classified based on deboning data with appropriate statistics, further suggesting weight of cut of beef might be the standard for gender classification.
Objective: Cattle were some of the first animals domesticated by humans for the production of milk, meat, etc. Long noncoding RNA (lncRNA) is defined as longer than 200 bp in nonprotein coding transcripts. lncRNA is known to function in regulating gene expression and is currently being studied in a variety of livestock including cattle. The purpose of this study is to analyze the characteristics of lncRNA according to sex in Hanwoo cattle. Methods: This study was conducted using the skeletal muscles of 9 Hanwoo cattle include bulls, steers and cows. RNA was extracted from skeletal muscle of Hanwoo. Sequencing was conducted using Illumina HiSeq2000 and mapped to the Bovine Taurus genome. The expression levels of lncRNAs were measured by DEGseq and quantitative trait loci (QTL) data base was used to identify QTLs associated with lncRNA. The python script was used to match the nearby genes Results: In this study, the expression patterns of transcripts of bulls, steers and cows were identified. And we identified significantly differentially expressed lncRNAs in bulls, steers and cows. In addition, characteristics of lncRNA which express differentially in muscles according to the sex of Hanwoo were identified. As a result, we found differentially expressed lncRNAs according to sex were related to shear force and body weight. Conclusion: This study was classified and characterized lncRNA which differentially expressed by sex in Hanwoo cattle. We believe that the characterization of lncRNA by sex of Hanwoo will be helpful for future studies of the physiological mechanisms of Hanwoo cattle.
The relationships of scrotal circumference (SC) to semen characteristics and the conception rate (70 days-nonreturn rate) of artificial insemination in farm were studied with 137 heads of bull in Hanwoo. The average and range of SC were $38.27{\pm}3.90$ cm and 26.0~52.5 cm, respectively. Hanwoo bulls were classified with SC, divided into 34 cm below group, 34~39 cm group, and 39 cm over group. The 5,487 semen records of 43 heads of bull from July. 1. 2007 to June. 30. 2008. were used to determine the relationships between SC and semen characteristics. The semen concentration and total sperm number of each group were 11.18, 16.68, and $17.4{\times}10^8/ml$, and 69.83, 101.64 and $114.40{\times}10^8$/ejaculate. The bulls with 34 cm or more SC were higher than the bulls with 34 cm below in semen concentration and total sperm number (p<0.01). But between SC and semen volume have no significant relationship (p>0.05). The 9,862 mating records of 44 farm with 137 heads of bull were used to determine the relationships between SC and conception rate. The conception rate of 1st artificial insemination were 73.31, 74.16, and 77.33 % in each group. Also SC was positively correlated with pregnancy rate (r=0.12, p=0.17). These results indicate that SC correlates positively with semen characteristics, and maybe with pregnancy rate in Hanwoo.
This study was conducted to examine the effect of sustained release recombinant bovine somatotropin(SR-rBST) with different dosage(0, 0.03mg/kg body weight/14 day, 0.06mg/kg body weight/14 day, 0.09mg/kg body weight/14 day) on the economical traits of the semen characteristics and sexual behaviors and blood chemical values with sixteen Holstein dairy young bulls in Dairy Cattle Improvement Center of National Livestock Co-operatives Federation. Sensual testings of collected semens of Holstein young bulls were not different among treatments (p>0.05). Ejaculated semen volumes of control, SR-rBST 0.03mg, 0.06mg and 0.09mg groups were 5.6ml, 5.6ml, 6.0ml and 6.7m, respectively, but the result of SR-rBST 0.09mg group significantly increased semen volume(P<0.05). In sperm concentration, SR-rBST 0.06mg and 0.09mg groups significantly increased the total number of sperms than the other groups(P<0.05). In otherwise, SR-rBST treatments did not affect on pH, osmotic pressure, anti forzen rate, abnormality and motility of collected semen during whole experimental period. Reaction time(RT), sexual aggressiveness(SA) and tactile stimulation(TS) were not different among treatments. The libido scores(LS) of control, 0.03mg, 0.06mg and 0.09mg groups were 68.0, 80.5, 73.9 and 80.8, respectively, and LS were significantly improved by SR-rBST administration(P<0.05). The effect of SR-rBST on scrotal circumference measurement that is important factor to determine the ability of semen production, was not different among treatments. These studies indicates that SR-rBST treatments favourably affect on semen quality and sexual libido in Holstein young bull.
This study was conducted to determine the effect of breed on the chemical composition of first (PUR1) and second (PUR2) generations of $Purun{\tilde{a}}$ young bulls, and to compare both generations with different genetic groups: CAN vs. ANG; CAR; and CHA vs. CAR. Thirty bulls were used. The animals from the PUR2 and CHA vs. CAR genetic groups featured higher (p<0.05) moisture percentage in comparison to the PUR1, CAR and CAN vs. ANG groups. The moisture percentage was similar (p>0.05) between PUR2 and CHA vs. CAR animals. The same was observed among the PUR1, CAR, CAN vs. ANG and CHA vs. CAR animals. There was no difference (p>0.05) among genetic groups for ash, total lipids and total cholesterol. The PUR1, CAR and CAN vs. ANG specimens had higher (p<0.05) crude protein percentages as compared to PUR2 and CAN vs. CAR animals. The fatty acid profile was different (p<0.05) among genetic groups. However, the percentages of SFA, MUFA, PUFA, n-6, n-3, PUFA:SFA, and n-6:n-3 were similar (p>0.05) among genetic groups.
The objective of this study was to investigate the effect of rapid chilling (RC) on beef quality and the degradation of cytoskeletal proteins. Twenty Chinese Yellow crossbred bulls were selected and randomly divided into two groups. RC and conventional chilling (CC) were applied to left and right sides of the carcasses respectively after slaughtering. To determine whether electrical stimulation (ES) treatment can alleviate the potential hazard of RC on meat quality, ES was applied to one group. The effects of RC and ES were determined by meat color, shear force and cytoskeletal protein degradation postmortem (PM). The results showed that RC decreased beef tenderness at 1 d and 3 d postmortem, but had no detrimental effect on meat color. Western blotting showed that RC decreased the degradation rate of desmin and troponin-T, but the effects weakened gradually as postmortem aging extended. Degradation rates of both desmin and troponin-T were accelerated by ES. The combination of RC and ES could improve beef color, accelerate degradation rate of cytoskeletal protein and improve beef tenderness.

Objective: The aim of study was to investigate the effects of season and single layer centrifugation (SLC) before cryopreservation on post-thaw bull sperm quality in Thailand. Methods: Semen was collected from 6 bulls (Bos indicus) in summer, rainy season and winter 2014 through 2016. Semen characteristics, sperm morphology, sperm kinematics, viability, chromatin structure and mitochondrial membrane were evaluated. Meteorological data were available from the local meteorological station; Results: Season had an effect on semen characteristics in the raw ejaculate, with higher proportions of normal spermatozoa and lower abnormalities in winter than in the other two seasons. Sperm kinematics, viability, DNA fragmentation index, and mitochondrial membrane potential were not different between seasons. Sperm samples selected by SLC had greater normal morphology and a lower proportion with bent tails than controls and higher values of progressive motility (PRO), beat cross frequency, linearity, straightness, wobble (WOB), and lower values of slow motility, velocity average path (VAP), velocity curved line, and amplitude of lateral head displacement than controls. In addition, SLC-selection had a favorable effect on PRO, VAP, and WOB that differed among seasons. Conclusion: Our results suggested that these bulls were well adapted to their location, with season having an effect on sperm morphology. Moreover, SLC could be used prior to cryopreservation, regardless of season, to enhance normal morphology and kinematics of bull sperm samples without adversely affecting other parameters of sperm quality. However, there was considerable variation among bulls in DNA fragmentation index, mitochondrial membrane potential and sperm viability. In addition, SLC had a positive effect on sperm morphology and sperm kinematics, which could be expected to influence fertility.

Four ruminally cannulated Nili-ravi buffalo bulls were used in a $4{\times}4$ Latin Square design to determine the influence of varying levels of ruminally degradable protein (RDP) on ruminal characteristics, digestibility, blood pH, blood urea nitrogen (BUN) and nitrogen (N) balance. Four isonitrogenous and isocaloric diets were formulated (NRC, 2001). The control diet contained 50% RDP. The medium (MRDP), high (HRDP) and very high (VHRDP) ruminally degradable protein diets had 66, 82 and 100% RDP, respectively. Increasing the level of dietary RDP resulted in a linear decrease in ruminal pH. A quadratic effect of RDP on ruminal pH was also observed with quadratic maxima at the 66% RDP diet. Dietary RDP had a quadratic effect on total bacterial and protozoal count with maximum microbial count at the 82% RDP diet. Increased microbial count was due to increasing level of ruminal ammonia nitrogen ($NH_3-N$). Increasing dietary RDP resulted in a linear increase in dry matter digestibility. Provision of an adequate amount of RDP caused optimum microbial activity, which resulted in improvement in DM digestibility. Increasing the level of dietary RDP resulted in a linear decrease in crude protein (CP) and neutral detergent fiber digestibility. Blood pH remained unaltered across all diets. A linear increase in ruminal $NH_3-N$ and BUN was noted with increasing level of dietary RDP. The increase in BUN was due to increased ruminal $NH_3-N$ concentrations. A positive N balance was noted across all diets. The results are interpreted to suggest that buffalo bulls can utilize up to 82% RDP of total CP (16%) with optimum results.
