한우에서 BVDV 지속감염우의 정액 성상에 관한 연구 (Semen Properties of a Hanwoo bull persistently infected by BVDV)

  • 소에 있어서 BVDV는 반추류에서 중대한 감염을 야기하여 번식장애, 설사 및 유량감소를 야기하여 농가에 막대한 피해를 야기하고 있다. 특히 지속감염우의 확인과 도태는 농가 내 감염원을 제거하는 중요한 일이나 아직까지 PI개체의 바이러스의 감염과 전파에 관한 연구는 미진한 것으로 추정된다. 본 연구에서는 도입된 수컷에서 PI를 확인하였으며 전혈 검사를 실시 하면, PI 혈액 성상에서 림프구가 낮게 관찰되며 전체 WBC의 수는 정상범위에 속하나 낮게 나타나는 것을 알 수 있었다. PI 수컷에서 생산된 정액은 정자 수가 매우 낮으며, 신선 정액의 생존성도 불량한 것으로 조사되었다. 정자 기형율 또한 증가되었으며 특히 공포와 중편부의 소적을 가진 기형정자의 비율이 높았다. PI수컷은 나이가 들수록 정액 성상이 불량하여 불임화되었고 이는 BVDV가 정소의 정자 생산능력을 낮추어 사출된 정액내 정자수 감소 현상을 야기한다는 것을 알 수 있었다. 그럼에도 불구하고 확인된 PI 개체에 의한 전체군의 감염 현상은 관찰할 수 없었는데, 이는 적절한 백신 프로그램에 의하여 PI에 의한 간접적인 전파의 위험성이 낮다고 판단된다.

한우 종모우에 있어 정액 채취시 의빈대 혹은 의빈우 이용이 정액성상에 미치는 영향 (Effects of Dummy Cow or Teaser Bull for Semen Collection on Semen Characteristics in Hanwoo Proven Bull)

  • 한우 종모우에 있어 정액 채취시 승가 조건(의빈우 혹은 의빈대)이 정액성상에 미치는 영향을 구명하기 위하여 의빈우 승가 종모우 21두, 의빈대승가 종모우 9두 등 보증 종모우 30두를 공시하여 연구하였다. 정액은 $3{\sim}6$일 간격으로 채취하였으며 채취 당일 1시간 간격으로 총 2회 정액 채취하여 6개월간 정액성상을 조사하였다. 의빈대 승가 종모우가 의빈우 승가 종모우보다 1차 및 2차 정자농도에 있어 유의적으로 높아(p<0.05) 1차 및 2차 총 정자수에서도 많았으며 특히 l차 총 정자수는 유의적으로 많게 나타났다(p<0.05). 종모우 승 가 조건(의빈대, 의빈우)에 상관없이 5일 간격으로 정액 채취시가 정액량이 많아(p<0.05) 총 정자수가 많았으며 의빈대를 이용하여 5일 간격으로 정액 채취시가 총 정자수가 다른 모든 처리군보다 많았다. 의빈대 승가 종모우는 $4{\sim}5$세에서 총 정자 수가 높게 나타났으며 의빈우 승가 종모우는 $6{\sim}7$세에서 총 정자수가 높게 나타났다(p<0.05). 이러한 결과로 미루어 볼 때 한우 종모우에서 의빈대를 이용하여 정액을 채취하는 것이 의빈우를 이용 할 때 보다 정액 생산성이 좋을 것으로 사료된다.

Evaluation of genotype by environment interactions on milk production traits of Holstein cows in southern Brazil

  • Objective: This study assessed the possible existence of genotype by environment interactions for milk, fat and protein yields in Holstein cattle raised in one of the most important milk production basins in Brazil. Methods: Changes in the genetic parameters and breeding values were evaluated for 57,967 animals from three distinct regions of southern Brazil, divided according to differences in climate. The genotype by environment interaction was determined by genetic correlations between regions, estimated by the restricted maximum likelihood, considering the animal model. Bull rankings were investigated to verify the ratio of coincident selected animals between regions for each trait. Results: The estimates of heritability coefficients were similar between two regions, but were lower in the third evaluated area, for all traits. Genetic correlations between regions were high, ranging from 0.91 to 0.99 for milk, fat and protein yields, representing the absence of a genotype by environment interaction for productive traits. The percentage of selection error between regions for the top 10% of animals ranged from 0.88% to 2.07% for milk yield, 0.99% to 2.46% for fat yield and 0.59% to 3.15% for protein yield. Conclusion: A slight change in genotype between areas was expected since no significant genotype by environment interactions were identified, facilitating the process of selecting Holstein cattle in southern Brazil.

Genomic selection through single-step genomic best linear unbiased prediction improves the accuracy of evaluation in Hanwoo cattle

  • Objective: Genomic selection (GS) is becoming popular in animals' genetic development. We, therefore, investigated the single-step genomic best linear unbiased prediction (ssGBLUP) as tool for GS, and compared its efficacy with the traditional pedigree BLUP (pedBLUP) method. Methods: A total of 9,952 males born between 1997 and 2018 under Hanwoo proven-bull selection program was studied. We analyzed body weight at 12 months and carcass weight (kg), backfat thickness, eye muscle area, and marbling score traits. About 7,387 bulls were genotyped using Illumina 50K BeadChip Arrays. Multiple-trait animal model analyses were performed using BLUPF90 software programs. Breeding value accuracy was calculated using two methods: i) Pearson's correlation of genomic estimated breeding value (GEBV) with EBV of all animals (rM1) and ii) correlation using inverse of coefficient matrix from the mixed-model equations (rM2). Then, we compared these accuracies by overall population, info-type (PHEN, phenotyped-only; GEN, genotyped-only; and PH+GEN, phenotyped and genotyped), and bull-types (YBULL, young male calves; CBULL, young candidate bulls; and PBULL, proven bulls). Results: The rM1 estimates in the study were between 0.90 and 0.96 among five traits. The rM1 estimates varied slightly by population and info-type, but noticeably by bull-type for traits. Generally average rM2 estimates were much smaller than rM1 (pedBLUP, 0.40 to0.44; ssGBLUP, 0.41 to 0.45) at population level. However, rM2 from both BLUP models varied noticeably across info-types and bull-types. The ssGBLUP estimates of rM2 in PHEN, GEN, and PH+ GEN ranged between 0.51 and 0.63, 0.66 and 0.70, and 0.68 and 0.73, respectively. In YBULL, CBULL, and PBULL, the rM2 estimates ranged between 0.54 and 0.57, 0.55 and 0.62, and 0.70 and 0.74, respectively. The pedBLUP based rM2 estimates were also relatively lower than ssGBLUP estimates. At the population level, we found an increase in accuracy by 2.0% to 4.5% among traits. Traits in PHEN were least influenced by ssGBLUP (0% to 2.0%), whereas the highest positive changes were in GEN (8.1% to 10.7%). PH+GEN also showed 6.5% to 8.5% increase in accuracy by ssGBLUP. However, the highest improvements were found in bull-types (YBULL, 21% to 35.7%; CBULL, 3.3% to 9.3%; PBULL, 2.8% to 6.1%). Conclusion: A noticeable improvement by ssGBLUP was observed in this study. Findings of differential responses to ssGBLUP by various bulls could assist in better selection decision making as well. We, therefore, suggest that ssGBLUP could be used for GS in Hanwoo proven-bull evaluation program.

Selection of Sahiwal Cattle Bulls on Pedigree and Progeny

  • The objective of the study was to compare ranking of Sahiwal bulls selected on the basis of highest lactation milk yield of their dams with their estimated breeding values (EBVs) using an animal model. Data on 23,761 lactation milk yield records of 5,936 cows from five main Livestock Experiment Stations in Punjab province of Pakistan (1964-2004) were used for the study. At present the young A.I bulls are required to be from A-category bull-dams. Dams were categorized as A, B, C and D if they had highest lactation milk yield of ${\geq}$2,700, 2,250-2,699, 1,800-2,249 and <1,800 litres, respectively. The EBVs for lactation milk yield were estimated for all the animals using an individual animal model having fixed effect of herd-year and season of calving and random effect of animal. Fixed effect of parity and random effect of permanent environment were incorporated when multiple lactation were used. There were 396 young bulls used for semen collection and A.I during 1973-2004. However, progeny with lactation yields recorded, were available only for 91 bulls and dams could be traced for only 63 bulls. Overall lactation milk yield averaged 1,440.8 kg. Milk yield was 10% heritable with repeatability of 39%. Ranking bulls on highest lactation milk yield of their dams, the in-vogue criteria of selecting bulls, had a rank correlation of 0.167 (p<0.190) with ranking based on EBVs from animal model analysis. Bulls' EBVs for all lactations had rank correlation of 0.716 (p<0.001) with EBVs based on first lactation milk yield and 0.766 (p<0.001) with average EBVs of dam and sire (pedigree index). Ranking of bulls on highest lactation yield of their dams has no association with their ranking based on animal model evaluation. Young Sahiwal bulls should be selected on the basis of pedigree index instead of highest lactation yield of dams. This can help improve the genetic potential of the breed accruing to conservation and development efforts.

소 동결-융해 정자에 있어서 체외수정능력과 정자 기능 및 성상 분석법간의 상관관계 (Correlations between the Capacity of In Vitro Fertilization and the Assays of Sperm Function and Characteristics in Frozen-thawed Bovine Spermatozoa)

  • 본 연구는 종모우의 정자수정능력 평가방법의 개발과 정자 기능 및 성상 변화에 영향을 미치는 요인을 알아보기 위하여 시행하였다. 동결-융해된 종모우 정액을 대상으로 정자의 운동성과 정자의 형태를 분석하였고, 정자의 기능 검 사 항목으로서 체외수정(IVF), HOST, Ca-ionophore에 의한 첨체반응율, 정자의 ROS 측정을 위한 luminol, lucigenin-dependent chemiluminescence, LPO 분석을 위한 malondialdehyde의 측정 및 TUNEL (terminal deoxynucleotidyl transferase(TdT) dUTP nick end labelling) 기법을 이용한 정자의 DNA fragmentation를 측정하였으며 이들 각각의 조사 항목들의 분석치들과 체외수정율 및 배발생율과의 상관관계를 조사하여 다음과 같은 결과를 얻었다. 1. 고수정군과 저수정군의 체외수정율과 배반포 발생율의 평균은 각각 64.4%와 34.3%, 18.50%와 6.2%였으며 두 군간에 통계학적으로 유의한 차이를 보였다(P<0.05). 고수정군과 저수정군의 정자운동성과 첨체반응률은 각각 평균79.0 %와 66.2%, 40.7%와 22.9%로 두 군간에 통계학적으로 유의한 차이를 보였으나(P<0.05), 정상형태 정자의 비율과 HOST는 각각 평균 94.6%와 92.7%, 69.4%와 59.8%로 두 군간 유의한 차이를 보이지 않았다. 2. Luminol dependent chemiluminescence, LPO 및 DNA fragmentation의 평균은 고수정군과 저수정군에 있어서 각각 6.4와 6.5, 2.Onmol와 3.Inmol 및 2.6%와 7.4%로 두 군간 통계학적으로 유의한 차이를 보였으나(P<0.05), lucigenin dependent chemiluminescence는 4.7와 4.6로 두 군간 유의한 차이를 보이지 않았다. 3. 체외 수정율은 정자의 운동성 및 첨체반응율과 통계학적으로 유의한 정(positive)의 상관관계(r=0.87, p<0.01; r=0.81, p<0.05)를 나타내었으며, luminol dependent chemiluminescence, lipid peroxldation 및 DNA fragmentation과는 통계학적으로 유의한 부(negative)의 상관관계 (r= -0.81, p<0.05; r: -0.74, p<0.05; r : 0.81, p<0.05)를 나타내었다. 그러나 체외수정율은 정상형태 정자의 비율, HOST 및 lucigenin dependent chemiluminescence와는 유의한 상관 관계를 나타내지 않았다. 4. 배반포 발생율은 첨체반응율과 통계학적으로 유의한 정의 상관관계(r=0.71, p<0.05)를 나타내었으며, luminol dependent chemiluminescence, lipid peroxidation 및 DNA fragmentation과는 통계학적으로 유의한 부의 상관관계(r= -0.71, p<0.05; r= -0.89, p<0.01; r= -0.71, P<0.05)를 나타내었다. 배반포 발생율은 정자의 운동성, 정상형태 정자의 비율 및 HOST, lucigenin dependent chemilumihescence와는 유의한 상관관계를 나타내지 않았다. 이상의 결과를 종합해 보면 정액질의 저하에 ROS의 영향이 밀접히 연관되어 있음을 알 수 있으며, 또한 본 연구에서 적용된 기법들은 정액질의 평가 및 정자 수정능력 향상을 위한 기술개발에 있어서 유용한 평가 방법으로 이용될 수 있을 것으로 사료된다.

햄스터 적출자궁에서 전배양한 소 정자의 난포란에의 침입시기 및 체외수정 초기상에 관한 연구 (Timing of Fertilization In Vitro of Follicular Oocytes by Bull Spermatozoa Preincubated in the Uteri Isolated fom Estrous Hamsters)

  • The cattle follicular oocytes matured for 26~28h in culture condition were examined at 4, 5, 6, 7, 8, 10, 12, 14, 16 and 18h after insemination with bull spermatozoa preincubated for 4.5h in the uter isolated from estrous hamsters. After further culture with spermatozoa for 4~18 h, 73~89% of the total oocytes had matured to the second metaphase. None of the follicular oocytes matured in culture, were fertilized 5h after insemination. But when the oocytes were examined at 6, 8, 10, 14 and 18h after insemination, 60, 73, 82, 80 and 87% of oocytes were fertilized, respectively. The majority of the fertilized oocytes had enlarged sperm head at 6h after insemination and a part of the fertilized oocytes begun to develop from enlarged sperm head to male pronuclear stage at 8h after insemination, and most of them developed to male and female pronuclear stage at 10h after insemination. The results suggest that the penetration of spermatozoa into the oocytes may occur earlier than 6h after insemination and development of their pronuclear stage may occur at 8h after insemination.

비발정 재귀율을 이용한 한우 보증씨수소 수정능력의 상대적 추정치 분석 (Analysis of Estimation of Relative Conception Rate on Korean Proven Bull Number Semen using Non-return Rate)

  • The decreased fertility is frequently thought to be problem of cattle production. However, studies figure out that number of these problems is related to bull factors especially in artificial insemination setting. Therefore, this study was designed to investigate the fertility status of bull by their estimated relative conception rate of cows that were inseminated by frozen semen from Korean proven bulls. Here we use the non-return rate (NRR) to access the bull fertility whereas, the NRR was define as the proportion of bulls that semen were used to inseminate cows and the number of cows that did not return for another service within 60 days. The data from 54,388 artificial inseminations (AI) were analyzed from 88 KPN semen. The NRRs of highest and lowest fertile bull were 83.81 and 51.33%, respectively. And mean NRR was 68.27%. 점토 광물질의 급여가 비거세 우육의 저온저장 중 품질에 미치는 영향 (Effect of Dietary Clay Mineral on Meat Quality of Hanwoo (Korean Cattle) Bull Beef during Refrigerated Storage)

  • 점토 광물질을 비거세 한우에 1.25%첨가 급여시 육질에 미치는 영향을 조사하였다. 실험처리구는 대조구(무첨가구)와 1.25% 점토 광물질 급여구(CT-1.25%)로 나누어 실험하였다. 일반성분과 지방산 조성은 처리구간의 유의적인 차이를 나타내지 않았으며, pH는 저장기간에 따라 유의적으로 변화하는 대조구와 달리 1.25% 점토 광물질 급여구의 PH는 저장 기간에 따른 유의적인 차이를 보이지 않고 안정적이었다. CIE a*값, chroma값 및 R630-R580값은 모든 처리구에서 저장기간이 경과함에 따라 유의적으로 감소하는 경향을 나타내었으며(p<0.05), 특히 대조구가 저장기간이 경과함에 따라 현저한 감소 현상을 보였다. Metmyoglobin(%)의 함량은 저장기간이 경과함으로써 대조구의 menyoglobin 형성율이 현저하게 증가하여 1.25%점토 광물질 급여구에 비해 갈색 축적현상이 급속하게 진행됨을 알 수 있었다. 지질의 산패 정도를 나타내는 TBARS는 1.25% 점토 광물질 급여구가 대조구보다 유의적으로 낮은 값을 나타내어 산화 안정성을 보였다. 보수력은 모든 처리구에서 저장기간이 경과함에 따라 유의적으로 증가하는 경향을 나타내었으며(p<0.05) 1.25% 점토 광물질 급여구가 대조구에 비해 높은 보수력을 보였다. 결과적으로 1.25% 점토 광물질 급여구(CT-1.25%)의 경우 대조구에 비해 육색, 지질 산화 및 보수력에 대해 안정성을 가지는 것으로 나타났다.

The Effects of Dietary Sulfur and Vitamin E Supplementation on the Quality of Beef from the Longissimus Muscle of Hanwoo Bulls

  • This study was carried out to investigate the effects of dietary sulfur (S) and vitamin E (Vit E) supplementation on the quality of beef from longissimus muscle of Hanwoo bulls. Eleven, 29 months-aged Hanwoo bulls were randomly divided into three feed supplementation groups (S (n = 4), Vit E (n = 4) and S+Vit E (n = 3)). S was given as much as 12 g/head/d and Vit E was given as much as 1,200 IU/head/d; both supplements were given for 3 months prior to slaughter. At 24 h post-slaughter, the carcasses were weighed and evaluated by official grader for carcass traits. At 48 h post-slaughter, the M. longissimus from each carcass was collected and stored at $4{\pm}0.2^{\circ}C$ for 10 days. There were no significant differences in yield and quality grades of carcass and proximate composition, physical properties and aroma pattern of meat among all groups. At 5 days of storage, the TBARS value of meat from cattle with S+Vit E supplementation was significantly lower (p<0.05) than other groups; and at 10 days of storage, the TBARS value of meat from cattle with Vit E and S+Vit E supplementations was significantly lower (p<0.05) than from cattle with S supplementation. At 5 days of storage, MetMb concentration of meat from cattle with S+Vit E supplementation was significantly lower (p<0.05) than from cattle with Vit E supplementation; and at 10 days of storage, MetMb concentration of meat from cattle with S+Vit E supplementation was significantly lower (p<0.05) than from other groups. At 10 days of storage, the redness value of meat from cattle with S supplementation was significantly higher (p<0.05) than from cattle with Vit E supplementation and the hue-angle value of meat from cattle with S and S+Vit E supplementations was significantly lower (p<0.05) than from cattle with Vit E supplementation. Dietary Vit E supplementation had a better effect on lipid stability whereas dietary S supplementation had a better effect on OxyMb stability. The dietary combination of S and Vit E created the highest protection for beef from myoglobin oxidation and thus improved the color stability of meat.
