Research for Deposition of $CeO_2$ Buffer Layer on Coated Conductor by Electron Beam Evaporation (전자빔 증발법에 의한 박막형 고온초전도체의 $CeO_2$ 버퍼층 증착 연구)

  • The properties of buffer layer for thermal and chemical stability in coated conductor is a very important issue. $CeO_2$ has desirable thermal and chemical stability as well as good lattice match. In this study, $CeO_2$ was deposited by electron beam deposition. The MgO(001) single crystal and LMO buffered IBAD substrate(LMO/IBAD-MgO/$Y_2O_3/Al_2O_3$/Hastelloy) were used as substrates, which have $\Delta\phi$ values of ${\sim}8.9^{\circ}$. The epitaxial $CeO_2$ films was deposited with high deposition rate of $12{\sim}16\;{\AA}/sec$. During deposition, the change of oxygen partial pressure(${\rho}O_2$) does not cause change in c-axis texture. In case of $CeO_2$ on MgO single crystal, the substrate temperature was optimized at $750^{\circ}C$ with superior $\Delta\phi$ and $\Delta\omega$ value. Otherwise, In case of LMO buffered IBAD substrate, It was optimized at $650^{\circ}C$ with increasing its deposition thickness of $CeO_2$, which was finally obtained with best $\Delta\phi$ value of $5.5^{\circ}$, $\Delta\omega$ value of $2^{\circ}$ and Ra value of 2.2 nm.

  • Recently, the occurrence of typhoons and heavy rainfall is increasing due to climate change. This causes increase in possibility of landslide damages in rural areas. However, in reality, the precise engineering stability assessment studies are still insufficient. Therefore, in order to reduce the landslide damages and effectively manage mountainous areas, the development of disaster prevention techniques is needed. In this study, to analyze the shock absorbing effect of the buffer-spring during application of dynamic impact load in the debris flow protection system, numerical analysis is carried out for each free field of the buffer-spring and the load sharing ratio of the buffer-spring is also examined. In addition, the field applicability is verified by comparison of the tensile strength of the conventional buffer-spring and the wedge type buffer-spring on various magnitudes of dynamic impact load. As a result of the study, it is found that the net-type debris protection system is effective to mitigate loss of properties and human lifes during landslide.

  • The optimal pH of the extraction buffer was 7.5 considiering AChE stability and its buffer capacity when AChE was isolated and extracted from the housefly(Musca domesitca L.)and three other insect species with 0.01 M sodium phosphate buffer. Also, the optimal pH of the reaction buffer was 7.5 considering enzyme activity and its buffer capacity when AChE activity was measured with the substrate in 0.1 M sodium phosphate buffer. The Potter Elvehjem type homogenizer with Teflon pestle was used to homgenize the tissues. When preparing a AChE suspension by centrifuging the homogenate, 700 g supernatant of adult head for the housefly, 700 g supernatnat of 5th instar nymphal whole body for the brown planthopper, lipid-eliminated 10,000 g supernatant of 5th instar larval whole body for the diamondback moth, and 700 g supernatant of 4th instar larval head for the tobacco cutworm were considered satisfactory as enzyme sources in view of mass preparation, extraction efficiency and stability of enzyme activity during evaluation. When AChE suspensions of 4 insect species were stored at $-18^{\circ}C$, more than 90% of activity was maintained up to 3 weeks. Km values of AChEs of the housefly, the brown planthopper, and the diamondback moth were 0.042, 0.037 and 0.043 mM, respectively and AChE-specific substrate inhibition was observed at high concentration. Km value of the tobacco cutworm ChE was 1.15 mM and BuChE characteristics was observed, though further study is needed. The optimal substrate concentration for the AChE inhibition tests was 0.5 mM for the housfly, the brown planthopper, and the diamondback moth and 12 mM for the tobacco cutworm.

A Numerical Analysis to Estimate Disposal Spacing and Rock Mass Condition for High Efficiency Repository Based on Temperature Criteria of Bentonite Buffer (벤토나이트 완충재 설계 기준 온도에 따른 고효율 처분시스템 처분 간격 및 암반 조건 산정을 위한 수치해석적 연구)

  • This study conducts coupled thermo-hydro-mechanical numerical modeling to investigate the maximum temperature and conditions for securing mechanical stability of the high-level radioactive waste repository when temperature criteria of bentonite buffer are 100℃ and 125℃, respectively. In case of temperature criterion of buffer as 100℃, the maximum temperatures at the interface between canister and buffer are calculated to be 99.4℃ and 99.8℃, respectively for a case with disposal tunnel spacing of 40 m and deposition hole spacing of 5.5 m and for the other case with disposal tunnel spacing of 30 m and deposition hole spacing of 6.5 m. In case of temperature criterion of buffer as 125℃, spacings of disposal tunnel and deposition hole could be decreased to 30 m and 4.5 m, respectively, which reduces the disposal area up to 55% compared to the disposal area of KRS+. According to analysis of mechanical stability for various disposal spacings, RMR of rock mass for KRS+ should be larger than 72.4 which belongs to good rock in RMR classification to prevent failure of rock mass. As disposal spacing is decreased, required RMR of rock mass is increased. In order to prevent failure of rock mass for a case with disposal tunnel spacing of 30 m and deposition hole spacing of 4.5 m, RMR larger than 87.3 is needed. However, mechanical stability of the repository is secured for all cases with RMR over 75 considering the enhancement of rock strength due to confining stress induced by swelling of the bentonite buffer and backfill.

The Adhesion Strength and Interface Chemical Reaction of Cu/Ni/Polyimide System (Cu/Ni/Polyimide 시스템의 접착력 및 계면화학반응)

  • The magnetron sputtering was used to deposit Ni buffer layers on the polyimide surfaces to increase the adhesion strength between Cu thin films and polyimide as well as to prevent Cu diffusion into the polyimide. The Ni layer thickness was varied from 100 to $400{\AA}$. The adhesion strength increased rather significantly up to $200{\AA}$ of Ni thickness, however, there was no significant increase in strength over $200{\AA}$. The XPS analysis revealed that Ni thin films could increase the adhesion strength by reacting with the polar C=O bonds on the polyimide surface and also it could prevent Cu diffusion into the polyimide. The Cu/Ni/ polyimide multilayer thin films showed a high stability even at the high heating temperature of $200^{\circ}C$, however, at the temperature of $300^{\circ}C$, Cu diffused through the Ni buffer layer into polyimide, resulting in the drastic decrease in adhesion strength.

Effect of Trehalose on Biological Membranes with Respect to Phase of the Membranes

  • The effect of the trehalose incorporation on the biological membranes was investigated with respect to the phase of the membranes using the fluorescence intensity change. Spherical phospholipid bilayers, vesicles, were prepared only with the variation in the phase of each layer via a double emulsion technique. In the aqueous inside of the vesicles, 8-Aminonaphthalene-1,3,6-trisulfonic acid disodium salt(ANTS) was encapsulated. As a quencher, p-Xylene-bis(N-pyridinium bromide)(DPX) was included in the buffer where the vesicles were dispersed. The fluorescence scale was calibrated with the fluorescence of ANTS vesicles in p-Xylene-bis(N-pyridinium bromide)(DPX)-included-buffer taken as 100% fluorescence and the mixture of ANTS and DPX in the buffer as 0% fluorescence. Trehalose injection into the vesicle solution led the distortion of the membrane. It was found that the distortion was related to the phase of each layer the vesicle up on the ratio of trehalose to lipid. In the identical measurements at glucose, the behavior of the distortion was completely different from that of trehalose. These results seem to depend on the stability of the vesicles, due to the osmotic and volumetric effects on the headgroup packing disruption.

Stability and drug release properties of liposomes containing cytarabine as a drug carrier

  • Liposomes were studied as a drug delivery system. Multilamellar vesicles, small unilamellar vesicles and large unilamellar vesicles containing cytarabine were prepared using egg yolk lecithin and cholesterol. Large unilamellar vesicles showed the highest encapsulation efficiency of all and their encapsulation efficiency increased as the buffer volume decreased. Cholesterol increased the stability of liposomal drug products as drug carriers and reduced the permeability of drug across the liposomal membrane. The release rate of cytarabine increased with incubation temperature and decreased with cholesterol incorporation in liposomal membrane. The release mechanism of cytarabine from large unilamellar vesicles in vitro was chiefly due to simple diffusion across the liposomal membrane rather than liposomal rupture.

Synthesis of Size Controllable Silk Fibroin Microparticles and Their Stability on Different Solutions

  • Silk fibroin microparticles were fabricated using a phase separation technique between silk fibroin solution and polyvinyl alcohol. We found that the concentration of polyvinyl alcohol determines the size of microparticles. The mean diameter of the silk fibroin microparticles varied from 3.48 ㎛ to 4.05 ㎛. The silk fibroin microparticle size increased as a function of the concentration of PVA in aqueous silk solution. The resulting silk fibroin microparticles have narrow size distribution (i.e. monodisperse) and smooth/spherical surface. Also, we studied the effects of mouse serum, sodium phosphate buffer (PBS), and pH on the stability of the silk fibroin microparticles. Overall, we demonstrated the simple method to fabricate and to control the silk fibroin microparticles that makes our silk microparticles to be usable for a potential drug delivery carrier.

Development of antigen for the microplate latex agglutination test on toxoplasmosis in animals (Latex 응집반응을 이용한 동물의 톡소플라즈마병 진단액 개발에 관한 연구)

  • This study was conducted to develop a sensitized latex-antigen for serodiagnosis of toxoplasmosis in animals. Tachyzoites of T gondii(RH-strain) harvested from mouse peritoneal cavity were purified through the filtraton of polycarbonate membrane(pore size, $3.0{{\mu}m}$, Costar Co.) and disrupted by ultrasonicator. The tachyzoite suspension was ultracentrifuged for 30 min at $60,000{\times}g(4{^{\circ}C})$ and the supernatant was used as a water-lysate antigen. Polystyrene latex particles of $0.8{{\mu}m}$ in diameter(Sigma) were used for the preparation of sensitized latex-antigen suspension. The several parameters including the preparation conditions, incubation buffer. serum dilution buffer and stability of agglutination reactions were evaluated and the results obtained were summarized as follows : 1. The antigen consisting of a water-lysate of T gondii tachyzoites was adsorbed onto polystyrene latex particles of $0.8{{\mu}m}$ in diameter by adding a latex suspension to an equal volume of diluted antigen solution and by incubating the mixture at $37{^{\circ}C}$ under different conditions. 2. The optimum incubation buffer used for the antigen sensitization was 0.1M Tris-HCl buffer(pH 8.0). 3. The optimum serum dilution buffer used for the latex agglutination test was 0.1M Tris-HCl-NaCl buffer(pH 7.4) containing 300 mM NaCl. But 0.1M Tris-HCl-NaCl buffer(pH 7.4) containing 300-600 mM NaCl, 0.5% BSA and 0.01% Tween-20 improved the agglutination pattems and cleared the background of microplate well without the effects on L.A titer. 4. The time required for antigen sensitization was 40 and 60 min in incubation buffer(pH 8.0) at $37{^{\circ}C}$. But the optimun time for antigen sensitization was min at $37{^{\circ}C}$. 5. The optimun quantity of antigen absorbed on latex particles for proper agglutination was the range of 20 to $32{\mu}g$ of latex particles. 6. The optimun concentration of the latex-antigen suspension for the proper agglutination reaction was determined as 0.2%(w/v). 7. The specificity, rapidity and simplicity of the latex-particle agglutination test suggested that it might be adaptable to large scale serum screening.

A Hybrid Transmission Scheme for Efficient Video Streaming (네트워크 및 버퍼 상태를 모두 고려한 혼합형태의 비디오 스트림 전송기법)

  • Existing streaming mechanisms have no consideration for the characteristics of streaming applications because they only consider network stability. In this paper, in order to overcome limitations of the previous work on video streaming, we propose a new video streaming mechanism called "HAViS(Hybrid Approach for Video Streaming)". The proposed mechanism includes more sophisticated features that consider both network and user requirements. Therefore, the HAViS mechanism improves the network stability by adjusting the sending rate of video stream based on the network state and it also provides the smooth playback by preventing the buffer underflow or overflow. Moreover, it is designed to take into consideration the streaming video content. Through the simulation, we prove that the HAViS mechanism efficiently uses the buffer resources and provides improved network stability and smooth playback.
