Changes of Glycosylation Pattern in Aging Rat Kidneys as Revealed with Lectin Conjugates (성장과정 중 흰쥐 신장의 복합당질 변화에 대한 연구)

  • The changes of glycoconjuagates (GCs) in rat kidney due to maturation were studied from samples of fetal and postnatal kidneys by lectin histochemistry. Rat kidneys of perinatal ages and adults were fixed in 4% paraformaldehyde and were stained with nine kinds of biotinylated lectins. The immature forms of the renal developmental stage such as vesicles and ureteric bud were observed in the cortex as late as day 14 of postnatal life, but the histological appearance of the weaning kidney was similar to that observed in adults. As for histochemical properties of GCs in the glomeruli, Con A affinity tended to increase with aging, but both RCA-1 and LCA affinities showed a transient increase in immature glomeruli of neonatal rats. DBA affinity with SBA, PNA, BSL-1 and RCA-1, additional Con A one in proximal tubule, were increased in both proximal and distal tubules according to maturation. In contrast to this, transient intensive LCA affinity were demonstrated in immature proximal and distal tubule of neonatal rats. In the collecting tubules, DBA, SBA, PNA and sWGA affinities tended to increase according to maturation, but transient increase for BSL-1, RCA-1 and LCA affinities were detected in neonatal rats. The present results suggest that the mature glycosylation pattern of the kidney undergoes profound changes during maturation and is probably associated with functional maturation of the kidney.

Breeding of 'Daewang' Strawberry for Forcing Culture with Good Taste and Fragrance (맛과 향이 좋은 촉성재배용 딸기 '대왕' 품종의 육성)

  • A new cultivar 'Daewang' strawberry was developed by the National Institute of Horticultural & Herbal Science for forcing culture in 2010. The cultivar 'Daewang' was originated from the cross between 'Maehyang', a high firmness cultivar and 'Wongyo 3111', a high sugar content in 2006. The cultivar shows erect plant type, vigorous growth habit, early flower bud differentiation with 12-15 flowers per cluster from planting on healthy nursery. Fruits of 'Daewang' are conical type having a bright red skin color, and 16-17 g in an average weight. 'Daewang' is suitable for forcing culture as time required for dormancy breaking ranged between 50 and 100 hours. This cultivar has excellent taste for high sugar/acid ratio as sugar content of $11.1^{\circ}Brix$, acidity of 0.39% with abundant texture and can be harvested by late spring because the fruit firmness of 'Daewang' was $18.2g{\cdot}mm^{-2}$ that was about $7.9g{\cdot}mm^{-2}$ higher than $10.3g{\cdot}mm^{-2}$ of 'Akihime' cultivar. But although total yield is not significantly different from 'Akihime' cultivar, its marketable yield is remarkably higher than that of 'Akihime' cultivar. Disease and pest resistance of 'Daewang' have a tendency to sensitive powdery mildew, anthracnose and spotted spider.

  • The embryos of Pinus taeda, P. rigida, and P. taeda ${\times}$ rigida were cultured for adventitious shoot regeneration in vitro. Culture media were modified from Gresshoff and Doy (MGD), Murashige and Skoog (MMS), Lloyd and McCown (MLM), and Schenk and Hildebrandt (MSH). NAA was added to initiation media at a concentration of 0.1 or 0.01 mg/l. BAP was used at the concentrations of 0.1. 0.5, 1, 2, or 5mg/l. Each explant was induced for 3-4 weeks on solid medium. All explants were cultured up to 16 weeks. Illumination was about $1506{\pm}540lux$ at the level of the tissues in the growth room with a temperature of $25{\pm}2^{\circ}C$. A 16-hour photoperiod per 24 hours was used. Half-strength medium was used for all the subcultures. For shoot production by loblolly pine, MMS, MLM, or MSH is preferred with 5 mg/l BAP with either 0.1 or 0.01 mg/l NAA. For shoot production by pitch pine, MMS, MLM, or MSH is recommended with 2 or 5 mg/l BAP with 0.1 mg/l NAA. For shoot production by the hybrid pine, MMS or MLM is more effective with 1, 2 or 5 mg/l BAP with 0.1 mg/l NAA. There were no differences recognized among the species tried in the patterns of bud formation and shoot development. Different composition of media, in major and minor salts or possibly in vitamins, should be tested for the two developmental stages of adventitious shoots ; the induction of shoot buds and the elongation of them into shoots.

Breeding of Self-compatible Pear "Wonkyo Na-jasoojung 2" (배 자가결실성 "원교 나-자수정 2호")

  • Pear has a gametophytic self-incompatibility (SI) system and its SI reaction is controlled by a single multi-allelic S-locus. 'Wonkyo Na-jasoojung 2' was selected from a cross between 'Wonwhang', early season major pear cultivar with high fruit quality and self-incompatible, and 92-18-79 (${S_4}^{sm}{S_4}^{sm}$) obtained from self cross of 'Osa-nijisseiki' (${S_2S_4}^{sm}$) (SM, stylar-part mutant), self-compatible bud mutant that originated from self-incompatible 'Nijisseiki' ($S_2S_4$) made in 2001 at the National Institute of Horticultural and Herbal Science, Rural Development Administration in Korea. '92-18-79' was selected as a self-compatible source through field investigation. It bloomed 1 day earlier than 'Osa-Nijisseiki' and similar to 'Wonwhang' in 2008. It is medium in tree vigor and spreading in tree habit. 'Wonkyo Na-jasoojung 2' is classified as highly susceptible to pear scab (Venturia nashicola) similar to 'Osa-Nijisseiki' and as resistant to black spot (Alternaria kikuchiana) similar to 'Wonwhang'. It had 65.7% fruiting rate by self pollination. The average optimum harvest time of 'Wonkyo Na-jasoojung 2' was 148 days after full bloom and it matured 2 days earlier than 'Osa-Nijisseiki' and 11 days later than 'Wonwhang'. The fruit is roundish oblate in shape and yellowish brown in skin color. Average fruit weight was 445 g and soluble solids content was 13.3 $^{\circ}Brix$. The flesh had abundant juice and negligible grit.

Plant regeneration through multiple-shoot induction and ex vitro rooting in Vaccinium oldhamii Miq. (정금나무(Vaccinium oldhamii Miq.)의 다신초 유도 및 기외발근을 통한 식물체 재분화)

  • In vitro techniques were developed for propagating Vaccinium oldhamii using shoots with apical buds. Explants having an apical bud were cultured on Murashige and Skoog (MS) medium supplemented with 1.0, 2.0, and 5.0 mg/L of each zeatin, thidiazuron, 6-benzylaminopurine (BA), and 6-(γ,γ-dimethylallylamino)purine (2-iP) in order to induce multiple shoots. Among the tested treatments, the 2.0 mg/L of 2-iP proved to be most suited for the multiplication and growth of shoots; the multiple shoot induction rate was 100.0%, the average number of shoots was 7.4 per explant, and the average shoot length was 51.7 mm. The in vitro elongated shoots were rooted on half-strength MS medium containing various concentrations of indole-3-butyric acid (IBA) and 1-naphthaleneacetic acid (NAA). However, overall callus overgrowth was observed in all treatments and resulted in necrosis and abnormal shoot growth in root formation. A low concentration (0.5 mg/L) of IBA was appropriate for normal root development and the in vitro rooting rate was 30%. Ex vitro treatments on root formation using various concentrations of IBA with Talc powder and two types of rooting substrates (Flexi-Plugs or Horticultural soil) were examined. The ex vitro rooting rate (80%) and length of roots (32.9 mm) were obtained when the cut ends of the shoots were treated with 1.0 mg/L IBA and cultivated in Horticultural soil for 2 months. These findings suggest that ex vitro rooting is the more effective method for improving root formation in Vaccinium oldhamii than in vitro rooting.

Comparison of Regeneration Conditions in Seven Pepper (Capsicum annuum L.) Varieties (7종의 고추(Capsicum annuum L.) 재분화 조건 비교)

  • Pepper (Capsicum annuum L.) is an important vegetable and spice crop that has been cultivated worldwide. Pepper fruits have unique taste and aroma, providing a variety of antioxidants and compounds important for human health, which makes a high economic value. In addition, there is a high demand for new pepper varieties, according to consumer's preference. However, pepper is a recalcitrant plant for in vitro tissue and organ differentiation and plant regeneration, which makes it difficult to develop demanded varieties using newly developed technologies such as genetic engineering and gene editing. In this study, tissue culture and regeneration conditions were investigated using seven pepper varieties that were obtained from the core-collection of Seoul National University. We observed callus and bud induction and shoot formation using several media composition composed of different cytokinins and auxin concentrations. As a result, it was found that there were differences in callus induction and shoot formation of each variety depending on the hormone composition, and the highest regeneration was shown when the medium containing Zeatin Riboside and the petioles of seedlings were used. In particular, out of seven pepper varieties, CMV980 exhibited a higher regeneration efficiency (approximately 48%) than other varieties, followed by Yuwolcho. Therefore, this study provides CMV980 and Yuwolcho as good candidates that can be used for pepper transformation, which might contribute to the development of various varieties through gene editing technology in the future.

Effects of the Daylight Disturbance on the Growth and Yield of Spray Chrysanthemum 'Yellow Cap' and 'Peach PangPang' (일조방해가 스프레이 국화 '옐로우캡'과 '피치팡팡'의 생육 및 수량에 미치는 영향)

  • This study was carried out to analyze the effect of changes in the light environment caused by the daylight disturbance on the growth, flowering, and cut flower quality of spray chrysanthemums. The spray chrysanthemum 'Yellow Cap' and 'Pitch PangPang' cultivars for cut flowers were artificially shaded to interfere with 66% of sunlight compared to the non-shading, and then the growing and flowering characteristics, and cut flower yield were investigated accordingly. There was no significant difference in the cut flower yield per unit area between the shading and the non-shading treatments. However, the number of days to flowering was 72.1 days for the 'Yellow Cap' and 65.2 days for the 'Pitch PangPang', which were delayed by 14.1 and 8.9 days, respectively, compared to the non-shading light. In the shading treatment, the flower diameter and the number of flowers also decreased by 10% and 15%, and 30% and 28% for both 'Yellow Cap' and 'Pitch PangPang', respectively. The stem length also decreased by 10% and 20%, the stem diameter by 23% and 37%, and fresh weight by 32% and 33%, respectively. The shading treatment delayed the flowering of chrysanthemums and reduced the growth such as flower diameter, number of flowers, and the length and weight of cut flowers. Based on these results, the daylight disturbance by artificial buildings is expected to reduce the productivity and quality of cut flowers by limiting the light intensity needed for chrysanthemum growth, flower bud differentiation, and flower development. Therefore, further research is needed on the rate of decrease in yield and market value according to the degree of shading to relieve damage to chrysanthemum growers caused by the daylight disturbance.

Assessment of Substantial Equivalence and Environmental Risk for Event Selection of Genetically Modified Chrysanthemum (국화의 GM 이벤트 선발을 위한 실질적 동등성 및 환경위해성 평가)

  • As an assessment of substantial equivalence and environmental risks of branchless chrysanthemum GM lines (LeLS69, LeLS76, LeLS80) developed in Korea, we investigated their degree of branchlessness and developmental characteristics including growth and flowering, as well as the stability of populations of insect pests and soil microbes. The GM lines were compared with a control variety, 'Jinba'. LeLS80 showed the highest level of branchlessness among the three GM lines, whereas abnormal axillary buds developed in lines LeLS76 and LeLS69. There were no significant differences between the growth characteristics of 'Jinba' and the GM lines. The branchlessness LeLS80 was increased during the summer season, suggesting that it is a useful germplasm for developing elite branchless cultivars. These GM lines showed no significant differences in the occurrence of five insect pests including aphids, mites, leaf miners, thrips, and whiteflies. Similarly, for soil microbes including bacteria and actinomycetes, there were no significant differences between 'Jinba' and the GM lines. However, there was a higher occurrence of fungi in the GM lines compared to 'Jinba'. Our findings suggest that 'Jinba' and the GM lines are substantially equivalent, although further monitoring studies are needed to confirm this.

Effects of Water Temperature and Salinity on the Oxygen Consumption Rate of Juvenile Spotted Sea Bass, Lateolabrax maculatus (점농어, Lateolabrax maculatus 치어의 산소 소비율에 미치는 수온과 염분의 영향)

  • An experiment was conducted to investigate the effects of three water temperatures (15, 20 and $25^{\circ}C$) in combination with three salinities (0, 15 and 30 psu) on the oxygen consumption rate of juvenile spotted sea bass, Lateolabrax maculatus (mean body weight $5.5{\pm}0.3g$). The oxygen consumption rates of L. maculatus were measured in triplicate for 24 hours using a continuous flow-through respirometer. Water temperature resulted in significant differences in the mean oxygen consumption rate of L. maculatus (p<0.001), but salinity and combinations of salinity and water temperature did not have (p>0.05). The oxygen consumption increased with increasing water temperatures in all experimental salinity regimes (p<001). Mean oxygen consumption rates at 15, 20 and $25^{\circ}C$ ranged 328.8~342.3, 433.9~441.0 and 651.5~659.9 mg $O_2\;kg^{-1}\;h^{-1}$, respectively. $Q_{10}$ values did not vary with salinity, bud varied with water temperature. $Q_{10}$ values ranged 1.63~1.75 between 15 and $20^{\circ}C$, 2.24~2.26 between 20 and $25^{\circ}C$, and 1.92~1.98 over the full temperature range. The energy loss by metabolic cost increased with increasing water temperatures in all experimental salinity regimes (p<0.001) Mean energy loss rates at 15, 20 and $25^{\circ}C$ ranged 224.6~233.8, 296.3~301.2 and $444.9{\sim}450.7kJ\;kg^{-1}\;d^{-1}$, respectively. These data suggest that the culture of juvenile spotted sea bass is possible without energy loss by salinity difference in freshwater as well as seawater after salinity acclimation. Thus, this result has an application for culture management and bioenergetic model for growth of this species.

Current status and prospects of molecular marker development for systematic breeding program in citrus (감귤 분자육종을 위한 분자표지 개발 현황 및 전망)

  • Citrus is an economically important fruit crop widely growing worldwide. However, citrus production largely depends on natural hybrid selection and bud sport mutation. Unique botanical features including long juvenility, polyembryony, and QTL that controls major agronomic traits can hinder the development of superior variety by conventional breeding. Diverse factors including drastic changes of citrus production environment due to global warming and changes in market trends require systematic molecular breeding program for early selection of elite candidates with target traits, sustainable production of high quality fruits, cultivar diversification, and cost-effective breeding. Since the construction of the first genetic linkage map using isozymes, citrus scientists have constructed linkage maps using various DNA-based markers and developed molecular markers related to biotic and abiotic stresses, polyembryony, fruit coloration, seedlessness, male sterility, acidless, morphology, fruit quality, seed number, yield, early fruit setting traits, and QTL mapping on genetic maps. Genes closely related to CTV resistance and flesh color have been cloned. SSR markers for identifying zygotic and nucellar individuals will contribute to cost-effective breeding. The two high quality citrus reference genomes recently released are being efficiently used for genomics-based molecular breeding such as construction of reference linkage/physical maps and comparative genome mapping. In the near future, the development of DNA molecular markers tightly linked to various agronomic traits and the cloning of useful and/or variant genes will be accelerated through comparative genome analysis using citrus core collection and genome-wide approaches such as genotyping-by-sequencing and genome wide association study.
