Metal Concentrations in some Brown Seaweeds from Kongsfjorden on Spitsbergen, Svalbard Islands (스발바드군도 스피츠베르겐섬 콩스피요르드에 서식하는 갈조류의 중금속 농도)

  • Concentrations of Al, As, Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb, Zn were determined in four arctic brown algae (Laminaria saccharina, L. digitata, Alaria esculenta, Desmarestia aculeata) in an attempt to examine for their metal accumulation capacity and also to assess their contamination levels. Macroalgae were collected from shallow subtidal waters (<20m) of Kongsfjorden (Kings Bay) on Spitsbergen during the period of the late July to early August 2003. Metal concentrations highly varied between sampling sites, species and tissue parts. Input of melt-water laden with terrigenous sediment particles seemed to have a large influence on baseline accumulations of some metals (Al, Fe, Mn, Pb etc.) in the macroalgae, causing a significant spatial variation. There were also significant concentration differences between the young and old tissue parts in L. saccharina, L. digitata and A. esculenta. While Al, Fe, Mn, Pb were higher in the perennial parts (stipes and holdfast below meristematic region), Cd and As concentrations were significantly higher in the young blades above the meristematic region. Zn and Cr, on the other hand, showed little differences between the tissue parts. The highest metal concentrations were found in D. aculeata, which seems to be due to its filamentous fine branches leading to high surface/volume ratios. The lowest concentrations were found in the two Laminaria spp., the blades of which are thicker than D. aculeata and A. esculenta. No distinct signs of contamination were detected in the brown algal species analyzed. Added to this, the results of the present studies suggest the potential utility of L. saccharina, L. digitata and A. esculenta as biomonitors for metal pollution monitoring in this area.

Preparation of Cellular Liquid from Brown Seaweeds for Functional Tonic Products (기능성 음료의 개발을 위한 갈조류 생세포액의 제조)

  • 기능성 해조음료를 개발하기 위하여 미역, 톳 및 다시마를 저온하에서 마쇄, 원심분리, 한회여과를 거쳐 생해조 세포액을 얻었으며, 각 해조 생세포액의 성분 분석과 색차 측정 및 관능검사를 통하여 품질평한 결과 다음과 같았다. 원심분리 후 한외 여과에 의한 생세포액의 수율은 생해조에 대하여 6.8~56.5% 였으며, 톳과 미역의 수율은 높았으나 다시마는 수율이 낮았다. 해조액을 한회여과한 다음 동결건조한 시료의 회분 합량은 28~60%였으며, 조단백 함량은 12.7%~6.7%정도 였는데 미역과 톳은 다시마 보다 각 성분의 함량이 많았다. 알긴산과 chlrophyll 및 carotenoid 은 한회여과막에 의하여 대부분 제거되었으나 톳의 polyphenol은 30K 한외여과막에 의해서도 효과적으로 제거되지 않았다. 다시마와 톳 생세포액의 주요 아미노산은 glutamic acid와 aspartic acid 였으며, 미역의 경우는 alanine, aspartic acid, valine이었다. 해조 생세포액중의 무기 질은 칼륨의 함량이 특히 높았으며 양이온의 경우 나트륨, 마그네슘, 칼슘, 철, 아연, 망간, 구리순으로 함유 되어있었다. 음이온으로는 $Cl^{-}$,$SO_{4}^{2-}$,$HPO_{4}^{2-}$,$NO_{3}^{-}$의 함량이 높았으며, I와 Br의 함량은 다시마가 톳과 미역에 비하여 높았다.

Summer Seaweed Flora and Community Structure of Uninhabited Islands in Goheung, Korea (한국 고흥군 무인도서 하계 해조상 및 해조류 군집구조)

  • The intertidal benthic macroalgal floras of 19 uninhabited islands were investigated in the Goheung area, South Sea, Korea, in June 2008. Seaweed community structures on the rocky shores of four (Naemaemuldo, Ceoldo, Araedombaeseom, and Jinjioedo) of the 19 islands were also examined. Eighty macroalgal species were identified, including 13 green, 19 brown, and 48 red algae. The maximum number of species was found at Naemaemuldo, with 35 species, and the minimum was at Aredombaeseom, with 21 species. Seaweed biomass ranged from 21.39-76.22 g dry wt/$m^2$, with a maximum at Naemaemuldo, and minimum at Jinjioedo. Sargassum thunbergii was a representative species, distributed widely in the intertidal zone of the four islands. Subdominant seaweeds were Corallina pilulifera and Ulva pertusa at Naemaemuldo and Jinjioedo, respectively. Also, Ishige okamurae was dominant at Ceoldo and Araedombaeseom. Six functional seaweed forms were found at each study site, except for Araedombaeseom, which had four functional groups. On the rocky shores of the four sites, a coarsely-branched form was the most dominant functional group ranging from 44.44-61.90% in species number and 72.42-91. 09% in biomass. In conclusion, among the four study sites, the shore of Naemaemuldo Island had the best ecological status, with the highest number of species, and greatest biomass (mainly brown and red algae) and functional form diversity of seaweeds. Furthermore, on the rocky shore of Naemaemuldo, coarsely branched- and joint calcareous-form seaweeds, which grow in clean and undisturbed environmental conditions, were the representative functional forms.

Quality Factors and Functional Components in the Edible Seaweeds I. Distribution of n-3 Fatty Acids in 10 Species of Seaweeds by Their Habitats (식용 해조류의 품질구성요인과 그 기능성 성분 I. 서식지에 따른 10종 해조류의 n-3 지방산의 분포)

  • Total lipid contents from 10 species of marine algae(2 green, 5 brown, and 3 red algae) collected from different places in Korea were examined and their fatty acid compositions were compared among species and habitats. Total lipid(TL) was prominent in green laver(about 7.3~10.1%) of the green algae, in sea mustard and seaweed fusiforme(about 3.1~4.8%) of the brown algae and purple laver(about 4.9~6.4%) of the red algae. Sea mustard and seaweed fusiforme collected at Chungmu contained a relatively high level of TL than that at Yosu and Kijang. The TL content of purple laver showed the highest portion in that collected at Nakdong. Green algae comprised the majority of n-3 fatty acids(29.0~66.3%), which mainly consisted of 16 : 4(n-3) (or 16 : 3 (n-3)), 18 : 3(n-3) and 18 : 4(n-3). Brown algae accounted for a low level of n-3 fatty acids(17.9~36.5%) mainly 18 : 4(n-3), 18 : 3(n-3) and 20 : 5(n-3), whereas the brown algae contained a significant level of n-6 fatty acids(7.23~26.5%) such as 20 : 4(n-6) and 18 : 2(n-6). In the case of red algae, the n-3 fatty acids consisted mostly of 20 : 5(n-3) which scored 53% of polyenoic acids in purple laver collected at Nakdong. The proportion of n-3 fatty acids in algae belonging to the same species was higher in algae of high TL contents. Consequently, TL and n-3 fatty acid levels from the seaweeds studied in this paper were different from their habitats.

Effects of Water and Ethanol Extracts from Four Types of Domestic Seaweeds on Cell Differentiation in 3T3-L1 Cell Line (국내산 해조류 4종의 물과 에탄올 추출물이 3T3-L1에서 지방세포 분화에 미치는 영향)

  • The aim of this study was to examine the cytotoxicity and potential inhibitory effects from four types of edible domestic brown seaweeds, Undaria pinnatifida (UP), Laminaria japonica (LJ), Sargassum fulvellum (SF), and Hizikia fusiforme (HF), on preadipocyte differentiation in 3T3-L1 cell line. Water and ethanol extracts from the four types of seaweeds were prepared and tested for cell viability in the 3T3-L1 cell line by using MTT assay. In addition, various doses of the water extract of seaweeds (WES) and ethanol extract of seaweeds (EES) were treated at the beginning of 3T3-L1 differentiation and continued until the cells were fully differentiated to adipocytes. Oil Red-O staining was performed to determine the potential cell differentiation inhibitory effects of the WES and EES by measuring the levels of lipid droplet accumulation in 3T3-L1 adipocytes. $PPAR{\gamma}$ mRNA expression levels were significantly reduced by WESs of UP, LJ, and HF as well as EESs of LJ and HF. As a result, we observed the superior cell differentiation inhibitory effects of WES compared to that of EES in a dose-dependent manner without any significant cytotoxicity in mouse adipocytes.

Seasonal Variations in the Macroalgal Flora and Community Structure in Hallyeohaesang National Park on the South Coast of Korea (한국 남해안 한려해상국립공원의 계절별 해조상 및 군집구조 변화)

  • Seasonal variability in the marine seaweed community structure was examined in the intertidal zones at nine study sites in Hallyeohaesang National Park, on the southern coast of Korea from March to October 2014. A total of 145 seaweeds were indentified, comprising 15 green, 41 brown and 89 red algae. Coarsely branched seaweeds were the dominant functional group, comprising 58.95% in species number, whereas filamentous, sheet, thick leathery, crustose and jointed calcareous forms comprised 2.63-17.72% each. The seaweed biomass averaged 358.00 g dry wt/m2 and it was maximal at Somaemuldo (847.64 g dry wt/m2) and minimal at Gamam (56.51 g dry wt/m2). Based on biomass, the dominant and subdominant seaweeds were Ulva australis at Gamam, Sargassum thunbergii at Sangju, Ulva australis and S.fulvellum at Neukdo, S.horneri at Dala-Bijindo-Somaemuldo, S. thunbergii at Dapo, and Corallina pilulifera at Songdo. Community indices were as follows: dominance index (DI), 0.43-0.71; richness index (R), 8.26-16.50; evenness index (J'), 0.36-0.54; and diversity index (H'), 1.57-2.19. In conclusion, we found that both biomass and the community structure of seaweeds in Hallyeohaesang National Park were similar to those in other studies of the Southern Sea along the Korean peninsula, and that Hallyeohaesang National Park is a relatively favorable habitat for seaweeds. Future studies should examine the changes in seaweed composition and biomass as they relate to climate change and environmental pollution.

Structure of Intertidal Macroalgal Community at the Yeonpyeongdo Islets, Korea

  • Intertidal benthic algal communities on the shores of Gujido and Daeyeonpyeongdo islets, Korea, were examined in October 2007. At both sites, 45 seaweeds including 7 green, 6 brown and 32 red algae were identified. The number of species at Gujido(38 species) was 1.5 times higher than at Daeyeonpyeongdo(25 species), but seaweed coverage was very similar with about 20%. Dominant seaweeds in terms of coverage and importance value were Hildenbrandtia sp., Caulacanthus okamurae, Ulva pertusa, and Gelidium amansii at Gujido and U. pertusa, Hildenbrandtia sp., Gelidium divaricatum at Daeyeonpyeongdo. The vertical distribution pattern of the seaweeds was G. divaricatum-U. pertusa, Hildenbrandtia sp.-U. pertusa, Hildenbrandtia sp., Ishige okamurae from upper to lower intertidal zone but seaweed zonations were not observed on the Gujido rocky shore. At both sites, coarsely-branched forms were the dominant functional group in species number and percent cover(among benthic algal species). The rocky shores of the two sites were dominated by crustose coralline and green algae, whose presence generally results in decreased seaweed biodiversity and community stability. Therefore, the shores of the Yeonpyeongdo islets are of considerable environmental concern and should be monitored for seaweed species composition and community structure.

The Correlation of Antioxidative Effects of 5 Korean Common Edible Seaweeds and Total Polyphenol Content (한국산 5종 해조류의 항산화효과와 총 폴리페놀 함량과의 관련성)

  • Antioxidative activities of 5 common edible seaweeds in Korea, three brown algae (seaweed fusiforme, sea mustard, sea tangle), one green algae (sea lettuce) and one red algae (laver), were examined. The antioxidative activities of ethanol extracts from these seaweeds were examined by measuring of inhibition rates against iron-induced linoleate peroxidation, DPPH (1,1-diphenyl -2-picrylhydrazyl) radical generation and MDA-BSA (malondialdehyde-bovine serum albumin) conjugation. Sea lettuce ethanol extract showed the strongest anti-oxidative activity among them, especially in inhibition against conjugation of lipid peroxide and protein. Second to sea lettuce, laver and sea tangle ethanol extracts showed high DPPH radical scavenging activity and inhibition against MDA-BSA conjugation. However, seaweed fusiforme and sea mustard ethanol extracts did not show antioxidative activities. Sea mustard contained the highest total flavonoids (11.33 mg/g dry wt) and sea lettuce contained the highest total polyphenol (8.97 mg/g dry wt) among these seaweeds. In addition, there was strong positive correlation between the antioxidative activity and total polyphenol content in these seaweeds, suggesting polyphenol compounds may contribute to antioxidative effect of seaweeds. From these data, it is suggested to consume much of seaweeds such as sea lettuce, laver and sea tangle to prevent age-related chronic diseases, and also develope neutraceutical products using polyphenol rich fraction from sea lettuce.

Comparision of antioxidant and anti-inflammatory activities of enzyme assisted hydrolysate from Ecklonia maxima blades and stipe

  • Marine brown seaweeds are a source of functional ingredients with various biological properties. They have been used in the food and functional food industries. Brown seaweeds are divided into three parts of blades, stipe, and root. Normally seaweed blades were used as raw materials for biological research. However, there are limited uses on stipes of Ecklonia maxima (E. maxima) depending on the physicochemical, nutritional, and biological properties. Besides, the comparative studies of two structures of E. maxima, blades and stipe didn't discover previously. This study aimed to compare the potent antioxidant and anti-inflammatory activities of the two structures of E. maxima, blades and stipe in vitro studies to increase the utilization of the two structures of E. maxima. The enzyme-assisted hydrolysate from E. maxima showed significant antioxidant and anti-inflammatory activities. Among them, celluclast-assisted hydrolysate from E. maxima blades (EMBC) and viscozyme-assisted hydrolysate from E. maxima stipe (EMSV) expressed significant protection on hydrogen peroxide-induced oxidative stress. Moreover, EMBC and EMSV treatment remarkably reduced nitric oxide production by downregulation of pro-inflammatory cytokine expressions in lipopolysaccharide-stimulated Raw 264.7 cells. Especially EMBC showed strong inhibition on pro-inflammatory cytokine production compared to EMSV. Taken together research findings suggest that EMBC and EMSV possessed potent antioxidant and anti-inflammatory properties and may be utilized as functional ingredients in the food and functional food sectors.

Screening Anti-inflammatory Actinomycetes Isolated from Seaweeds and Marine Sediments (해조류 및 해양저질에서 항 염증성의 방선균 선발)

  • The anti-inflammatory activities of marine actinomycetes were surveyed. In total, 363 strains were isolated from marine sediments, seaweed tissue, and seaweed rhizosphere. Of these, strains 16 and 291-11 showed the most potent anti-inflammatory activity in phorbol-ester-induced mouse ear edema and erythema assays. Strains 16 and 291-11 were isolated from the rhizosphere of the brown seaweeds Sargassum thunbergli and Undaria pinnatifida, respectively, and were identified as Streptomyces macrosporeus and St. praecox, respectively, using 165 rDNA sequence analysis.
