Intraspecific Protoplast Fusion of Brevibacterium and Intergeneric Protoplast Fusion between Brevibacterium flavum and Corynebacterium glutamicum and the Metabolic Control of L-Lysine Biosynthesis in Improved Bacterial Strains (Brevibacterium flavum의 동종간 및 Corynebacterium glutamicum과의 이속간 원형질체 융합 및 개량균주의 L-Lysine 생합성의 대사제어)

  • As a trial method of breeding L-lysine producing strains, the intraspecific protoplast fusion bet-ween Brevibacterium flavum ATCC 21528R and Brevibacterium flavum ATCC 21529S and the intergeneric protoplast fusion between Brevibacterium flavum ATCC 21528R and Corynebacterium glutamicum ATCC 13058S were performed. The optimum conditions for protoplast formation of these strains were examined and the effect of plasma expander on regeneration and/or fusion was also observed. Both fusants No. CH23 and No. CH4l showed higher productivity of L-lysine than those of parental cells under the optimum cultural conditions at a rate of 21% and 8.9%, respectively. And, activity of several enzymes in L-lysine biosynthetic pathway including aspartokinase, a rate-limiting enzyme, was determined. Besides, metabolic control mechanism of L-lysine biosynthesis in fusant No. CH23 and in No. CH41 was investigated to compare with that of parental strains.

Optimum Culture Conditions of Brevibacterium sp. CH2 for Production of Nitrile Hydratase

  • Optimum culture conditions for the formation of nitrile hydratase by Brevibacterium sp. CH2 were investigated. Addition of ferric and ferrous ions greatly increased the nitrile hydratase formation. The effects of nitriles, amides, and acids as an inducer on the formation of nitrile hydratase were investigated. Isobutyramide was the best inducer among the tested compounds. When Brevibacterium sp. CH2 was cultivated for 23 h at $30^{\circ}C$ in a optimized medium containing 15 g of glucose, 5 g of bacto peptone, 3 g of yeast extract, 3 g of malt extract, 1 g of $KH_2$$PO_4$, 1 g of $K_2$$HPO_4$, 1 g of NaCl, 0.5 g of isobutyramide, 0.2 g of MgSO$_4$ㆍ7$H_2O$, and 0.02g of $FeSO_4$$7H_2$O per liter of distilled water with pH controlled at 7.1, the maximum total activity was 665 units/ml of the culture broth and the specific activity was 70 units/mg of the dry cells. The medium optimization increased the specific activity of Brevibacterium sp. CH2 2.2 times.

Effects of Acrylonitrile and Acrylamide on Nitrile Hydratase Action of Brevibacterium sp. CH1 and CH2

  • The effects of acrylonitrile and acrylamide on the enzyme action of nitrile hydratase of Brevibacterium sp. CH1 and CH2 strains used for the biotransformations of nitriles were studied. The excessive substrate (acrylonitrile) and product (acrylamide) inhibited the enzyme activity competitively. In comparison with 0.2 mol/l of CH1 strain, the substrate inhibition of CH2 strain began to appear only at a high acrylonitrile concentration of 0.91 mol/l. In a packed bed reactor, dispersed plug flow model was proposed and this model was proved to be valid by the experiment. Also acrylamide productivity decreased sharply when acrylamide concentration in the substrate solution exceeded 20% (wt/v).

  • Seven new hydroxyamine derivatives of 2,2'-methylene bis(3,4,6-trichloroacetoxy benzene) were synthesized by the Mannich reactions. 13 strains of microorganisms were tested for sensitivity to these derivatives by both paper disk method and tube dilution method. Of these compounds, -NHOH compound displays the most effective antimicrobial activity in vitro against Brevibacterium ammoniagenes, Staphylococcus aureus and Bacillus subtilis. Its minimal inhibitory concentration is $1.6{\mu}g/ml$ for Brevibacterium ammoniagenes, and $5{\mu}g/ml$ for Staphylococcus aureus and Bacillus subtilis.

  • The effect of concentrations of phosphate buffer and acrylonitrile, pH, and various salts on the operational stability of the immobilized cells of Brevibacterium CH2 in a packed bed reactor were investigated. The effects of salts and carriers on the swelling of the immobilized beads during hydrolysis in a columnreactor were also investigated. Immobilization of the cells in Ba-alginate was more desirable than those in polyacrylamide and Ca-alinate for the swelling of the immobilized beads and the desired quality of the acrylamide produced. High quality acrylamide was produced using the Ba-alginate beads in a recycle fed-batch reactor without using an isotonic substrate. The conversion yield was nearly 100%, including a trace amount of acrylic acid produced as a by-product.
