Effect of Glycine on L-Ornithine Production by a Citrulline Auxotroph of Brevibacterium ketoglutamicum and Stoichiometric Analysis

  • The effects of glycine on cell growth and L-omithine production were investigated in shake-flask and jar fermentor cultures of a citrulline auxotrophic mutant, Brevibacterium ketoglutamicum BK 1046. In the shake-flask culture, the optimal concentration of glycine for L-ornithine production was found to be 20 g/l. In the jar fermentor culture with the glycine at an initial concentration of 20 g/l, L-ornithine production increased by 28%, compared to that of the culture with no glycine added. 37 g/l of L-ornithine was produced when additional feeding of glycine (5 g/l) was made. This was a significant improvement in L-ornithine production compared to that (ca. 24 g/l) of the corresponding batch culture conducted without glycine. According to the stoichiometric analysis with the batch fermentation results, the experimental and theoretical L-ornithine yields based on the glucose consumption were 0.24 and 0.59, respectively. This indicates that the performance of L-ornithine fermentation can further be improved by the supplementation of glycine and the development of a mutant strain possessing a higher growth yield.

Brevibacterium sp. 로부터 유도된 PFP 내성 변이주에 의한 L-Tyrosine 생성 (Production of L-Tyrosine by PFP Resistant Mutant Induced from Brevibnrcterium sp.)

  • This study was attempted to investigate the production of L-tyrosine by Brevibacterium flavum ATCC 14067. To select the strain which produce more L-tyrosine, mutants were induced by N-methyl-N'-nitro-nitrosoguanidine (NTG) treatment and phenylalanine auxotrophic mutants were induced by NTG and penicillin treatments. PFP resistant mutant was isolated from a phenylalanine auxotroph by retreatment with NTG and screened for increase of L-tyrosine production. PFP-326 mutant resistant to PPP (100ug/ml) was derived from phenylalanine auxotroph by mutagenesis with NTG and PFP-106 mutant resistant to PFP (1201g/ml) was derived from PFP-326 by mutagenesis with NTG. The composition of media for L-tyrosine production in strain PFP-106 was studied. PFP-106 mutant strain produced 50mg 11 of L-tyrosine while the parent strain produced 0.56mg 11 of L-tyrosine. The optimum composition of medium for L-tyrosine by strain PFP-106 was 10cA sucrose as carbon source, 3% ammonium sulfate as nitrogen source. The optimum cultural condition for producing L-tyrosine by strain PFP-106 was L-phenylalanine at a concentration of 1000g/mg.

Brevibacterium ammoniagenes에 의한 글루탐산 제조에 관한 연구 (Studies on the Production of L-Glutamic Acid by Brevibacterium ammoniagenes)

  • A bacterium strain (K-173-10) which was isolated from waste soil of Korea brewing factory, could be grown on acetate as the sole carbon source and accumulated a considerable amount of L-glutamic acid in the medium. This strain was identified as the new species Brevibacterium ammoniagenes. This study was concerned not only with the culture condition for the production of L-glutamic acid and the cell growth, but also with the effects on concentration of various kind of organic substances, growth factors and penicillin. The results obtained were summarized as follow; 1. It was found that the concentrations of acetate and ammonium ions affected the growth of the bacterium as well as its L-glutamate accumulation. The optimum conditions of the composition of grown media for the growth of the bacterium and its glutamic acid production was found to be 40 g/l of total acetate, $100\;{\mu}g/l$ thiamine, $0.5\;{\mu}g/l$ biotin and $1{\sim}2g/l$ corn steep liquor as the growth factors. 2. Organic acid such as succinic acid, malic acid and ${\alpha}-ketoglutaric$ acid inhibited the cell growth as well as its L-glutamic acid production. 3. The penicillin (20 units/ml) stimulated the production of glutamic acid at appropriate incubation period. 4. It was found that this strain could grow in the presence of urea and ammonium acetate but not in other nitrogen sources.

Brevibacterium Flavum의 Auxotrophic Mutants에 의한 L-Threonine 생성(生成)에 관한 연구(硏究) (Studies on the Formation of L-Threonine by auxotrophic mutants of Brevibacterium flavum)

  • 본(本) 실험(實驗)은 세균(細菌)에 의한 L-threonine의 효율적인 생성(生成)을 검토할 목적(目的)으로 Brevibacterium flavum ATCC 14067을 사용하여 L-threonine 생성능(生成能)이 우수한 균수(菌株)를 선발(選拔)하기 위해 변이원인 N-methyl-N'-nitro-N-nitrosoguanidine (NTG)로 처리하여 돌연변이주(突然變異株)를 유도(誘導)한 후(後), 다시 methionine 영양요구주, lysine 영양요구주, isoleucine 영양요구주, lysine 영양요구주, isoleucine 영양요구주, methionine 및 isoleucine 영양요구주를 선발(選拔)하였다. 또한 선발(選拔)된 영양요구성 변이주들 중에서 L-threonine 생성능(生成能)이 원균(原菌)에 비(比)해 $3{\sim}4$배(倍)정도 우수한 B-13균주(菌株)$(met^-)$를 선발(選拔)하여 L-threonine 생성력(生成力), 배지(培地) 조성(組成) 및 배양(培養)에 따른 몇가지 요인(要因)들에 대하여 실험한 결과 다음과 같은 결과(結果)를 얻었다. 1. L-threonine생성량은 원균주가 1.4mg/ml에 비해 methionine 영양요구성 변이주인 B-13은 4.86mg/ml로서 약 3.5배(倍)의 높은 생성량을 나타내었다. 2. B-13에 의한 L-threonine 생성(生成)에 적당한 배지조성(培地組成)은 glucose 10%, ammonium sulfate 2%, potassium phosphate monobasic 0.2%, magnesium sulfate 0.05%, biotin $200{\mu}g/l$ thiamine $300{\mu}g/l$이였으며 nicotinic acid 0.05% 첨가시 더욱 증가 되었다. 3. B-13에 있어서 유기영양원에 대한 효과는 yeast extract와 Peptone이 양호하였으며 영양요구물질인 metionine은 $100{\mu}g/ml$가 적당하였으며 aspartic acid와 homoserine 첨가시 L-threonine 생성이 증가되었으며 lysine 첨가시에는 감소하는 경향이 나타났다. 4. B-13에 의한 L-threonine 생성에 가장 적절한 pH는 $7.0{\sim}8.0$이였으며 배양일수(培養日數)는 4일(日)이 적당하였다.

  • By the treatment of various mutagens, a number of 5'-guanylic acidproducing from 5'-xanthylic acid were obtained from Brevibacterium ammoniagenes ATCC 6871. The indispensable genetic characters of the mutants were adenine requirement, lack of GMP-reductase and mutation to adenosine resistance from adenosine sensitiveness. Main product of these mutants from 5'-xanthylic acid was 5'-guanylic acid. The substance was isolated in a crystalline form the culture broth of BA 17-2, and identified as 5'-guanylic acid by means of paper chromatography, ultra violet, absorption spectra, and infra red spectrum.

  • 전국 각지대의 279개소의 토양시료로 부터 초산자화성세균 383균주를 분리하였으며 이들 초산자화성세균중 L-GA생성 능이 우수한 5균주를 선정하고 균학적성질을 조사하여 Brebacterium flavum nov. sp. D1005B, Corynebacterium glutamicum nov. sp. D1025A, Brevibacterium. flavum nov. sp. D2209B, Corynebacterium acetoacidophilum nov. sp. D2212B 및 Corynebacterium acetoacidophilum nov. sp. D2349A로 동정 명명하였다

L-Glutamic Acid 생성균에 관한 연구 1 (Studies on L-Glutamic Acid-Producing Bacteria(I))

  • Three strains which accumulated large quantities of L-hlutamic acid as the chief product of metabolism in the presence of glucose and inorhanic ammonium salt under the aerobic condition sere newly isolated. These strains have the general characters as follows: Gram-positive, short fod-shaped, non-sporulating, non-motile, and facultative anaerobe. A change of cell shates was not almost observed in their life cycles and neither a phenomenon of cell-elongation nor a pleomorphism was recognized in any cases. Together with rigid stability in Gram-stain, these characters as above indicated evidences that newly isolates would belong to genus Brevibacterium clearly differentiated from genus Corynebacterium. At the same time the newly isolates, in addition to nutritional requirement of biotin and thiamine, showed the distinctive character of requirement of special amino acid such as histidine or cysteine for their considerable growth. These physiological characters including nitrates reduction, reaction on milk, and slow formation of acid from sugar also were useful in confirming that these bacteria would be Brevibacterium ammoniagenes.

Effects of Acrylonitrile and Acrylamide on Nitrile Hydratase Action of Brevibacterium sp. CH1 and CH2

  • The effects of acrylonitrile and acrylamide on the enzyme action of nitrile hydratase of Brevibacterium sp. CH1 and CH2 strains used for the biotransformations of nitriles were studied. The excessive substrate (acrylonitrile) and product (acrylamide) inhibited the enzyme activity competitively. In comparison with 0.2 mol/l of CH1 strain, the substrate inhibition of CH2 strain began to appear only at a high acrylonitrile concentration of 0.91 mol/l. In a packed bed reactor, dispersed plug flow model was proposed and this model was proved to be valid by the experiment. Also acrylamide productivity decreased sharply when acrylamide concentration in the substrate solution exceeded 20% (wt/v).

Quantitative Analysis of Corynomycolic Acids in Fermentation Broth

  • The mycolic acids and fatty acids of mycolic acid- containing bacteria in various types of fluids were analyzed using capillary gas chromatography and mass spectrometry. As model strains, Brevibacterium and Coryebacterium species, which have corynomycolic acids ill the range of $C_{32}C_{36}$ in the whole cell, were investigated. Optimized solvents extraction procedures for the mycolic acids and fatty acids from the culture fluids were: chloroform/methanol (1:2, v/v) as the first extraction solvents fur 4 h; and chlorofunuwater (1:1, v/v) as the second extraction solvents far 1 h. These conditions gave above 95% recovery yields fur mycolic acids from the culture fluids. The mycolic acid profile for the whole cells and the culture fluids were similar fur all the media tested. Thus, the procedure described here could be applied for the identification of mycolic acid-containing bacteria in fermentation broth or liquid from of foods.

  • 1) 자연체에서 분리한 세균의 한 균주(No.1214)는 초산염을 자화하여 L-glutamic acid를 상당량(22g/l) 생산하였다. 2) 분리된 균주는 형태학적, 생리학적, 배양학적인 특징으로 보아 Brevibacterium flavum속으로 간주되었다.
