A simple model for selection and rapid advancement of transgenic progeny in sorghum

  • To select agronomically useful transgenic plants, a large number of transgenic events are initially produced, gene transfer confirmed, and advanced to obtain homozygous lines for testing in field trials. Direct in planta assays for identifying the transgene carriers in the segregating progeny are based on the activity of selectable marker gene and are easy, simple and inexpensive. For this purpose, expression of bar gene as measured by tolerance to damage by glufosinate ammonium, the active ingredient in the herbicide BASTA, was investigated. Dose damage curves were generated by leaf paint tests with BASTA on four genotypes of sorghum. Transgenic plants were characterized in terms of sensitivity to the concentration of glufosinate ammonium. In transgenics, symptoms of BASTA swab tests at different growth stages and PCR analysis for cry1B were carried out and correlated. Germination tests could not be employed for large scale evaluation of transgenic progeny because of mortality of tolerant seedlings after transplantation to soil. Based on the above findings, a simple, inexpensive, time-saving, two-step scheme for effective evaluation of transgenics and their progeny containing bar gene as selection marker using BASTA swab tests is described.

Neutronics modelling of control rod compensation operation in small modular fast reactor using OpenMC

  • The small modular liquid-metal fast reactor (SMFR) is an important component of advanced nuclear systems. SMFRs exhibit relatively low breeding capability and constraint space for control rod installation. Consequently, control rods are deeply inserted at beginning and are withdrawn gradually to compensate for large burnup reactivity loss in a long lifetime. This paper is committed to investigating the impact of control rod compensation operation on core neutronics characteristics. This paper presents a whole core fine depletion model of long lifetime SMFR using OpenMC and the influence of depletion chains is verified. Three control rod position schemes to simulate the compensation process are compared. The results show that the fine simulation of the control rod compensation process impacts significantly the fuel burnup distribution and absorber consumption. A control rod equivalent position scheme proposed in this work is an optimal option in the trade-off between computation time and accuracy. The control position is crucial for accurate power distribution and void feedback coefficients in SMFRs. The results in this paper also show that the pin level power distribution is important due to the heterogeneous distribution in SMFRs. The fuel burnup distribution at the end of core life impacts the worth of control rods.

한우 ADSF/resistin 유전자의 단일 염기 다형과 육질관련형질 상관 분석 (Analysis of the ADSF/resistin Gene Polymorphism Associated with Carcass Traits in Hanwoo)

  • 본 연구는 후대검정 한우 295두의 혈액으로부터 genomic DNA를 추출하여 PCR 방법에 의한 증폭과 염기서열 분석을 통하여 ADSF/resistin 유전자의 단일염기다형을 발굴하고 이들과 한우 육질관련형질과의 상관관계를 분석하기 위하여 실시하였다. 확보된 DNA로부터 염기서열을 결정한 결과 promoter와 4개의 exon영역에서는 SNP를 찾지 못하였으나 intron 영역에서 7개의 SNP를 발굴하였다. 발굴된 SNP의 출현 빈도는 0.027에서 0.16까지 그 차이가 많았다. 육질형질과의 상관분석에서 이들 SNP 중 intron 2에서 발굴된 764A ins 만이 근내지방도와 상관관계가 발견되었다(P<0.05). 근내지방도는 유전력이 매우 높기 때문에 이번에 발굴된 ADSF/resistin 유전자의 SNP 764A ins와 같이 유전표지인자를 이용하는 것이 근내 지방도의 개량을 위해 우수한 결과를 가지는 것으로 사료된다. 가축의 경제형질의 경우 다수의 유전자가 관여하기 때문에 한 개의 유전자를 이용한 가축의 선발 또는 개량에 이용한다는 것은 제한적일 수 있다. 따라서 다수의 관련 유전자를 이용한 다형현상과 경제형질과의 연관성 연구가 동반될 때 육질개선 및 가축개량에 실질적인 증대효과가 있을 것으로 사료된다.

Holstein 보증종모우 및 후보종모우의 선천성 장애 유전좌위 검색에 관한 연구 (Studies on the Detections of Congenital Genetic Disorder in Holstein Proven and Candidate Bulls)

  • 본 연구는 국내 홀스타인 젖소 보증종모우 16두와 후보종모우 93두를 이용하여 선천성 장애 유전자의 검색을 통하여 불량 유전자의 존재 유무를 판별함과 동시에 가축의 선발 및 육종, 개량시 기초자료로 제공하고자 하는데 그 목적이 있으며, 본 연구의 결과를 요약하면 아래와 같다. 공시재료(홀스타인 젖소 보증종모우 16두, 후보 종모우 93두) 109두에 대하여 DUMPS (deficiency of uridine monophophate synthase)의 검색결과 모든 개체에서 DUMPS 유전자를 보유하는 개체는 없는 것으로 판명되었다. 또한 PCR-RFLP(Ava I) 방법에 의해 조기 검색이 가능하게 되었다. 한편, BLAD(bovine leukocyte adhesion deficiency) 검색결과, 보증종모우 16두에서는 검출되지 않았으나, 후보우 93두중 5두에서 BLAD 잠재성 보유개체(carrier)로 판명되었고, 혈통확인을 통하여 BLAD 유전자의 전이 경로를 추정할 수 있었으며, PCR 증폭산물에 대한 제한효소 처리시 HaeⅢ 보다는 TaqⅠ 제한효소를 사용하였을 때 더 효율적으로 판명할 수 있는 것으로 나타났다. Citrullinemia 검색결과 보증종모우 16두 및 후보종모우 93두 모두에서 잠재성 보유개체는 없는 것으로 판명되었으나 citrullinemia에 대한 폭넓고 다양한 조사 및 분석이 이루어져야 할 것으로 사료된다. 본 연구의 결과로 미루어 볼 때 가축의 유전성 질환에 대한 다양하고 폭넓은 연구가 이루어 져야 할 것으로 사료되며, 가축의 선발과 육종, 개량에 있어서 지속적이며 혈통의 철저한 관리를 통한 개량의 방향을 설정하여야 할 것으로 판명되었다.

Survey of genetic structure of geese using novel microsatellite markers

  • Objective: The aim of this study was to create a set of microsatellite markers with high polymorphism for the genetic monitoring and genetic structure analysis of local goose populations. Methods: Novel microsatellite markers were isolated from the genomic DNA of white Roman geese using short tandem repeated probes. The DNA segments, including short tandem repeats, were tested for their variability among four populations of geese from the Changhua Animal Propagation Station (CAPS). The selected microsatellite markers could then be used to monitor genetic variability and study the genetic structures of geese from local geese farms. Results: 14 novel microsatellite loci were isolated. In addition to seven known loci, two multiplex sets were constructed for the detection of genetic variations in geese populations. The average of allele number, the effective number of alleles, the observed heterozygosity, the expected heterozygosity, and the polymorphism information content were 11.09, 5.145, 0.499, 0.745, and 0.705, respectively. The results of analysis of molecular variance and principal component analysis indicated a contracting white Roman cluster and a spreading Chinese cluster. In white Roman populations, the CAPS populations were depleted to roughly two clusters when K was set equal to 6 in the Bayesian cluster analysis. The founders of private farm populations had a similar genetic structure. Among the Chinese geese populations, the CAPS populations and private populations represented different clads of the phylogenetic tree and individuals from the private populations had uneven genetic characteristics according to various analyses. Conclusion: Based on this study's analyses, we suggest that the CAPS should institute a proper breeding strategy for white Roman geese to avoid further clustering. In addition, for preservation and stable quality, the Chinese geese in the CAPS and the aforementioned proper breeding scheme should be introduced to geese breeders.

  • 1. This report is on the outcome of observation made up the breeding process of Narcissus flycatcher, Muscicapa narcissina zanthopygia, using 15 ness-boxes at Seoul and Kwang Nung Experimental forest in 1964, and the relevant feeding habit was studied by the collar method of wrapping the necks of thenestlings for 50-60 minutes or more in some cases from 1963 to 1964. 2. The Narcissus flycatcheris a common summer resident throughout Korea arriving at the end of April. The earliest data of arrival of this species in Seoul, is April 26, 1964. They migrate southward from the later part of September to early part of October. 3. In a week after their arrival , they start to build nest after a brief courtship. Male and female prepare their nest ina nestboxes in 4 or 5 days. Nests are usually found in the moss, which provide definite frame-work. The inner side of the nest is usually composed of roots of Gramineae or leaves of pine, oak or other broad leave tree. 4. The laying period is generally from the later part of April to the middle of June : and the number of eggs laid is from 3 to 8 , therefore 5 in average : eggs are pink with traces of milky yellow blending of light auburn and orange red spots and chicken-egg-like in shape. 5.The incubation takes place for 7-12 days from the middle part of May to the middle of June, in the order of male and female . The hatching rate tends to increase in proportion to clutch size. 6. The feeding period usually lasts 15 days in average from the early part of June to the middle of July and the feeding urge tends to increase in proportion to brood size. 7. The animals invading into their nest-boxes are red ants, chipmunks, bees , barkbeetles and philippine red-tailed shirkes, etc. 8. On the basis of analysis covering 714 birds in 31 nests, the food they consumed during the nesting period of nestlings were mostly of animal matters composing of : insect larvae form-39. 88% insect adult form 36.26%, insect pupae-0.19%, spider-17.75% and other animal matters 5.92% , Besides a small amount of plant pieces were found . As to the insect larvae, Lepidoptera was the largest in the content followed by Hymenoptera , Orthoptera and Coleoptera. As to the insect adult, Diptera was the largest in the content followed by Coleoptera, and Hymenoptera. In view of forest protection, noxious insect was of 69.38%, useful insect 4.39%, and spider 17.75%. 9. The rich assortment of its food, the ability of acquiring various insect (adults as well as larvae), its destruction of great numbers of harmful insects , and its quick assimilation artificial nesting with the nest-boxes of 2.5-3cm entrance hole employed, allow us to conclude that the flycatcher may be successfully employed in the elimination of fores pests. The above results are only that part obtained near Seoul in the middle of Korea, however, if we would extend the scheme to other broad areas using similar techniques, the nest-boxes could not only be well used by the birds, but also it would help to prevent the increase of harmful. insects.

흑소의 얼굴을 이용한 개체인식 (Identification of Japanese Black Cattle by the Faces for Precision Livestock Farming)

  • Recent livestock people concern not only increase of production, but also superior quality of animal-breeding environment. So far, the optimization of the breeding and air environment has been focused on the production increase. In the very near future, the optimization will be emphasized on the environment for the animal welfare and health. Especially, cattle farming demands the precision livestock farming and special attention has to be given to the management of feeding, animal health and fertility. The management of individual animal is the first step for precision livestock farming and animal welfare, and recognizing each individual is important for that. Though electronic identification of a cattle such as RFID(Radio Frequency Identification) has many advantages, RFID implementations practically involve several problems such as the reading speed and distance. In that sense, computer vision might be more effective than RFID for the identification of an individual animal. The researches on the identification of cattle via image processing were mostly performed with the cows having black-white patterns of the Holstein. But, the native Korean and Japanese cattle do not have any definite pattern on the body. The purpose of this research is to identify the Japanese black cattle that does not have a body pattern using computer vision technology and neural network algorithm. Twelve heads of Japanese black cattle have been tested to verify the proposed scheme. The values of input parameters were specified and then computed using the face images of cattle. The images of cattle faces were trained using associate neural network algorithm, and the algorithm was verified by the face images that were transformed using brightness, distortion, and noise factors. As a result, there was difference due to transform ratio of the brightness, distortion, and noise. And, the proposed algorithm could identify 100% in the range from -3 to +3 degrees of the brightness, from -2 to +4 degrees of the distortion, and from 0% to 60% of the noise transformed images. It is concluded that our system can not be applied in real time recognition of the moving cows, but can be used for the cattle being at a standstill.

Three-dimensional thermal-hydraulics/neutronics coupling analysis on the full-scale module of helium-cooled tritium-breeding blanket

  • Blanket is of vital importance for engineering application of the fusion reactor. Nuclear heat deposition in materials is the main heat source in blanket structure. In this paper, the three-dimensional method for thermal-hydraulics/neutronics coupling analysis is developed and applied for the full-scale module of the helium-cooled ceramic breeder tritium breeding blanket (HCCB TBB) designed for China Fusion Engineering Test Reactor (CFETR). The explicit coupling scheme is used to support data transfer for coupling analysis based on cell-to-cell mapping method. The coupling algorithm is realized by the user-defined function compiled in Fluent. The three-dimensional model is established, and then the coupling analysis is performed using the paralleled Coupling Analysis of Thermal-hydraulics and Neutronics Interface Code (CATNIC). The results reveal the relatively small influence of the coupling analysis compared to the traditional method using the radial fitting function of internal heat source. However, the coupling analysis method is quite important considering the nonuniform distribution of the neutron wall loading (NWL) along the poloidal direction. Finally, the structure optimization of the blanket is carried out using the coupling method to satisfy the thermal requirement of all materials. The nonlinear effect between thermal-hydraulics and neutronics is found during the blanket structure optimization, and the tritium production performance is slightly reduced after optimization. Such an adverse effect should be thoroughly evaluated in the future work.

Semi-domesticated and Irreplaceable Genetic Resource Gayal (Bos frontalis) Needs Effective Genetic Conservation in Bangladesh: A Review

  • Several studies arduously reported that gayal (Bos frontalis) is an independent bovine species. The population size is shrinking across its distribution. In Bangladesh, it is the only wild relative of domestic cattle and also a less cared animal. Their body size is much bigger than Bangladeshi native cattle and has prominent beef type characters along with the ability to adjust in any adverse environmental conditions. Human interactions and manipulation of biodiversity is affecting the habitats of gayals in recent decades. Besides, the only artificial reproduction center for gayals, Bangladesh Livestock Research Institute (BLRI), has few animals and could not carry out its long term conservation scheme due to a lack of an objective based scientific mission as well as financial support. This indicates that the current population is much more susceptible to stochastic events which might be natural catastrophes, environmental changes or mutations. Further reduction of the population size will sharply reduce genetic diversity. In our recent investigation with 80K indicine single nucleotide polymorphism chip, the $F_{IS}$ (within-population inbreeding) value was reported as $0.061{\pm}0.229$ and the observed ($0.153{\pm}0.139$) and expected ($0.148{\pm}0.143$) heterozygosities indicated a highly inbred and less diverse gayal population in Bangladesh. Prompt action is needed to tape the genetic information of this semi-domesticated bovine species with considerable sample size and try to investigate its potentials together with native zebu cattle for understanding the large phenotypic variations, improvement and conservation of this valuable creature.

한우의 도체 형질이 경제성에 미치는 영향 (The Effect of Carcass Traits on Economic Values in Hanwoo)

  • 본 연구는 한우 농가의 소득에 직접적인 영향을 미치는 경매단가(AP) 및 도체판매가(CP)에 영향을 미치는 도체 특성(근내지방도(MS), 배최장근단면적(LMA), 등지방 두께(BFT), 도체중(CW) 및 육량 지수(MYI))와의 상관관계를 조사하였다. AP의 경우는 근내지방도(MS)가 가장 높은 상관(r=0.73)을 나타내었으며, CP는 도체중이 0.71로 가장 높고, 다음으로 근내지방도가 영향력이 높게 나타났다. 하지만, LMA, MYI는 비교적 낮은 상관을 나타내었다. 이상의 결과 농가의 소득의 향상과 한우의 MS 및 CW 향상이 유의적인 상관도가 있었다.
