Barbigerone Inhibits Tumor Angiogenesis, Growth and Metastasis in Melanoma

  • Tumor angiogenesis, growth and metastasis are three closely related processes. We therefore investigated the effects of barbigerone on all three in the B16F10 tumor model established in both zebrafish and mouse models, and explored underlying molecular mechanisms. In vitro, barbigerone inhibited B16F10 cell proliferation, survival, migration and invasion and suppressed human umbilical vascular endothelial cell migration, invasion and tube formation in concentration-dependent manners. In the transgenic zebrafish model, treatment with $10{\mu}M$ barbigerone remarkably inhibited angiogenesis and tumor-associated angiogenesis by reducing blood vessel development more than 90%. In vivo, barbigerone significantly suppressed angiogenesis as measured by H and E staining of matrigel plugs and CD31 staining of B16F10 melanoma tumors in C57BL/6 mice. Furthermore, it exhibited highly potent activity at inhibiting tumor growth and metastasis to the lung of B16F10 melanoma cells injected into C57BL/6 mice. Western blotting revealed that barbigerone inhibited phosphorylation of AKT, FAK and MAPK family members, including ERK, JNK, and p38 MAPKs, in B16F10 cells mainly through the MEK3/6/p38 MAPK signaling pathway. These findings suggested for the first time that barbigerone could inhibit tumor-angiogenesis, tumor growth and lung metastasis via downregulation of the MEK3/6/p38 MAPK signaling pathway. The findings support further investigation of barbigerone as a potential anti-cancer drug.

강도다리(Platichthys stellatus) 변태 과정에서 안구 이동의 방향에 관여하는 환경적 요인 (Effects of Environmental Factors on the Eye Direction in Juvenile Starry Flounder Platichthys stellatus)

  • The direction of eye movement in Pleuronectiformes is species-specific. In farmed flounder, deviations in eye direction can reduce marketability. Therefore, we explored the effects of genetic and environmental factors on the direction of eye movements in the starry flounder Platichthys stellatus to address this economic challenge. Four pairs of female and male starry flounder broodstock, with eyes positioned to the right or left, were used in a mating experiment. The experimental groups were established with breeding seawater temperatures set at 10, 14, and 18℃ for the water temperature experiment. Whole-body samples of juveniles were collected to analyze the expression of genes related to eye movement and direction. The mating experiment results showed no significant differences in abnormal eye movement. In the water temperature experiment, the rate of abnormal eye movement was significantly higher at 32.19±1.33% in the 18℃ group than in the 10 and 14℃ groups. Genetic analysis of eye movement related to water temperature revealed that the expression of eye migration genes was significantly higher at 10℃. Moreover, no significant differences were observed in mating experiments. In conclusion, water temperature and not mating affected the eye movement of starry flounder.

대구 불로고분 자연마당 조류군집 및 서식지 이용 특성 (Characteristics of Bird Community and Habitat Use in Daegu Bulnogoboon Jayeon Madang)

  • This study was carried out four times on 2005 and 2006, respectively May and October to understand the bird community and habitat using condition in the Daegu Bulnogoboon Jayeon Madang (DBJM). In this study, 34 species and 332 individuals were observed. The dominant species were Passer montanus 22.0%, Pica pica 18.1%, Hypsipetes amaurotis 6.02% and Sinosuthora webbiana 5.72%, and the species diversity was 2.797. With migration, the highest number and percentage to the lowest was residents 61.7%, summer visitors 26.5% and winter visitors 11.8%. Among 24 species which bred(check or possibility) in the DBJM, canopy was 50.0%, hole was 37.5% and bush was 12.5% in their nesting guilds. In foraging guilds in breeding season was canopy was 58.1%, water was 19.3% and bush was 16.1%, and in non-breeding season, canopy was 54.8%, water 22.6% and bush 19.3%. For the habitats of the birds in the DBJM, wetlands including reservoir area were used by herons, ducks and Hirundo rustica, grassland area was used by Sinosuthora webbiana, buntings and Passer montanus, and forest area was used by Streptopelia orientalis, woodpeckers, Hypsipetes amaurotis and tits. The number of species and individuals of birds observed in the DBJM was higher than other urban parks. We suggest that this was attributed to various man-made habitats such as wetland with reservoir, grassland and forest area constructed in the DBJM.

길동자연생태공원에서 조류의 군집 특성과 서식지 이용 현황 (Characteristics of Bird Community and Habitat Use in Gildong Natural Ecological Park)

  • This study was carried out to understand the bird community and habitat using condition in the Gildong natural ecological park, from April 2001 to March 2002. In this study, 63 species and 2,075 individuals were observed respectively. The dominant species were Paradoxornis webbianus(33.6%), Emberiza elegans 18.5%), Parus major(6.4%), Pica pica(6.0%), and the diversity of the species was 2.511. With migration, the highest number and percentage to the lowest was residents, summer visitors, winter visitors and passage migrants. Among 17 species which bred in the Gildong natural ecological park, hole(H) was 47.1%, canopy(C) was 41.1% and bush(B) was 11.8% in their nesting guilds. In foraging guilds in breeding season was canopy(c) was 45.7%, water(w) was 23.9% and bush(c) was 15.2%, and, in non-breeding season, canopy was 43.5%, water 23.9% and bush 21.7%. For the habitats of the birds in the Gildong natural cological park, reservoir area was used by herons, ducks and sandpiper, wetland area was used by Lanius bucephalus, Paradoxornis webbianus and buntings, grassland area was used by Paradoxornis webbianus and buntings, and forest area was used by Streptopelia orientalis, woodpeckers and tits. The number of species and individuals of birds observed in the Gildong natural ecological park was higher than other urban parks. We suggest that this was attributed to different habitats such as reservoir, wetland, grassland and forest area constructed in the Gildong natural ecological park.

Association of farmers' knowledge, attitude and practices with bovine brucellosis seroprevalence in Myanmar

  • Objective: This study aimed to identify the relationship between bovine brucellosis prevalence, farmers' knowledge, attitude, practice (KAP), and social factors on migratory draft cattle and smallholder dairy farms in the central dry zone of Myanmar. Methods: This cross-sectional study was conducted on 54 migratory and 38 dairy cattle farms between August 2020 and February 2021. A structured questionnaire was used to identify farmers' behaviors. Bulk milk was sampled and tested using indirect enzyme-linked immunosorbent assay (I-ELISA). STATA 17 was used for all the analyses. Results: Migratory cattle farms had a higher farm level brucellosis prevalence (14.8%) than dairy farms (2.6%; χ2 = 3.75; df = 1; p = 0.05). Only 2.8% of the farmers had knowledge about brucellosis, while 39.1% and 41.6% had attitudes and farm practices with respect to brucellosis, respectively in the study area. Socio-economic attribute of training in animal husbandry (p<0.01), raising system (p<0.01), practice of separating the aborted cow (p<0.01) were negatively associated to brucellosis. The overall farm level brucellosis prevalence was strongly associated with cattle herd size (p = 0.01), free movement grazing practices (p<0.01), practice of self-removal of placental debris without using personal protective equipment (p<0.01) and farmers' attitudes towards eating cow placenta debris (p<0.01). Conclusion: Farmers had little knowledge of brucellosis. Attitudes and practices differed significantly between migratory and dairy farmers. Training and extension programs are necessary to make farmers aware of their KAP situation since livestock migration and the custom of eating cow placental debris contribute to the spread of brucellosis. Persistent efforts are required to reduce the adverse effects of brucellosis. Therefore, the study suggests that a feasible control intervention and public awareness campaigns need to be conducted regarding methods of preventing human exposure to brucellosis.

생태통로 식재수종의 현황 및 문제점 고찰 (The Existing Conditions and Problems of Ecological Corridor in Korea - Focusing on Planting Species -)

  • Ecological corridor is the facility to connect two habitats fragmented by roads, dam, housing and industrial area. Critics of faunal corridors have suggested that they are simply an untested application of unproved island biogeography theory. But, everyone agree that ecological corridor is important useful structural components that help animal movement in fragmented landscapes. There are 52 ecological corridors in Korea. We investigated 43 ecological corridors of these and recorded the size of corridor, planting species, management and growth condition. We compared planting species to nearby exiting vegetation species and structure. We found that there are no considerations for many aspects of planting, for example, planting species, structure, animal migration and behavior, surrounding vegetation etc. Expecially, It was ignored management of plants, difference of surrounding vegetation, lacks of diverse planting species and structure. We suggest planting the native species of tall tree, shrub and ground cover, edge planting for transitional space between road and natural habitat, planting for foraging and breeding of target animal, diverse layer vegetation, and open space of center for rapid animal dispersal.

Jewel of Thar Desert: Case study of a hidden wetland

  • Wetlands are very critical for the conservation of aquatic ecosystems, while also serving as the breeding/ nesting/ resting grounds for water birds. Generally, wetlands support both resident and migratory birds, thus serving as connecting dots in the global flyways. The Rajasthan state of India has two Ramsar sites (Keoladeo National Park and Sambhar Lake) and many other water bodies/wetlands. However, most of these areas are segregated in the eastern, southeastern, southern, and northern parts. In the western part of Rajasthan, where lies the Great Indian or Thar desert, there are no such reported prominent wetlands drawing attention towards a substantial number of resident and migratory water birds. The author is an Indian Forest Service officer, who was posted in the Thar Desert region and during that time had identified a hidden wetland in the desert landscape. This study deliberates on the wetland location and its faunal diversity with prospects of developing the area as a proper wetland conservation zone. India is a signatory to the Central Asian Flyway of migratory species and serves as an important member in terms of having significant wetlands and reported migratory birds count. The need of preserving and bring the arid zone's hidden wetlands to the forefront can serve as an important tool to conserve water birds and comply with worldwide bird migration conservation efforts.

Seasonal fluctuations and changing characteristics of a temperate zone wetland bird community

  • Background: The composition of wild bird populations in temperate zones greatly varies depending on phenological changes rather than other environmental factors. Particularly, wild birds appearing in wetlands fluctuate greatly due to the crossover of species arriving for breeding during the summer and for wintering. Therefore, to understand the changes to species composition related to phenology, we conducted this basic analysis of populations to further the cause of the protection of wetland-dependent wild birds. Methods: It is wrong to simply divide a wild bird population investigation into seasons. This study identifies species composition and indicator species that change along with seasons. Wetlands to be surveyed are protected by natural monuments and wetland inventory and are in a state close to nature. In order to identify as many species as possible in wetlands, a survey was conducted in both shallow and deep wetlands. The water depth varied in these areas, ranging from 0.2 to 2.0 m, allowing for both dabbling and diving ducks to inhabit the area. Surveys were conducted using line-transect and distance sampling methods and were conducted at intervals of 2 weeks. The survey was conducted under the following three categories: the eco-tone and emergent zone, the submergent zone, and the water surface. The survey was conducted along a wetland boundary by observing wild birds. A PC-ord program was used for clustering, and the SAS program was used to analyze the changes in species composition. The data strongly indicates that day length is the main factor for seasonal migration periods, despite the fact that climate change and increasing temperatures are often discussed. Results and conclusions: The indicator species for determining seasons include migrant birds such as Ardea cinerea, Alcedo atthis, Anas penelope, and Poiceps ruficollis, as well as resident birds such as Streptopelia orientalis and Emberiza elegans. Importantly, increases in local individual counts of these species may also serve as indicators. The survey results of seasonal fluctuations in temperate zones shows that spring (April to June), summer (July to September), autumn (October), and winter (November to March) are clearly distinguishable, even though spring and summer seasons tend to overlap, leading to the conclusion that additional research could more clearly identify fluctuation patterns in species composition and abundance in the study area.

칠산도 노랑부리백로 유조의 이소 후 하절기 행동권 (Home Range of Juvenile Chinese Egrets Egretta eulophotes during Post-fledging Stage in Chilsan Archipelago, Republic of Korea)

  • 번식지에서 이소하여 분산한 유조의 이동 패턴과 서식지를 파악하는 것은 개체 보전과 관리를 위해 매우 중요하다. 노랑부리백로는 국제적인 보호종으로 필리핀, 말레이시아, 대만 등지에서 월동하며, 중국, 러시아, 우리나라 서해 무인도서에서 번식한다. 본 연구에서는 2018년도와 2019년도에 영광 칠산도에서 번식하여 이소한 노랑부리백로 유조 총 6개체를 대상으로 야생동물위치추적장치를 부착하여 이소 후 하절기 동안 이동 경로 및 행동권 분석을 통해 주요 서식지 이용 양상을 파악하였다. 2018년도에 번식한 노랑부리백로 유조 3개체 중 CE1801 개체는 이소 후 북상을 하다가 다시 영광 백수 갯벌 지역으로 회귀하여 서식하였으며, 나머지 CE1802, CE1803 개체는 태안 지역으로 북상하여 서식하였다. 2019년도의 경우 CE1901 개체는 CE1801 개체와 유사한 패턴을 보였으며, CE1902 개체는 완도 지역으로 남하하여 서식하였고, CE1903 개체는 신안 지역으로 남하하다가 다시 영광 백수 갯벌 지역으로 회귀하여 핵심 행동권을 보였다. 본 연구에서 노랑부리백로 유조는 하절기 동안 안정적인 서식지를 찾기 위해 서해안 갯벌 지역을 중심으로 폭 넓게 이동을 하였다. 향후 노랑부리백로 유조가 이용하는 핵심 서식지에 대해 개발과 출입을 제한하는 등 서식지 보호를 위한 노력이 필요하다.

Origin and evolution of Korean ginseng revealed by genome sequence

  • 인삼은 세계에서 가장 중요한 약용식물 중 하나이다. 본 연구실에서는 국립종자원에 최초로 등록된 인삼 품종 '천풍'을 이용하여 대략 3Gbp의 완성도 높은 유전체 서열과 60,000여개의 유전자를 동정하여 공개하였다. 인삼속 근연종들과의 비교유전체연구를 통해 종의 분화 시기 등을 추정하였고, 이를 통해 고려인삼의 기원과 두 번의 대륙이동을 통한 인삼속의 진화와 분포모델을 확립하였다. 인삼속 18종 중 2종 (고려인삼, 화기삼)은 24쌍의 염색체를 가지는 사배체 식물이며 나머지 16종은 12쌍의 염색체를 가지는 이배체 식물이다. 인삼속과 두릅나무속은 두릅나무과에 속하는 가장 가까운 식물로서 약 8백만년 전에 분화하였다. 인삼속은 약 6백만년 전 베트남 등의 동남아시아에서 러시아와 같은 동북아시아에 이르는 지역의 깊은 숲 속 서늘한 기후와 숲 속의 음지조건에 적응하며 음지식물로 진화했다. 그 기간은 빙하기와 간빙기가 반복되는 시기로 월동 능력이 없는 이배체 인삼종은 대부분 동북아시아 지역에서 멸종하였고 이 과정에 이배체간 종간 교잡종인 이질사배체가 약 2백만년전 만들어졌으며 한반도를 위시한 동북아시아를 중심으로 월동능력을 가진 고려인삼이 태동되었다고 추정된다. 북미에 분포하는 화기삼은 동북아시아 전역에 분포하던 고려인삼이 약 1백만년전에 빙하의 이동과 더불어 대륙간 이주를 통해 새로운 생태 환경에 적응하면서 분화되었다고 판단된다. 반면 대부분의 이배체 인삼종은 고온을 견디지 못하고 월동능력도 없어 동남아시아 지역에서 1,600미터 이상의 고산 지역으로 쫓겨 올라가 연중 서늘한 기후에서 생존하고 있다. 유전체 해독 정보는 인삼의 기원과 진화기작을 추정하는 학문적 성과 뿐 아니라 인삼산업을 보호하고 우수 인삼을 개발하기 위한 실용적인 분자육종 수단에도 매우 효율적으로 활용될 수 있다.
