배추(Brassica campestris L. ssp. pekinensis) 형질전환 세포로부터의 식물체 재분화를 위한 선발체계를 확립하기 위하여 kanamycin, hygromycin, carbenicillin, cefotaxime등 4가지 항생제와 제초제 phosphinotricin가 자엽 및 배축 절편체로부터의 신초형성에 미치는 영향을 조사하였다. 자엽절편체는 kanamycin $1mg{\cdot}L^{-1}$처리에서 신초형성에 아무 영향을 받지 않았으나 $2mg{\cdot}L^{-1}$ 처리부터 형성수가 감소하여 $6mg{\cdot}L^{-1}$ 이상 처리구부터는 신초가 전혀 형성되지 않았다. 배축 절편체도 자엽과 비슷한 결과를 나타내어 kanamycin의 경우 $7mg{\cdot}L^{-1}$의 농도가 배추 형질전환체의 선발에 적당하다고 생각되었다. Hygromycin은 $4mg{\cdot}L^{-1}$ 처리부터 신초형성을 억제하여 배추형질전환을 위하여 낮은 농도로 사용할 수 있는 선발 마커로 생각된다. Cephalosporin type의 항생제인 carbenicillin과 cefotaxime은 신초형성에 아무 영향을 주지 않아 배추 절편체에 독성을 주지 않는 결과를 보여 주어, Agrobacterium과의 공동배양 후 이를 제거하는데 적당한 항생제라 할 수 있었다. 제초제인 phosphinotricin의 경우 $1mg{\cdot}L^{-1}$ 처리부터 신초 형성이 감소하기 시작하여 $2mg{\cdot}L^{-1}$ 이상부터는 완전히 억제되어 항생제와 더불어 형질전환 세포의 선발에 이용될 수 있다고 생각된다. 배추는 다른 배추속의 작물과 비교할 때 신초 재분화가 어렵기 때문에 비록 escape율이 높아진다고 해도 낮은 농도의 항생제나 제초제를 사용하여 선발압을 낮추어 주는 것이 형질전환에 효과적이라 생각된다.
본 연구에서는 국내에서 다소비 되는 십자화과 새싹채소 7종(콜라비, 적무, 브로콜리, 배추, 유채, 무순 및 다채)의 항산화 및 지방세포 내 ROS 생성 억제 활성을 측정하였다. 총 페놀 및 총 플라보노이드 함량을 측정한 결과, 총 페놀 함량은 콜라비 새싹과 무순 추출물이 $23.97{\pm}0.46mg\;TAE/g$ 및 $24.40{\pm}1.24mg\;TAE/g$으로 유의적으로 가장 높은 값을 나타내었고, 총 플라보노이드 함량은 무순 추출물이 $15.30{\pm}1.35mg\;CE/g$으로 다른 새싹채소 추출물에 비해 높은 값을 나타내었다. DPPH radical 소거능과 ORAC value를 측정한 결과, 콜라비가 DPPH radical 소거능($IC_{50}=1.95mg/mL$)과 ORAC value($79,032.5{\mu}M\;TE/g$)에서 모두 항산화능이 뛰어난 것으로 나타났다. 한편, 다채를 제외한 새싹채소 추출물 6종 모두에서 세포 내 ROS 생성량을 감소시킴을 확인하였다. 이상의 결과로부터, 십자화과 새싹채소 중 콜라비 새싹과 무순 추출물의 경우 항산화활성 및 지방세포 내 ROS 생성 억제 활성을 가지며, 천연물 유래 항산화 소재로써 활용 가능성이 높은 것으로 기대된다.
본 연구결과 독도의 식물종은 총 1문 3강 21목 29과 50속 48종 1아종 9변종 1품종으로 총 59종류가 생육하는 것으로 밝혀졌다. 동도의 경우 유채와 소리쟁이 등의 귀화식물이 넓은 면적에 분포하고 있으며. 독도 식생으로 보이는 갯제비쑥, 돌피, 땅채송화, 섬갯장대, 섬기린초 등이 피해를 받고 있다. 서도의 경우 등산로가 풍화작용으로 많이 훼손되어 상당히 위험하며, 섬정상부에서 북서쪽 아래로 왕호장근이 군락을 이루고 있다. 섬에 있는 나무로는 사철나무, 섬괴불나무 등이 있으며, 식재된 수종으로 해송과 무궁화가 현재 생육하고 있다. 독도에서 조사된 특정식물종은 참나리, 갯까치수영, 왕호장근 3종이며, 그 중 정밀생태종은 왕호장근 한 종으로 조사되었다. 독도는 귀화식물, 무분별한 건물의 건립과 조림사업으로 인하여 많이 훼손이 되었으며, 이에 대한 복원방안이 필요하다. 이를 위해서는 장, 단기적인 식생복원계획과 학제간의 연구. 지역단체의 관심이 필요하다.
Pectobacterium, which causes soft rot disease, is divided into 18 species based on the current classification. A total of 225 Pectobacterium strains were isolated from 10 main cultivation regions of potato (Solanum tuberosum), napa cabbage (Brassica rapa subsp. pekinensis), and radish (Raphanus sativus) in South Korea; 202 isolates (90%) were from potato, 18 from napa cabbage, and five from radish. Strains were identified using the Biolog test and phylogenetic analysis. The pathogenicity and swimming motility were tested at four different temperatures. Pectolytic activity and plant cell-wall degrading enzyme (PCWDE) activity were evaluated for six species (P. carotovorum subsp. carotovorum, Pcc; P. odoriferum, Pod; P. brasiliense, Pbr; P. versatile, Pve; P. polaris, Ppo; P. parmentieri, Ppa). Pod, Pcc, Pbr, and Pve were the most prevalent species. Although P. atrosepticum is a widespread pathogen in other countries, it was not found here. This is the first report of Ppo, Ppa, and Pve in South Korea. Pectobacterium species showed stronger activity at 28℃ and 32℃ than at 24℃, and showed weak activity at 37℃. Pectolytic activity decreased with increasing temperature. Activity of pectate lyase was not significantly affected by temperature. Activity of protease, cellulase, and polygalacturonase decreased with increasing temperature. The inability of isolated Pectobacterium to soften host tissues at 37℃ may be a consequence of decreased motility and PCWDE activity. These data suggest that future increases in temperature as a result of climate change may affect the population dynamics of Pectobacterium.
십자화과 작물중 양배추, 배추 및 유채의 자엽과 엽육조직을 재료로 하여 원형질체 나출과 배양 실험을 실시하여 다음과 같은 결과를 얻었다. 1. 양배추, 배추 및 유채의 자엽과 엽육조직은 품종과 사용부위의 차이없이 공히 1 % Macerozyme 'R-10' 과 1.5% Onozuka 'R-10'이 포항된 E3 효소 용액에서 4 시간 처리하였을 때 우량한 원형질체를 나출할 수 있었다. 2. MS, B5 및 $GK_2$ 배지에서는 갈변현상이 관찰되어 세포분열 유기에는 적합하지 않았으며, NN 배지에서 갈변현상 없이 양호한 세포분열이 이루어졌다. 3. 배추와 유채의 자엽에서 단리한 원형질체는 9.1% D-mannitol, 1% glucose, 1% sucrose, $1mg/{\ell}$ 2,4-D, $0.5mg/{\ell}$ NAA 및 $0.5mg/{\ell}$ BA를 첨가한 NN배지에서 세포분열과 budding을 하였다. 4. 양배추, 배추 및 유채의 엽육조직에서 단리한 원형질체는 9.1% D-mannitol, 1% grucose, 1% sucrose, $1mg/{\ell}$ 2,4-D, $0.5mg/{\ell}$ NAA 및 $0.5mg/{\ell}$ BA를 첨가한 배지에서 세포분열과 budding을 하였고, 특히 유채의 나출원형질체는 cell cluster를 거쳐 colony까지 분화하였다.
PDRN(Polydeoxyribonucleotide)은 조직재생 활성물질로 손상된 세포 및 조직의 자가 재생을 촉진하는 DNA 유래의 중합물질이다. PDRN은 DNA를 다양한 물리적 또는 화학적 방법으로 작은 크기로 절단한 DNA 조각으로 체내 투여시 조직세포 표면의 adenosine A2A receptor 수용체를 자극하여 세포 재생을 촉진하며 상처를 빠르게 회복시키고, 통증도 감소시키는 효과가 있다. 보통 어류의 정소나 정액으로부터 PDRN 추출을 하지만 본 연구에서는 다양한 식물에서 PDRN 추출 실험을 진행하였다. 실험 결과, 7종의 식물에서 PDRN 수율과 순수도는 단위 식물 중량 당 쑥갓이 가장 높았고, 브로콜리가 그 다음으로 우수했다. 이 두 식물의 PDRN을 대상으로 시험관에서 wound healing assay를 진행하여 PDRN의 효능을 분석한 결과, ㎍/ml 수준의 쑥갓과 브로콜리의 PDRN가 유의하게 wound healing 활성이 높음을 확인하였다. 본 연구의 결과는 이들 식물 유래 PDRN이 연어와 같은 어류 유래의 PDRN의 대체제로 사용할 수 있음을 의미한다.
BACKGROUND/OBJECTIVES: Glutathione S-transferase (GST) forms a multigene family of phase II detoxification enzymes which are involved in the detoxification of reactive oxygen species. This study examines whether daily supplementation of kale juice can modulate blood pressure (BP), levels of lipid profiles, and blood glucose, and whether this modulation could be affected by the GSTM1 and GSTT1 polymorphisms. SUBJECTS/METHODS: 84 subclinical hypertensive patients showing systolic BP over 130 mmHg or diastolic BP over 85 mmHg received 300 ml/day of kale juice for 6 weeks, and blood samples were collected on 0-week and 6-week in order to evaluate plasma lipid profiles (total cholesterol, triglyceride, HDL-cholesterol, and LDL-cholesterol) and blood glucose. RESULTS: Systolic and diastolic blood pressure was significantly decreased in all patients regardless of their GSTM1 or GSTT1 polymorphisms after kale juice supplementation. Blood glucose level was decreased only in the GSTM1-present genotype, and plasma lipid profiles showed no difference in both the GSTM1-null and GSTM1-present genotypes. In the case of GSTT1, on the other hand, plasma HDL-C was increased and LDL-C was decreased only in the GSTT1-present type, while blood glucose was decreased only in the GSTT1-null genotype. CONCLUSIONS: These findings suggest that the supplementation of kale juice affected blood pressure, lipid profiles, and blood glucose in subclinical hypertensive patients depending on their GST genetic polymorphisms, and the improvement of lipid profiles was mainly greater in the GSTT1-present genotype and the decrease of blood glucose was greater in the GSTM1-present or GSTT1-null genotypes.
xBrassicoraphanus is an intergenic breed crossed between Brassica campetris L. ssp. pekinensis and Raphanus sativus L. that have been daily consumed. xBrassicoraphanus was known to have good tastes and biological activities. Nevertheless, its constituetnts were not elucidated yet. Thus, in the present study, to indirectly evaluate the biological activity of xBrassicoraphanus, 12 compounds were isolated from leaves and roots of xBrassicoraphanus. On the basis of spectroscopic evidences, the structures of these compounds isolated from leaves of xBrassicoraphanus. were identified as ${\beta}-sitosterol$, indole-3-acetonitrile, ferulic acid, methyl ferulate, linolenic acid methyl ester, linolenic acid and coniferyl alcohol, while the chemical structures of compounds isolated from the roots of were xBrassicoraphanus were characterized as ${\beta}-sitosterol$, indole-3-acetonitrile, ferulic acid, methyl ferulate, linolenic acid methyl ester, 1-methoxyindole-3-acetonitrile, goitrin, 4-hydroxycinnamyl alcohol, coniferyl alcohol, palmitic acid and daucosterol. These can be classified as three steroids, two indole cyanides, two cinnamic acid derivatives, one cinnamyl alcohol derivative, three fatty acid derivatives one isothiocyanate. These results suggest that the compounds isolated from xBrassicoraphanus were almost identical with known components of Brassica campetris L. ssp pekinensis or Raphanus sativus L. However, it is necessary to investigate more about the difference of amounts of constituents according to harvest time and variant species amounts.
The major substrates used in soilless growing media are peat moss and perlite, where peat moss is a limited and expensive natural resource. Determination of appropriate substrates based on technical and economic feasibility is the vital aspect of research and the key to success in any soilless production system. This research work was performed to evaluate different low-cost and sustainable alternative substrate as soilless growing media for horticulture plant. The objective of this study was to compare the effect of sawdust species and steam treatment, for physico-chemical properties and growth of horticultural plant. This study involves the physical and chemical characterization and growth test of four substrate (pine sawdust, oak sawdust, steamed pine sawdust and steamed oak sawdust) in order to evaluate their use as components of growing media. Steamed oak sawdust ($121^{\circ}C$, 30 min) showed adequate physical and chemical properties compared to peat moss for their use as growing media. The growing media were prepared using different mixture proportion to grow Brassica campestris L., Festuca arundinacea and Lespedeza cyrtobotrya Miq. The highest germination, stem length and leaf area of Brassica campestris L., Festuca arundinacea and Lespedeza cyrtobotrya were observed in 30 minute steamed oak sawdust mixture growing media. The steam treatment condition of sawdust used in the growing media significantly positive affected the germination, the stem length and the leaf area.

Chromium (Cr) toxicity is hazardous to the seed germination, growth, and development of plants. ${\gamma}$-Aminobutyric acid (GABA) is a non-protein amino acid and is involved in stress tolerance in plants. To investigate the effects of GABA in alleviating Cr toxicity, we treated eight-d-old mustard (Brassica juncea L.) seedlings with Cr (0.15 mM and 0.3 mM $K_2CrO_4$, 5 days) alone and in combination with GABA ($125{\mu}M$) in a semi-hydroponic medium. The roots and shoots of the seedlings accumulated Cr in a dose-dependent manner, which led to an increase in oxidative damage [lipid peroxidation; hydrogen peroxide ($H_2O_2$) content; superoxide ($O{_2}^{{\cdot}-}$) generation; lipoxygenase (LOX) activity], MG content, and disrupted antioxidant defense and glyoxalase systems. Chromium stress also reduced growth, leaf relative water content (RWC), and chlorophyll (chl) content but increased phytochelatin (PC) and proline (Pro) content. Furthermore, supplementing the Cr-treated seedlings with GABA reduced Cr uptake and upregulated the non-enzymatic antioxidants (ascorbate, AsA; glutathione, GSH) and the activities of the enzymatic antioxidants including ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), glutathione reductase (GR), glutathione peroxidase (GPX), superoxide dismutase (SOD), catalase (CAT), glyoxalase I (Gly I), and glyoxalase II (Gly II), and finally reduced oxidative damage. Adding GABA also increased leaf RWC and chl content, decreased Pro and PC content, and restored plant growth. These findings shed light on the effect of GABA in improving the physiological mechanisms of mustard seedlings in response to Cr stress.
