인터넷중독 위험 청소년에 대한 단기 산림치유 효과: 생물학적, 신경인지적 및 심리-사회적 측면을 중심으로 (The Effect of Short-term Forest Therapy Camp on Youths with Internet Addiction Risk Group: Focused on the Biological, Neurocognitive and Psychosocial Aspects)

  • 본 연구에서는 인터넷중독 위험 청소년들에 대한 '산림치유(Forest Therapy)'의 생물학적, 신경인지적 및 심리사회적 영역에서의 효과를 살펴보았다. 청소년 인터넷중독 위험군을 대상으로 2박 3일 혹은 3박 4일 형태의 산림치유 캠프를 진행했고, 캠프 전-후로 참가자들의 혈청 뇌유래신경영양인자(BDNF), 종합주의력 검사(CAT) 그리고 우울(CDI) 및 상태 불안(STAI)을 포함한 다양한 심리사회적 변인들을 측정했다. 그 결과, 산림치유 캠프에 참가한 이후에 참가자들의 혈청 BDNF 수치 및 CAT의 수행점수가 증가했고, 인터넷/게임 욕구가 감소했으며, 회복탄력성 및 대인관계가 긍정적인 방향으로 변화하였다. 또한, 산림치유 캠프 노출량에 따라 집단을 구분하여 분석한 결과, 집단내에서 일부 불안 완화를 보고하기도 하였다. 전반적으로 인터넷중독 위험 청소년의 증상 개선 및 적응력 증가를 위한 산림치유의 적용가능성을 확인하였다.

알츠하이머 질환 마우스에서 중첩주파수를 활용한 미세전류가 인지능력 개선에 미치는 효과 (Effect of Microcurrent Wave Superposition on Cognitive Improvement in Alzheimer's Disease Mice Model)

  • 본 연구에서는 Alzheimer's disease(AD) 마우스 모델에서 미세전류의 적용을 통한 인지능력 개선 효과를 확인하였다. ICR 마우스에 amyloid beta($A{\beta}$)를 뇌 내 주입하여 인지능력 손상을 유도한 후, 4가지 파형의 미세전류를 각각 적용하여 손상된 인지능력에 미치는 미세전류의 영향을 검토하였다. AD 마우스의 공간 및 물체 인지능력을 확인하기 위해 행동실험을 실시한 결과, novel object recognition test와 Morris water maze test에서 $A{\beta}$로 인해 손상되었던 인지능력이 미세전류 적용군에서 유의적으로 개선됨을 확인하였으며, 지질과산화 반응으로 인한 malondialdehyde의 뇌 내 생성량 또한 감소하였다. 뇌 조직에서 AD 관련 단백질 발현을 측정한 결과, 특히 미세전류 Wave4 [STEP FORM 파형(0, 1.5, 3, 5V), 중첩Hz 적용] 적용군에서 $A{\beta}$ 생성 관련 단백질인 ${\beta}$-secretase, presenilin 1, presenilin 2의 발현이 감소하였고 신경영양인자인 brain-derived neurotrophic factor 단백질 발현이 증가하였다. 이 결과를 바탕으로 AD 마우스에서 미세전류를 이용한 손상된 인지능력에 대한 개선 효과를 확인하였으며, AD 예방 및 치료를 위한 비약물적인 방법으로서 적용할 수 있을 것으로 기대된다.

Effects of Acupuncture at GB30, GB34, and BL40 on Functional Recovery after Sciatic Crushed Nerve Injury in Rats

  • Background: Peripheral nerve injuries are a commonly-encountered clinical problem and often result in a chronic pain and severe functional deficits. Objectives: The aim of this study was to evaluate the effects of acupuncture on the descending pain and the recovery of the locomotor function that follows sciatic crushed nerve injury in rats. Method: In order to assess the effects of acupuncture on the descending pain and functional recovery, we investigated the walking track analysis, brain-derived neurotrophic factor (BDNF) and its receptor tyrosine receptor kinase B (TrkB) expression in the sciatic nerve, and on the expressions of c-Fos and nitric oxide synthase in the paraventricular nucleus (PVN) of the hypothalamus and in the ventrolateral periaqueductal gray (vlPAG) region resulting from sciatic crushed nerve injury in rats. Results: Acupuncture treatment at Huantiao (GB30), Yanglingquan (GB34), and Weizhong (BL40) facilitated functional recovery. C-Fos and nitric oxide synthase expressions in the brain and BDNF and TrkB expressions in the sciatic nerve were decreased by acupuncture treatment. The most potent effects of acupuncture were observed at the GB30 acupoint. Conclusion: It is possible that acupuncture can be used for pain control and functional recovery from sciatic nerve injury.

Bortezomib Is Toxic but Induces Neurogenesis and Inhibits TUBB3 Degradation in Rat Neural Stem Cells

  • Bortezomib (BTZ) is a proteasome inhibitor used to treat multiple myeloma (MM). However, the induction of peripheral neuropathy is one of the major concerns in using BTZ to treat MM. In the current study, we have explored the effects of BTZ (0.01-5 nM) on rat neural stem cells (NSCs). BTZ (5 nM) induced cell death; however, the percentage of neurons was increased in the presence of mitogens. BTZ reduced the B-cell lymphoma 2 (Bcl-2)/Bcl-2 associated X protein ratio in proliferating NSCs and differentiated cells. Inhibition of βIII-tubulin (TUBB3) degradation was observed, but not inhibition of glial fibrillary acidic protein degradation, and a potential PEST sequence was solely found in TUBB3. In the presence of growth factors, BTZ increased cAMP response element-binding protein (CREB) phosphorylation, brain-derived neurotrophic factor (Bdnf) transcription, BDNF expression, and Tubb3 transcription in NSCs. However, in the neuroblastoma cell line, SH-SY5Y, BTZ (1-20 nM) only increased cell death without increasing CREB phosphorylation, Bdnf transcription, or TUBB3 induction. These results suggest that although BTZ induces cell death, it activates neurogenic signals and induces neurogenesis in NSCs.

안면신경 압박손상 후 Adenovirus 매개 BDNF 유전자 전달을 통한 신경손상 회복에 관한 연구 (ADENOVIRAL VECTOR MEDIATED IN VIVO GENE TRANSFER OF BDNF PROMOTE FUNCTIONAL RECOVERY AFTER FACIAL NERVE CRUSH INJURY)

  • Objectives Despite considerable advances in technique, experience and skill, the precise place of surgery in the treatment of facial nerve injury remains uncertain. We designed a facial nerve crush injury model in rats and evaluated the recovery of crushed nerve which is the most common injury type of facial nerve using adenovirus vector mediated in vivo gene transfer of Brain derived neurotrophic factor(BDNF). Materials and methods In 48 Sprague Dawley rats, we made a facial nerve crush injury model to main trunk before the furcation, and injected a $10^{11}$pfu adenoviral BDNF in experimental group(BDNF adenoviral injection group; ad-BDNF) and $3{\mu}l$ saline in control group(Saline injection group; saline). After a period of regeneration from 10 to 40 days, nerve regeneration was evaluated with functioinal test (vibrissae and ocular movement), electrophysiologic study(threshold, peak voltage, conduction velocity) and histomorphometric study of axon density. Results Vibrissae and ocular movement, threshold and conduction velocity improved as time elapse in both group, however axon density was increased significantly only in experimental group. Functional test in 10 days and 20 days showed no difference between experimental group and control group. Vibrissae movement, threshold, conduction velocity and axon density in 30 days revealed that the regeneration in quality of experimental group was significantly superior to that of control group. Conclusion In general, there is tendency for nerve regeneration in experimental group (BDNF-adenovirus injection group) during 40 days, functional recovery was detected successfully after facial nerve crush in 30 days postoperatively.

3-Acetylpyridine에 의한 운동실조 동물모델에서 로타로드 운동과 전침이 근활성도와 혈청 BDNF에 미치는 영향 (Effects of Rotarod Exercise and Electroacupuncture on Muscle Activity and Serum BDNF Level in the Ataxic Rats by the 3-Acetylpyridine)

  • 3-Acetylpyridine(3-AP)는 아래올리브핵을 선택적으로 파괴함으로써, 소뇌 손상을 유발하는 신경독소이다. 본 연구에서는 3-AP의 투여로 운동실조가 유발된 동물모델에서 로타로드 운동과 전침이 후지의 근활성도와 혈청 BDNF에 미치는 영향을 알아보고자 하였다. 본 연구를 위해 실험동물을 대조군, 3-AP군, 운동군, 전침군 그리고 운동전침군으로 무작위 배치하였다. 3-AP군의 최대점핑수직높이는 대조군에 비해 유의하게 감소하였고, 로타로드 운동과 전침의 적용 이후 점차 증가하는 것으로 나타났다. 또한 후지의 근활성도는 3-AP의 투여로 유의하게 증가하였고, 치료적 중재 이후 약간의 감소를 보였다. 3-AP군의 혈청 BDNF 농도는 대조군보다 유의하게 증가하였고, 운동군, 전침군 그리고 운동전침군에서는 3-AP군보다 감소하는 것으로 나타났다. 이러한 결과를 통해 로타로드 운동과 전침은 운동실조 동물모델의 기능적 회복에 긍정적인 치료효과를 가지는 것으로 생각된다.

우울증의 후성유전기전: BDNF 유전자의 히스톤 변형 및 DNA 메틸화의 역할 (Epigenetic Mechanisms of Depression: Role of Histone Modification and DNA Methylation in BDNF Gene)

  • 우울증은 심각하며 재발하는 흔한 정신질환이다. 우울증은 환경 요인과 유전 요인, 그리고 신경생물학적 체계의 구조 및 기능의 변화로 발병한다. 후성유전학적 변화가 우울증과 관련 된다는 여러 연구들이 보고되었다. 후성 유전은 환경 요인이 크로마틴 구조를 변화시켜 DNA 염기 서열 변화 없이 유전자 발현을 조절하는 기전으로 설명된다. DNA 메틸화와 히스톤 아세틸화 및 메틸화를 포함하고 있는 히스톤 변형이 주요 후성유전기전으로 알려져 있다. 우울증 동물모델연구에서는 생애 초기 스트레스 같은 스트레스 환경이 게놈에 지속적으로 후성유전표지를 남기게 되고 이로 인해 유전자 발현이 변화되고 결국 성체가 되었을 때 신경 기능이나 행동 기능에 영향을 미치게 된다고 설명하고 있다. BDNF는 우울증과 관련된 대표적인 유전자로 알려져 있다. 설치류가 출생 전, 후, 그리고 성체 기간에 스트레스에 노출되면 해마에서 BDNF 유전자의 히스톤 변형과 DNA 메틸화 패턴이 변화되고 이로 인해 BDNF 발현이 변화된다. 이러한 과정은 불안과 우울 행동에도 영향을 미치게 된다. 본 종설에서는 BDNF 유전자의 히스톤 변형 및 DNA 메틸화와 같은 우울증 발병에 관여하는 후성유전기전의 최신 지견에 대해 논의하여 우울증 치료의 새로운 타겟 개발에 도움이 되고자 한다.

Smoking Habits and Neuropeptides: Adiponectin, Brain-derived Neurotrophic Factor, and Leptin Levels

  • This study aimed to identify changes in the level of neuropeptides among current smokers, former smokers, and individuals who had never smoked, and how smoking habits affect obesity and metabolic syndrome (MetS). Neuropeptide levels, anthropometric parameters, and metabolic syndrome diagnostic indices were determined among male workers; 117 of these had never smoked, whereas 58 and 198 were former and current smokers, respectively. The total sample comprised 373 male workers. The results obtained from anthropometric measurements showed that current smokers attained significantly lower body weight, body mass index, waist circumference, and abdominal fat thickness values than former smokers and those who had never smoked. Current smokers' eating habits proved worse than those of non-smokers and individuals who had never smoked. The level of brain-derived neurotrophic factor (BDNF) in the neuropeptides in the case of former smokers was $23.6{\pm}9.2pg/ml$, higher than that of current smokers ($20.4{\pm}6.1$) and individuals who had never smoked ($22.4{\pm}5.8$) (F = 6.520, p = 0.002). The level of adiponectin among former smokers was somewhat lower than that of current smokers, whereas leptin levels were higher among former smokers than current smokers; these results were not statistically significant. A relationship was found between adiponectin and triglyceride among non-smokers (odds ratio = 0.660, ${\beta}$ value=-0.416, p < 0.01) and smokers (odds ratio = 0.827, ${\beta}$ value=-0.190, p < 0.05). Further, waist circumference among non-smokers (odds ratio = 1.622, ${\beta}$ value=0.483, p < 0.001) and smokers (odds ratio = 1.895, ${\beta}$ value=0.639, p < 0.001) was associated with leptin. It was concluded that cigarette smoking leads to an imbalance of energy expenditure and appetite by changing the concentration of neuropeptides such as adiponectin, BDNF, leptin, and hsCRP, and influences food intake, body weight, the body mass index, blood pressure, and abdominal fat, which are risk factors for MetS and cardiovascular disease.

A possible mechanism to the antidepressant-like effects of 20 (S)-protopanaxadiol based on its target protein 14-3-3 ζ

  • Background: Ginsenosides and their metabolites have antidepressant-like effects, but the underlying mechanisms remain unclear. We previously identified 14-3-3 ζ as one of the target proteins of 20 (S)-protopanaxadiol (PPD), a fully deglycosylated ginsenoside metabolite. Methods: Corticosterone (CORT) was administered repeatedly to induce the depression model, and PPD was given concurrently. The tail suspension test (TST) and the forced swimming test (FST) were used for behavioral evaluation. All mice were sacrificed. Golgi-cox staining, GSK 3β activity assay, and Western blot analysis were performed. In vitro, the kinetic binding analysis with the Biolayer Interferometry (BLI) was used to determine the molecular interactions. Results: TST and FST both revealed that PPD reversed CORT-induced behavioral deficits. PPD also ameliorated the CORT-induced expression alterations of hippocampal Ser9 phosphorylated glycogen synthase kinase 3β (p-Ser9 GSK 3β), Ser133 phosphorylated cAMP response element-binding protein (p-Ser133 CREB), and brain-derived neurotrophic factor (BDNF). Moreover, PPD attenuated the CORT-induced increase in GSK 3β activity and decrease in dendritic spine density in the hippocampus. In vitro, 14-3-3 ζ protein specifically bound to p-Ser9 GSK 3β polypeptide. PPD promoted the binding and subsequently decreased GSK 3β activity. Conclusion: These findings demonstrated the antidepressant-like effects of PPD on the CORT-induced mouse depression model and indicated a possible target-based mechanism. The combination of PPD with the 14-3-3 ζ protein may promote the binding of 14-3-3 ζ to p-GSK 3β (Ser9) and enhance the inhibition of Ser9 phosphorylation on GSK 3β kinase activity, thereby activating the plasticity-related CREBeBDNF signaling pathway.

Fermented Saccharina japonica (Phaeophyta) improves neuritogenic activity and TMT-induced cognitive deficits in rats

  • Marine organisms are frequently used to be harmful and have lower side effects than synthetic drugs. The cognitive improving efficacy of gamma aminobutyric acid-enriched fermented Saccharina japonica (FSJ) on the memory deficient rats, which were induced by trimethyltin chloride (TMT), was investigated by assessing the Morris water maze test and by performing choline acetyltransferase (ChAT), cAMP response element binding protein (CREB), and brain derived neurotrophic factor (BDNF) immunohistochemistry. The neurite outgrowth of Neuro2a cells was assessed in order to examine the underlying mechanisms of the memory enhancing effects of FSJ. Treatment with FSJ tended to shorten the latency to find the platform in the acquisition test of the Morris water maze at the second and fourth day compared to the control group. In the probe trial, the FSJ treated group increased time spent in the target quadrant, compared to that of the control group. Consistent with the behavioral data, these treatments recovered the loss of ChAT, CREB, and BDNF immunepositive neurons in the hippocampus produced by TMT. Treatment with FSJ markedly stimulated neurite outgrowth of the Neuro2a cells as compared to that of the controls. These findings demonstrate that FSJ may be useful for improving the cognitive function via regulation of neurotrophic marker enzyme activity.
