Abrogation of the Circadian Nuclear Receptor REV-ERBα Exacerbates 6-Hydroxydopamine-Induced Dopaminergic Neurodegeneration

  • Parkinson's disease (PD) is a neurodegenerative disease characterized by progressive degeneration of dopaminergic (DAergic) neurons, particularly in the substantia nigra (SN). Although circadian dysfunction has been suggested as one of the pathophysiological risk factors for PD, the exact molecular link between the circadian clock and PD remains largely unclear. We have recently demonstrated that $REV-ERB{\alpha}$, a circadian nuclear receptor, serves as a key molecular link between the circadian and DAergic systems. It competitively cooperates with NURR1, another nuclear receptor required for the optimal development and function of DA neurons, to control DAergic gene transcription. Considering our previous findings, we hypothesize that $REV-ERB{\alpha}$ may have a role in the onset and/or progression of PD. In the present study, we therefore aimed to elucidate whether genetic abrogation of $REV-ERB{\alpha}$ affects PD-related phenotypes in a mouse model of PD produced by a unilateral injection of 6-hydroxydopamine (6-OHDA) into the dorsal striatum. $REV-ERB{\alpha}$ deficiency significantly exacerbated 6-OHDA-induced motor deficits as well as DAergic neuronal loss in the vertebral midbrain including the SN and the ventral tegmental area. The exacerbated DAergic degeneration likely involves neuroinflammation-mediated neurotoxicity. The $REV-erb{\alpha}$ knockout mice showed prolonged microglial activation in the SN along with the over-production of interleukin $1{\beta}$, a pro-inflammatory cytokine, in response to 6-OHDA. In conclusion, the present study demonstrates for the first time that genetic abrogation of $REV-ERB{\alpha}$ can increase vulnerability of DAergic neurons to neurotoxic insults, such as 6-OHDA, thereby implying that its normal function may be beneficial for maintaining DAergic neuron populations during PD progression.

  • Brain-computer interface is (BCI) is a communication device that the brain activity is directly input to the computer without input devices, such as a mouse or keyboard. As the brain wave interface hardware technology evolves, expensive and large EEG equipment has been downsized cheaply. So it will be applied to various multimedia applications. Among BCI studies, we suggest the domestic and foreign research trend about how the BCI is applied about the game almost people use. Next, look at the problems of the game with the BCI, we would like to propose the future direction of domestic BMI research and development.

  • In this study we suggest that the spatial correlation structure of the brain fMRI data be used to characterize the functional connectivity of the brain. For some concussion and recovery data, we examine how the correlation structure changes from one step to another in the data analyses, which will allow us to see the effect of each analysis to the spatial correlation or the functional connectivity of the brain. This will lead us to spot the processes which cause significant changes in the spatial correlation structure of the brain. We discuss whether or not we can decompose correlation matrices in terms of its causes of variations in the data.

  • The brain is an organ that consists of various cell types. As our knowledge of the structure and function of the brain progresses, cell type-specific research is gaining importance. Together with advances in sequencing technology and bioinformatics, cell type-specific transcriptome studies are providing important insights into brain cell function. In this review, we discuss 3 different cell type-specific transcriptome analyses i.e., Laser Capture Microdissection (LCM), Translating Ribosome Affinity Purification (TRAP)/RiboTag, and single cell RNA-Seq, that are widely used in the field of neuroscience.   • In this paper, we proposed a brain-computer interface system using EEG signals. It can generate 4 direction command signal from EEG signals captured during imagination of subjects. Bandpass filter used for preprocessing to detect the brain signal, and the power spectrum at a specific frequency domain of the EEG signals for concentration status and non-concentration one is used for feature. In order to generate an adequate signal for controlling the 4 direction movement, we propose a new interface system implemented by using a support vector machine and a time-multiplexing method. Moreover, bio-feed back process and on-line adaptive pattern recognition mechanism are also considered in the proposed system. Computer experimental results show that the proposed method is effective to recognize the non-stational brain wave signal.

  • Boron neutron capture therapy is a precision treatment technology that selectively destroys only tumor cells by irradiating thermal neutrons after accumulating boron drugs in tumor cells. Brain tumor is difficult to diagnose and treat due to the low permeability and targeting of drugs caused by the blood-brain-barrier. Crossing the BBB is essential for drug delivery to the brain. In this study, we designed and synthesized a novel compound incorporating benzothiazole to develop a boron drug with high BBB permeability and selectivity for brain tumor cells. In addition, their potential as a BNCT drugs was evaluated.

  • This study examines the possibility of chronic brain disease rehabilitation treatment using VR/AR technology and raises the research need for the development of digital therapeutics. In addition, by proposing a digital therapeutic research and development process this study intends to contribute to the development of VR/AR rehabilitation treatment. To this end, this study identified research trends of VR/AR technology, neurophysiology, and chronic brain disease and proposed a method for applying VR/AR technology to treat chronic brain disease patients in three stages. The first stage is to prepare a neurophysiological basis for rehabilitation treatment of brain disease patients using VR technology. The second stage is to provide a treatment method using VR/AR technology and systematize the contents characteristics. The third stage is to conduct clinical trails and validate that the treatment method and contents utilization is effective for the patient. It is hope that this study serves as a guide for developing media production base technology for treating patient with a chronic brain disease.

  • As an essential part of the urban transportation system, precise perception of the traffic flow parameters at the traffic signal intersection ensures traffic safety and fully improves the intersection's capacity. Traditional detection methods of road traffic flow parameter can be divided into the micro and the macro. The microscopic detection methods include geomagnetic induction coil technology, aerial detection technology based on the unmanned aerial vehicles (UAV) and camera video detection technology based on the fixed scene. The macroscopic detection methods include floating car data analysis technology. All the above methods have their advantages and disadvantages. Recently, indoor location methods based on wireless signals have attracted wide attention due to their applicability and low cost. This paper extends the wireless signal indoor location method to the outdoor intersection scene for traffic flow parameter estimation. In this paper, the detection scene is constructed at the intersection based on the received signal strength indication (RSSI) ranging technology extracted from the wireless signal. We extracted the RSSI data from the wireless signals sent to the road side unit (RSU) by the vehicle nodes, calibrated the RSSI ranging model, and finally obtained the traffic flow parameters of the intersection entrance road. We measured the average speed of traffic flow through multiple simulation experiments, the trajectory of traffic flow, and the spatiotemporal map at a single intersection inlet. Finally, we obtained the queue length of the inlet lane at the intersection. The simulation results of the experiment show that the RSSI ranging positioning method based on wireless signals can accurately estimate the traffic flow parameters at the intersection, which also provides a foundation for accurately estimating the traffic flow state in the future era of the Internet of Vehicles.

  • Recent advances in magnetic resonance imaging and identification of causative genes led to the recognition of a new group of disorders named neurodegeneration with brain iron accumulation (NBIA). NBIA is a group of inherited disorders characterized by abnormal iron deposition in the brain, usually in the basal ganglia. The disorder shares the clinical features of movement disorders and is accompanied by varying degrees of neuropsychiatric abnormalities. In this review, the causative genes, clinical presentations, neuroimaging features, and pathological findings are summarized.
