Comparison of Quality of Life of Turkish Breast Cancer Patients Receiving Breast Conserving Surgery or Modified Radical Mastectomy

  • Purpose: The aim of this study was to investigate the differences in quality of life in patients who received breast conserving surgery (BCS) or modified radical mastectomy (MRM) for breast cancer. Materials and Methods: A total of 100 women with breast cancer who underwent either BCS or MRM between September 2011 and April 2012 at a private health center and completed their chemotherapy and radiation therapy cycles were included in the study. To assess the quality of life, we used a demographic questionnaire, the European Organization for the Research and Treatment of Cancer Quality of Life Questionnaire (EORTC QLQ-C30) and the Quality of Life assessment in Breast Cancer (EORTC QLQ-BR23). Results: Using QLQ-C30, we found that patients who underwent BCS had better functional status and fewer symptoms than patients who underwent MRM. In QLQ-BR23, independent factors improving the functional scales were BCS, higher level of education and marital status (married); independent factors improving symptoms were BCS, higher level of education, younger age and low and normal body mass index (BMI). In QLQ-C30, independent factors affecting the functional and symptom scales were only BCS and higher level of education. Conclusions: We determined that patients who received BCS had better functional status and less frequent symptoms than patients who underwent MRM.

  • Plasmid Achromobacter secretion (PAS) factor is a putative secretion factor that induces the secretion of periplasmic proteins. PAS factor from Vibrio vulnificus was crystallized at 294 K by the hanging drop vapor-diffusion method. It was isolated as a monomer during the purification procedures. The native crystal belongs to the F222 space group with unit cell parameters a = 56.1, b = 74.4, $c=80.0{\AA}$, ${\alpha}={\beta}={\gamma}=90^{\circ}$. The crystal was soaked in cryoprotectant containing 1 M NaBr for 1 h for MAD phasing. The diffraction limit of the Br-MAD data set was $1.9{\AA}$ using synchrotron X-ray irradiation at beam line BL-18B at the Photon Factory, Japan.

제주흑우와 한우에서 Extension, Agouti 유전자형과 모색 출현 양상 (Coat Color Patterns and Genotypes of Extension and Agouti in Hanwoo and Jeju Black Cattle)

  • 소의 Extension (E), Agouti (A) 유전자형과 모색유전양상의 연관관계를 이해하기 위해, 한우와 제주흑우(JBC), 이들 사이에서 생산된 교배후손에서 melanocortin-1 receptor (MC1R)과 agouti signaling protein (ASIP)의 유전자형을 분석하였다. 흑모색의 JBC에서는 세 가지 MC1R 대립인자들($E^D$, $E^+$, e)이 모두 발견되었다. JBC 집단은 한우에서 우점적으로 나타나는 열성동형접합자(e/e)가 발견되지 않았다. 반면, MC1R $E^+$/e 한우는 흑모색이 아닌 갈색이나 적색의 모색을 나타내었다. ASIP 유전자에서, 한우에서는 L1-BT 전위인자의 삽입/결실에 의해 세 가지 유전자형(A/A, A/$A^{Br}$, $A^{Br}/A^{Br}$)이 모두 관찰되었다. JBC 집단에서는 ASIP $A^{Br}$ 대립인자가 드물게 관찰되고, ASIP $A^{Br}/A^{Br}$ 동형접합인 개체들은 발견되지 않았다. ASIP $A^{Br}$ 대립인자를 보유하더라도 한우와 JBC, 이들의 교배후손에서도 agouti-유사 얼룩무늬 모색은 발견되지 않았다. 교배후손의 모색은 Agouti 유전자형과는 무관하고, Extension 유전자형과 직접적으로 연관되어 황-갈색(e/e)과 암-갈/흑색($E^+$/e)으로만 구분되었다. ASIP 유전자형은 아마도 한우 또는 교배 후손에서 모색의 발달에 영향이 없는 것으로 추정된다. 본 연구 결과는 한우, 제주흑우, 이들의 교배후손에서 MC1R 유전자형이 모색 표현형을 결정하며, ASIP 유전자형은 모색의 변이에는 핵심적인 역할을 수행하지 않는다는 것을 시사한다.

Lactobacillus plantarum K-1 BR 유래 박테리오신의 여드름균에 의한 염증성 사이토카인 억제 효과 (The Suppression of Inflammatory Cytokines Induced by Propionibacterium acnes Using Bacteriocin Isolated from Lactobacillus plantarum K-1 BR)

  • 본 실험은 유산균 유래의 박테리오신이 여드름균에 의한 염증성 사이토카인의 발현을 억제하는 효능을 가지는지를 확인하기 위해 수행되었다. 본 실험을 위해 인간의 각질형성세포인 HaCaT 세포에 열처리한 여드름균인 P. acnes을 농도별로 처리하여 염증성 사이토카인인 TNF-α, IFN-γ, IL-8의 발현을 유도하였고, 여드름균과 박테리오신을 HaCaT 세포에 함께 처리하여 여드름균에 의해 증가된 사이토카인의 양이 박테리오신에 의해 어떻게 변화하는지를 확인하였다. P. acnes 균을 1×105, 1×106 그리고 1×107 CFU/ml와 같이 농도별로 처리한 결과, 사이토카인의 발현량이 농도 의존적으로 증가하였는데 TNF-α와 IFN-γ의 발현량은 여드름균을 1×107 CFU/ml로 처리했을 때 두 배 이상 증가하였고 IL-8의 발현량은 네 배 이상 증가하였다. 그러나 여드름균에 의해 증가되었던 TNF-α, IFN-γ 그리고 IL-8의 발현량은 박테리오신을 처리하였을 때 농도 의존적으로 유의하게 감소하였다. 이상의 결과로 보아 김치유산균인 L. plantarum K-1 BR 균주에서 분리한 박테리오신은 여드름균에 의한 염증유발을 완화시키는 효과가 있다고 할 수 있다.

  • 단백질 인산화는 세포의 활동을 조절하는 보편적인 과정이다. 브라시노스테로이드(brassinostreoid)에 의해 매개되는 신호전달은 브라시노스테로이드에 의해 활성화된 세포막상의 protein kinase 로부터 인산화되어 있는 전사인자들을 탈인산화하는 연속적인 인산화/탈인산화 과정이다. 브라시노스테로이드에 의해 매개되는 신호전달의 연구는 인산화에 관여하는 kinase 기질상의 아미노산을 밝히고, 그와 관련된 돌연변이체의 표현형을 알아봄으로써 급속하게 발전하였다. BRI1과 BAK1의 자기인산화(autophosphorylation), 상호인산화(transphosphorylation), 타이로신 인산화(tyrosine phosphorylation)를 밝힘으로써 그들의 조절작용을 식물의 생리학적, 발생학적 과정을 더 이해할 수 있는 장이 열렸다. 브라시노스테로이드에 의한 인산화는 수용체에 의해 매개되는 세포 내 함입(endocytosis)과 그에 뒤따르는 수용체의 파괴현상에서도 볼 수 있다. 인산화/탈인산화 과정에 관련하여 브라시노스테로이드에 의해 매개되는 신호전달은 더 연구할 여지가 많이 남아 있다. 이 총설은 단백질의 인산화/탈인산화 과정을 통한 브라시노스테로이드의 신호전달 연구의 최근 상황을 기술하였다.

환자교육을 위한 유방암 환자의 지식측정 도구개발 (Development and Validation of a Knowledge Scale for Patients with Breast Cancer (KS-Br))

  • Purpose: The purpose of this study was the development and validation of knowledge scale for patients with breast cancer (KS-Br) in Korea. Methods: The process included the construction of a conceptual framework, generation of preliminary items, and the test of validity and reliability of the scale. Thirty-seven items were developed through an evaluation process by 10 experts and 24 items were finally confirmed through item analysis. Psychometric testing was performed with a convenient sample of 303 women with breast cancer. The data was analyzed using independent t-test, Pearson's correlation, and calculation of KR-20. Results: Participants averaged 70.8% correct on the test. The KS-Br has 24 items consisting of 5 categories: incidence of breast cancer, diagnosis and treatment, symptom management, sexuality, and maintenance of daily life. Validity was supported by the use of content validity, known-group technique, and criterion-related validity. Women who had undergone education intervention scored significantly higher than women who had not (p<.001). KS-Br scores were significantly correlated with those of Mishel's Illness Uncertainty Scale (r=-.214, p<.001). Internal consistency of the KS-Br was appropriate (KR20=.805). Conclusion: This study reveals that the KS-Br is reliable and valid scale to measure the knowledge of breast cancer. Therefore, this scale can be effectively utilized to assess the knowledge of patients with breast cancer regarding their disease.

Expression of BrD1, a Plant Defensin from Brassica rapa, Confers Resistance against Brown Planthopper (Nilaparvata lugens) in Transgenic Rices

  • Plant defensins are small (5-10 kDa) basic peptides thought to be an important component of the defense pathway against fungal and/or bacterial pathogens. To understand the role of plant defensins in protecting plants against the brown planthopper, a type of insect herbivore, we isolated the Brassica rapa Defensin 1 (BrD1) gene and introduced it into rice (Oryza sativa L.) to produce stable transgenic plants. The BrD1 protein is homologous to other plant defensins and contains both an N-terminal endoplasmic reticulum signal sequence and a defensin domain, which are highly conserved in all plant defensins. Based on a phylogenetic analysis of the defensin domain of various plant defensins, we established that BrD1 belongs to a distinct subgroup of plant defensins. Relative to the wild type, transgenic rices expressing BrD1 exhibit strong resistance to brown planthopper nymphs and female adults. These results suggest that BrD1 exhibits insecticidal activity, and might be useful for developing cereal crop plants resistant to sap-sucking insects, such as the brown planthopper.

Metabolic Engineering of Rational Screened Saccharopolyspora spinosa for the Enhancement of Spinosyns A and D Production

  • Spinosyns A and D are potent ingredient for insect control with exceptional safety to non-target organisms. It consists of a 21-carbon tetracyclic lactone with forosamine and tri-Omethylated rhamnose which are derived from S-adenosyl-methionine. Although previous studies have revealed the involvement of metK1 (S-adenosylmethionine synthetase), rmbA (glucose-1-phosphate thymidylyltransferase), and rmbB (TDP-D-glucose-4, 6-dehydratase) in the biosynthesis of spinosad, expression of these genes into rational screened Saccharopolyspora spinosa (S. spinosa MUV) has not been elucidated till date. In the present study, S. spinosa MUV was developed to utilize for metabolic engineering. The yield of spinosyns A and D in S. spinosa MUV was $244mgL^{-1}$ and $129mgL^{-1}$, which was 4.88-fold and 4.77-fold higher than that in the wild-type ($50mgL^{-1}$ and $27mgL^{-1}$), respectively. To achieve the better production; positive regulator metK1-sp, rmbA and rmbB genes from Streptomyces peucetius, were expressed and co-expressed in S. spinosa MUV under the control of strong $ermE^*$ promoter, using an integration vector pSET152 and expression vector pIBR25, respectively. Here-with, the genetically engineered strain of S. spinosa MUV, produce spinosyns A and D up to $372/217mgL^{-1}$ that is 7.44/8.03-fold greater than that of wild type. This result demonstrates the use of metabolic engineering on rationally developed high producing natural variants for the production.

Analysis of Phosphorylation of the BRI1/BAK1 Complex in Arabidopsis Reveals Amino Acid Residues Critical for Receptor Formation and Activation of BR Signaling

  • The plasma membrane-localized BRASSINOSTEROID-INSENSITIVE1 (BRI1) and BRI1-ASSOCIATED KINASE1 (BAK1) are a well-known receptor pair involved in brassinosteroids (BR) signaling in Arabidposis. The formation of a receptor complex in response to BRs and the subsequent activation of cytoplasmic domain kinase activity share mechanistic characteristics with animal receptor kinases. Here, we demonstrate that BRI1 and BAK1 are BR-dependently phosphorylated, and that phosphorylated forms of the two proteins persist for different lengths of time. Mutations of either protein abolished phosphorylation of the counterpart protein, implying transphosphorylation of the receptor kinases. To investigate the specific amino acids critical for formation of the receptor complex and activation of BAK1 kinase activity, we expressed several versions of BAK1 in yeast and plants. L32E and L46E substitutions resulted in a loss of binding of BAK1 to BRI1, and threonine T455 was essential for the kinase activity of BAK1 in yeast. Transgenic bri1 mutant plants overexpressing BAK1(L46E) displayed reduced apical dominance and seed development. In addition, transgenic wild type plants overexpressing BAK1(T455A) lost the phosphorylation activity normally exhibited in response to BL, leading to semi-dwarfism. These results suggest that BAK1 is a critical component regulating the duration of BR efficacy, even though it cannot directly bind BRs in plants.

First Recorded Family Odiidae (Crustacea, Amphipoda, Gammaridea) from the Korean Coast, with Description of Newly Recorded Species

  • A newly recorded species of gammaridean amphipod, Cryptodius kelleri (Br$\ddot{u}$ggen, 1907) belonging to the family Odiidae Coleman and Barnard, 1991 collected from the east coastal area of Korea. Cryptodius kelleri (Br$\ddot{u}$ggen) is characterized by the medially expanded maxillipedal palp article 2, smooth dorsal pereonites, propodochelate gnathopod 1, subchelate gnathopod 2, lacking the posterior process on the basis of pereopods 5-7, dorsal carination of pleonite 3, posterolateral upturned process of pleonite 3, and an entire telson. The species is described and fully illustrated in the present study. A key is also provided to the species of genus Cryptodius. This is the first record of the family Odiidae from Korea.
