This study was carried out to investigate the effects of Salvia plebeia R. Br. ethanolic extract with different aspects (stem/leaf and whole plant) on differentiation and lipid accumulation in 3T3-L1 preadipocytes. The morphological changes and the degrees of lipid accumulation in 3T3-L1 cells were measured by Oil Red O staining and intra-cellular triglyceride (TG) assay. The mRNA expressions of special peroxisome proliferation activated receptor- genes (PPAR), CCAAT/ enhancer-binding protein (C/$EBP{\alpha}$), fatty acid synthase (FAS) and lipoprotein lipase (LPL) were detected by reverse transcriptase polymerase chain reaction (RT-PCR). The 50% ethanolic extracts ($100{\mu}g/mL$) of stem and leaf (SALE) and 30% ethanolic extracts (100 g/mL) of whole plant (SAE) from Salvia plebeia R. Br. were significantly attenuated lipid accumulation during adipogenesis in 3T3-L1 cells. Ethyl acetate-soluble fractions ($50{\mu}g/mL$) significantly inhibited lipid droplet accumulation in 3T3-L1 cells. In addition, SALE induced down-regulation of specific adipogenic transcriptional factors (C/$EBP{\alpha}$ and $PPAR{\gamma}$) and target genes (FAS and LPL) during adipogenesis. Salvia plebeia R. Br. may be used as a safe and efficient natural substance to manage obesity.
In this study the effect of heartwood-coating (HCO), vapor-dam (VD), bark-remaing (BR) and bark-remaining- coating (BRC) treatments were evaluated during air circulating oven-drying of hollowed round-post of Korean red pine. Hollowed round-post could be dried from the green condition to approximately 6% MC within 84 to 156 hours for the UC, HCO and VD specimens and it took 72 to 240 hours to about 8% MC, which is recommended as the indoor in-use MC. The temperatures in the hole of the Control (UC) and HCO specimens constantly showed a higher-leveled distribution pattern than those inside wood. The vapor pressure in the hole of the UC, HCO and BR specimens was higher than that inside wood and showed difference as highest value of about 20 mbar. The surface checks of all specimens were mild and were observed in increasing order of BRC, BR, UC, VD and HCO specimens.

The drying times of the hollowed round-posts for control, VD, HCO and BR specimens were 72, 168, 204 and 240 hours, respectively, from the initial MC to about 8% MC, which was recommended as the indoor in-use MC. The temperature in the hole of the VD specimen was lower than that of wood and the difference between air temperature in the hole and wood temperature became large during drying period. The vapor pressure of air in the hole was higher than that of inside wood for all specimens except VD specimen. The surface checks on all specimens were observed in increasing order of BR, BRC, UC, HCO and VD specimens.
The photodissociation dynamics of 1,2-bromochloroethane ($C_2H_4BrCl$) was investigated near 234 nm. A two-dimensional photofragment ion-imaging technique coupled with a [2+1] resonance-enhanced multiphoton ionization scheme was utilized to obtain speed and angular distributions of the nascent Br($^2P_{3/2}$) and Br${\ast}($^2P_{1/2}$) atoms. The total translational energy distributions for the Br and Br${\ast}$ channels were well characterized by Gaussian functions with average translational energies of 100 and 84 kJ/mol, respectively. The recoil anisotropies for the Br and Br${\ast}$ channels were measured to be ${\beta}$ = 0.49 ${\pm}$ 0.05 for Br and 1.55 ${\pm}$ 0.05 for Br${\ast}$. The relative quantum yield for Br${\ast}$ was found to be ${\Phi}_{Br{\ast}}$ = 0.33 ${\pm}$ 0.03. The probability of nonadiabatic transition between A' states was estimated to be 0.46. The relevant nonadiabatic dynamics is discussed in terms of interaction between potential energy surfaces in Cs symmetry.
본 연구는 실용계의 육질특성을 조사하고자 수행하였다. 재래계와 육용계의 교잡종 6개구(RcDR, RcLR, BcBR, BcDR, BcLR, RcBR)와 전용육계 1개구(Arbor Acre), 재래계 3개구(적갈색종-D, 황갈색종-L, 흑색종-B)에 대한 지방산 조성 및 관능적 특성을 조사하였다. 출하주령에서의 지방산 조성에서 재래계는 일반적으로 다른 종보다 불포화지방산함량이 더 높았다. B, BcBR, BcDR은 가장 높은 불포화지방산 함량을 나타낸 반면 포화지방산 함량은 다른 종에 비해 가장 낮은 함량을 나타내었다. 관능검사결과를 살펴보면 다리육의 경우 다즙성(juiciness), 연도(tenderness), 기호성(acceptibility)에서 유의적인 차이를 보였다(P<0.05). 다즙성은 흑색종과 RcBR에서 가장 우수하였으며 Arbor Acre 가 가장 낮았다. 연도는 흑색종이 가장 우수하였고, 그다음은 RcBR이었으며, RcDR은 가장 낮았다. 풍미와 냄새에서는 각각 유의적인 차이가 없었으며, 기호성은 재래계 특히 흑색종에서 가장 우수하였고, 시험구중에서는 RcBR이 가장 우수하였다.
GC-MS/SIM를 통하여 옥수수 유식물 신초로부터 해 castasterone (CS)과 6-deoxocastasterone (6-deoxoCS)을 동정하였다. 또한 BRs의 생합성 전구물질인 24$\alpha$-methylcholesterol과 24$\alpha$-methylcholestanol이 동정하였다. 이들 BRs와 생합성 전구물질은 BRs의 생합성과정 중 후기 C6산화과정에 속하는 화합물로서, 옥수수 유식물 줄기에서는 BRs의 생합성 과정으로 후기 C6산화과정이 주된 생합성 과정임을 알 수 있었다. 다음으로 옥수수 유식물 줄기내의 BRs가 발아와 더불어 종자에서부터 이동해 온 것인지, 아니면 줄기 생장 시 줄기 자체가 생합성된 것인지를 조사하기 위하여 몇몇 중요한 후기 C6산화과정의 반응을 촉매하는 효소의 활성이 유식물 줄기에 존재하는지를 조사하였다. 그 결과 후기 C6산화과정에 포함되는 24$\alpha$-methylcholestanol에 서 6-deoxocathasterone (6-deoxoCT), 6-deoxoteasterone (6-deoxoTE) 에서 6-deoxo-3-dehydroteasterone (6-deoxo-3-DHT)을 거쳐 6-deoxotyphasterol (6-deoxoTY),그리고 6-deoxoCS에서 CS로의 과정을 촉매하는 24$\alpha$-methylcholestanol 22(R)-hydroxlyase, 6-deoxoTE dehydrogenase/6-deoxo-3-DHT reductase, 6-deoxoCS oxidase의 활성이 옥수수 유식물 줄기에 존재하고 있음이 확인되었다. 이는 유식물 줄기 생장 시 필요한 BRs가 발아종자로부터 이동되는 것이 아니라, 유식물 줄기 자체에서 생합성 될 가능성이 높음을 나타내는 결과라 할 수 있다. 또한 옥수수 유식물 줄기에서 CS를 brassinolide(BL)로 전환하는 BL synthase의 활성이 검출되지 않아, 옥수수 유식물 줄기에서의 활성형 BR은 BL이 아닌 CS임을 밝혔다.
본 연구에서는 식물뿌리의 굴중성 반응에 있어서 식물호르몬과 AtEXPA3 유전자와의 관계를 밝히고자 하였다. AtEXPA3 유전자의 RT-PCR을 통한 발현분석 결과 잎, 근출엽, 뿌리 꽃 등 생장이 활발한 조직에서 발현률이 높게 나타났으며, 굴중성 자극과 BRs, IAA에 의해서도 발현이 증가되었다. 또한 ethylene 생합성 저해제인 AVG를 처리하면 굴중성 반응이 현저히 억제되었는데 이는 ethylene 그 자체도 BR과 IAA처럼 굴중성을 촉진시키는 활성을 갖고 있음을 의미한다. 한편 호르몬을 처리하지 않은 AtEXPA3 RNAi mutant에서 굴중성 반응이 억제되는 현상은 애기장대 뿌리의 생장에 관여하는 AtEXPA3의 조절 인자로 BRs, auxin, ethylene 등의 식물호르몬이 관여하고 있음을 나타낸다. 아울러 BRs signaling mutant에서 변화된 굴중성(bri1-301, bak1에서 감소, BRI-GFP에 서 증가) 반응의 감소와 증가는 굴중성 반응이 BRs의 신호전달 과정을 통하여 일어남을 의미한다. 결론적으로 애기장대 뿌리의 굴중성 반응은 식물호르몬에 의한 AtEXPA3 유전자의 발현 증가의 결과로 인해 애기장대 뿌리의 생장이 촉진되어 나타나는 결과라 하겠다.
Type III polyketide synthase (PKS) found in bacteria is known as 1,3,6,8-tetrahydroxynaphthalene synthase (THNS). Microbial type III PKSs synthesize various compounds that possess crucial biological functions and significant pharmaceutical activities. Based on our sequence analysis, we have identified a putative type III polyketide synthase from Nocardia sp. CS682 was named as ThnA. The role of ThnA, in Nocardia sp. CS682 during the biosynthesis of 1,3,6,8 tetrahydroxynaphthalene(THN), which is the key intermediate of 1-(α-L-(2-O-methyl)-6-deoxymannopyranosyloxy)-3,6,8-trimethoxynaphthalene (IBR-3) was characterized. ThnA utilized five molecules of malonyl-CoA as a starter substrate to generate the polyketide 1,3,6,8-tetrahydroxynaphthalene, which could spontaneously be oxidized to the red flaviolin compound 2,5,7-trihydroxy-1,4-naphthoquinone. The amino acid sequence alignment of ThnA revealed similarities with a previously identified type III PKS and identified Cys138, Phe188, His270, and Asn303 as four highly conserved active site amino acid residues, as found in other known polyketide synthases. In this study, we report the heterologous expression of the type III polyketide synthase thnA in S. lividans TK24 and the identification of THN production in a mutant strain. We also compared the transcription level of thnA in S. lividans TK24 and S. lividans pIBR25-thnA and found that thnA was only transcribed in the mutant.
Statement of problems: There are few studies which reported the survival rates of the specific dental implant systems in the Korean population with the follow-up periods longer than 5 years. Purpose: This retrospective clinical study was aimed to evaluate cumulative survival rate (CSR) of $Br{\aa}nemark$ implants followed for 10 years and to determine risk factors for implant failure. Material and methods: A total of 271 $Br{\aa}nemark$ implants in 83 patients were investigated with several identified risk factors. Life table analysis was undertaken to examine the CSR. Cox regression method was conducted to assess the association between potential risk factors and overall CSR. Results: Thirty implants failed. The 10-year implant CSR was 82.5%. Cox regression analysis demonstrated a significant predictive association between overall CSR and implant length (P<.05). Conclusion: An acceptable long-term result of $Br{\aa}nemark$ implant was achieved and implant length showed a significant association with the CSR.
TEMPO와 NaBr의 수준에 따른 쌀가루의 반응 특성을 조사한 결과, 반응시간은 TEMPO와 NaBr의 수준이 낮을수록 증가하였고, 수율은 TEMPO의 수준이 증가할수록 증가하였다가 다시 약간 감소하였으며 NaBr의 수준이 증가할수록 감소하는 경향을 나타내었다. 선택성은 NaBr의 수준이 증가할수록 감소하는 경향을 보였다. 이에 따라 반응시간, 수율 및 선택성을 고려하여 산화 쌀가루 제조를 위한 TEMPO와 NaBr의 적정 수준을 각각 0.9 mM과 44 mM/100 mM AGU로 결정하였다. 산화 반응을 통해 쌀가루의 물과 결합능력 및 점도가 증가하는 것으로 나타났으며, 산화 쌀가루 수용액은 의가소성 유체의 성질을 나타내었다. 산화 쌀가루로 일부 대체된 쌀가루 현탁액은 초기 점성 증가 온도가 낮아지고 최고 점도는 증가하였으며 setback은 감소하는 것으로 나타나, 산화 쌀가루의 첨가에 따른 쌀가루의 노화 지연 가능성을 보여주었다. 산화 쌀가루를 2% 첨가하고 가수량을 증가시켜 제조한 절편의 텍스처 특성을 평가한 결과, 저장에 따른 경도 증가 현상이 억제되는 것으로 나타났다. 따라서 산화 쌀가루를 절편에 첨가할 경우 노화가 억제되어 저장기간의 증진 효과를 기대할 수 있으리라 본다.
