Effects of Several Insecticides on the Biology and Population of the Brown Planthopper, Nilaparvata lugens STAL (몇가지 수도용 살충제 처리가 벼멸구 차세대 밀도에 미치는 영향)

  • The insecticides MIPC, BPMC, carbofuran and diazinon were treated to the brown planthopper (BPH) , Nilaparvata lugens STAL, at the dose levels of $LD_{10}\;and\;LD_{50}$(by topical application)/at the rate of 40kg/ha (product base) (by dusting or broadcasting on potted rice). The BPH population in the first generation was significantly decreased in the topical treatments compared to the untreatment. The number of BPH offsprings was relatively greater at the dose level of $LD_{10}\;than\;of\;LD_{50}$. The number of nymphs, however, were greatly varied with the insecticides. The offsprings from the BPH treated with the rate of 40kg/ha showed longer nymphal periods and higher adult emergence, but except diazinon treatment, rather less egg-numbers were observed. The BPH population density was significantly decreased in general up to 38 days after treatment(DAT) in the treated pots. Among the insecticides tested, however, only diazinon induced greater number of insects at the 54 DAT compared to the untreated pots.

  • The aromatic dioxygenase system in Sphingomonas yanoikuyae strain Bl consists of three components, an oxygenase, a ferredoxin, and a reductase. The insertional knockout of the bphA4 gene encoding a reductase and subsequent complementation experiments showed that the reductase encoded by bphA4 in S. yanoikuyae strain Bl is associated with multiple dioxygenase components including that of toluate dioxygenase (XyIXY).

Marker Assisted Selection of Brown Planthopper Resistance and Development of Multi-Resistance to Insect and Diseases in Rice (Oryza sativa L.) (DNA 마커를 이용한 벼멸구 저항성 선발 및 복합내병충성 벼 계통 육성)

  • The main objective of this study was to develop the multi-resistance lines to insects(brown planthopper; BPH, rice green leafhopper; GRH) and disease(blast; BL, bacterial blight; BB and rice stripe virus disease;RSV) with good grain quality and plant type by combining conventional breeding and marker assisted selection(MAS) and to eliminate the linkage drag effects between Bph1 gene and culm length, we conducted MAS of Bph1 gene in advanced backcross and double cross progenies. 'Nampyeong', 'Junam' and 'Milyang220' were used as the parent in this study. 'Milyang220' was used as the donor of brown planthopper resistance gene Bph1 with tall culm length. Two backcross progenies were developed using two recipients 'Nampyeong' carrying GRH resistance gene Grh3(t) with good grain appearance and 'Junam' harboring bacterial blight resistance gene Xa3 with short culm length. Two $BC_1$ generations were resulted from the backcrossing of the $F_1$ plants with recurrent parents 'Nampyeong' and 'Junam'. The second rounds of backcrossing($BC_2$) were derived from the cross of selected resistant $BC_1F_1$ plants based on heterozygous genotype of RM28493 linked to Bph1 gene. The double crossed population was constructed from the cross of between each heterozygous $BC_2F_1$ plants at RM28493 locus of '$Nampyeong^*3$ / Milyang220' and '$Junam^*3$ / Milyang220'., The homozygous alleles in Bph1 gene were selected using co-dominant DNA marker RM28493 in double crossed population. Eighty-five lines with multi-resistance to BL, BB, RSV, GRH and BPH were selected by bio-assay and MAS in generation of double crossing. The culm length, head rice ratio and yield of the selected multi resistance lines was ranged from 71 to 88 cm, from 51 to 93%, from 449 to 629 kg/10a. respectively. We can select a promising multi resistance line similar with 'Nampyeong' of major agronomic traits such as culm legnth, head rice ratio and yield. It was designated as Milyang265. Finally this study was developed the multi resistant varieties against to insects and diseases with the good grain quality 'Milyang265' by the advanced backcross and double cross combining MAS and it can be used as genetic resources of multi-resistance to insect and diseases in rice breeding programs.

Human Papilloma Virus Detection by INNOLiPA HPV in Prostate Tissue from Men of Northeast Mexico

  • Background: Prostatic adenocarcinoma by Prosate cancer (PCa) is the most prevalent cancer and the second cause of cancer-related death among men in the Western world. Human papilloma virus (HPV) may be considered as a preventable risk factor. In this study, we assessed the frequencies of HPV infection in prostatic adenocarcinoma and benign prostatic hyperplasia (BPH) cases in Northeast Mexico. Materials and Methods: A total of 87 paraffin-embedded blocks (from 25 and 62 patients with definite diagnoses of BPH and adenocarcinoma, respectively) were selected and subjected to INNOLiPA HPV Genotyping to detect 28 high- and low-risk HPV types. The rates of infection were compared in the two studied groups. Results: INNOLiPA HPV demonstrated great sensitivity for HPV detection on paraffin-embedded tissue. Global prevalence was 14.9% (13/87). HPV infection was positive in 19.4% (12/62) of patients with adenocarcinoma and 4.0% (1/25) of patients with BPH. HPV-11, which is considered to be low risk, was more prevalent. Interestingly, one patient with BPH and six with prostate cancer showed examples considered to be high risk (HPV-18, -51, -52, and -66). Conclusion: A higher rate of HPV infection among Mexican patients with prostatic carcinoma than among those with BPH was observed. HPV infections may thus contribute to the risk of prostate cancer. Further studies are required to elucidate any roles of HPV infection in prostate disease in Mexico and the effect of prevention and treatment of HPV infection on prostatic adenocarcinoma.

The Predatory Behavior of Green Mirid Bug, Cytorhinus lividipennis Reuter, on Brown Planthopper Eggs in Different Temperature Conditions (온도에 따른 등검은황록장님노린재 (Cyrtorhinus lividipennis Reuter)의 벼멸구 알에 대한 포식습성)

  • The functional responses of the female Cyrtorhinus lividipennis on brown planthopper (BPH), Nilaparvata lugens, eggs and their predation behavior were investigated at six temperature conditions; 20, 23, 26, 29, 32, and $35^{\circ}C$. C. lividipennis was found to prefer young BPH eggs, especially 3-day-old eggs the most. The functional responses for female C. lividipennis on BPH eggs fitted the Holling's Type-I true predator-prey Rogers' (1972) model at all temperatures tested except $35^{\circ}C$ at which the negative Th value was produced. With the temperature increased up to 32$^{\circ}C$, the instantaneous attacking rate(a) increased from 0.1923 at $20^{\circ}C$ to 0.5085 at $32^{\circ}C$, while the handling time (Th) was gradually decreased as low as 0.0151 at $32^{\circ}C$. C. lividipenis preferred the BPH eggs laid on the upper part of rice stem when the BPH egg density was high, but there was no significant difference in the preference when the egg density was low. The preference was more obvious in high temperature conditions such as above 29$^{\circ}C$.

  • These studies were conducted to investigate the biological effects of buprofezin 25% WP on the adult of the brown planthopper (BPH) and its control effects on the BPH population, when buprofezin was treated on different days after the introduction of the BPH adult to the potted rice. Also age structures of the BPH population at the time of buprofezin treatment were observed. The results were as follows: Buprofezin (7.0 g a.i./l0 a) could reduce the longevity and fecundity of the newly emerged female but did not affect the biology of older female, and it did not affect the biology of the male. The larval densities at the time of buprofezin treatment was reduced to nearly zero after treatment. The higher the residual effect and the proportion of larvae at the time of buprofezin treatment were, the higher its con¬trol effect on the BPH population was. In case of buprofezin 7.0 g a.i./l0a treatment, the time interval of treatment that could show the highest control effect was about 10 days (from 18 days after the adult introduction to the adult emergence days of next generation).

Comparison of Selection Efficiency between Marker-Assisted Selection and Phenotypic Selection for Development of Brown Planthopper Resistance Lines in Rice (벼멸구 저항성 계통선발을 위한 MAS의 선발효율 비교)

  • This study presents a case study designed to compare the selection efficiency between phenotypic selection (PS) and marker-assisted selection (MAS) in breeding of resistance lines to brown planthopper (BPH). The efficiency between PS and MAS were compared with four population such as the $F_2$, RILs ($F_6$), DH, and backcrosse ($BC_6F_5$) population, derived from a cross 'Samgang / Nagdong'. The resistance lines were selected using two markers, RM28493 and BpE18-3, related to BPH resistance were screened as resistance lines over 95% in PS. The costs required for BPH screening in the MAS system account for approximately 32% of the total costs of PS. The period needed to select the resistance plants was 30 days in PS and 7 days in MAS. Based on the results, we could establish the breeding system for selection of BPH resistance lines by MAS.

Investigation into Whether Agronomic Traits Are Fixed for the Breeding of Drought Tolerance and BPH-resistant Crosses (내건성 및 BPH 내성 계통의 육성을 위한 농업형질 고정여부 조사)

  • Late in the 1960s, South Korea established cultivation technology and began breeding new "Tongil" type rice cultivars by crossing indica and japonica. To date, this cultivation technique has been used to produce a wide variety of cultivars to meet consumer preferences and adapt to extreme weather conditions. Once major genetic traits are no longer segregated through advanced generations, varieties obtained from a cross between different crops may become a new variety. In this study, we confirmed the agronomic traits of F4, F5, and F6 by advanced generation a population in which drought tolerance and brown planthopper (BPH) resistance were crossed. HV23, Ilmi/HV23, and Drimi2ho/HV23 were used. HV23 was drought-tolerant, and Drimi2ho was resistant to BPH. As a result, it was possible to consider that the agronomic traits were fixed because none of them showed a significant difference from the others. While it takes more than 10 years for the pedigree method of breeding, this study confirmed that the agronomic traits were fixed in 4-6 years. In the future, we will investigate the homology of the CaMsrB2 gene and the Bph1 gene, to confirm that both genes are closely related to each other, and analyze the stable inheritance of the introduced gene for multiple successive generations.

Inhibition of Proliferation of LNCaP Prostate Cells by Corni Fructus Extract Is Associated with a Decrease in the Expression of Benign Prostatic Hyperplasia-Causing Factors (산수유 추출물에 의한 LNCaP 전립선 세포의 증식 억제 및 양성 전립선 비대증 유발 인자의 발현에 미치는 영향)

  • Objectives: Benign prostatic hyperplasia (BPH) is a progressive pathological condition characterized by excessive proliferation of the prostate. In this study, we evaluated the effect of Corni Fructus water extract (CF) on the promotion of prostate cell proliferation by dihydrotestosterone (DHT). Methods: The effect of CF on the proliferation of LNCaP prostate cells was evaluated, and DHT was treated to induce an in vitro BPH model. To study the mechanism of inhibition of cell proliferation and BPH by CF, changes in the expression of key factors related to cell cycle and BPH were investigated. We further investigated the effect on the production of reactive oxygen species (ROS) and nitric oxide (NO) to evaluate the antioxidant and anti-inflammatory efficacy of CF. Results: Inhibition of LNCaP cell proliferation by CF was associated with decreased expression of cyclin D1 and cyclin A and increased expression of cyclin-dependent kinase inhibitor p21. CF also suppressed expression of BPH inducing factors such as 5α-reductase type 2 and androgen receptor (AR) as well as prostate specific antigen (PSA). Furthermore, CF significantly blocked DHT-induced LNCaP cell proliferation and effectively attenuated DHT-induced expression of BPH mediators and cyclins. In addition, CF inhibited DHT-induced oxidative and inflammatory reactions by inhibiting production of ROS and NO. Conclusion: Our results demonstrated that CF probably acted as 5α-reductase type 2 inhibitor, preventing the 5α-reductase type 2-AR signaling pathway, thereby reducing the conversion of testosterone to DHT and the expression of PSA, which is at least correlated with the antioxidant and anti-inflammatory activities of CF.

  • The relative toxicity of some of the insecticides which have been used for the control of paddy rice insect pests in Korea was evaluated in the laboratory with the brown planthopper (BPH) Nilaparvata lugens, and a predaceous paddy spider Pirata subpiraticus. In order of the relative toxicity (LD5O value to spider/LD50 value to BPH) were PAP (0.4), MPP(0.1), MEP(1.8), diazinon(2.8), carbofuran(7.5), NAC(11.3), BPMC(17.5), Pyridaphenthion(35.9) and MIPC(65.7). MIPC and Pyridaphenthion were considered as having the desirable selective toxicity for the spider and the BPH.
