국내에서 분리된 Borrelia burgdorferi Sensu Lato의 병원성 연구 (Study on Pathogenicity of Borrelia burgdorferi Sensu Lato Isolated in Korea)

  • Spirochetes were isolated from the midgut of Ixodes persulcatus ticks captured at Chungju, Korea and identified as Borrelia afzelii strains by polymerase chain reaction. To determine the pathogenicity of the B. afzelii strains isolated in Korea, the microbiological and pathological features of Lyme disease were observed in C3H/He mice after intraperitoneal inoculation of the fresh isolate of B. afzelii strain. The results are summarized as follows 1) The Borrelia were detected in the tissues of heart, spleen, kidney, urinary bladder and knee joint within 7 days after inoculation of infection by dark field microscopic examination. The isolation rate from heart, urinary bladder and joint was significantly higher than the rate from spleen, kidney, and blood samples. 2) The Borrelia was detected in heart muscle by indirect immunofluorescent antibody test. 3) Antibody to the Borrelia was detected as early as one week after inoculation. 4) The marked tropism of the Borrelia was observed in myocardial, urinary tract and joint tissue. The main pathological features are inflammation in tissues of heart, kidney, joint and urinary bladder. From these results, the Borrelia afzelii strain isolated in Korea were determined as pathogenic strain.

실험적으로 감염시킨 토끼에서 Borrelia burgdorferi 분포 및 면역반응 양상 (Dissemination of Borrelia burgdorferi and immunological responses after experimental infection in rabbits)

  • The visceral dissemination of Borrelia burgdorferi in New Zealand White rabbits was evaluated following intradermal inoculation of $1{\times}10^8$ spirochetes. We inoculated Borrelia burgdorferi B31, B garinii KW1 and B afzehlii S13, respectively, and monitored the dissemination in the experimentally infected rabbits for 28 days. In the B burgdorferi B31-challenged group, the spirochetes were completely cleared in rabbits at day 1 and visceral dissemination was not demonstrated. However, B garinii KW1 and B afzelii S13 were found to successfully disseminate in visceral organs of rabbits during the experiment period of 28 days. And experimentally infection-derived immunological responses in rabbits were identified with enzyme-linked immunosorbent assay and immunoblot analysis. Based on these results, the differences in the virulence of Lyme borrelial strains were proved in rabbit model.

Molecular detection of Borrelia theileri in cattle in Korea

  • Bovine borreliosis, caused by Borrelia theileri which is transmitted via hard tick bites, is associated with mild clinical symptoms, such as fever, lethargy, hemoglobinuria, anorexia, and anemia. Borrelia theileri infects various animals, such as cattle, deer, horses, goats, sheep, and wild ruminants, in Africa, Australia, and South America. Notably, no case of B. theileri infection has been reported in Korean cattle to date. In this study, 101 blood samples were collected from a Korean indigenous cattle breed, among which 1.98% tested positive for B. theileri via nested PCR. The obtained sequences exhibited high homology with B. theileri strains identified in other regions. Phylogenetic analysis of 16S rRNA confirmed the B. theileri group affiliation; however, flagellin B sequences exhibited divergence, potentially due to regional evolutionary differences. This study provides the first molecular confirmation of B. theileri infection in Korean livestock. Further isolation and nucleotide sequence analyses are necessary to better understand the presence of B. theileri strains in cows in Korea.

Survey of Ehrlichia canis and Borrelia burgdorferi antibodies in dogs (German shepherd) reared in Korea

  • This study was conducted to survey of Ehrlichia (E.) canis and Borrelia (B.) burgdorferi antibodies among clinically healthy German shepherd dogs (116 females and 120 males) using a ELISA kit (SNAP 3Dx; IDEXX Laboratories, USA) in Korea. Whole blood samples are collected from the 236 dogs and are tested to detect E. canis and B. burgdorferi antibodies by using ELISA kit (SNAP 3Dx; IDEXX Laboratories, USA). Confidence interval comparisions revealed that dogs of 4-6 years have higher prevalence with a seropositive result (CI=0.17-0.45) in E. canis than the other age groups but there are no differences among age groups in B. burgdorferi. Also, no differences with a seropositive result were found among different regions in E. canis and B. burgdorferi antibodies. In conclusion, this study was the first large scale survey of canine E. canis and B. burgdorferi antibodies in Korea and provide an useful reference for clinicians.

라임병 원인 스피로헤타 Borrelia burgdorferi의 운동성과 주화성: 발병기전에서의 역할 (Motility and Chemotaxis in the Lyme Spirochete Borrelia burgdorferi: Role in Pathogenesis)

  • 운동성 및 주화성은 스피로헤타를 포함한 많은 병원성 세균의 병원인자로 작용한다. 라임병은 스피로헤타인 Borrelia burgdorferi에 의해 발병하며, 검은다리 참진드기에 물린 상처를 통해 사람에게 전염되는 미국 및 유럽 내에서 가장 유행하는 벡터-매개성 질병이다. Borrelia를 포함한 스피로헤타 균들은 다른 일반적인 편모를 가지는 균들과 달리 주변세포질에 그 편모를 가지며, 운동성이 결여된 돌연변이주의 경우 야생주와 같은 병원성을 가지지 못한다고 알려져 있다. 또한 대장균에 비해 더욱 다양한 종류의 주화성 관련 유전자를 지니고 있어, 편모를 통한 이 균의 운동성이 매우 복잡한 메커니즘을 가질 것으로 예상할 수 있다. 최근 초저온 전자현미경 및 새로운 유전자 조작기술의 발달로 인해 베일에 싸여 있던 스피로헤타의 운동성 및 주화성, 특이한 편모의 구조가 밝혀지고 있다. 본 리뷰 논문에서는 이러한 최첨단 기술의 이용으로 현재까지 밝혀진 Borrelia burgdorferi의 새로운 편모 모터 구조를 소개하고, 균의 병원성과 운동성 및 주화성의 상관관계에 대해 설명하고자 한다.

PCR-Restriction Fragment Length Polymorphism 방법에 의한 Borrelia burgdorferi Sensu Lato의 분류 (Molecular Typing of Borrelia burgdorferi Sensu Lato by PCR Restriction Fragment Length Polymorphism Analysis)

  • 라임병의 원인균인 Borrelia burgdorferi에 대하여 각 균종의 표준균주와 진드기에서 추출한 DNA를 template로 PCR을 실시한 후 그 증폭산물을 Alu I으로 처리한 restriction fragment length polymorphism (RFLP) 방법으로 각 균종의 연관성을 조사하고자 하였다. 표준균주로 RFLP를 실시한 결과 B. burgdorferi sensu stricto와 B. garinii의 RFLP 형태 (50 bp, 70 bp, 150 bp)가 유사하였으며 B. afzelii에서는 다른 RFLP 형태 (50 bp,110 bp,150 bp)를 관찰하였다. 그 중 B. afzelii KK-1과 B. garinii HPI은 새로운 RFLP형태를 보여 B. afzelii자 B. garinii는 각각 2 type의 subgroup으로 분류할 수 있었다. 진드기 DNA에서는B. afzelii를 포함한 각 균종에 대하여 모두 유사한 RFLP형태를 보였는데, 진드기 DNA에서 확인된 B. afzelii는 KK-1과 같은 군에 속하는 것으로 사료되었다.

국내에서 채집한 진드기에서 중합효소연쇄반응을 이용한 라임병균 및 Ehrlichiosis 원인체의 검출 (Detection of Borrelia burgdorferi and Ehrlichiosis Agent in Ticks Collected in Korea Using Polymerase Chain Reaction)

  • 국내에서 채집한 진드기의 라임병 및 ehrlichiosis 원인체 보균 상태를 조사하기 위하여 총 516마리 (Ixodes spp. 22마리 , Haemaphysalis spp. 494마리)의 ixodid 진드기를 봄과 가을에 걸쳐 강원도 고산지대 일원에서 채집하였다. 수집한 진드기에서 DNA를 추출·정제한 후 추출한 DNA를 template로 이용하여, Borrelia burgdorferi sensu lato 및 Ehrlichia spp.에 특이하게 반응하도록 제작한 primer를 이용한 중합효소연쇄반응 (polymerase chain reaction, PCR)을 실시하였으며, 이 결과를 oligonucleotide probe를 사용한 southern blotting을 통하여 다시 확인하였다. 총 516마리의 진드기중 B. burgdorferi sensu lato DNA 양성인 진드기는 68 (13.2%)마리 (B. burgdorferi sensu stricto, 2; B. afzelii, 1; B. garinii, 33; B. tanukii, 8; B. turdae, 4)로 나타났으며 이 중 37 (7.2%)마리의 진드기는 southern blot analysis에서도 양성으로 확인되었다. 또한 101 (19.2%)마리의 진드기가 Ehrlichia spp.에 대한 PCR에서 양성이었으나, 이들 중 25 (4.8%)마리만이 southern blot analysis에서 양성으로 확인되었다. 그러나 사람의 병인체로 추정되는 human granulocytic ehrlichiosis (HGE) agent DNA를 보균한 진드기는 확인되지 않았다. 한편 라임병균과 ehrlichiosis원인체를 동시에 보균한 것으로 밝혀진 진드기가 3마리에서 (0.6%) 확인되어, 국내에서도 진드기 교상시 이들 두 가지 열성질환이 동시에 감염될 가능성이 있는 것으로 사료됨으로 진드기 매개성 열성질환에 대한 적절한 진단법 등을 보다 체계적으로 연구하여야 할 것으로 판단된다.

Horses as a Potential Reservoir of Lyme Borreliosis in Jeju-do, Korea

  • Lyme borreliosis (LB) is the most common tick-borne infectious disease in North America, and it was designated as a national notifiable infectious disease in Korea in December 2010. While no cases in Jeju-do were recorded from 2012 to 2016, a recent survey reported that the seroprevalence of Borrelia burgdorferi using enzyme-linked immunosorbent assays in horses in Jeju-do was 19.0% (95% confidence interval, 12.0 to 28.3%). This fact suggests that horses may be a potential reservoir of LB in Jeju-do and that individuals in close contact with horses may be a high-risk group. Thus, a serological study in this high-risk group is urgently needed.

  • Lyme disease is a tick-borne zoonotic infectious disease caused by Borrelia burgdorferi. The present study assessed the infection status of B. burgdorferi among horses reared in Korea using ELISA and PCR. Between 2009 and 2013, blood samples were collected from 727 horses throughout Korea. Data for each animal including age, gender, breed, and region of sample collection were used for epidemiological analysis. Overall, 38 (5.2%; true prevalence: 5.5%) of 727 horses were seropositive by ELISA. There were statistically significant differences according to breed and region (P<0.001) whose differences might be attributed to the ecology of vector ticks and climate conditions. Using 2 nested PCR, none of the samples tested positive for B. burgdorferi. Thus, a positive ELISA result can indicate only that the tested horse was previously exposed to B. burgdorferi, with no certainty over the time of exposure. Since global warming is likely to increase the abundance of ticks in Korea, continuous monitoring of tick-borne diseases in Korean horses is needed.
