인간 중간엽 줄기세포로부터 골아세포로의 분화시 관찰되는 유전자 발현 분석 (Gene Expression Profile Associated with the Differentiation of Osteoblasts from Human Mesenchymal Stem Cells)

  • 인체의 골수내에 존재하는 중간엽 줄기세포는 성장인자나 환경적 요인에 의해 지방세포, 골아세포, 연골모세포 및 연골세포 등으로 분화됨이 알려져 있다. 본 연구에서는 정상 인체의 골수으로 얻어진 중간엽 줄기세포로부터 골아세포의 분화 가능성을 알아보고 이에 관여하는 유전자의 발현을 조사하였다. 정상 골수의 중간엽 줄기세포를 골유도성 자극보조제로서 $\beta$-glycerol phosphate, L-ascorbic acid 및 dexamethasone을 첨가하여 골아세포로의 분화를 유도한 세포와 골유도성 자극보조제를 첨가하지 않은 세포를 배양하여 일정 간격으로 cDNA microarray를 이용하여 각각의 단계에서 발현되는 유전자를 검사하고 이로 인해 얻어진 유전자의 발현량을 분석하기 위해 real time quantitative RT-PCR 을 시행하였다. 골유도성 자극보조제를 첨가한 군에서 첨가하지 않은 군에 비하여 정상적인 골아세포로의 성장이 유도되었고, 분화과정에서 36개의 유전자의 발현이 증가되었고, 59개의 유전자의 발현이 억제되었다. 주로 골 생성 과정과 연관이 있다고 알려진 osteoprotegerin, LRP5 및 metallothionein 2A 등의 유전자들이 분화과정에서 발현 증가되어 나타났고, 줄기세포로부터 분화될 수 있는 조직들 중 근육, 지방, 연골, 혈관 및 신경 조직과 연관된 유전자들은 분화 후기에 감소하거나 혹은 전분화 과정 동안 발현이 억제되었다. 본 연구에서는 골아세포의 분화와 연관된 유전자 발현을 확인함으로써 특정 조건하에서 중간엽 줄기세포로부터 골아세포로의 분화가 가능함을 확인할 수 있었고, 이 과정에 관련된 특정 유전자의 발현 양상을 밝힐 수 있었다.

PSII를 이용한 마그네슘 이온 주입 임플란트에 대한 MC3T3-E1 골모양 세포 반응 연구 (Cell study on the Magnesium ion implanted surface with PSII)

  • 임플란트와 골 반응을 개선하기 위한 생화학적 표면 처리 방법으로 다양한 이온을 이용한 이온주입법에 대한 관심이 높아지고 있다. 본 연구는 플라즈마 상태의 마그네슘 이온을 임플란트 표면에 주입하여 이온 피막을 형성하는 방법으로 표면 처리를 한 임플란트에 대한 MC3T3-E1 골모양 세포의 초기 반응을 평가해 보고자 하였다. 티타늄 디스크를 네 가지 군으로 표면처리를 달리하였다. A군은 연마만 하였고 B군은 연마 후 마그네슘 이온을 주입하였다. C군은 알루미늄 입자분사 하였고, D군은 알루미늄 입자분사 후 마그네슘 이온을 주입하였다. 조골세포의 반응을 세포 부착, 증식, 분화의 단계별로 평가하였다. 세포 부착을 평가하기 위해 MC3T3-E1 골모양 세포를 4시간, 24시간, 48시간 금속 표면에서 배양하여 주사현미경으로 관찰하였다. 세포분화도평가는 세포를 4일간 배양 후 알칼리성 인산분해효소 활성도 분석을 통해 시행하였다. 세포외기질의 세포내 발현은 RT-PCR을 통해 평가하였다. 이상의 실험에서 다음과 같은 결과를 얻었다. 1. 주사현미경 관찰시 시간의 흐름에 따라 세포 부착량은 증가하였으며, 마그네슘 이온을 주입한 시편에서 더 많은 양 세포 증식이 관찰되었으며 분화정도도 더 높은 것으로 관찰되었다. 2. RT-PCR 분석시 알루미늄 입자분사 후 마그네슘 이온을 주입한 시편에서 c-fos와 osteonectin의 발현이 증가된 소견을 보였다. 3. 알칼리성 인산분해효소 활성도 분석시 금속 표면처리 방법에 따른 차이는 발생하지 않았다. 이상의 결과를 종합하면 Mg 이온이 주입된 군의 세포가 Mg 이온이 주입되지 않은 군보다 초기의 세포반응이 더 우수하다는 것을 알 수 있다.

저산소 상태로 인한 조골세포 고사사기전에서 p-38 MAP kinase의 역할에 관한 연구 (Role of p-38 MAP Kinase in apoptosis of hypoxia-induced osteoblasts)

  • 교정력에 의한 치아 이동은 기계적인 힘에 의하여 압박측에는 다양한 구조를 가진 치주 조직에 혈류의 변화가 생기며 국소적으로 산소 장력에도 변화가 생겨 저산소 상태 가 유발됨은 이미 확인한 바 있다. 본 연구는 치아 주위 골격을 형성하는 조골세포를 대상으로 교정적 치아 이동과 유사한 시험관내 조건을 설정하여 저산소 상태 시 유발되는 조골세포 고사조절 기전을 규명하고자 시행하였다. 생리적인 저산소증의 실험조건으로 $2\%$ 산소상태를 설정하여 저산소 하에서 세포가 고사(apoptosis) 됨을 확인하였고, stress유발 시 많은 관련을 가진 것으로 알려진 p-38 MAPK의 활성을 관찰하였다. 또한 p-38 MAPK의 억제제인 SB203580의 전처치로 인하여 세포의 죽음이 억제됨을 확인하였고, 저산소 상태 시 활성형태로 분절되는 caspase-3, -6및 9등의 세포고사관련 효소들의 활성 형태로의 분절이 억제됨을 확인하였으며 이러한 caspase의 기질인 Lamin-A등의 분절 또한 억제됨을 밝혔다. 또한 마이토콘드리아 내의 cytochrome c의 세포질내로의 이동 또한 조절됨을 확인함으로써 p-38 MAPK의 조절단계를 시사하여 주고 있다. 본 연구로 치아 이동 시 유발되는 저산소 상태 하에서 발생하는 조골세포의 고사 조절에 p-38 MAPK가 관여함을 확인하였다.

치수 치근단 병소의 전구 위험요인으로서의 제 2 형 당뇨의 역할에 관한 소고 (THE ROLE OF TYPE 2 DIABETES AS A PREDISPOSING RISK FACTOR ON THE PULPO-PERIAPICAL PATHOGENESIS: REVIEW ARTICLE)

  • 당뇨(Diabetes Mellitus)란 혈당을 조절하는 인슐린의 분비나 기능에 장애를 야기하는 질환으로 인슐린 의존성 여부에 따라 제 1 형과 제 2 형으로 분류된다. 본 종설은 최근 증가 추세에 있는 제 2 형 당뇨가 치수 치근단 병소의 병인 과정에 전구 위험요인으로 작용할 수 있는지를 평가 하고자 문헌고찰을 통해 당뇨의 병인 과정에서 특징적으로 나타나는 혈관 합병증에 관해 알아보고, 부가적으로 제 2 형 당뇨 쥐 모델에서 인위적인 치수 감염 후 얻은 치근단 조직의 조직병리학적 분석을 시행하였다. 조직학적 관찰 결과 제 2 형 당뇨 쥐에서 대조군에 비해 치수 치근단 병소의 크기가 증가하였고, 치수 염증 반응도 심하게 나타난 것으로 보아 당뇨 자체가 숙주를 감염에 취약한 상태로 만드는 전구 위험요소로 작용하였음을 알 수 있었다. 이러한 이유로는 첫째, 당뇨 시 전반적으로 나타나는 혈관 내 죽상 침착(atheromatous deposits)에 의해 혈관내벽의 두께가 두꺼워져 미세 순환의 장애는 물론 탐식 세포의 기능 저하, 면역 세포의 혈류 이동이 차단되어 치수 감염 시 쉽게 치근단 병소로 이환될 가능성이 높고, 둘째 치수 혈관에서 특징인 측부 순환(collateral circulation)의 부재에 따른 살균성 다형핵 백혈구의 활동 억제를 포함한 미세 혈관계의 취약성으로 인해 치수 조직의 재생능이 저하되어 추가적인 감염원의 공격에 대한 방어 및 치유 저하를 더욱 심화시키기 때문인 것으로 사료된다. 따라서 제 2 형 당뇨 환자의 수복치료 시 치과의사는 당뇨조절 하에서 치수 조직의 자극을 최소화하기 위한 세심한 처치가 필요하다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. Our department has developed an information system for routine hematology using down-sized computer system. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer. The polymer valve showed smaller regurgitation volume and transvalvular pressure drop compared to the mechanical valve at higher heart rate. Because the point of maximun temperature of laser-irradiated gelatin exists below the surface due to surface cooling, evaporation at the boiling temperature is made explosively from below the surface. The important parameters of this process are the conduction loss to laser power absorption (defined as the conduction-to-laser power parameter, Nk), the convection heat transfer at the surface to conduction loss (defined as Bi), dimensionless extinction coefficient (defined as Br.), and dimensionless irradiation time (defined as Fo). Dependence of Fo on Nk and Bi has been observed by experiment, and the results have been compared with the numerical results obtained by solving a 2-dimensional conduction equation. Fo and explosion depth (from the surface to the point of maximun temperature) are increased when Nk and Bi are increased.To find out the minimum laser power for explosive evaporative removal process, steady state analysis has been also made. We obtained a two-dimensional current density distribution within a phantom by processing the real and imaginary MR images from a 4.77 NMR machine. Optimal stimulus parameters, which can minimize muscle fatigue and can achieve effective muscle contraction were determined in slow and fast muscles in Sprague-Dawley rats. Stimulus patterns of our stimulator were designed to simulate electromyographic activity monitored during locomotion of normal subjects.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm.   • 기계적 응력은 정상적인 발달과정 동안 조직의 항상성에 있어 중요한 역할을 한다. 기계적 응력은 치아 이동과 저작과 같은 상황을 포함한다. 치아 이동과 저작 중에 치주인대 섬유모세포는 기계적 자극을 감지하고 주위의 세포밖 물질과 생체분자 대사의 변동에 의해 반응을 보인다. 그러나 아직까지 기계적 응력 하에서 치주인대 세포에서 발현된 유전자에 관한 연구는 미비한 실정이다. 최근 기계적 응력이 초파리에 존재하는 UNC-50 유전자에 영향을 줄 수 있다고 보고되었다. 또한 UNC-50은 치은 섬유모세포와 비교해 치주인대 섬유모세포에서만 발현된다고 보고되었다. 본 연구에서는 간헐적 교정력 적용 시에 백서의 치아에서 일어나는 치근 및 치주 조직의 조직학적 변화와, UNC-50의 발현 양상을 면역조직학적 염색으로 조사하여 치주인대에서 기계적 응력과 UNC-50의 관련성을 알아보고자 하였다. Sprague-Dawely계 수컷 백서 12마리를 4마리씩 세 군으로 나누어 상악 우측 구치부에 NiTi closed coil spring을 사용하여 40 g 정도의 견인력이 발생하도록 하여 하루에 1시간씩 간헐적인 교정력을 적용한 후 1, 3, 5일 후 치주인대의 조직학적 변화를 관찰하여 다음과 같은 결과를 얻었다. 조직학적 소견에서 상악 제1대구치 근심구개치근의 치근부 1/3에서 압박측은 안장측보다 더 좁은 치주인대공간을 보였고 교정력을 적용시킨 후 3일 후부터 인장측에서 백악모세포 활성으로 인한 백악질 침착이 관찰되었다. UNC-50은 인장측의 분화 중인 백악모세포에서 강한 발현을 보였다. Osteocalcin은 인장측에서 압박측에 비해 신생 백악질에 존재하는 분화 중인 백악모세포를 따라 강한 발현을 보였다. 이상의 연구결과는 UNC-50이 간헐적 교정력 즉 기계적 응력의 변화에 따른 백악모세포의 분화과정에 중요한 역할을 함을 나타낸다. 그러나 이를 명확히 하기 위해서는 교정력 적용 후 UNC-50의 세포내 신호전달과정에 대한 보완연구가 필요할 것이다.

중합효소연쇄반응을 이용한 HLA-class I, II 유전자군의 유전적 다형성에 관한 연구 (A Study of Genetic Polymonhisms of HLA-class I and II Genes Using Polymerase Chain Reaction)

  • HLA 유전자군은 인간의 유전자 중에서 가장 높은 유전적 다형성을 보이며, 분석방법에 따라 판정할 수 있는 대립유전자의 수에 많은 차이가 있다. 현재까지 혈청학적 방법을 이용하여 HLA항원형 구분을 하였으나, 최근 골수이식 등 여러 HLA활용분야에서 HLA유전자형 분석이 요구되고 있어, 많은 수의 HLA유전자형을 쉽고 정확하게 구분할 수 있는 HLA DNA typing 방법이 필요한 실정이다. 본 연구에서는 HLA-A, B, C, DRBI 유전자형 구분은 PCR-SSP 방법을, HLA-DQAl, DQBl, DPBl 유전자형 구분은 PCR-RFLP 방법을 사용한 HLA DNA typing 방법으로 한국인에서 HLA 유전자형을 구분하고자 하였다. 본 방법을 이용하여 HLA형이 규명된 B-임파아구 표준세포에서 DNA typing을 실시하였을 때, 11차 국제 조직적 합성학회에서 발표된 결과와 모두 일치하였다. 한국인에서 HLA-A, -B, -C 대 립 유전자는 17종, 23종, 16종이 확인되었으며, HLA-DQAl, -DQBl, -DPBl, -DRBl 대립유전자는 8종, 16종, 13종, 37종이 확인되었다. 한국인에서 빈도가 높은 HLA-class I 유전자는 HLA-A 유전자에서 $A^*$02가 27.0%였으며 HLA-B 유전자에서 는 $B^*$40이 17.6%를 나타내 었고 HLA-C 유전자에서는 Cw$^*$0101이 19.2%로 가장 높은 빈도를 나타내었다. 한국인에서 가장 빈도가 높은 HLA-class II 유전자는 DQAl 유전자에서 DQAl$^*$0301이 32.1%였고, DQBl 유전자에서는 DQBl$^*$0303이 12.9%를 나타내었으며, DPBl 유전자에서는 DPBl$^*$0501이 31.3%였고 DRBl 유전자에서는 DRBl$^*$1501이 9.2%를 나타내었다. 본 연구에서 실시한 HLA DNA typing 방법은 비교적 빠른 시간 내에 많은 종류의 HLA 대립 유전자형을 정확하게 구분할 수 있으므로 앞으로 tissue typing 실험실에서 유용하게 활용될 수 있을 것으로 생각된다. 또한 DNA typing방법을 이용하여 분석한 한국인의 HLA-class I, II 유전자군의 유전자형 빈도는 골수이식을 비롯한 각종 이식검사, 특수 질환 관련검사나 인류유전학연구 등 HLA 유전자의 임상적 활용을 위한 자료로 사용될 수 있을 것으로 기대된다.

  • The treatment for oral and maxillofacial carcinoma with chemotherapeutic agents is evaluated by many effective methods to reduce the tumor mass and cancer cell proliferation. However these chemotherapy have many serious side effects, such as bone marrow suppression, renal toxicity, G-I troubles. Therefore a possible approach to develop a clinically applicable chemotherapeutic agent is to screen anticancer activity of Taxol which is known to have very little side effect and have been used to breast cancer and ovarian carcinoma. Taxol is a new anti-microtubular anti-cancer agent extracted from the bark of the Pacific yew, Taxus brevifolia. Paclitaxel(Taxol) acts by promoting tubulin polymerization and over stabilizing microtubules agianst depolymerization. Despite the constant improvements of methods of the cancer treatment especially chemotherapy, the rate of cancer metastasis and recurrent are not decreased. Thus the investigation of new drug which have very little side effect and a possible clinically application continues to be a high priority. Considering that the Taxol have shown very effective chemotherapeutic agent with relatively low toxicity in many solid tumors, it deserves to evaluate its efficacy in oral squamous cell carcinoma. In this study, to investigate the in-vivo and in-vitro anti-cancer efficacy of Taxol in oral squamous cell carcinoma and lastly, the potency of Paclitaxel in the clinical application for oral cancer was evaluated. In vivo study, after HN22 cell line were xenografted in nude mice, the growth of tumor mass was observed, 3 mg/Kg taxol was injected intraperitoneally into nude mice containing tumor mass. The methods of these study were measurement of total volume of tumor mass, histopathologic study, immunohistochemical study, drug resistance assay, growth curve, MTT assay, flow cytometry, cDNA microarray in vivo and in vitro. The results were obtained as following. 1. The visual inspection of the experimental group showed that the volume of the tumor mass was slightly decreased but no significant difference with control group. 2. Ki-67 index was decreased at weeks 4 in experimental group. 3. Microscopic view of the xenografted tumor mass showed well differentiated squamous cell carcinoma and after Taxol injection, some necrotic tissue was seen weeks 4. 4. The growth curve of the tumor cells were decreased after 1day Taxol treatment. 5. According to the MTT assay, HN22 cell line showed relative drug resistancy above $5\;{\mu}g/ml$ concentrations of Taxol. 6. In drug resistance assay, the decrease of cell counts was seen relatively according to concentration. 7. In Flow cytometry, G2M phase cell arrests were seen in low concentration of the Taxol, while S phase cell arrests were seen in high concentration of the Taxol. 8. Using cDNA microarray technique, variable gene expression of ANGPTL4, TXNRD1, FAS, RRAGA, CTGF, CYCLINEA, P19, DUSP5, CEBPG, BTG1 were detacted in the oral squamous cell carcinoma cell after taxol treatment. In this study paclitaxel is effective against oral squamous cell carcinoma cell lines in vitro, but week effect was observed in vivo. So we need continuous study about anticancer effect of taxol in vivo in oral squamous cell carcinoma.

성견의 실험적 치아이동시 치주조직내 대식세포와 T-림파구의 활성에 관한 면역조직화학적 연구 (AN IMMUNOHISTOLOGIC STUDY ON THE ACTIVITY OF MACROPHAGE AND T-CELL IN THE PERIODONTIUM DURING TOOTH MOVEMENT OF DOG)

  • 교정력에 의한 성견의 치아이동시 발생하는 조직변화 중 염증과 면역반응에 중요한 역할을 하는 대식세포와 T-림파구의 치주조직내 활성도를 교정력의 크기와 기간에 따라 알아보고자 하였다. 생후 1년 6개월된 성견 6마리 중 5마리의 실험군과 1마리의 대조군이 연구되었는데 실험군에서 하악 좌측에는 강한 힘(250-300g)을, 하악 우측에는 약한 힘(50-75g)을 제1소구치와 제2소구치 사이에 open-coil spring으로 적용하였으며, 실험군의 성견을 각각 12시간, 24시간, 3일, 1주, 2주에 희생시켜 얻은 조직에 $\alpha$-1-antichymotiypsin과 CD3를 이용한 면역조직화학적 염색을 시행하였다. 결과적으로 염증세포침윤은 약한 힘을 준 경우보다 강한 힘을 준 경우에서 3일째까지 더 많이 나타나다가 7일이후에는 차이가 없어졌고 견인측보다는 압박측에서 많이 나타났으며, 압박측의 파골세포 활성은 12시간째에서부터 관찰되어 증가하다가 7일째부터 감소되었는데 약한 힘을 준 경우에서보다 강한 힘을 준 경우에서 더 많이 나타났다. 견인측 치주인대의 단절과 혈관확장은 12시간째 관찰되어 증가하다가 3일째 이후에는 감소되었는데 약한 힘을 준 경우보다는 강한 힘을 준 경우에서 더 많았으나 14일째에는 차이가 없었다. 대조군의 $\alpha$-1-antichymotiypsin발현은 열구상피에서 주로 발현되었고 치주인대, 치수, 골세포에서는 음성반응을 보였으며, 실험군의 $\alpha$-1-antichymotiypsin발현은 견인측보다 압박측에서 많았으며, 3일째까지는 치경부쪽이 약간 많았고, 7일째 이후에는 주로 치경부보다는 치근단쪽에서 더 많이 발현되었다. 약한 힘을 준 경우는 $\alpha$-1-antichymotiypsin 염색이 12시간째부터 나타나서 7일째에 가장 많이 나타나다가 이후 감소되었고, 강한 힘을 준 경우에서는 3일째에서 최고에 달하다가 이후에는 감소되었다. 약한 힘을 준 경우나 강한 힘을 준 경우 모두에서 CD3에 대한 발색은 전치주조직을 통해 거의 음성으로 나타났다.
