Background and Objectives: Osteoarthritis (OA) is a degenerative disease that leads to the progressive destruction of articular cartilage. Current clinical therapeutic strategies are moderately effective at relieving OA-associated pain but cannot induce chondrocyte differentiation or achieve cartilage regeneration. We investigated the ability of wedelolactone, a biologically active natural product that occurs in Eclipta alba (false daisy), to promote chondrogenic differentiation. Methods and Results: Real-time reverse transcription-polymerase chain reaction, immunohistochemical staining, and immunofluorescence staining assays were used to evaluate the effects of wedelolactone on the chondrogenic differentiation of mesenchymal stem cells (MSCs). RNA sequencing, microRNA (miRNA) sequencing, and isobaric tags for relative and absolute quantitation analyses were performed to explore the mechanism by which wedelolactone promotes the chondrogenic differentiation of MSCs. We found that wedelolactone facilitates the chondrogenic differentiation of human induced pluripotent stem cell-derived MSCs and rat bone-marrow MSCs. Moreover, the forkhead box O (FOXO) signaling pathway was upregulated by wedelolactone during chondrogenic differentiation, and a FOXO1 inhibitor attenuated the effect of wedelolactone on chondrocyte differentiation. We determined that wedelolactone reduces enhancer of zeste homolog 2 (EZH2)-mediated histone H3 lysine 27 trimethylation of the promoter region of FOXO1 to upregulate its transcription. Additionally, we found that wedelolactone represses miR-1271-5p expression, and that miR-1271-5p post-transcriptionally suppresses the expression of FOXO1 that is dependent on the binding of miR-1271-5p to the FOXO1 3'-untranscribed region. Conclusions: These results indicate that wedelolactone suppresses the activity of EZH2 to facilitate the chondrogenic differentiation of MSCs by activating the FOXO1 signaling pathway. Wedelolactone may therefore improve cartilage regeneration in diseases characterized by inflammatory tissue destruction, such as OA.
본 연구는 제대혈과 골수로부터 얻은 조혈모세포를 재조합 retrovirus로 감염시킬 때의 최적조건을 human growth hormone (hGH)과 $\beta$-galactosidase를 발현하는 두 가지의 다른 retroviral vector를 이용하여 찾았다. Retrovirus는 자라는 세포에만 감염하는 것으로 알려져 있어 이에 대한 최적조건을 구하기 위해 세포 배양을 통해 조혈모세포의 성장곡선을 얻었으며, 또한 감염된 세포를 환자에게 다시 넣는 유전자요법에서는 이 세포가 체내에서 가능하면 조혈모세포의 기능을 가지는 것이 요구되어 이 때 얻어진 세포의 분열능을 나타내는 집락형성 세포분율을 구하였다. 우선, 세포성장에 대해 조사한 결과 초기에 넣은 세포농도가 5$\times$$10^4$세포/mL일 때 세포성장속도가 가장 빠른 것으로 나타났다. 그러나, 배양시간이 지남에 따라 집락을 형성할 수 있는 능력은 급격하게 감소하여 유전자요법을 위한 최적조건을 구하기 위해서는 이를 고려한 최적화가 필요하였다. 이를 위한 예비실험으로 감염이 잘 된다고 알려진 NIH3T3 세포에 retrovirus 상층액으로 감염시킨 결과 성공적으로 유전자가 전달된 것을 배지에 분비되는 hGH을 측정하여 확인하였다. 이러한 결과로부터 hGH을 발현하는 재조합 retrovirus는 정상적으로 작동하는 것을 확인하였다. 그러나, 조혈모세포와 retrovirus를 분비하는 packaging cell을 동시 배양하는 방법을 채택하였다. 제대혈로부터 얻은 조혈모세포와 대장균 lacZ 유전자로부터 $\beta$-galactosidase를 분비하는 packaging cell을 이용한 경우 동시배양의 경우 조혈모세포를 3일 동안 세포배양을 한 후 이 증식된 세포를 48시간 동안 동시배양하면서 감염시켰을 때 최대의 감염율을 나타내었다. 한편, 골수로부터 얻은 조혈모세포와 hGH을 분비하는 packaging cell과 동시배양시켰을 때 세포농도가 다름에도 불구하고 제대혈에서와 마찬가지로 조혈모세포를 3일 동안 세포배양한 후 48시간 동안 동시배양하는 경우에 hGH이 최대로 분비되었다. 이러한 결과로부터 세포의 source나 세포농도와 관계없이 유전자전달을 통한 단백질의 발현에 있어서 최적조건이 존재하였다. 그러나, 이러한 경우에 유전자전달이 완료되는 시점이 배양을 시작한지 5일이 되므로 집락을 형성할 수 있는 세포의 분율이 약 1/3로 감소하였다. 따라서, 이러한 결과를 유전자요법에 적용하는 경우에는 그 목적에 따라 적절한 실험조건을 선정하는 것이 필요하리라 사료된다.
4주령의 돼지에서 여러 용량의 방사선 조사를 하고 경시적으로 혈액 수치의 변화, helper T-cell 및 cytotoxic T-cell에 대한 변화를 관찰하였다. 6 Gy 이상을 조사한 돼지에 3일 이후 백혈구와 혈소판의 수의 감소되어 회복되지 않았으며, 식욕 부진,설사 등의 부작용을 보이며, 조사 후 10일부터 15일 사이에 모두 폐사 하였다. 4 Gy 전신 단일 조사한 돼지에서는 조사 후 임상적인 부작용을 보이지 않았으며, 9일 이후 백혈구, 혈소판이 증가하여 회복되는 경향을 보였다. 본 연구의 결과는 돼지를 사용한 방사선 생물학적 실험에서 혈액변화 및 면역억압 관찰에 대한 기초 자료로 활용 될 수 있을 것이다.

Graft-versus-host disease (GVHD) is a fatal complication that occurs after allogeneic hematopoietic stem cell transplantation. To understand the dynamics of CD4 and CD8 T cell production of IFN-${\gamma}$ and IL-17 during GVHD progression, we established a GVHD model by transplanting T cell-depleted bone marrow (TCD-BM) and purified T cells from B6 mice into irradiated BALB.B, creating an MHC-matched but minor histocompatibility (H) antigen-mismatched transplantation (B6 ${\rightarrow}$ BALB.B GVHD). Transplantation-induced GVHD was confirmed by the presence of the appropriate compositional changes in the T cell compartments and innate immune cells in the blood and the systemic secretion of inflammatory cytokines. Using this B6 ${\rightarrow}$ BALB.B GVHD model, we showed that the production of IFN-${\gamma}$ and IL-17 by CD4 T cells preceded that by CD8 T cells in the spleen, mesenteric lymph node, liver, and lung in the BALB.B GVHD host, and Th1 differentiation predated Th17 differentiation in all organs during GVHD progression. Such changes in cytokine production were based on changes in cytokine gene expression by the T cells at different time points during GVHD development. These results demonstrate that both IFN-${\gamma}$ and IL-17 are produced by CD4 and CD8 T cells but with different kinetics during GVHD progression.

Background: Matrix metalloproteinase -2 (gelatinase-A, Mr 72,000 type IV collagenase, MMP-2) and -9 (gelatinase-B, Mr 92,000 type IV collagenase, MMP-9) are key molecules that play roles in tumor growth, invasion, tissue remodeling, metastasis and stem-cell regulation by digesting extracellular matrix barriers. MMP-2 and -9 are well known to impact on solid cancer susceptibility, whereas, in hematological malignancies, a paucity of data is available to resolve the function of these regulatory molecules in bone marrow mononuclear cells (BM-MNCs) and stromal cells of myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML). Objectives: The present study aimed to investigate mRNA expression and gelatinase A and B secretion from BM-MNCs in vitro and genotypic associations of MMP-2 (-1306 C/T; rs243865), MMP-9 (-1562 C/T; rs3918242), tissue inhibitor of metalloproteinase -1 (TIMP-1) (372T/C; rs4898, Exon 5) and TIMP-2 (-418G/C; rs8179090) in MDS and AML. Results: The study covered cases of confirmed MDS (n=50), AML (n=32) and healthy controls (n=110). MMP-9 mRNA expression revealed 2 fold increased expression in MDS-RAEB II and 2.5 fold in AML M-4 (60-70% blasts). Secretion of gelatinase-B also revealed the MMP-9 mRNA expression and ELISA data also supported these data. We noted that those patients having more blast crises presented with more secretion of MMP-9 and its mRNA expression. In contrast MMP-9 (-1562 C/T) showed significant polymorphic associations in MDS (p<0.02) and AML (p<0.02). MMP-9 mRNA expression of C/T and T/T genotypes were 1.5 and 2.5 fold increased in MDS and AML respectively. In AML, MMP-2 C/T and T/T genotypes showed 2.0 fold mRNA expression. Only MMP-9 (-1306 C/T) showed significant 4 fold (p<0.001) increased risk with chemical and x-ray exposed MDS, while tobacco and cigarette smokers have 3 fold (p<0.04) risk in AML. Conclusions: In view of our results, MMP-9 revealed synergistic secretion and expression in blast crises of MDS and AML with 'gene' polymorphic effects and is significantly associated with increased risk with tobacco, cigarette and environmental exposure. Release and secretion of these enzymes may influence hematopoietic cell behavior and may be important in the clinical point of view. It may offer valuable tools for diagnosis and prognosis, as well as possible targets for the treatments.
본 연구는 사람의 다양한 세포주를 이용하여 활성산소종(과산화수소수)이 세포의 노화에 미치는 영향을 비교 조사하였다. 여러 농도의 과산화수소수에 세포주를 일주일 동안 배양하여 MTT 방법으로 과산화수소수에 대한 세포 성장의 반억제농도를 구하였다. 그 결과, 50대에서 유래하는 피부 섬유아세포와 10대의 노화 유도 피부 섬유아세포와 비교하여 10대에서 유래하는 피부 섬유아세포에서 과산화수소수에 대한 반억제농도의 값이 유의적으로 더 높았고, 10대의 피부 섬유아세포보다는 10대의 여러 조직 기원하는 성체줄기세포에서 반억제농도의 값이 유의적으로 더 높게 관찰되었다. 또한, 50 ppm 과산화수소수를 1주일 동안 처리한 후, 50대의 피부 섬유아세포에서 다른 세포주에 비해 세포 성장이 현저히 억제되었고, 노화 관련 베타-갈락토시다아제의 활성이 증가되는 것을 관찰하였다. 또한, 활성산소의 세포 독성을 중화시키는 두 유전자, 글루타티온 과산화효소(GPX)와 카탈라아제(CAT)의 발현을 각 세포주에서 조사하였을 때, CAT의 발현은 모든 세포주에서 대체로 낮았지만, GPX 유전자의 발현이 50대의 피부 섬유아세포보다 10대의 피부 섬유아세포와 성체줄기세포에서 현저히 높게 발현되는 것을 관찰하였다. 이상의 결과에서 활성산소는 세포 노화를 유도하고, GPX의 발현이 높은 10대의 피부 섬유아세포와 줄기세포보다는 50대의 피부 섬유아세포와 노화된 피부 섬유아세포에서 활성산소종에 대해 더 큰 민감성을 가지고 있는 것을 알 수 있었다.
중간엽 줄기세포(MSC)를 체외배양할 때 사용하는 소태아혈청 (FBS)의 생물학적 불안전성은 이를 임상적으로 사용하는데 있어 제한점으로 작용한다. 본 연구에서는 소태아혈청 및 인간의 제대혈청을 사용하여 인간의 양막유래 중간엽 줄기세포 (HAM)를 각각 배양한 후, 세포의 성장속도와 유전자 및 단백질의 발현 양상을 비교하였다. 제왕절개 후 얻은 양막으로부터 HAM을 분리하여 10% FBS, 5% HCS 혹은 10% HCS가 각각 첨가된 DMEM 배양액에서 배양하였으며, 초기와 후기 계대의 세포를 얻어 이들의 생물학적 특성을 비교 분석하였다. 역전자 중합효소반응 결과, 혈청의 종류에 상관없이 배양된 세포들은 모두 OCT-4, Rex-1, SCF, FGF-5, BMP-4, nestin, NCAM, GATA-4, HLA-ABC 유전자를 발현하였으며, 이러한 발현은 초기 및 후기 계대의 세포에서도 마찬가지로 나타났다. 세포면역화학 반응 결과, FBS 혹은 HCS를 첨가한 배양액에서 배양된 HAM은 4번째 계대에서 collagen I, II, III, XII, fibronectin, $\alpha$-smooth muscle actin, vimentin, CK18, CD54, FSP, TRA-1-60, SSEA-3, -4, HLA ABC 단백질을 뚜렷하게 발현하였다. 그러나 desmin 단백질은 FBS가 첨가된 배양액에서 배양된 HAM에서만 발현되었고 vWF 단백질은 HCS가 첨가된 배양액에서 배양된 HAM에서만 발현되었다. 결론적으로 유전자와 단백질의 발현양상을 살펴본 결과, HCS가 첨가된 배양액에서 배양된 HAM은 전형적인 인간성체줄기세포의 특징을 나타내고 있으며, FBS가 첨가된 배양액과 비교하여 동등한 성장 촉진 효과를 가지는 것으로 보인다.
Objective : This study aimed to evaluate the hypotheses that administration routes [intra-arterial (IA) vs. intravenous (IV)] affect the early stage migration of transplanted human bone marrow-derived mesenchymal stem cells (hBM-MSCs) in acute brain infarction. Methods : Male Sprague-Dawley rats (n=40) were subjected to photothrombotic infarction. Three days after photothrombotic infarction, rats were randomly allocated to one of four experimental groups [IA group : n=12, IV group : n=12, superparamagnetic iron oxide (SPIO) group : n=8, control group : n=8]. All groups were subdivided into 1, 6, 24, and 48 hours groups according to time point of sacrifice. Magnetic resonance imaging (MRI) consisting of T2 weighted image (T2WI), $T2^*$ weighted image ($T2^*WI$), susceptibility weighted image (SWI), and diffusion weighted image of rat brain were obtained prior to and at 1, 6, 24, and 48 hours post-implantation. After final MRI, rats were sacrificed and grafted cells were analyzed in brain and lung specimen using Prussian blue and immunohistochemical staining. Results : Grafted cells appeared as dark signal intensity regions at the peri-lesional zone. In IA group, dark signals in peri-lesional zone were more prominent compared with IV group. SWI showed largest dark signal followed by $T2^*WI$ and T2WI in both IA and IV groups. On Prussian blue staining, IA administration showed substantially increased migration and a large number of transplanted hBM-MSCs in the target brain than IV administration. The Prussian blue-positive cells were not detected in SPIO and control groups. Conclusion : In a rat photothrombotic model of ischemic stroke, selective IA administration of human mesenchymal stem cells is more effective than IV administration. MRI and histological analyses revealed the time course of cell migration, and the numbers and distribution of hBM-MSCs delivered into the brain.
제대혈 조혈모세포 이식은 악성 종양 및 혈액 질환의 치료에 있어 중요한 치료법인 조혈모세포 이식의 하나로 이식 편대 숙주병의 발생이 적어 비혈연간 환아에서의 제대혈 이식이 활발히 이루어지고 있다. 이에 저자들은 급성 골수구성 백혈병인 환아에서 성공적으로 호중구 및 혈소판 생착을 보인 비혈연간, 주조직 적합 항원-3개 불일치 제대혈 조혈모세포 이식을 경험하였기에 이에 보고하는 바이다.
Background: Chronic myeloproliferative diseases are clonal stem cell diseases which occur as a result of uncontrollable growth and reproduction of hematopoietic stem cells, which are the myeloid series source in bone marrow. Recent studies have suggested that chronic inflammation can be a triggering factor in the clonal change in chronic myeloproliferative neoplasia (CMPN). In our study, we evaluated the existence of a chronic inflammation process in our Philadelphia negative (Ph-)CMPN patients using inflammation parameters in combination with demographic, laboratory and clinical characteristics of the patients. Materials and Methods: Demographic characteristics, clinical and laboratorial data, and thrombosis histories of 99 Ph-CMPN patients, who were diagnosed at our outpatient clinic of hematology in accordance with WHO 2008 criteria, were analyzed retrospectively,with 80 healthy individuals of matching gender and age included as controls. Complete blood counts, sedimentation, C reactive protein (CRP), JAK V617F gene mutations, abdomen ultrasound images and previous thrombosis histories of these patients were retrospectively analyzed. Results: Ph-CMPN and healthy control groups included 99 and 80 cases, respectively. PV, ET and MF diagnoses of patients were 43 (%43.4), 44 (44.4%) and 12 (12.1%), respectively. JAK V617F gene mutation was found to be positive in 64 (71.1%) of all cases and in 27(65.8%), 32 (82%), 5 (50%) of the cases in PV, ET and PMF groups, respectively. Thrombosis was determined as 12 (12%) in the entire group, 12.5% in the JAK V617F negative and 15.3% in the positive patients, with no statistical significance (p=0.758). No significant difference was observed between patients with and without previous thrombosis history in respect to hemogram parameters, sedimentation and CRP (p>0.05), neutrophil to lymphocyte ratio (NLR), erythrocyte distribution width (RDW), mean platelet volume (MPV) and sedimentation levels of the patient.

④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
