Evaluation of Antibacterial Efficacy of Certain Botanicals Against Bacterial Pathogen Bacillus sp. of Silkworm, Bombyx mori L.

  • An in vitro and in vivo studies were conducted to evaluate the antibacterial efficacy of certain botanicals viz., rhizomes of turmeric (Curcuma longa) and leaves of amla (Phyllanthus emblica), asparagus (Asparagus racemosus), bael (Aegle marmelos), boerhavia (Boerhavia diffusa), garlic (Allium sativum) and basil (Oscimum basicilum) against bacterial pathogens viz., Staphylococcus sp., Bacillus sp. and Klebsiella cloacae, of silkworm, Bombyx mori. Asparagus and basil, amla and boerhavia, basil and bael at concentration of 20, 000 ppm showed higher antibacterial activity against Staphylococcus sp., Bacillus sp., K. cloacae respectively, both in vitro and in vivo studies.

  • Studies on extended egg preservation schedule from 120 days to 180 days was taken up with 20 germplasm accessions of mutant silkworm genetic stocks of Bombyx mori L. Statistical analyses of the data collected over three trials revealed no significant changes both in the qualitative and quantitative traits of the genetic stocks between treatment (6 months egg preservation) and control (4 months egg preservation), except for fifth instar larval duration in TMS-61, TMS-62, TMS64, TMS-31 and TMS-34 shell weight in TMS-62, TMS-64 and TMS-66. Thus, the results indicate that extended schedule of 6 months egg preservation can safely be adopted, which will reduce the cost of conservation and minimize the genetic erosion owing to reduced crop cycle.

PCR-Based Detection of Densovirus Infection in Silkworm (Bombyx mori L.)

  • Two pairs of DNA primers were designed for the detection of the Zhenjiang (China) strain of Bombyx mori densonucleosis virus (BmDNV-Z). These primers were designed from the nucleotide sequence of major structural protein gene (putative VD1-ORF2). PCR amplification was attempted from different issues (including silk gland, blood, skin and midgut) and feces of the silkworm which infected wit BmDNV-Z were amplified by PCR. Both of the primers gave expected size of in the DNA bands from midgut and feces, but not in the DNA of silk gland, blood and skin. The two bands were sequenced, and their sequence were same as the sequence designed for. BmDNV-Z could be successfully detected in single silkworm after it was infected for 12 hrs, and could not be detected before 9 hrs after infected.

  • 10-3, 10-4, 10-5으로 희석한 인세가 희석액을 배자발생 초기, 중기, 말기의 누에씨 표면에 각각 분무한 후, 그 부화비율을 조사하였다. 부화비율은 인세가를 분무한 모든 시험구에서 저하하였는데 약제의 농도가 높을수록 또한 처리시기가 빠를수록 그 정도가 심했다.

  • The silkworm, Bombyx mori, is prone to infection of various pathogenic organisms. Pebrine, one of the deadliest disease of silkworm caused by highly virulent parasitic microsporidian, Nosema bombycis has been understood since long. Infections of the disease range from chronic to highly virulent and can result in complete loss to the sericulture industry. Several strains and species of microsporidians have since been isolated from the infected silkworms; the disease is becoming increasingly more and more complex. Epizootiology, development of immunodiagnostic kit, use of chemotherapy and thermotherapy techniques has been addressed for identification and control of the disease. A technique of delayed mother moth examination, which plays a decisive role in the detection of the disease and harvestation of stable cocoon crop, has been described. An attempt has been made to review briefly the literature available on various aspects of the pebrine disease in order to develop efficient model(s) for the prevention and control of the disease and to suggest future avenues of investigation in the field of pebrine disease management.

Electrophoretic Analysis of Nonspecific Esterases in Silkworm (Bombyx mori L.) Female Genital Organs and Eggs

  • By using PAGE, a study was made on the nonspecific esterase spectra of female genital organs and eggs in Bombyx mori L. The expression of 11 esterase bands was detected during ontogenesis of races and inter-races hybrids kept in Bulgaria. The gene activity of 9 esterase loci was assumed. Esterases specific for the spectrum of diapausing eggs were observed. In two esterase zones, intra- and inter-breed polymorphism was found. Based on the same breed specific expression, the existence of correspondence between esterase bands from spectra of different silkworm tissues and organs was suggested. Stage-specific expression of esterases in female genital glands, indicative of differentiated gene activity during ontogenesis, was established.

Study on Productivity and Genotype Structure by Several Enzyme Loci of Silkworm(Bombyx mori L.) Parthenoclones Obtained by Thermal and Combined(Low-High Temperature) Method

  • The purpose of this study was to establish whether there are differences in the productivity of the same silkworm(Bombyx mori L.) parthenoclones, obtained by two different methods-thermal and combined, as well as to study their genotype structure by several enzyme loci. It was established that all individuals of parthenoclones Joana, Joana(${\downarrow}{\uparrow}$), Pohi and Pohi(${\downarrow}{\uparrow}$), are homozygous by the studied esterase and phosphoglucomutase loci, which substantiated the clones' genetic stability. By comparative analysis of some biological and technological properties, it was found that parthenoclone Pohi(${\downarrow}{\uparrow}$) obtained by low-high temperature activation is characterized by higher values of these properties as compared to parthenoclone Pohi obtained by thermal parthenogenesis. Comparing the two methods of inducing ameiotic parthenogenetic development, we would recommend that parthenoclone Joana is sustained by thermal parthenogenesis, and parthenoclone Pohi-by the combined method (low-high temperature).

  • 누에의 성충체액 주단백질(AMHP)의 생리·생화학적 기능을 구명하기 위한 연구의 하나로 AMHP의 생체내 합성과정을 autoradiography, 효소 혈청학적 방법 및 지방체의 in vitro 배양실험 등에 의해 조사하였다. 그 결과 성충의 지방체는 AMHP의 subunnit인 18K및 20K의 polypeptide를 합성하여 체액중으로 방출하며, 방출된 이들은 체액중에서 서로 결합하여 천연의 AMHP를 형성하는 것으로 확인되었다.

Transcription of Some Early and Late Genes of Bombyx mori Nuclear Polyhedrosis Virus in the Cells

  • To understand expression of some early and late genes of Bombyx mori nuclear polyhedrosis virus (BmNPV) in the B. mori-derived BmN cell line, the transcripts were analyzed by polymerase chain reaction with synthetic primers. After infection, the transcript of early genes, which include p35, IE1 and helicase p143, was immediately detected in the infected cells. In addition, the transcript of late genes, which include p10 and polyhedrin, was also detected in just-infected cells. In conclusion, our results revealed that transcripts of early and late genes of BmNPV are immediately expressed from the cells after infection.

  • Mating is an essential behavioural social event in the life cycle of silkworm, Bombyx mori (L.) for the perpetuation of population. A number of intrinsic and extrinsic factors and events of significant importance are involved in successful mating and egg deposition by an adult silk moth which besides biochemical, physiological and environmental factors also includes attraction of reproductively competent male and female moth for mating, duration and frequency of mating, age of moth at the time of mating, reuse of male moth in the production of eggs etc. An attempt has been made in this review article to elucidate briefly the behaviour of male towards female moth after eclosion, impact of duration and frequency of mating on egg deposition and oviposition, reuse of mated male moth in the production of quality and quantity eggs etc. in the silk-worm, B. mori and its significance in silkworm seed production.
