Evolutionary Relationship of Liobagrus mediadiposalis (Teleostei: Amblycipitidae) Populations in Korea Inferred from Cytochrome b DNA Sequences (한국 고유종인 자가사리(Liobagrus mediadiposalis) 지역개체군의 분자진화적 유연관계)

  • Phylogenetic relationships and DNA polymorphism among local populations of the Korean native catfish species, Liobagrus mediadiposalis, have been investigated based on mitochondrial cytochrome b DNA sequences. As a result, three genetically distinct groups of local populations were recognized based on the phylogenetic tree constructed. The first group was called "Nakdong-river group" that included the local populations from Geumho-river, Gyeongho-river, and Deokcheon-river; the second one was "Geum-river group"; the third one was represented as "Seomjin-river group" that included the samples from Seomjin-river, Dongjin-river and Geokum-do. The phylogeny also implied that the ancestral group of L. mediadiposalis have first evolved to Nakdong-river group, and later two local populations (Geum-river and Seomjinriver group) were diverged from the other lineage. DNA polymorphisms we observed were 4.4~4.7% between Geum-river group and Seomjin-river group, 5.1~5.5% between Seomjinriver group and Nakdong-river group, and 5.5~5.7% between Nakdong-river group and Geumriver group. These results indicated the long period of geographic isolation due to the river system in Korea caused such high degrees of DNA polymorphisms between local populations of L. mediadiposalis.

Chemical Component Analysis of Red Pepper (Capsicum annuum L.) Seeds with Various Cultivars (품종별 고추씨의 화학적 성분 분석)

  • This study was carried to investigate the proximate compositions, capsacinoids, organic acid of various red pepper (Capscium annuum L.) seeds. Twenty domestic varieties of red pepper seeds cultivated in Korea and four foreign varieties of samples cultivated in New Mexico, USA were analyzed. In proximate composition of the various red pepper seeds, ash content showed a range of $3.11{\pm}0.03{\sim}3.77{\pm}0.01%$, and protein content showed a range of $13.25{\pm}0.01{\sim}16.53{\pm}0.08%$. On the other hand, crude lipid content showed wide range distribution of $18{\sim}30%$ and total dietary fiber showed a range of $40{\sim}65%$. Capsaicin content of various red pepper seeds showed $0.09{\pm}0.04{\sim}5.32{\pm}0.14\;mg%$ and dihydrocapsaicin was $0.00{\sim}2.17{\pm}0.18\;mg%$. Capsaicin content was higher than dihydrocapsaicin content. In the organic acid content, Dangchan, Chohyang and Mixed red pepper seed showed a low range of $1.6{\sim}1.9%$; in contrast, Chunhamujuk, Samgang, Ganggun, Wangdaebak, Chunhailpum, New Mexico 6 and LB-25 showed higher values of over 5%.

DC ∼ 45 GHz CPW Wideband Distributed Amplifier Using MHEMT (MHEMT를 이용한 DC ∼ 45 GHz CPW 광대역 분산 증폭기 설계 및 제작)

  • In this paper, CPW wideband distributed amplifier was designed and fabricated using 0.1 $\mum$ InGaAs/InAlAs/GaAs Metamorphic HEMT(High Electron Mobility Transistor). The DC characteristics of MHEMT are 442 mA/mm of drain current density, 409 mS/mm of maximum transconductance. The current gain cut-off frequency(fT) is 140 GHz and the maximum oscillation frequency(fmax) is 447 GHz. The distributed amplifier was designed using 0.1 $\mum$ MHEMT and CPW technology. We designed the structure of CPW curve, tee and cross to analyze the discontinuity characteristics of the CPW line. The MIMIC circuit patterns were optimized electromagnetic field through momentum. The designed distributed amplifier was fabricated using our MIMIC standard process. The measured results show S21 gain of above 6 dB from DC to 45 GHz. Input reflection coefficient S11 of -10 dB, and output reflection coefficient S22 of -7 dB at 45 GHz, respectively. The chip size of the fabricated CPW distributed amplifier is 2.0 mm$\times$l.2 mm.

Physiological Activities of Korean and Chinese Viola mandshurica Extracts (국내산과 중국산 자화지정(Viola mandshurica) 추출물의 생리활성)

  • This study was carried out to compare the physiological activities of the extracts from Korean and Chinese Viola mandshurica W. Baker. The water extract from leaves of Chinese V. mandshurica exhibited the highest extraction yields of 30.45 g/100 g and the highest content of total flavonoids as 102.30 mg/g. Also, its ethanol extract showed the best content of polyphenol compounds as 136.16 mg/g. The leaf extract of Korean V. mandshurica produced higher electron donating abilities (EDA) of 92.69% (KLW) and 93.61% (KLE) than the other fractions. The strongest SOD-like activity was shown in the ethanol extract from Korean leaves of 17.28% at the concentration of 1.0 mg/mL. The nitrite scavenging abilities (NSA) of the leaf extracts of V. mandshurica from China were intense over 50% at pH 1.2 and 3.0. In the results of inhibitory rates of xanthine oxidase (XO), both ethanol extracts from Korean and Chinese leaves were higher than the other fractions as 98.67% and 93.80% respectively. Effect of tyrosinase inhibition was the highest in the water extract (45.04%) of Chinese leaves, followed by its ethanol extract (31.36%). The results of EDA, SOD-like activity and XO inhibition of the leaf extracts from Korean V. mandshurica were higher than those of Chinese, on the other hand, determinations on total polyphenol contents, NSA and tyrosinase inhibition were higher in those of Chinese.

Growth and Yield of Strawberry $(Fragaria\;\times\;ananassa\;Duchesne)$ 'Nyoho' and Salt Accumulation in PE Film House Soil as Affected by Fertilization Program (시비방법이 무기염의 토양집적과 딸기 생육 및 수량에 미치는 영향)

  • This research was conducted to determine the effect of fertilization programs on crop growth, nutrient uptake and yield of strawberry and salt accumulation in PE film house soil. To achieve these, experiments were conducted through two years with various fertilization program such as traditional fertilization(A), recommended fertilization of Rural development administration (B), Tochigi prefectural experiment station (C) and Nonsan strawberry experiment station (D), and control (E). In 1st year, statistical differences in growth characteristics were not observed among treatments except (E). Yield per 10 was the highest in (D) followed by (A), (B), (C) and (E) and there was the statistical difference between (D) and (E). In the 2nd year experiment, growth characteristics and yield showed similar trends to those of 1st year experiment. In the results of soil analysis of 2nd year, the soil pH of all treatments were in the acceptable range, while electrical conductivity of (A) and (D) were 2.36 and 2.19 %$dS{\cdot}m^{-1}$, respectively. After finishing of strawberry cultivation, nitrate concentration in soil solution of (A) and (D) were 74.6 and 65.0%$mg{\cdot}L^{-1}$ and Na in those were 3.71 and 3.53 $cmol+kg^{-1}$, respectively. Above results indicated that (A) and (D) were good fertilization program for strawberry cultivation, but those also resulted in accumulation of $NO_{3-}N$ and Na in PE house soil.

Influences by position of node and existence of leaf on microtuberization in node culture of potato (감자절간배양에서 마디위치 및 잎의 부착유무에 따른 소괴경형성의 차이)

  • Single-node stem pieces ca. 1 cm in length containing a axillary bud were obtained from in vitro plants of potato (Solanum tuberosum L.). The influences by a position of the node and the existence of a leaf at the node were observed in the single-node culture on the 8% sucrose MS medium. The effect of CCC was also investigated for the microtuberization. The apical part node was excellent in the tuberization not to mention shoot length, fresh weight, diameter, the number of node on the in vitro culture of a single-node than the lower part. The differences in the diameter of a tuber formed in the part of the axillary bud on all treatments including the cultivation of the apical part node were not recognized. However, the fresh weight of the tuber showed high value in the tuber formed at the axillary bud of shoot apex part. At 20 days after cultivation, tuberization was promoted in the new stolen that developed from the bud of node with a leaf under SD condition of 8 hours at $20^{\circ}C$. The tuberization from axillary bud of the single-node without leaf was inhibited at high temperature of $28^{\circ}C$ regardless of daylength. Whereas, tuberization at $20^{\circ}C$ and $28^{\circ}C$ was similar without the difference under SD condition but the tuber formation ratio were low. CCC 500 mg/L promoted tuberization and the effect was also showed even under LD condition at $28^{\circ}C$. The inhibiton of tuberization under LD and high temperature condition could be solved by treatment with CCC.

Effects of Soil Texture, Irrigation System, and Soil Ameliorators on the Cadmium Adsorption in Soil and Uptake in Rice Plant (토양의 카드뮴흡착과 수도체 카드뮴흡수에 미치는 토성, 물관리 및 개량제 효과)

  • This study was conducted to investigate the adsorptive characteristics with cadmium in different levels of organic matter and lime in soil. And in order to identify the effect of soil ameliorators on cadmium uptake in rice plants, compost and lime were treated. Plants were grown at two soil textures(sandy loam and clay loam) with irrigation water containing $0.01\;mg\;L^{-1}$ of cadmium and treatments of two irrigation systems(intermittent irrigation and continuous submersion). The adsorption capacity of cadmium by soil was increased in proportion to initial concentration of solution, and it was higher at clay soil compared to loamy soil. The adsorption rate of cadmium by soil was increased with increasing the concentration of organic matter and lime in soil, highly increased at the both organic matter and lime treatment. Soil pH was negatively correlated with the cadmium contents of the both shoot and brown rice while Eh was positively correlated with those. In the harvest season, cadmium contents in the both leaves and brown rice were lower in the clay sail plots than sandy soil plots, and the continuous submersion plots were lower than intermittent irrigation plots. Cadmium uptake was highly reduced at the compost and lime mixture plot compared to other treatments among the continuous submersion plots. The cadmium content of shoot was positively correlated with that of brown rice in the harvest season.

Effect of Band Spotty Fertilization for Reduction of Nitrogen Fertilizer on Chinese Cabbage(Brassia campestris L.) in Plastic Film Mulching Cultivation (비닐피복 배추 재배시 국소시비에 의한 질소질비료의 절감 효과)

  • This study was conducted to establish a low-input fertilization technique and increase of fertilization efficiency using the band spotty applicator(1999-2000) during the cultivation of mulching for chinese cabbage(Brassia campestris L.). The obtained results such as nitrogen efficiency, yield and soil improvement after cultivation of chinese cabbage were as follows. The content of $NO_3-N$ in soil increased in band spotty fertilization(BSF) by increasing application rate from the beginning stage to the middle stage. The content of total nitrogen increased but content of organic matter, available phosphate and exchangeable potassium decreased in comparison with the soil before experiment. Growth rate of Chinese cabbage increased in band spotty fertilization plot and uptake amount of nitrogen fertilized for chinese cabbage increased by increasing of the application rate. N use efficiency was higher by 5-21% in band spotty fertilization plot than in conventional fertilization(CF) plot. Yield of chinese cabbage increased by 16% in 70% band spotty fertilization plot and increased by 20% in 100% band spotty fertilization plot. It was found that 70% band spotty fertilization was more effective as fertilization method to reduce both environmental pollution and chemical nitrogen fertilizer in plastic film mulching cultivation.

Investigation of Bacterial Contamination of Liquid Soaps Used in Public Restroom (공공 화장실에서 사용하는 액체 손세정제의 세균 오염도 조사)

  • Handwashing with soap is an important practice to reduce the transmission of potentially pathogenic microorganisms, but liquid soaps with refillable dispensers are prone to extrinsic bacterial contamination. This study investigated the bacterial contamination of liquid soaps in 58 public restrooms in six buildings. The bacteria were identified by a biochemical test and MALDI-TOF mass spectrophotometry. The antimicrobial susceptibility was determined using the Vitek II system. Of the 58 restrooms examined, 27(46.55%) were using a refill dispenser, of which 25(92.59%) were contaminated with bacteria. The bacteria recovered from the soaps ranged from 1.6×103 to 2.7×105 CFU/mL. Serratia liquefaciens (12), Achromobacter xylosoxidans (9), S. marcescens (4), Staphylococcus pastueri (1), and Achromobacter spanius (1) were isolated. Except for one A. xylosoxidans, bacteria of the same species isolated in the same building showed a unique resistance pattern. In conclusion, handwashing with contaminated soap may play a role in the transmission of bacteria in public health settings. Therefore, it is necessary to limit the use of refillable liquid soaps in the restrooms of hospitals used by patients with reduced immunity.

Development of Value-added Ketchup Products with Korean Chile Peppers(Capsicum annuum L.) and Their Sensory Evaluation (국내산 고추를 이용한 고추 케찹 개발 및 관능적 특성 평가)

  • The purposes of this study were to develop value-added sauce(ketchup) products with Korean advanced chile peppers (Capsicum annuum L.), determine their physicochemical characteristics, and conduct a sensory evaluation. American chile ketchup products were collected from American local favorites and analyzed based on their compositions. The Korean chile ketchup contained tomato paste(37.5%), Korean chile pepper(14.1%), sugar(14.8%), vinegar(14.1%), garlic(8.5%), herbs, plum extract, and oligosaccharide. Its physiochemical analysis showed: moisture $59.61{\pm}0.28%$, crude protein $2.18{\pm}0.11%$, crude lipid $1.99{\pm}0.04%$, crude ash $9.26{\pm}0.13%$, crude carbohydrate $26.97{\pm}0.48%$, reducing sugar $35.19{\pm}0.97%$, salt $3.04{\pm}0.04%$, acidity $2.22{\pm}0.01%$, pH $3.7{\pm}0.01$, and $^{\circ}brix\;36.3{\pm}0.14$. Korean chile ketchup showed higher overall acceptability compared to American local favorite chile ketchup. This result suggests the possibility for replacing chile ketchup products imported from foreign countries(USA and Europe).
