Polymorphisms in the uncoupling protein 3 gene and their associations with feed efficiency in chickens

  • Objective: The uncoupling protein 3 (UCP3) is a member of the mitochondrial anion carrier superfamily and has crucial effects on growth and feed efficiency in many species. Therefore, the objective of the present study was to examine the association of polymorphisms in the UCP3 gene with feed efficiency in meat-type chickens. Methods: Six single nucleotide polymorphisms (SNPs) of the UCP3 gene were chosen to be genotyped using matrix-assisted laser desorption-ionization time-of-flight mass spectrometry in meat-type chicken populations with 724 birds in total. Body weight at 49 (BW49) and 70 days of age (BW70) and feed intake (FI) in the interval were collected, then body weight gain (BWG) and feed conversion ratio (FCR) were calculated individually. Results: One SNP with a low minor allele frequency (<1%) was removed by quality control and data filtering. The results showed that rs13997809 of UCP3 was significantly associated with BWG and FCR (p<0.05), and that rs13997811 had significant effects on BW70 and BWG (p<0.05). Rs13997812 of UCP3 was strongly associated with BW70, FI, and FCR (p<0.05). Furthermore, individuals with AA genotype of rs13997809 had significantly higher BWG and lower FCR (p<0.05) than those with AT genotype. The GG individuals showed strongly higher BW70 and BWG than AA birds in rs13997811 (p<0.05). Birds with the TT genotype of rs13997812 had significantly greater BW70 and lower FCR compared with the CT birds (p<0.05). In addition, the TAC haplotype based on rs13997809, rs13997811, and rs13997812 showed significant effects on BW70, FI, and FCR (p<0.05). Conclusion: Our results therefore demonstrate important roles for UCP3 polymorphisms in growth and feed efficiency that might be used in meat-type chicken breeding programs.

Suppression of Anaphylactic Reaction in Murine by Siegesbeckia pubescens

  • The aqueous extract of Siegesbeckia pubescens (SPAE) inhibited compound 48/80-induced systemic anaphylaxis 100% with the dose of 1.0, 0.5 mg/g body weight (BW) at 1 h before or 5 min, 10 min after intraperitoneal injection of compound 48/80. The passive cutaneous anaphylaxis (PCA) reaction also inhibited to 78.5% by oral administration of SPAE(1.0 mg/g BW). When SPAE pretreated on mice at concentrations ranging from 0.0001 to 1.0 mg/g BW, the serum histamine levels were reduced in a dose-dependent manner. Moreover, SPAE ($100-800{\mu}g/ml) dose-dependently inhibited the histamine release from peritoneal mast cells (RPMC) by compound 48/80 $(5{\mu}g/ml)$. Analysis by microscopic appearance observation revealed that SPAE $(500{\mu}g/ml) stabilized the RPMC membrane. Therefore, these findings indicate that SPAE inhibits anaphylactic reactions through stabilization of mast cell membrane.

Antidiabetic Effect of Auricularia auricula Mycelia in Streptozotocin-induced Diabetic Rats

  • Auricularia auricula (Heterobasidiae) called 'tree ear' or 'ear mushroom' was investigated to know its antidiabetic effect. Administration of dried powder of Auricularia auricula mycelia (AAM) (0.5 g/kg bw and 1.0 g/kg bw) caused a statistically significant reduction of plasma glucose (35% and 39%, respectively), total cholesterol (18% and 22%, respectively), triglyceride (12% and 13%, respectively), GOT (53% and 56%, respectively) GPT (26% and 37%, respectively), levels and the rate of liver weight against final total body weight (6% and 7%, respectively) compared with diabetic control group. These results support the antidiabetic effect of AAM.

The Body Weight-related Differences of Leptin and Neuropeptide Y (NPY) Gene Expression in Pigs

  • To determine if body weight change is directly related to altered leptin and neuropeptide Y (NPY) gene expression, we assessed adipose tissue weight, percent body fat, leptin and NPY mRNA levels and serum leptin concentration in pigs at weights of 1, 20, 40, 60, and 90 kg. The results indicated that the weight of adipose tissues and the percent body fat of pigs significantly increased and correlated with body weight (BW) from 1 to 90 kg (p<0.01). Serum leptin concentrations and leptin mRNA levels in omental adipose tissue (OAT) increased from 1 to 60 kg, and then decreased from 60 to 90 kg. At 60 kg, the serum leptin concentration and leptin mRNA level significantly increased by 33.5% (p<0.01) and 98.2% (p<0.01), respectively, as compared with the levels at 1 kg. At 60 kg, the amount of leptin mRNA in subcutaneous adipose tissue (SAT) was significantly higher than that of 1 and 40 kg animals (p<0.05). NPY gene expression in the hypothalamus also changed with BW and at 60 kg the NPY mRNA level significantly decreased by 54.0% (p< 0.05) as compared with that in 1 kg. Leptin mRNA in OAT was correlated with serum leptin concentrations (r = 0.98, p<0.01), body weight (r = 0.82, p<0.05) and percent body fat (r = 0.81, p<0.05). This is the first report of the developmental expression of leptin in porcine OAT, peritoneal adipose tissue (PAT) and SAT, and proves that the expression of leptin in OAT could reflect the levels of circulating leptin. These results provide some information for nutritional manipulation of leptin secretion which could lead to practical methods of controlling appetite and growth in farm animals, thereby regulating and improving efficiency of lean meat production and meat production quality.

Estimation of Daily Exposure to 3-Monochloropropane-1,2-diol from Commercial Soy Sauces in Korea

  • To assess the dietary exposure to 3-monochloropropane-1,2-diol (3-MCPD) from soy sauces, the levels of 3-MCPD in commercial soy sauces were analyzed with gas chromatography/mass spectrometry. Sixty nine out of the 72 soy sauces tested contained a level of 3-MCPD below $0.3\;{\mu}g/g$, the maximum limit in Korea. The average concentration of the 72 samples was $0.080\;{\mu}g/g$ and the highest concentration was $3.131\;{\mu}g/g$. On the basis of the consumption data, 3-MCPD concentration and body weight, the estimated daily exposures to 3-MCPD were ranged from 0.037 to $0.146\;{\mu}g/kg$ body weight (bw)/day for 95th percentile exposed population groups. Our estimated daily exposures are significantly lower than the provisional maximum tolerable daily intake (PMTDI) of $2\;{\mu}g/kg$ bw/day, which was established by the Joint Food and Agriculture Organization/World Health Organization Expert Committee on Food Additives (JECFA).

  • The purpose of this study was to estimate the standard of normal spleen size and its variations in relation to age, body weight(BW) and 1st lumbar vertebra($L_1$) length using radiographs. The Maximum transverse length (MTL) and maximum vertical length(MVL) of the spleen were obtained from ventrodorsal abdominal radiographs of 134 dogs which underwent radiography for indications unrelated to splenic disease. Splenic size did not significantly correlated with the age. MTL and MVL increased with BW according to the nonlinear correlation. Also, MTL and MVL increased with $L_1$ length according to the linear correlation.

Evaluation of seasonal effects on production performance of lactation sows reared in two different environments in South Korea

  • This study was conducted to evaluate seasonal effects on production performance of lactation sows reared in two different environments in South Korea. A total of 76 sows ($Landrace{\times}Yorkshire$) from the Dankook University experimental farm and 120 sows ($Landrace{\times}Yorkshire$) from a commercial farm were collected. In the current study, reduced (p < 0.01) feed intake and energy intake during lactation were observed in warm season compared with cool season in both farms. Sows in cool season had higher backfat thickness at weaning but lower back fat loss (p < 0.01) than those in warm season. The weaning to estrus interval was lower (p < 0.01) in cool season than in warm season. Piglets weaned in warm season had lower (p < 0.01) body weight (BW), body weight gain (BWG), and average daily gain (ADG) at weaning than did those weaned in cool season on the Dankook University farm, whereas the reduction effect was only observed during 0 - 21 d on the commercial farm (p < 0.01). Moreover, piglets weaned in cool season had higher BW, BWG, and ADG (p < 0.05) on both farms. In conclusion, our results indicate that warm season had very negative effects on feed intake and production performance of lactating sows and piglets.

Influence of enzyme mixture supplementation on growth performance, nutrient digestibility, and fecal score in growing pigs

  • This study evaluated the influence of enzyme mixture supplementation on the growth performance, nutrient digestibility, and fecal score of growing pigs. A total of 72 pigs with an initial body weight of 20.23 ± 1.46 kg were randomly assigned to two treatments consisting of a basal diet and the basal diet supplemented with 0.5% enzyme mixture. During a 19-day trial, no significant difference was observed in the body weight (BW) and average daily feed intake (ADFI) of the pigs. However, a gradual increase in the average daily gain (ADG) was observed during the period from day 14 to day 19 and the overall period in pigs fed a diet supplemented with the 0.5% enzyme mixture (p < 0.10) as compared to the pigs that were fed the control diet. From days 4 to 14 and in the overall experiment, a gradual increase in the feed conversion ratio (FCR) (p < 0.10) was observed with the inclusion of 0.5% enzyme mixture supplementation. The nutrient digestibility of dry matter (DM), nitrogen (N), and energy were not affected by enzyme mixture supplementation. In addition, dietary supplementation with the enzyme mixture had no significant effects on the fecal score of growing pigs. In summary, supplementation with the enzyme mixture had beneficial effects on the ADG performance but failed to have a significant effect on growth performance (BW), nutrient digestibility, and fecal score.

한우의 육성 및 비육기간중 배합사료 급여 수준에 따른 사료섭취량 조사 (Feed Intake Evaluation of Korean Cattle (Hanwoo) Fed Diets Containing Different Levels of Compound Fattening Periods)

  • 배합사료와 볏짚을 사료원으로 하여 한우 수소와 거세우를 비육 할 때의 사료섭취량을 조사하고자 5~7개월된 한우 수송아지 20두와 거세 송아지 60두를 공시하여 육성기(체중 300kg 이하), 비육 전기 (체중 300~450kg), 비육 후기 (체중 450kg 이후) 3단계로 나누어 육성우 배합사료 (CP 14.1%, TDN 70.0%), 비육 전기 배합사료(CP 12.1%, TDN 70.6%), 비육 후기 배합사료(CP 11.2%, TDN 71.9%)를 각각 성장단계별로 급여하면서 1) 수소와 거세우의 무제한 급여시의 사료 섭취량 (원물 기준)과 2) 거세우를 성장단계에 따라 배합사료를 ① 전기간 무제한 급여 ② 1.0%-1.5%-무제한급여, ③ 1.5%-2.0%-무제한급여의 3유형으로 사양관리 하면서 사료 섭취량 (원물 기준)의 변화를 조사하였다 1. 수소의 1일 사료섭취량은 직선적으로 증가된 반면에 거세우의 사료섭취량은 521 kg 까지는 증가되다가 감소되었다. 2. 체중대비 사료 섭취량은 시험 시작시인 150 kg 에서는 체중의 3.5% 정도를 섭취하였으나 600 kg에서 수소는 체중비 2.0% 내외로 거세우는 1.5% 정도로 감소되었다 3. 거세우의 1일 사료 섭취량은 전기간 무제한 급여시는 비육 전기까지는 완만하게 증가 하다가 비육 후기에는 다시 완만하게 감소된 반면 육성기와 비육 전기에 배합사료를 체중비 1.0%와 1.5%로 제한하여 급여 하였을 때는 사료 섭취량이 직선적으로 증가하였으며 1.5%와 2.0%로 제한하여 급여하였을 때는 비육 전기까지는 비교적 빠르게 증가되고 비육 후기(체중 455 kg)에는 비교적 빠른 속도로 감소되었다. 4. 배합사료 급여수준에 따른 거세우의 단위체중당 사료 섭취량은 무제한 급여시는 시험 시작시 (체중 150 kg 내외)에는 3.5% 내외에서 체중 600kg 내외에서는 1.5%까지 감소되었고 배합사료 제한 급여시는 육성기에는 2.0%에서 3.0%로 증가되다가 다시 1.5-2.0% 정도로 감소되었다 5. 거세우에 배합사료 급여수준에 따른 볏짚 섭취량은 배합사료 제한급여시 무제한 급여에 비하여 육성기에는 2-3배로 비육 전기에는 2배로 증가되었으나 비육 후기에는 차이가 없었다.

Performance and Carcass Composition of Broilers under Heat Stress : I. The Effects of Dietary Energy and Protein

  • An experiment was conducted to determine the effects of dietary energy and protein on performance and carcass composition of broilers under heat stress during the growing period (3-6 weeks). A factorial arrangement of three levels of energy (3.0, 3.2 and 3.4 kcal/g), three levels of protein (18, 20 and 22%), an two rearing temperatures were used in this study. Groups of birds were kept under moderate temperature ($24{\pm}1^{\circ}C/24h$) or hot cycling temperature ($26-34^{\circ}C/6h$, $34{\pm}1^{\circ}C/12h$, and $34-26^{\circ}C/6h$). Body weight (BW), weight gain (WG), feed intake (FI), feed conversion (feed : gain) (FC), carcass weight (CW), carcass yield (YP), breast meat (BM), abdominal fat (AF), drumsticks (DS), and thighs (TH) percentages were determined at the end of the experiment. Hot cycling temperature significantly (p<0.05) decreased BW, WG, FI, CW, and BM, increased FC, YP, DS, and TH but did not affect AF. High energy significantly (p<0.05) increased BW, WG, CW, YP, AF, and TH, decreased FI and FC but did not affect BM or DS. This improvement was observed only under moderate temperature resulting in significant (p<0.05) energy by temperature interaction. High protein significantly (p<0.05) increased BW, WG, CW and BM, decreased AF but did not affect FI, FC, TP, DS, or TH. There were no significant protein by temperature interactions for any of the parameters tested except CW. It is concluded, under the conditions imposed in this experiment, that increasing dietary energy did not alleviate the depressing effect of heat stress while increasing dietary protein up to 22% improved the performance of broilers irrespective of rearing temperature.
