Effects of yeast and dried kratom leaves (Mitragyna speciosa [Korth] Havil.) supplementation on digestibility, rumen fermentation, blood metabolites and nitrogen balance in goats

  • Objective: The objective of the experiment was to study yeast supplementation (yeast, Y) and dried kratom leaves (DKTL) on the digestibility, ruminal fermentation, blood metabolites and nitrogen balance in goats. Methods: Four of 7 to 8 months old male crossbred (50% Thai Native-Anglo Nubian) goats with average liveweight 20±0.13 kg were randomly assigned according to a 2×2 factorial arrangement in a 4×4 Latin square design to receive four diets ad libitum basis. The study investigated the effects of two levels of yeast (Y) supplementation (Y, 0 and 0.5g/kg dry matter [DM]) along with two levels of DKTL supplementation (DKTL, 0 and 4.44g/kg DM). The experimental groups were as follows: T1 = control group with 0Y+0DKTL, T2 = 0Y+4.44 DKTL, T3 = 0.5Y+0DKTL, and T4 = 0.5Y+4.44 DKTL. Results: The results showed that there were no interactions between Y levels and DKTL levels with respect to total DM intake, but there were significant effects (p<0.05) by levels of Y; goats receiving 0.05 g/kg DM Y had higher than goats fed 0.0 g/kg DM on average (kg/d). A percentage of body weight (% BW) and grams per kilogram of metallic weight (g/kg w0.75) had no influence on yeast levels and DKTL, but there was a difference (p<0.05) by yeast level Y at 0.5 g/kg DM, being higher compared to the non-supplemented group. Apparent digestibility coefficient of nutrition in the form of (DM, organic matter, crude protein, neutral detergent fiber, and acid detergent fiber) was an increased trend in the Y-level complementary group at 0.5 g/kg DM and DKTL at 4.44 g/kg DM, respectively. Protozoa populations decreased in the group receiving Y levels at 0.5 g/kg DM and DKTL levels at 4.44 g/kg DM when compared to group T1. The acetic acid concentration and methane gas generation decreased (p<0.05) in the group receiving Y levels of 0.5 g/kg DM and DKTL levels of 4.44 g/kg DM, while the amount of propionic acid increased (p<0.05). Conclusion: Effects of feeding combinations of Y and DKTL supplementation on feed showed no interaction effect (Y×DKTL) on feed intake, rumen fermentation, bacterial and fungi population. The effect on protozoal populations was lower in the group that was supplemented with DKTL at 4.44 g/kg DM related to synthetic CH4 was reduced.

인천 연안에 출현하는 쥐노래미(Hexagrammos otakii)의 식성 (Feeding Habits of Fat Greenling Hexagrammos otakii off the Incheon Coast of Korea)

  • 이번 연구에 사용된 쥐노래미는 2020년 3월부터 2021년 2월까지 매월 인천 연안에서 통발과 개량안강망에 의해 어획되어 위판된 376개체를 이용하였으며, 전장(TL: Total length, 0.1 cm)과 체중(BW: Body weight, 0.1 g)을 측정한 뒤, 위를 적출한 후 현미경을 이용하여 위내용물을 가능한 종 수준까지 분석하고, 분류된 먹이생물들은 계수하고 무게를 측정하였다. 위내용물 분석 결과는 상대중요도지수비를 이용하였으며, 시기별 및 크기군에 따른 먹이조성 변화를 알아보았다. 쥐노래미의 가장 중요한 먹이생물은 복족류(Gastropoda)였으며, 그 다음으로 중요한 먹이생물은 어류(Teleostei)였다. 그 밖에 단각류(Amphipoda), 집게류(Anomura), 이매패류(Bivalvia) 등이 출현하였으나, 0.9% 이하의 상대중요도지수비를 보여 그 값은 매우 낮았다. 시기별 위 내용물 조성을 분석한 결과, 춘계는 두족류, 하계는 갯지렁이류, 추계는 어류, 동계는 복족류가 중요한 먹이생물로 나타났다. 크기군별 위내용물 조성을 알아본 결과, <25.0 cm, 25.0~30.0 cm 크기군에서는 어류가 우점한 먹이생물이었고, 30.0~35.0 cm, ≥35.0 cm 크기군에서는 복족류가 중요한 먹이생물로 나타났다. 쥐노래미는 어류를 주로 섭식하며 큰 크기군으로 갈수록 어류의 비율은 감소하고 복족류의 비율이 점차 증가하는 양상을 보였지만, 크기군별 쥐노래미의 개체당 평균 먹이생물 개체수와 습중량은 통계적으로 유의한 차이를 보이지 않았다.

조피볼락, Sebastes schlegeli 자치어의 갑상선 분화 및 갑상선호르몬의 변화 (Differentiation of Thyroid Gland and Changes of Thyroid Hormone Concentration during Early Development of Black Rockfish, Sebastes schlegeli)

  • 조피볼락 갑상선 여포의 원시세포는 모체내 부화자어의 구강 아래쪽에 존재하는 때의 원형 여포 안에 존재하였다. 갑상선 여포는 구강 연골조직 아래쪽 제 1, 2,3 도입새동맥 부근에 흩어져 존재하고, 출산 직후에는 1${\sim}$3개 정도로 분화되어 있었다. 갑상선 여포의 수는 출산 직후(전장 6.3 mm)에 개체당 1.6${\pm}$0.8개였으며, 치어기로 전환하는 30일째에는 17.0${\pm}$2.1개, 50일째(전장 13.2 mm)에는 32.5${\pm}$6.9개로 그 수가 매우 많아졌다. 갑상선 여포의 직경은 출산 직후에 18.1${\pm}$0.6 ${\mu}$m였던 것이 출산후 30일째 30.5${\pm}$2.2 ${\mu}$m, 50일째 41.5${\pm}$1.7 ${\mu}$m로 그 크기가 증대되었다. 갑상선 여포세포의 높이는 출산직후 4.1${\pm}$0.2 ${\mu}$m, 20일째 5.7${\pm}$0.2 ${\mu}$m, 50일째 6.0${\pm}$0.2 ${\mu}$m로 성장하였다. 자치어의 전어체 내 갑상선호르몬을 분석한 결과, 난황을 가진 출산 직후에 호르몬을 함유하고 있었으며, 난황이 완전 흡수되는 5일째에는 그 농도가 급격히 감소하다가, 20일${\sim}$30일에 급격히 상승하여 50일째 최고치를 나타냈다. L-thyroxine (T$_4$)에 대한 3,5,3'-triiodo-L-thyronine (T$_3$) 농도는 출산 직후를 제외하고, 전 기간 동안 상대적으로 낮은 값을 나타냈다.

내독소로 유도된 급성폐손상에서 Moxifloxacin의 효과 (The Effects of Moxifloxacin in Endotoxin-induced Acute Lung Injury)

  • 배경: 패혈증에 동반된 급성호흡곤란증후군의 병태생리는 내독소에 의해 유발된 급성폐손상이다. 이 연구에서 우리는 내독소로 인해 유발된 급성 폐손상에서 moxifloxacin의 영향을 알아보고자 하였다. 대상 및 방법: 백서를 대조군(C), 내독소처리군(L), 내독소-moxifloxacin처리군(L-M)으로 나누었다. 대장균의 내독소를 기관 내 분무하여 급성폐손상을 유발하였고 L-M군에서는 내독소 분무 30분 후 moxifloxacin을 투여하였다. 내독소 투여 5시간 후 폐질량/체중비, 폐세척액 내의 단백함량, 폐세척액 내의 호중구 수, 폐장 내 myeloperoxidase (MPO) 활성도, 폐장 내 malondialdehyde (MDA) 함량을 측정하였고, 폐장 내 세포질형 $phospholipaseA_2$ ($cPLA_2$)와 분비형 $phospholipaseA_2$ ($sPLA_2$)의 발현을 관찰하고 광학현미경을 이용하여 형태학적인 분석을 시행하였다. 결과: 폐질량/체중비, 폐세척액 내의 단백함량, 폐세척액 내의 호중구 수, 폐장 내 myeloperoxidase (MPO) 활성도, 폐장 내 malondialdehyde (MDA) 함량은 L군에서 대조군 보다 유의하게 증가되어 있었고, L-M군은 L군보다 의미있게 낮은 것으로 나타났다. L군에서 $cPLA_2$$sPLA_2$의 발현은 증가되어 있었고 L-M군에서는 $cPLA_2$의 발현이 감소되어 있었다. 그러나 L-M군에서 $sPLA_2$의 발현은 L군과 차이가 없었다. 형태학적 분석으로 L군에서 많은 염증성 소견들을 관찰할 수 있었으나 L-M군에서는 관찰할 수 없었다. 결론: 내독소에 의해 유발된 급성폐손상의 많은 염증성 변화들이 moxifloxacin처리에 의해 소멸됨을 관찰할 수 있었다.

청보리 사일리지 및 목건초 급여가 엘크 사슴의 생산성 및 경제성에 미치는 영향 (The Effect of Feeding Whole-crop Barley Silage and Grass Hay on Productivity and Economic Efficiency in Elk (Cervus canadensis))

  • 본 연구는 사슴의 조사료 원으로 엘크사슴에 대한 청보리 사일리지 급여가 사슴의 사료섭취량, 체중변화 및 경제성에 미치는 영향을 구명하기 위하여 실시하였다. 엘크사슴 21두를 공시하여 건초구, 청보리 사일리지구 및 건초+청보리 사일리지구로 나누어 농후사료를 3.7 kg 정량급여 하고 조사료는 자유채식 시켰으며, 그 결과는 다음과 같다. 엘크사슴의 일당증체량은 건초구, 청보리 사일리지구 및 건초+청보리 사일리지구 혼합구가 각각 0.26, 0.36 및 0.44 kg으로 건초+청보리 혼합구가 가장 높았고, 엘크사슴의 청보리 사일리지 일일섭취량은 6.25~9.00 kg (풍건) 범위였으며, DM 섭취량은 건초구, 건초+청보리 사일리지 혼합구 및 청보리 사일리지 순으로 높은 경향을 보였다. 녹용 생산량은 건초구, 청보리 사일리지구 및 건초+청보리 사일리지구가 각각 8,037, 7,713 및 8,585 g으로 건초+청보리 사일리지 혼합구가 가장 많았다. 3개 처리구간 일당증체량, 녹용생산량 및 사료섭취량은 사슴의 개체차가 심하여 통계적으로 유의성은 인정되지 않았지만(p>0.05) 증체량 및 녹용산량은 증가하는 경향을 보였다. 경제성 분석에서는 건초+청보리 사일리지 혼합구가 건초 및 청보리 사일리지 단독급여구보다 소득지수가 29~43% 높게 나타났다. 이상의 결과를 종합적으로 고려할 때 사슴의 조사료 원으로 청보리 사일리지는 기호성에서 건초를 대체할 만큼의 섭취량을 가지므로 이용가능성이 있었으며, 조사료 원으로 연중 공급이 가능하고, 경제적인 측면에서도 건초보다 유리하여 농가에서 사슴의 조사료 원으로 사용이 가능할 것으로 사료된다. 또한 청보리 사일리지 급여 시에는 청보리 사일리지 단독 급여보다는 청보리와 건초를 각각 50%씩 혼합 급여하는 것이 일당증체량, 증체, 녹용생산량 및 경제성에서 가장 바람직한 것으로 사료된다.

Effect of Dietary β-1,3/1,6-glucan Supplementation on Growth Performance, Immune Response and Plasma Prostaglandin E2, Growth Hormone and Ghrelin in Weanling Piglets

  • The experiment was conducted to evaluate the effect of ${\beta}$-1,3/1,6-glucan on growth performance, immunity and endocrine responses of weanling piglets. One hundred and eighty weanling piglets (Landrace$\times$Large White, $7.20{\pm}0.25kg$ BW and $28{\pm}2$ d of age) were randomly fed 1 of 5 treatment diets containing dietary ${\beta}$-1,3/1,6-glucan supplemented at 0, 25, 50, 100 and 200 mg/kg for 4 wks. Each treatment was replicated in 6 pens containing 6 pigs per pen. On d 14 and 28, body weight gain, feed consumption and feed efficiency were recorded as measures of growth performance. Peripheral blood lymphocyte proliferation and serum immunoglobulin G (IgG) were measured to study the effect of dietary ${\beta}$-1,3/1,6-glucan supplementation on immune function. Plasma prostaglandin E2 (PGE2), growth hormone (GH) and ghrelin were measured to investigate endocrine response to ${\beta}$-1,3/1,6-glucan supplementation. Our results suggest that average daily gain (ADG) and feed efficiency had a quadratic increase trend with dietary ${\beta}$-1,3/1,6-glucan supplementation from d 14 to 28, whereas it had no significant effect on average daily feed intake (ADFI). The treatment group fed with 50 mg/kg dietary ${\beta}$-1,3/1,6-glucan supplementation showed a numerical increase in ghrelin, a similar change trend with ADG and no significant effect on GH. Lymphocyte proliferation indices, serum IgG and plasma PGE2 concentrations varied linearly with dietary supplementation levels of ${\beta}$-1,3/1,6-glucan on d 14. Higher levels of ${\beta}$-1,3/1,6-glucan may have a transient immuno-enhancing effect on the cellular and humoral immune function of weanling piglets via decreased PGE2. Taking into account both immune response and growth performance, the most suitable dietary supplementation level of ${\beta}$-1,3/1,6-glucan is 50 mg/kg for weanling piglets.

Effect of Roughage Sources on Cellulolytic Bacteria and Rumen Ecology of Beef Cattle

  • The effect of different tropical feed sources on rumen ecology, cellulolytic bacteria, feed intake and digestibility of beef cattle was investigated. Four fistulated, castrated male crossbred cattle were randomly allocated to a $4{\times}4$ Latin square design. The treatments were: T1) urea-treated (5%) rice straw (UTS); T2) cassava hay (CH); T3) fresh cassava foliage (FCF); T4) UTS:FCF (1:1 dry matter basis). Animals were fed concentrates at 0.3% of body weight on a DM basis and their respective diets on an ad libitum basis. The experimental period was 21 days. The results revealed that the use of UTS, CH, FCF and UTS:FCF as roughage sources could provide effective fiber and maintain an optimal range of ruminal pH and $NH_3-N$. Total viable and cellulolytic bacterial populations were enhanced (p<0.05) with UTS as the roughage source. Animals fed FCF had a higher rumen propionate production (p<0.05) with a lower cellulolytic bacteria count. Moreover, three predominant cellulolytic bacteria species, namely Fibrobacter succinogenes, Ruminococcus albus and Ruminococcus flavefaciens, were found in all treatment groups. Roughage intake and total DM intake were highest with UTS (2.2 and 2.5% BW, respectively) as the roughage source (p<0.05). Nutrient intake in terms of organic matter intake (OMI) was similar in UTS, CH and UTS:FCF treatments (8.0, 6.8 and 8.7 kg/d, respectively), while crude protein intake (CPI) was enhanced in CH, FCF and UTS:FCF as compared to the UTS treatment (p<0.05). Digestion coefficients of DM and organic matter (OM) were similar among treatments, while the CP digestion coefficients were similar in CH, FCF and UTS:FCF treatments, but were higher (p<0.05) in CH than in UTS. CP and ADF digestible intakes (kg/d) were highest (p<0.05) on the CH and UTS treatments, respectively. It was also observed that feeding FCF as a full-feed resulted in ataxia as well as frequent urination; therefore, FCF should only be fed fresh as part of the feed or be fed wilted. Hence, combined use of FCF and UTS as well as CH and FCF were recommended.

Evaluation of a Nutrition Model in Predicting Performance of Vietnamese Cattle

  • The objective of this study was to evaluate the predictions of dry matter intake (DMI) and average daily gain (ADG) of Vietnamese Yellow (Vang) purebred and crossbred (Vang with Red Sindhi or Brahman) bulls fed under Vietnamese conditions using two levels of solution (1 and 2) of the large ruminant nutrition system (LRNS) model. Animal information and feed chemical characterization were obtained from five studies. The initial mean body weight (BW) of the animals was 186, with standard deviation ${\pm}33.2$ kg. Animals were fed ad libitum commonly available feedstuffs, including cassava powder, corn grain, Napier grass, rice straw and bran, and minerals and vitamins, for 50 to 80 d. Adequacy of the predictions was assessed with the Model Evaluation System using the root of mean square error of prediction (RMSEP), accuracy (Cb), coefficient of determination ($r^2$), and mean bias (MB). When all treatment means were used, both levels of solution predicted DMI similarly with low precision ($r^2$ of 0.389 and 0.45 for level 1 and 2, respectively) and medium accuracy (Cb of 0.827 and 0.859, respectively). The LRNS clearly over-predicted the intake of one study. When this study was removed from the comparison, the precision and accuracy considerably increased for the level 1 solution. Metabolisable protein was limiting ADG for more than 68% of the treatment averages. Both levels differed regarding precision and accuracy. While level 1 solution had the least MB compared with level 2 (0.058 and 0.159 kg/d, respectively), the precision was greater for level 2 than level 1 (0.89 and 0.70, respectively). The accuracy (Cb) was similar between level 1 and level 2 (p = 0.8997; 0.977 and 0.871, respectively). The RMSEP indicated that both levels were on average under-or over-predicted by about 190 g/d, suggesting that even though the accuracy (Cb) was greater for level 1 compared to level 2, both levels are likely to wrongly predict ADG by the same amount. Our analyses indicated that the level 1 solution can predict DMI reasonably well for this type of animal, but it was not entirely clear if animals consumed at their voluntary intake and/or if the roughness of the diet decreased DMI. A deficit of ruminally-undegradable protein and/or a lack of microbial protein may have limited the performance of these animals. Based on these evaluations, the LRNS level 1 solution may be an alternative to predict animal performance when, under specific circumstances, the fractional degradation rates of the carbohydrate and protein fractions are not known.

Effects of phosalone consumption via feeding with or without sodium bentonite on performance, blood metabolites and its transition to milk of Iranian Baluchi sheep

  • Background: Transfer of pesticides from environment to animal products is inevitable, so the purpose of the present work was to evaluate phosalone consumption via feeding with or without sodium bentonite (SB) on performance, blood metabolites and its transition to milk of Iranian Baluchi sheep. Methods: Twenty Baluchi ewes were divided into four treatments (P1 as control, P2, P3, and P4) of five animals in which phosalone, an organophosphate pesticide, was given via diet (only for P2 and P3) at a dose of 280 mg/sheep/day for 63 consecutive days. The SB (32 g/sheep/day; for P3 and P4) was also evaluated for its ability to reduce deleterious effects of phosalone in the sheep diets. The control group (P1) did not receive any phosalone and SB during the experiment. Sampling was conducted in two periods of time including weeks 5 and 9. Results: Phosalone residues were observed in the milk samples of P2 and P3 groups during two sampling periods. During period 1, the transfer rate of phosalone from feed to milk was 0.23 and 0.02%, respectively for the contaminated diets (P2 and P3), which is relatively similar to period 2 (0.22 and 0.02%). Only 0.34 (period 1) and 0. 36% (period 2) of phosalone residue are excreted in the feces of P2 group following its daily consumption. Transfer of phosalone from feed to milk was affected (P < 0.05) by the dietary inclusion of a commercial SB, as it (SB) decreased excretion of phosalone via milk (P3). The phosalone and SB alone or together had no significant effect (P > 0.05) on the dry matter intake (DMI) and body weight (BW) gain, but feed efficiency, milk production, milk fat, dry matter (DM) and organic matter (OM) digestibility, acetylcholinesterase (AChE) inhibitory activity, hemoglobin (Hb), red blood cell (RBC), serum glutamic pyruvic transaminase (SGPT), serum glutamic oxaloacetic transaminase (SGOT), albumin and mean corpuscular hemoglobin concentration (MCHC) affected by the treatments in period 1 or 2 (P < 0.05). The Hb, RBC, and MCHC were significantly decreased (P < 0.05) by about 9.72, 20.77, and 9.71%, respectively in the group P2 as compared to those of the control group during period 1. The AChE inhibitory activity (period 1 and 2) significantly increased when phosalone administered via the diet (P < 0.05). Conclusions: Although there were no adverse effects on the performance of sheep following the intake of phosalone alone (P2 vs. P1), but other research on the long and short times to the phosalone in high and low doses with more animals is suggested. Overall, compared to the control group, addition of SB in the diet of sheep improved nutrient digestibility, animal performance, and milk health.

Influence of Lighting Schedule and Nutrient Density in Broiler Chickens: Effect on Growth Performance, Carcass Traits and Meat Quality

  • The study was conducted to evaluate the effect of lighting schedule and nutrient density on growth performance, carcass traits and meat quality of broiler chickens. A total of 576 day old Arbor Acre male chickens was used with a $4{\times}2$ factorial arrangement. The four lighting schedules were continuous (23 L:1 D, CL), 20 L:4 D (12 L:2 D:8 L:2 D), 16 L:8 D (12 L:3 D:2 L:3 D: 2 L:2 D) and 12 L:12 D (9 L:3 D:1 L:3 D:1 L:3 D:1 L:3 D) and provided by incandescent bulbs. The two nutrient densities were high (H, starter diet: 13.39 MJ/kg apparent metabolisable energy (AME), 23.00% crude protein (CP); finisher diet: 13.39 MJ AME/kg, 19.70% CP) and low energy and protein level (L, starter diet: 12.03 MJ AME/kg, 20.80% CP; finisher diet: 12.14 MJ AME/kg, 18.30% CP). Houses with dark curtains and solid sidewalls were used. Chickens were randomly allocated to the 8 treatments with each treatment comprising 6 replicates of 12 chickens. Feed and water were available ad libitum. Lighting schedules showed no difference (p>0.05) in growth performance at the end of the experiment. 12 L:12 D significantly reduced (p<0.05) the concentration of malondialdehyde (MDA) compared to 23 L:1 D treatment. Intermittent lighting (IL) schedules produced higher protein content (p<0.001) in breast meat. Birds on high density diets had higher body weight (BW), feed intake (FI) (p<0.001), and feed conversion ratio (FCR) (p<0.001) throughout the experiment with the exception of 36 to 42 d. High nutrient density increased (p<0.05) abdominal fat, decreased (p<0.05) the moisture loss of meat, and reduced percentage of wings and legs. There was a significant lighting schedule${\times}$diet interaction (p<0.001) on FCR for days 8 to 14 and 15 to 21. Results indicated that IL can give similar growth performance in comparison with CL, meanwhile with positive effects on meat quality by increasing protein content and decreasing the concentration of MDA. High nutrient density resulted in greater growth performance.
