Early Gonadogenesis and Sex Differentiation in the Korean Rose Bitterling, Rhodeus uyekii

  • This report describes the sex differentiation of the Korean rose bitterling, Rhodeus uyekii, from hatching to 170 days post-hatch (DPH) in relation to total length (TL), body weight (BW), and integral water temperature (IWT). The growth curve of TL from just hatching to 83 DPH was $5.144e^{0.045t}$ ($R^2=0.961$; t, time), and that of BW was $2.398e^{0.086t}$ ($R^2=0.725$). Primordial germ cells (PGCs) were observed at 17 DPH (7.9 mm TL, 3.74 mg BW, $374^{\circ}C$ IWT), and thereafter began to protrude into the peritoneal cavity. At 21 DPH ($9.2{\pm}0.14mm$ TL, $4.8{\pm}0.07mg$ BW, $462^{\circ}C$ IWT), some PGCs contained condensed chromatin and oocyte were observed in meiotic prophase. In contrast to the ovaries, which grew gradually after sexual differentiation, testes began multiplying at 25 DPH (10.1 mm TL, 5.42 mg BW, $550^{\circ}C$ IWT), when testicular differentiation was first identified, and multiplied continuously thereafter. At 33 DPH (11.2 mm TL, 10.5 mg BW, $726^{\circ}C$ IWT), the developing testes contained spermatogonia that exhibited mitotic activity. No spermatocyte or sperm cell was observed until 83 DPH (18.9 TL, 48.2 mg BW, $1,826^{\circ}C$ IWT). At 170 DPH (32.5 mm TL, 270.1 mg BW, $3,740^{\circ}C$ IWT), which was the end point of this study, the mature ovaries showed germinal vesicle breakdown, while the mature testes contained observable spermatocytes and sperm cells. These results allow us to identify the sex differentiation type of the Korean rose bitterling as differentiated gonochoristic.

Net Phosphorus Requirements of Dorper×Thin-tailed Han Crossbred Ram Lambs

  • A comparative slaughter trial was conducted to estimate the phosphorus (P) requirement for maintenance and growth of crossbred lambs of Dorper with a Chinese indigenous sheep breed, thin-tailed Han sheep. Thirty-five Dorper${\times}$thin-tailed Han crossbred, noncastrated ram lambs ($20.3{\pm}0.22kg$ of shrunk body weight (SBW)) were used. Seven lambs were randomly chosen and slaughtered at 20 kg SBW as the baseline group for measuring initial body composition. Another seven lambs were also randomly chosen and offered a pelleted mixed diet for ad libitum intake and slaughtered at 28 kg SBW. The remaining 21 sheep were randomly divided into 3 groups with 7 sheep each and subject to the same diet of either 70 or 40% of ad libitum intake. The 3 groups were slaughtered when the sheep fed ad libitum reached 35 kg of SBW. Body P contents were determined after slaughter. The results showed that the net P requirement for maintenance was 30.0 mg/kg of empty body weight (EBW) or 23.4 mg/kg body weight (BW), and the P requirement for growth decreased from 5.3 to 5.0 g/kg of EBW gain as the lamb grew from 20 to 35 kg. The net P requirement for growth of Dorper${\times}$thin-tailed Han crossbred ram lambs was lower than that of sheep adopted by the American nutritional system.

Expressional Changes of Connexin Isoform Genes in the Rat Caput Epididymis Exposed to Flutamide or Estradiol Benzoate at the Early Postnatal Age

  • Direct communication between neighboring cells through connexin (Cx)-based gap junction is a crucial biological manner to regulate functions of a tissue consisting of multi-cell types. The present research evaluated expressional changes of Cx isoforms in the caput epididymis of adult rat exposed to estradiol benzoate (EB) or flutamide (Flu) at the early postnatal age. A single subcutaneous administration of EB at a low-dose [$0.015{\mu}g/kg$ body weight (BW)] or a high-dose ($1.5{\mu}g/kg\;BW$) or Flu at a low-dose ($500{\mu}g/kg\;BW$) or a high-dose (5 mg/kg BW) was performed to an animal at 1 week of age. Quantitative real-time PCR analysis was employed to determine expressional changes of Cx isoforms. The transcript levels of Cxs30.3 and 37 were decreased by a low-dose EB treatment, while decreases of Cxs31, 31.1, 32, 40, and 45 transcript levels were observed with a low-dose EB treatment. The treatment of a high-dose EB resulted in expressional reduction of Cxs30.3, 31, 31.1, 37, 40, 43, and 45. The Flu treatment at a low dose caused increases of Cxs26, 37, and 40 transcript levels but decreases of Cxs31.1, 43, and 45 transcript levels. Increases of Cxs30.3, 31, 37, and 40 mRNA amounts were induced by a high-dose Flu treatment. However, exposure to a high-dose Flu produced expressional decreases of Cxs31.1, 32, and 43 in the adult caput epididymis. These observations suggest that exposure to EB or Flu at the neonatal period could lead to aberrant expression of Cx isoforms in the adult caput epididymis.

Changes in Sex Steroid Hormones and Ovarian Development during Artificial Maturation of Female Eel, Anguilla japonica

  • The present study demonstrates the changes in body weight (BW) and plasma sex steroid hormone profiles during artificial maturation induced by human chorionic gonadotropin (HCG) or salmon pituitary extract (SPE) injections in cultured eel, Anguilla japonica, kept in seawater for 3 months. In the weekly SPE-injected female group, BW was relatively stable during vitellogenesis. Following induction of vitellogenesis, females exhibited a rapid increase of BW, and the oocytes were observed to be in the migratory nucleus stage at the end of the experiment. Plasma testosterone (T) and $estradiol-17{\beta}$ ($E_2$) levels increased slightly during vitellogenesis and peaked at an average of 5.82 ng/mL and 4.76 ng/mL, respectively, at the end of the experiment. In the weekly control and HCG-injected female groups, BW slowly decreased during the experimental period, and the oocytes of the two groups were observed to be at the primary yolk globule stage. In the weekly HCG-injected female group, plasma T and $E_2$ levels increased slightly during vitellogenesis and decreased afterward. In the control female group, however, plasma T and $E_2$ levels were not altered during the experimental period. Furthermore, plasma $17{\alpha},20{\beta}-dihydroxy-4-pregnen-3-one$ (DHP) was not detected in all experimental groups. Fertility and hatching rates of SPE-injected females were significantly higher in those that ovulated 15 h after DHP injection than 18 h. These results indicate that long rearing in seawater increases responsiveness to SPE in ovarian maturation of the Japanese eel, resulting in shortened period from completion of vitellogenesis by sex steroid hormone production.

COMPARATIVE PERFORMANCE OF DAMASCUS GOATS AND CHIOS EWES IN OMAN

  • Twenty growing female Damascus (Shami) goats and twenty growing Chios ewes of two mating ages (10 and 12 months) were used to study the effect of mating age (MA) on reproductive performance (RP). In addition to (RP), The growth, milk production and nutrient requirements were also evaluated. A complete random design in a $2{\times}2$ factorial was used, with (MA) and species as the main factors. (MA) had no effects on (RP), milk production and mortality. No significant difference (p > 0.05) were observed between (MA) or between species, for age at kidding/lambing (K/L). Sheep reached puberty earlier than goats. Average age (months) at conception and at (K/L) was 15.8; 20.8 and l7.0; 22.0 for sheep and goats respectively. Body weight (BW), rather than age was the most important factor determining puberty. Both goats and ewes conceived when they reached about 67% of their mature (BW) for goats and 85% for ewes. Sheep grew faster than goats, but the latter had higher twinning (67.8% versus 25%). Chios sheep were suitable for meat and Shami goats for dairying or dual purpose systems. Nutrient requirements for growth of goats and ewes were similar with an average DM-intake of about 4% (BW); 12-13% CP and 65-69% TDN (DM-basis) of the total ration. It is recommended to breed Chios ewes at a (BW) of at least 50 kg and Shami does at 38 kg, irrespective of their age.

Changes in Serum Metabolites and Growth Characteristics of Korean Native Steers Fed Alcohol-fermented Feeds

  • This study was carried out to assess whether feeding of alcohol-fermented feeds (AFF) affects the nutritional metabolism and growth characteristics of Korean native steers. Ten steers were randomly assigned to one of two treatment groups. The dietary treatments were AFF (50% commercial beef cattle feed+30% alcohol-fermented soybean curd dregs+20% rice straw) and control (80% commercial beef cattle feed+20% rice straw). The change of serum metabolites and growth characteristics were measured every two months during the whole twelve months experimental period and the relationships between serum metabolites and growth characteristics were simultaneously analyzed. Four hours after feeding AFF, serum alcohol concentration reached its peak with a significantly higher value than that after control feeding (11.9 and 4.9 mg/dl, respectively). Serum glucose and inorganic phosphorus (IP) concentrations (63.1 and 8.4 mg/dl, respectively) of steers fed AFF were higher than those (56.6 and 7.0 mg/dl) fed the control diet. In both treatments, the serum glucose concentration rapidly increased when body weight (BW) of the steer reached about 600kg, while IP concentrations were rapidly diminished at that BW. Lower concentrations of both blood urea nitrogen (BUN) and cholesterol were observed in steers fed AFF up to 450 kg of BW. The IP concentration was correlated with concentrations of BUN, cholesterol and glucose in AFF fed cattle but not in the cattle fed control diets. Average daily gain was higher in steers fed AFF than steers fed control, particularly during the growing stage of cattle. These findings indicated a capability of AFF to improve BW gain of Korean native steers by decreased protein degradation as well as increased fat synthesis.

  • To investigate whether the rearing environmental factors (size heterogeneity, rearing density, and feeding frequency) affect the growth and cannibalism of fry in cultured puffer Takifugu obscurus, we conducted three experiments. To examine size effects, we used small (total length [TL] $26.0{\pm}0.5\;mm$, body weight [BW] $0.18{\pm}0.01\;g$), medium (TL $23.1{\pm}0.1\;mm$, BW $0.28{\pm}0.01\;g$), large (TL $26.0{\pm}0.2\;mm$, BW $0.48{\pm}0.01\;g$) and small-large size combination groups. For density, we used size-matched puffers (TL $25.0{\pm}0.5\;mm$, BW $0.53{\pm}0.03\;g$) and five density were examined ranging from 1.43 to 7.14 individuals/L. For the feeding frequency, we also used size-matched fry and feeding frequencies of three times/2 days, one time/day, two times/day, three times/day. We ran each experiment in triplicate and investigated the survival rate, daily food intake (DFI), feed efficiency (FE), and daily growth rate (DGR). The growth of the puffers increased with increasing size, density, and feeding frequency, while cannibalism increased with a greater size gap, density and lower feeding frequency. Therefore, we concluded that size, rearing density, and feeding frequency are major factors influencing growth and cannibalism of the puffer, T. obscurus.

A Review of the Optimum Feeding Rates in Olive Flounder (5 g through 525 g) Paralichthys olivaceus Fed the Commercial Feed

  • Ten feeding trials were conducted to determine and to make the guideline for the optimum feeding rates in olive flounder Paralichthys olivaceus fed extruded pellets at various growth stages at the optimum rearing water temperature. The entire trials were grouped into four growth stages, namely the first stage of juvenile (5 and 9 g), the second stage of juvenile (20, 30, 40 and 50 g), growing (240 and 317 g) and sub-adult (384 and 525 g) stages with 2, 4, 2 and 2 feeding trials, respectively. In the first and second experiments, results indicated that the optimum feeding rates for the first stage of juvenile olive flounder weighing 5 and 9 g could be >5.17 but <5.52% and >4.44 but <4.64% body weight (BW)/day, respectively. In the third, fourth, fifth and sixth experiments, results suggested that the optimum feeding rates for the second stage of juvenile olive flounder weighing 20, 30, 40 and 50 g could be >3.47 but <3.50, >2.85 but <3.53, >2.58 but <3.04 and >2.36 but <2.50% BW/day, respectively. In the seventh and eighth experiments, results showed the optimum feeding rates for growing olive flounder weighing 240 and 317 g to be >1.03 but <1.25 and >0.85 but <1.0 % BW/day, respectively. In the ninth and tenth experiments, the optimum feeding rates in sub-adult olive flounder weighing 384 and 525 g were shown to be >0.69 but <0.7 and >0.55 but <0.8% BW/day, respectively. We will compare these results with the previous studies done by other research groups.

Effects of Exposure to Estradiol Benzoate or Flutamide at the Weaning Age on Expression of Connexins in the Caudal Epididymis of Adult Rat

  • The present research was chiefly designed to determine the effect of the treatment of estrogenic agonist, estradiol benzoate (EB), or antiandrogenic compound, flutamide (Flu), at the weaning age on the expression of connexin (Cx) isoforms in the caudal epididymis of adult male rat. Animals were subcutaneously administrated with a single shot of either EB at a low-dose ($0.015{\mu}g$ of EB/kg body weight (BW)) or a high-dose ($1.5{\mu}g$ of EB/kg BW) or Flu at a low-dose ($500{\mu}g$ of EB/kg BW) or a high-dose (5 mg of EB/kg BW). Expressional changes of Cx isoforms in the adult caudal epididymis were examined by quantitative real-time PCR analysis. The treatment of a low-dose EB caused significant increases of Cx30.3, Cx31, Cx32, and Cx43 transcript levels but reduction of Cx31.1, Cx37, and Cx45 expression. Exposure to a high-dose EB resulted in very close responses observed in a low-dose EB treatment, except no significant expressional change of Cx37 and a significant induction of Cx40. Expression of all Cx isoforms, except Cx45, was significantly increased by a low-dose Flu treatment. Expressional increases of all Cx isoforms were detected by a high-dose Flu treatment. The current study demonstrates that a single exposure to estrogenic or antiandrogenic compound during the early postnatal developmental period is sufficient to disrupt normal expression of Cx isoforms in the adult caudal epididymis.

Aberrant Expression of Connexin Isoforms in the Corpus Epididymis of the Adult Rat by Exposure to Estradiol Benzoate or Flutamide at the Weaning Age

  • A proper development of the epididymis during the early postnatal development is required for successful fertility in the adult male. Direct cell-cell communication via connexin (Cx) molecules is a common way of cellular interactions to achieve normal development of a given tissue consisting of different cell types. The present research was attempted to determine the effect of exogenous exposure to estrogenic agonist or antiandrogen at the weaning age on expression of Cx isoforms in the adult corpus epididymis. Male rats were subcutaneously administrated with estradiol benzoate (EB) or flutamide (Flu) at the weaning age. The tissue was collected at 4 months of age. Expressional levels of Cx isoforms were determined by a quantitative real-time PCR. Statistical comparison showed significant increases of Cxs31, 32, 37, 40, and 43 transcript amounts by a treatment of $0.015{\mu}g$ of EB /kg body weight (BW). A treatment of $1.5{\mu}g$ of EB /kg BW caused a significant decrease of Cx43 gene expression but increases of Cxs26, 31, 32, 37, and 40 transcript levels. Exposure to $500{\mu}g$ of Flu/kg BW induced an increase of Cx37 expression but significant decreases of Cxs43 and 45 mRNA levels. Expression of Cx37 was increased by a treatment of 5 mg of Flu/kg BW, while transcript levels of Cxs26, 30.3, 31, 31.1, 32, and 43 were significantly decreased by same treatment. These results demonstrate that exposure to steroidal compounds at the early developmental age alters expression of Cx isoforms in the adult corpus epididymis.
