Clinical Application of Oclacitinib in Dogs with Atopic Dermatitis

  • This study evaluated the efficacy and safety to determine the therapeutic responses of oclacitinib in canine atopic dermatitis (CAD) and identified factors related to the therapeutic response. Overall, 13 dogs with CAD were treated with oclacitinib for 56 days. Owners and veterinarians assessed visual analog scale (VAS) scores of pruritus and dermatitis. The examined dogs were grouped according to treatment success and failure based on changes in the VAS scores. To identify factors related to the therapeutic responses of oclacitinib, signalments (breed, sex, age, and body weight), mean progression time of CAD, mean Owner and Veterinarian VAS scores at day 0, and preexisting infection rate were compared between the two groups. Among the 13 dogs, 7 (53.8%) met the criteria of treatment success. In the success group, the Owner VAS scores were significantly lower from day 6 to 56 compared with the score at day 0 (P < 0.05). Additionally, the Veterinarian VAS scores were significantly decreased at days 14 and 42 compared with the score at day 0 (P < 0.05). There were no changes in hematological indices after the administration of oclacitinib. The most common abnormality reported was otitis externa (30.8%), followed by pyoderma (23.1%), and vomiting (7.7%). Factors related to responses of oclacitinib were not identified. This study demonstrated that oclacitinib was safe and moderately effective in dogs with CAD. This is the first report of the clinical application of oclacitinib in South Korea.

General Pharmacology of LB71350, a New HIV-1 Pretense Inhibitor

  • Safety evaluation of LB71350, a new HIV-1 protease inhibitor, was performed in mice, rats and dogs. For the general behavior of mice, LB71350 at an oral dose of 200 mg/kg did not show any significant effects on muscle tone and locomotor activity. In terms of central nervous system, at oral doses of 200 mg/kg and 1000 mg/kg, LB71350 inhibited acetic acid-induced pain response approximately 41% and 83% of control. respectively. At oral doses of 200 mg/kg and 500 mg/kg, it reduced the rectal body temperature in rats. Pentylenetetrazole-induced seizure in mice was slightly potentiated by oral administration of LB71350 at doses ranging from 200 mg/kg to 1000 mg/Ag. Single or five day treatment of LB71350 doubled the hexobarbital- induced sleeping time in mice at oral doses ranging from 50 mg/kg to 500 mg/kg. It did not cause any effects on gastric secretion and acidity in rat at oral doses of 200 mg/kg and 1000 mg/kg and also it did not change intestinal motility in mice up to 1000 mg/kg. Blood coagulation indices such as prothrombin time (PT), activated partial thromboplastin time (aPTT), and thrombin time (TT) in rats were not affected by the treatment of LB71350 up to 500 mg/kg. LB71350 caused no significant effects on the cardiac output, stroke volume, heart rate, and mean blood pressure when infused intravenously to the anesthetized rats and dogs. Taken together, LB71350 at high oral doses caused significant pharmacological effects on the central nervous system and the hexobarbital-induced sleeping time.

산소중독에 대한 Vitamin E의 보호효과에 관한 실험적 연구 (An Experimental Study on the Efficacy of Vitamin E aganinst Oxygen Toxicity)

  • Since the wide spread application of hyperbaric oxygenation in clinical setting, the problems of oxygen toxicity have been attracting a deep interest from the researchers on hyperbaric medicine as a practical issue. Among extensive research trials, the study on the protective agents against oxygen toxicity occupied one of the most challenging field. As the mechanisms of oxygen toxicity, the role of the oxygen free radicals produced by peroxidation process are strongly accepted by the leading researchers on oxygen toxicity, the probable protective effects of antioxidant against oxygen toxicity are sustaining a sufficient rationale. In this study, the author attempted to evaluate the effect of vitamin E as a protective agent against oxygen toxicity through the observation of death rate, convulsion rate, time to convulsion, and macroscopic and microscopic pathological changes of experimental rats exposed to 100% oxygen at 5 ATA for 120 minutes. The findings observed are as follows: 1) The death rate, convulsion rate, time to convulsion, organ/body weight ratio and microscopic pathological findings were identified as reliable objective and quantitative indices for oxygen toxicity. 2) Vitamin E showed excellent protective effects against CNS and pulmonary oxygen toxicity as a strong antioxidant. The most effective dose seemed to be around 400 mg/kg 3) The results of this study are supporting the oxygen free radical hypothesis on oxygen toxicity.

서울지역 중년에 있어서 정상체중군과 과체중군의 영양상태 비교 (Comparison of Nutritional Status among Obese, Overweight and Normal Subjects in Seoul)

  • The nutritional status of middle-aged overweight and control normal subjects were evaluated for the purpose of providing the background information of the degenerative disease control. A survey was conducted with 293 healthy subjects (121 males and 172 females) between the ages 40-64. The average BMI (body mass index) of male subjects was 24.5 $\pm2.75$, and that of females was 23.5 $\pm2.87$. The average values of WHR (waist-hip ratio) were 0.88 $\pm0.04$, for males and 0.82 $\pm0.07$, for females. The normal BMI group (BMI 18.5-22.9) comprised 28.9% of males and 47.1% of females. The percentage of overweight subjects (BMI 23-24.9) was 26.4% of males and 30.8% of females. The obese group (BMI > 25) was 44.6% of males ana 22.1% of females, showing the greater rate of obese state among male subjects. The average energy intakes were 76.6 $\pm14.9$% for males and 77.8 $\pm12.6$, % for females, protein intakes were 108.0 $\pm24.6$% for males and 111.2 $\pm22.7$, % for females of the RDA levels. The average intakes of other nutrients were above the 75% of RDA levels except calcium. The average nutrient intakes of the three subgroups according to their BMI values were not different for both males and females. There were weak correlations between obesity and blood biochemical indices. There were positive correlations between BMI or WHR and hemoglobin, hematocrit, fasting glucose, total cholesterol, triglyceride, AST or ALT. There were negative correlations between BMI or WHR and HDL-cholesterol. These results suggest that the obesity rate of middle-aged is an influential factor of chronic disease. The middle-aged subjects of this study with higher educational and socioeconomic background tend to have desirable nutrition knowledge and attitude, but the application of their knowledge or attitude was relatively poor.

흡연과 Carotenoid 함유 식품 섭취빈도에 따른 지질과산화 및 항산화능 비교 (Effect of Smoking and Carotenoid-rich Food Consumption on Lipid Peroxidation and Antioxidant Status)

  • This study was performed to determine the effect of carotenoid-rich food consumption and smoking on the lipid peroxidation and antioxidant status in human. The subjects consisted of the health 210 middle-aged adults who visited health care center in the Inha University hospital. The blood and urine samples of the subjects were taken to analyze serum lipid profiles, plasma TBARS, total antioxidant status (TAS) and urinary 8-isoprostanes concentration. The anthropometric indices of the subjects were measured. The nutritional intake and the frequency of carotenoid-rich food consumption was determined by semi-quantitative food frequency questionnaire survey. HDL-cholesterol level of the smoking subjects was significantly lower than that of the non-smoking subjects in men. In the analysis of the carotenoid-rich food consumption, the frequency of pepper and tomato consumption of the non-smoking subjects was significantly higher than that of the smoking subjects in men. In women, the Sequency of pear and peach consumption of the non-smoking subjects was significantly higher than that of the smoking subjects. HDL-cholesterol level of the high carotenoid rich food consumption group was significantly higher than that of medium and low group in women. TAS of the high carotenoid rich food consumption group was significantly higher than that of medium and low group. In conclusion, carotenoid intakes seemed to be effective to reduce lipid peroxidation and increase antioxidant status in the body. The frequency of the carotenoid-rich food consumption seemed to be lower in the smokers than in the non-smokers. However, further researches consisted of large-scaled and randomized clinical trials are required to determine whether carotenoids have any other beneficial effect in human. The Prevalence of Obsersity and Nutritional Status in Adult Women Who Exercise Regularly

  • This study was conducted to investigate the relationship among prevalence of obesity, nutritional status and factors related to obesity of women who exercise regularly. The subjects of this study were 100 women who have been exercising regularly for more than 6 months. They were mostly housewives ages 24 to 63 years and had graduated middle or high school. Their average income was form 700 to 1,500 thousand won per month and most of them had 2 r 3 children. They sleep usually 6~8 hours a day. Most of physical indices of obesity were increased progressively with age. Among them , weight (61.5$\pm$1.4kg), subscapular (16.3$\pm$1.4mm) and abdomen(31.4$\pm$1.2mm) skinfold thickness were higher in 50 years than in others. BMI after exericse was decreased compared with BMI before exericise. WHR was 0.82 in 40 years and 0.85$\pm$0.01 in 50- to 60- year-old group. The percentage of body fat increased with advancing years and the highest values were shown in 50 - to 60-year old group (skyndex value : 38.6$\pm$0.6, BIA value : 28.7$\pm$0.8<0.05). The prevalence of obesity was measured by bioelectrical impedance fatness analyzer (BIA), obdy mass index (BMI) and skinfold thickness were 17.0%, 24.0% and 78.0% respectively. Most subjects were in good nutritional status, but in the subjects aged 24~49, energy (89.3%), iron (93.3%) and vit. A (97.4%) intake were slighltyl lower than the RDA. Energy intake was slightly insufficient tot he 50-63year old subjects (88.6%) compared with RDA. It seemed that they restricted calorie intake for the weight control. The energy percentage of carbohydrate, fat and protein was 65 : 20 : 15, the result of which came closed to the recommended calorei composition. There were positive correlations between obesity and other variable such as age. number of childeren and physical indics.

Changes in bioimpedance analysis components before and after hemodialysis

  • Background: This study compared nutritional parameters in hemodialysis (HD) subjects and controls using bioimpedance analysis (BIA) and investigated how BIA components changed before and after HD. Methods: This cross-sectional study included 147 subjects on maintenance HD from two hospitals and 298 propensity score-matched controls from one healthcare center. BIA was performed pre- and post-HD at mid-week dialysis sessions. Results: Extracellular water/total body water (ECW/TBW) and waist-hip ratio were higher in the HD patients; the other variables were higher in the control group. The cardiothoracic ratio correlated best with overhydration (r = 0.425, P < 0.01) in HD subjects. Blood pressure, hemoglobin, creatinine, and uric acid positively correlated with the lean tissue index in controls; however, most of these nutritional markers did not show significant correlations in HD subjects. Normal hydrated weight was predicted to be higher in the pre-HD than post-HD measurements. Predicted ultrafiltration (UF) volume difference based on pre- and post-HD ECW/TBW and measured UF volume difference showed a close correlation ($r^2=0.924$, P < 0.01). Remarkably, the leg phase angle increased in the post-HD period. Conclusion: The estimated normal hydrated weight using ECW/TBW can be a good marker for determining dry weight. HD subjects had higher ECW/TBW but most nutritional indices were inferior to those of controls. It was possible to predict UF volume differences using BIA, but the post-HD increase in leg phase angle, a nutritional marker, must be interpreted with caution.

  • Objectives: Recently, many studies have demonstrated that the waist-to-height ratio (WHtR) is more correlated with metabolic syndrome than the waist-hip ratio (WHR), body mass index (BMI). The aim of this study was to investigate and compare WHtR as a predictor of metabolic syndrome with other anthropometric indices. Methods: Approximately 3,800 data of subjects were used for the analysis, and data from Korean Medicine Data Centar). Receiver operating characteristic curve was performed and area under the curve (AUC) was calculated to find the usefulness of WHtR, WHR, and BMI. Results: WHtR was the most predictive factor with the highest diagnostic value for metabolic syndrome than WHR, BMI. AUC of WHtR was significantly higher in total subjects, men and women. Conclusions: WHtR may be the simple and effective anthropometric index for metabolic syndrome.

  • By-products produced from medicinal plants (MP) were evaluated as feed additives on growth performance, blood chemistry, and immune responses of the juvenile red lip mullet Liza haematocheila. A commercial diet was used as the control. Four experimental diets were prepared by combining 0.5% of MPs such as Panax ginseng (PG), Schisandra chinensis (SC), Angelica gigas (AG), and Rehmannia glutinosa (RG), via water absorption, to the control diet. Triplicate groups of fish averaging 61.8 g were fed one of experimental diets to apparent satiation for 8 weeks. Dietary supplementation of MPs did not affect the growth performance, biological indices and whole body composition of fish compared to the control. The fish fed with AG diet had an increased weight gain, and specific growth rate compared to fish in SC group. Decreased level of plasma glucose was observed in fish fed diet containing MPs. Lysozyme activity was significantly increased in the fish fed AG diet compared to that of fish in PG group. These results indicate that dietary supplementation of Angelica gigas may enhance growth performance and immune responses in juvenile red lip mullet.

Effect of increasing levels of threonine relative to lysine on the performance and meat quality of finishing pigs

  • Objective: The present study aimed to evaluate the effects of varying standardized ileal digestible lysine:threonine (SID Lys:Thr) ratio in the diet on the performance and meat quality of finishing pigs. Methods: In total 192 crossbred pigs ([Landrace×Yorkshire]×Duroc, 17 weeks old), with an initial body weight (BW) of 70.6±3.9 kg were used in an 8-wk trial. Pigs were randomly allotted to one of six dietary treatments based on their initial BW and sex (8 replications; 4 pigs per pen, 2 barrows and 2 gilts). The pigs in the 6 treatments were fed diets having different SID Lys:Thr ratios such as 1:0.65, 1:0.66, 1:0.67, 1:0.68, 1:0.69, and 1:0.70. Results: A linear increment (p<0.05) in average daily gain (ADG) and trends in reduction in feed conversion ratio (FCR) were observed during day 29 to 56 of the experiment and the apparent total tract digestibility (ATTD) of dry matter tended to increase linearly (p = 0.094) at the end of the experiment (day 56) with the increase in the dietary SID Lys:Thr ratios. The backfat thickness and lean percentage increased (linear effect, p<0.05) on day 28. In addition, at day 56, a linear (p<0.05) increment in lean percentage was observed. Significant quadratic responses (p = 0.02) for pH and drip loss at day 7 (p = 0.02), a linear increase (p<0.05) in cooking loss and drip loss at day 7, and a trend in quadratic response (p = 0.07) in the lightness of meat color (L*) were observed, whereas other meat quality indices were unaffected by varying the SID Lys:Thr ratios. Conclusion: The SID Lys:Thr ratio for maximum ADG, minimum FCR and enhanced digestibility was found to be 0.70. However, for carcass trait and meat quality, the SID Lys:Thr ratio of 0.65 was enough.
