Identification and Characterization of Bombyx mori LDH Gene through Bioinformatics Approaches

  • Lactate dehydrogenase (LDH) is a ubiquitous enzyme that plays a significant role in the clinical diagnosis of pathologic processes. Discovery of the LDH (BmLDH) gene in B. mori may shed light on its role in the biology of Lepidoptera species, and afford further understanding of the function of the enzyme. In this study, we used the bioinformatics tools to identify LDH gene in B. mori. Sequence analysis showed that BmLDH cDNA contains a 996 bp open reading frame, encoding 331 AA proteins, with seven introns. Compared with hHLDH (human heart LDH), BmLDH contained the same key active sites. Domain search and protein fold recognition analyses provide compelling evidences that the deduced protein is a LDH. Using the computer program MEGA3, we conducted a search for homologs of BmLDH among many eukaryotic species and confirmed that the BmLDH was conserved in all organisms investigated. This gene has been registered in GenBank under the accession number EU000385.

녹두(Phaseolus aureus L.) 급여가 당뇨성 흰쥐의 BUN 및 간 기능 효소 활성에 미치는 영향 (Effects of Mung Bean (Phaseolus aureus L.) Supplementation on BUN and Hepatic Functional Enzyme Activities in Streptozotocin -induced Diabetic Rats)

  • The purpose of this study was to investigate the improvement effect of mung bean (Phaseolus aureus L.) on the hepatic functional enzyme and catalase activity of streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley (SD) male rats were divided into four groups (n=6), and fed experimental diets containing mung bean meal [basal diet+5% mung bean (BM), basal diet+STZ+5% mung bean (SM)], and control (Basal Diet, BD), BS groups (basal diet+STZ). Serum concentrations of Blood Urea Nitrogen (BUN) and creatinine were significantly decreased (p<0.05) by 5% mung bean supplementation diet. The activities of aspartate transaminase (AST), alanine transaminase (ALT), akaline phosphatase (ALP), lactate dehydrogenase (LDH), amylase and lipase were decreased in the BD, BM and SM group than BS group. The catalase (CAT) activity was significantly increased (p<0.05) in mung bean supplementation diet (BM, SM group) than diabetic group (BS). In vivo experiments with diabetic rats showed that ingestion of mung bean supplementation diet were effective in BUN concentration, and hepatic functional enzyme activities.

고지혈증 유발 흰쥐에서 녹두가 혈청 BUN 및 효소활성에 미치는 효과 (Effects of Mung Bean on Serum BUN and Enzyme Activities in Hyperlipidemic Rats Induced by High Cholesterol Diet)

  • 본 연구의 목적은 5% 녹두(Phaseolus aureus L.)의 급여가 고지혈증 유발 Sprague-Dawley(SD)계 흰쥐의 혈청 blood urea nitrogen(BUN), creatinine 및 요산(uric acid)의 농도와 aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase(ALP), lactate dehydrogenase(LDH), amylase, lipase 및 catalase 활성에 미치는 영향을 규명하고자 시행하였다. 연구결과 녹두는 고지혈증으로 유발된 흰쥐의 혈청 BUN, creatinine, 요산 농도 및 AST, ALT, ALP, LDH, amylase, lipase의 활성 감소와 catalase 활성을 증가시키는 것으로 나타났다. 따라서 녹두는 신장과 간 등의 기능 개선과 예방에 효과적일 것으로 판단되어 기능성 소재로서의 이용 가능성이 있을 것으로 기대된다.
