Re-evaluation of green tide-forming species in the Yellow Sea

  • Green tides occur every year in the Yellow Sea (YS), and numerous investigations are proceeding on various aspects of the phenomenon. We have identified bloom-forming species collected from diverse locations in the YS using morphological traits and the chloroplast gene for the large subunit of ribulose-1,5-bisphosphate carboxylase (rbcL). Morphological and rbcL sequence data analyses characterized the blooming species on both sides of the YS as belonging to the Ulva linza-procera-prolifera (LPP) complex clade or U. prolifera of earlier reports. However, U. procera within the LPP complex must be regarded as synonym of U. linza. Moreover, U. prolifera in free-floating samples collected from the Qingdao coast in 2009 was clearly in a distinct clade from that of the blooming species. Therefore, U. linza is the main green tide alga in the YS and has the procera-morphology. The green drift mats in the southeastern part of the YS (southwest sea of Korea) consisted predominantly of U. linza and rarely of U. compressa or U. prolifera.

Distribution and Ecological Characteristics of Native Iris Odesanensis in Mt. Naeyon

  • Iris odesanensis Y.N. Lee is one kind of species among 54 endangered plant species designated by the Ministry of Environment in Korea. It is very rare native plant throughout the country. Therefore, this study was conducted to investigate the native Iris odesanensis in Mt. Naeyon for their distribution and ecological characteristics. They were mostly distributed mid-slope of the mountain from 443m to 498m altitude. They were located lower part of valley and provided enough humid condition. They had proper light and humidity conditions as they faced south south west, south south east, and south. They were also located at the places deciduous woody plants were distributed for proper light conditions for spring blooming. Iris flowered to use light at the period before the trees thickly covered with foliage. However this unfavorable light condition caused low percentage as 7.04% of blooming individuals in this study. In herbaceous, Vioia chaerophylloides(Regel) W. Hocker, Carex siderosticta Hance, Erythronium japonicum Decne., Oxalis obtriangulata Max., Hepatica asiatica Nakai, Lilium tsingtauense Gilg et cetera were appeared well. We propose that proper pruning of upper branches of the deciduous trees is needed for suitable light condition of management of native sites.

Microspore Development According to the Floral Budsize in Astragalus membranaceus Bunge

  • Astragalus membranaceus has flowers that are similar to that of the legume family, but shows poor bearing when self-pollination is induced. Thus, this study was carried out observing the ripening procedure of pistils and stamens and development stages of pollen in the context of the birth and growth of the flower. As to the bearing of the flower of A. membranaceus, few pod setting and 13% pod setting were observed when self-pollination is induced by paper-bag covering or artificial pollination treated respectively. The result indicates that A. membranaceus is a cross-pollination plant. A pistil grew faster than a stamen until just before blooming. The flower size was about 17.0mm$\times$4.0mm. Pistils and stamens had the same length after flowering. Pollen mother cells passed through meiosis and mitosis when its length reached around 3.5mm, thus creating the tetrade when 4 mm long. Pollen attained full growth when the bud was about 10mm long. An anther was found to tend to dehisce when the length of a bud reached around 12.0mm. As to the shape of pollen, about 70 % were normal. 1% and 30 % were small or empty pollen respectively. The result indicates that pollen of A. membranaceus attains full growth just before anther dehiscence which occurs before blooming while pistils grow faster than stamens until before flowering.

Analysis of Exclusive Causality between Environmental Factors and Cell Number of Cyanobacteria in Guem River (금강 주요지점에서의 환경 인자와 남조류 세포수의 배타적 인과성분석)

  • Algal blooming in 4 major rivers introduces substantial impacts to water front activity. Concentrations of algae are increasing at major points along the Geum River. Ecosystem food webs can be affected by algal blooming because blue-green algae release toxic materials. Even though there have been many studies on blue-green algae, its causality to environmental factors has not been completely determined yet. This study analyzed the exclusive correlation between various hydrometeorological, water quality, and hydrologic variables and the cell number of cyanobacteria to understand causality of blue-green algae in the Geum River. A prewhitening process was introduced to remove the autocorrelation structure and periodicity, which is useful to evaluate the effective relationship between two time series.

Characteristics of Photosynthetic Pigments during the Outbreak of Harmful Algal Bloom at the South Coastal Area in the Korean Sea Waters

  • Cochlodinium polykrikoides has occurs regularly during the summer in the South Sea of Korea. To investigate photosynthetic pigments concerned with phytoplankton community structure as bloom of Cochlodinium polykrikoides, the experiment was sampled at 20 stations three times between July and September 2002 in the Southern Sea of Korea. The distribution of peridinin, the biomarker of dinoflagellate, was higher at the blooming time than it was before and it disappeared after bloom. The correlative coefficients between Chl. a and peridinin at the blooming time and out of bloom were 0.9253 and 0.1613, respectively. This result indicated that the bloom was caused by dinoflagellate. The correlative coefficients between Chl. a and fucoxanthin were 0.3282 and 0.9759, respectively, and the correlative coefficients showed the succession from dinoflagellate to diatom. This result means that the bloom of Cochlodinium polykrikoides can be detected by Chl. a information from satellite remote sensing. Therefore, if the algorithm to detect peridinin in addition to Chl. a were to be developed, dinoflagellate red tide could be monitored more effectively.

Isolation of the Microbes Having Cyanobacteria Lytic Activity from Blooming Reservoirs (수화발생 저수지로부터 남조류 분해능을 가지는 미생물의 분리)

  • We have from water samples of Kwalim, Dochang, and Mulwang reservoirs in Kyonggi-Do, where cyanobacteria blooming occurred. Isolated microbes which have lytic activity for cyanobacteria. Water samples were smeared on the Anabaena cylindrica lawn and incubated in light chamber at $28^\circ{C}$, under 3000 lux for 13 days. A fungus having cyanobacterial lytic activity was isolated from the samples of Dochang reservoir. The isolate was identified as Cryptococcus laurentii by Vitek system. From the culture of the isolate, four major extracellular protein bands (29, 35.2, 40.9, 51.1 kDa) have been detected and the 29 kDa protein band was more thickly appeared in the culture with cyanobacteria.

Phytoplankton Community and the Evaluation of Water Quality Status in So-ok Stream, the Inflowing Stream to Daechung Lake (대청호의 유입지천인 소옥천의 식물플랑크톤 군집 분석 및 수질영양단계 평가)

  • The study analyzed the physico-chemical quality of water and the resident phytoplankton community, and evaluated the water trophic status in So-ok stream flowing into Daechung lake from October 2007 to July 2008. Algal blooming by cyanophytes and diatoms continued throughout the year, with Microcystis aeruginosa and Aulacoseira granulata being prevalent. Ten taxa that are indicators of polluted water, including Oscillatoria limosa appeared. Results based on (Trophic status index) TSI and (Lake trophic status index) LTSI evaluations of water quality status indicated mesotrophication.

Effects of Sediments on the Growth of Algae at Chusori Area in Daechung Reservoir (대청호 추소리 수역의 퇴적물이 조류 성장에 미치는 영향)

  • In order to investigate the effect of internal loading from sediment on algal blooming at Chusori area in Daechung Reservoir, the amount and contamination level of sediment and the release rate of total phosphorus were analyzed. The sedimentary layer was consisted with two layers, and the average depth of upper and lower ones were 0.35 and 1.44 m, respectively. The fraction of inorganic phosphorus in the sediment was higher than that of organic phosphorus, and the fractions of phosphorus which responsible for internal loading were very high as in the range of 72.7 and 80.2% of inorganic phosphorus. The C/N ratio of sediment taken with core sampler indicated the organic compounds are originated from settled algae from water body. The average release rate of total phosphorus from sediment was $6.74({\pm}0.50)mg/m^2/day$. These results indicated that the internal loading from sediment contributes the excessive algal growth at Churosi area, and the countermeasures to improve the quality of sediments are required to manage algal blooming in Daechung Reservoir.

Improved High-Performance Liquid Chromatographic Method for the Determination of N-vanillyl-n-nonivamide(nonivamide) in Korean Hot Peppers (HPLC 법에 의한 국산 고추에 함유되어 있는 Nonivamide의 신규 분석법 개발)

  • We report a novel HPLC method for analyzing the capsaicinoids and nonivamide. The following conditions were used: a total column and column temperature of $3^{\circ}C$, a detection wavelength of 280 nm, an eluent containing acetonitrile : 0.5% formic acid(31:69, v/v). The experimental data was compared with standard nonivamide and capsaicin. The results showed that nonivamide was eluted in 96.7 minutes with a recovery ratio of 96.5% after adding known nonivamide. In addition, the detection limit for nonivamide was 18.6 ng. The capsaicinoids from Korean pepper were extracted (after blooming 60th) from 3 breeds cultivated in Korea, Buchon, Chungyang and Hanbando. and crisis it analyzed sympathizes the nonivamide in the result nonivamide from the Buchon and the Chungyang which are not detected from the Hanbando $0.16{\sim}0.24{\mu}g/g$ it detects with very small amount it contains it confirmed the each capsaicinoid with the HPLC and the LC-MS. These results are expected contribute to the quality control of various product processes, which includes product delivery inspection as well as examining the capsaicinoids present in the product for possible medicine manufacture and various foodstuff, and the nonivamide content, which may be a feed for various manufacturing processes involving chemical synthesis.
