왕종개 Iksookimia longicorpus (Pisces, Cobitidae) 피부 구조와 세포화학적 특징 (Structure and Cytochemistry of Skin in Spined Loach, Iksookimia longicorpus (Pisces, Cobitidae))

  • 왕종개 Iksookimia longicorpus 피부 및 피부샘 (skin gland)의 구조와 그들의 세포화학적 특징을 조사하기 위해 등, 측면, 복부, 머리부분 등 4부분을 조사하였다. 왕종개의 표피 (epidermis)는 작은 점액세포 (mucous cell)와 커다란 club cell의 2 type의 gland가 분포하였다. 특히 mucous cell의 성분은 acidic sulfomucins이었으나 club cell은 점액물질 (mucosubstance) test에 전혀 반응을 나타내지 않았다. 표피의 기저층 (stratum germinativum)에는 림파구를 가지는 lymphocytic space가 잘 발달되어 있으며, 기저막 (basement membrane) 바로 아래에는 많은 양의 모세혈관들이 서로 가깝게 분포하였다. 진피 (dermis)의 stratum laxum에는 alcian blue와 PAS 반응에서 양성반응을 보이는 부분 (a definite area)이 존재하였다.

Biocompatibility of Poly(MPC-co-EHMA)/Poly(L-1actide-co-glycolide) Blends

  • Poly(L-lactide-co-glycolide)(PLGA) was blended with poly[$\omega$-methacryloyloxyethyl phospho-rylcholine-co-ethylhexylmethacrylate (PMEH)] (PLGA/PMEH) to endow with new functionality i.e., to improve the cell-, tissue- and blood-compatibility. The characteristics of surface properties were investigated by measurement of contact angle goniometer, Fourier-transform infrared spectroscopy with attenuated total reflectance (FTIR-ATR) and electron spectroscopy for chemical analysis (ESCA). NIH/3T3 fibroblast and bovine aortic endothelial cell were cultured on control and PLGA/PMEH surfaces for the evaluation of ceil attachment and proliferation in terms of surface functionality such as the concentration of phosphoryl-choline. Also, the behavior of platelet adhesion on PLGA/PMEH was observed in terms of the surface functionality. The contact angles on control and PLGA/PMEH surfaces decreased with increasing PMEH content from 75$^{\circ}$ to about 43$^{\circ}$. It was observed from the FTIR-ATR spectra that phosphorylcholine groups are gradually increased with increasing blended amount of MPC. The experimental P percent values from ESCA analysis were more 3.28∼7.4 times than that of the theoretical P percent for each blend films. These results clearly indicated that the MPC units were concentrated on the surface of PLGA/PMEH blend. The control and PLGA/PMEH films with 0.5 to 10.0 wt% concentration of PMEH were used to evaluate cell adhesion and growth in terms of phosphorylcholine functionality and wettability. Cell adhesion and growth on PLGA/PMEH surfaces were less active than those of control and both cell number decreased with increasing PMEH contents without the effect of surface wettability. It can be explained that the fibronectin adsorption decreased with an increase in the surface density of phosphorylcholine functional group. One can conclude the amount of the protein adsorption and the adhesion number of cells can be controlled and nonspecifically reduced by the introduction with phosphorylcholine group. Morphology of the adhered platelets on the PLGA/PMEH surface showed lower activating than control and the number of adhered platelets on the PLGA/PMEH sample decreased with increasing the phosphorylcholine contents. The amount of fibrinogen adsorbed on the PLGA/PMEH surface demonstrated that the phospholipid polar group played an important role in reducing protein adsorption on the surface. In conclusion, this surface modification technique might be effectively used PLGA film and scaffolds for controlling the adhesion and growth of cell and tissue, furthermore, blood compatibility of the PLGA was improved by blending of the MPC polymer for the application of tissue engineering fields.

  • This experiment was done to investigate the effects of Ojajiwhangeumja(OJWEJ) to the symptoms of senility. To prove the effects of OJWEJ we injected scopolamine(1mg/kg) to the vein of rats. And we measured the blood cells(WBC, RBC, Platelet), constituents of serum(BUN, creatinin, glucose, uric acid), endurance of films of the red blood cell to erythrocyte hemolises, the activity of cholinesterase in serum, TBA and the activity of catalase, SOD in the purified microsome of brain tissue of rats. The results were as follows; 1. The number of white blood cells, platelet was increased significantly in the group treated by OJWEJ in comparison with control group. 2. The number of BUN, creatinin, glucose of serum decreased significantly in the OJWEJ treated group compared with control group. 3. The erythrocyte hemolises in red blood cells restrained significantly in the group treated by OJWEJ in comparison with control group. 4. The activity of cholinesterase in OJWEJ treated group increased significantly compared with control group. 5. The amounts of malondialdehyde of serum decreased significantly in the OJWEJ treated group in comparison with control group. 6. The catalase in the microsome of rat brain was activated significantly in the group treated by OJWEJ compared with control group. 7. The superoxide dismutase in the group treated by OJWEJ activated significantly in comparison with control group. According to this experiment it is suggested that OJWEJ accelerates the activity of colinesterase and restrains the creation of erythrocyte hemolises and accumulation of senile substance. But I look forward to see the further research to be made.

유산소 운동이 남자 중학생의 면역기능에 미치는 영향 (Effects of Aerobic Exercise on Immune Function in Middle School Boys)

  • The purpose of this study was to evaluate the effect of moderate physical exercise program on a number of immune parameters in middle school boys($15.07{\pm}0.39$ yrs). 14 volunteers were included in this physical exercise program. They were randomly assigned to an experiment(7) and a control(7) group. Measurements were taken before program, after 4 weeks and 8 weeks of aerobic exercise for immune parameters such as number of circulating leukocytes, concentration of WEC subsets, lymphocyte subsets, immunoglobulins, complements, and number of blood adipose components. Aerobic exercise consisted of track running at a work intensity of 65% HRmax, $45{\sim}55$ min per day, 3 times per week, and for 8 weeks. The results are as follows; 1. There were no significant changes between and within groups in the number of circulating leukocytes and concentration of WEC subsets. 2. There were significant(p<.05) changes in concentration of B-cell between groups, and T-cell, helper T-cell, and B-cell within experimental group. 3. There were no significant changes between and within groups in concentration of complements. 4. There were significant(p<.05) changes in concentration of IgG between groups and within experimental group. 5. There were significant(p<.05) changes in concentration of TG between groups, and TG, HDL-C and LDL-C within experimental group. In conclusion, the moderate exercise training for 8 weeks can be beneficial on immune function and decrease the concentration of flood adipose components in adolescents.

율무 추출물이 마우스 면역세포 활성에 미치는 영향 (Effects of Job's Tear(Yul-Moo) Extracts on Mouse Immune Cell Activation)

  • Natural products are increasingly appreciated as a lead for drug discovery and development. A number of investigators have studied various activities of natural products and have found that they have not only nutritional effects but also beneficial properties to cure various diseases and to maintain good health. Job's Tear(Yul-Moo) is a grass crop that have long been used in traditional medicine and a nourishing food. Job's Tear has been reported to exhibit anti-inflammatory, stomachic, antiallergic activity, and antispastic effects and has been used in China for the treatment of warts, rheumatism, and neuralgia although its mechanism remains unclear. Previous results in our laboratory demonstrated that the ethanol extract and water extract of Job's Tear exerted an immune regulatory function on mice cells in vitro. The present study was performed to investigate the ex vivo effect of Job's Tear on immune function. Seven to eight weeks old mices(Balb/c) were fed ad libitum on chow diet and water extract of Job's Tear were orally administrated every other day for two or four weeks at two different concentrations (50 and 500mg/kg B.W.). Proliferation of mice spenocytes and antibody production to sheep red blood cells(SRBC) using hemolytic plague forming cell assay were used to indicate the immune activity. Splenocytes proliferation of Job's Tear with mitogen stimulation such as Con A and LPS was enhanced at 50 mg/kg B.W. concentrations compared to those of control group. In case of antibody production to sheep red blood cells, the number of antibody- secreting cells was increased by administration of 50mg/kg B.W. concentration in mice immunized as a T-dependent antigen. From the present study, Job's Tear water extracts may be suggested to stimulate the mice immune response by enhancing the splenocytes proliferation and the number of plague forming cells.

Immuno-enhancement effects of Korean Red Ginseng in healthy adults: a randomized, double-blind, placebo-controlled trial

  • Background: Most clinical studies of immune responses activated by Korean Red Ginseng (KRG) have been conducted exclusively in patients. However, there is still a lack of clinical research on immune-boosting benefits of KRG for healthy persons. This study aims to confirm how KRG boosts the immune system of healthy subjects. Methods: A total of 100 healthy adult subjects were randomly divided into two groups that took either a 2 g KRG tablet or a placebo per day for 8 weeks. The primary efficacy evaluation variables included changes in T cells, B cells, and white blood cells (WBCs) before and after eight weeks of KRG ingestion. Cytokines (TNF-α, INF-γ, IL-2 and IL-4), WBC differential count, and incidence of colds were measured in the secondary efficacy evaluation variables. Safety evaluation variables were used to identify changes in laboratory test results that incorporated adverse reactions, vital signs, hematological tests, blood chemistry tests, and urinalysis. Results: Compared to the placebo group, the KRG intake group showed a significant increase in the number of T cells (CD3) and its subtypes (CD4 and CD8), B cells, and the WBC count before and after eight weeks of the intake. There were no clinically significant adverse reactions or other notable results in the safety evaluation factors observed. Conclusion: This study has proven through its eight-week intake test and subsequent analysis that KRG boosts the immune system through an increase in T cells, B cells, and WBCs, and that it is safe according to the study's safety evaluation.

Attenuation of Brain Injury by Water Extract of Goat's-beard (Aruncus dioicus) and Its Ethyl Acetate Fraction in a Rat Model of Ischemia-Reperfusion

  • Ischemic stroke constitutes about 80% of all stroke incidences. It is characterized by brain cell death in a region where cerebral arteries supplying blood are occluded. Under these ischemic conditions, apoptosis is responsible for the cell death, at least in part. Goat's-beard (Aruncus dioicus var. kamtschaticus) is a perennial plant that grows naturally in the alpine regions of Korea. In the present study, we first determined whether water extract of goat's-beard (HY1646) and some of its fractions prepared by partitioning with organic solvents could improve the viability of human hepatocellular carcinoma cells (HepG2) cultured under hypoxic condition by blocking apoptotic pathways. Based on the in vitro findings, we subsequently investigated whether HY1646 and the ethyl acetate fraction (EA) selected from cell culture-based screening could attenuate brain injury in a rat middle cerebral artery occlusion (MCAO) model of ischemia (2 hr), followed by 22 hours of reperfusion. The cell number was sustained close to that initially plated in the presence of HY1646 even after 24 hr of cell culture under hypoxic condition (3% $O_2$), at which time the cell number reached almost zero in the absence of HY1646. This improvement in cell viability was attributed to the delay in apoptosis, identified by the formation of DNA ladder in gel electrophoresis. Of fractions soluble in hexane, ethyl acetate (EA) and butanol, EA was chosen for the animal experiments because EA demonstrated the best cell viability at the lowest concentration (10 ${\mu}g$/mL). HY1646 (200 mg/kg) and EA (10 and 20 mg/kg) significantly reduced infarct size, an index of brain injury, by 16.6, 40.0 and 61.0%, respectively, as assessed by 2,3,5-triphenyl tetrazolium chloride staining. The findings suggest that prophylactic intake of goat's beard might be beneficial for preventing ischemic stroke.

한국 재래 산양의 비교해부학적 연구 2. 장간막 비만세포에 대하여 (Comparative Anatomy of the Korean Native Goat 2. Mesenteric mast cell)

  • This study was carried out to investigate on the morphology, distribution and stainability of the mast cells in the Korean Native goat. For the study, the experimental animals were anesthetized with pentobarbital sodium and opened the anterior abdomianl wall to remove immediately the specimens with a minimum of mechanical effects. The mesenteries were fixed in 10% neutral formalin, 4% basic lead acetate, absolute alcohol and ethlene glycol monoethyl ether. Following 24 hours of fixation, the toto preparation stained with 0.4% toluidine blue, 1% methylene blue, 1.5% bismark brown, saturated thionine and thionlne-methylene blue complex solution. The preparation were observed from 10 microscopic field with 450 magnification. The results were as follows: 1. The form of the mesenteric mast cell was found 2 types. One was spindle form in larger number around vessels, the other was ovoid or spherical form in connective tissue far from blood vessels. 2. The average size was $18.63{\pm}5.75{\mu}m$ in length, $10.61{\pm}3.39{\mu}m$ in width and number was $105.50{\pm}18.45$. 3. Ethylene glycol monoethyl ether was particularly useful in preserving the mast cell granules. 4. Thione-methylene blue complex solution might be recommended to stain of granules.

Captafol 免疫毒性에 미치는 Ethanol의 영향 (Studies on the Effect of Captafol and Ethanol the Murine Immune System)

  • Captafol (1H-Isoindole-1.3(2H)-dione, 3a, 4, 7, 7a-tetrahydro-2-[1, 1, 2, 2-tetrahydroethyltkio]) is widely used as fungicide in agriculture. Immune modulatory effects of captafol and ethanol were studied in mice. Mice administered captafol intra peritoneally every other day for 5 times, and ethanol per os as captafol. Mice were sensitized and challenged with sheep red blood cells, serum antibody titer, foot pad swelling, and rosette forming cell number were mediated immune response. 1. The result show that humoral immune response and cell mediatea response were suppressed by captafol. 2. Especially effect of ethanol on the captafol immune response were significantly suppressed the humoral immune response and cell mediated immune response.

인태아 상경신경절내 소형의 과립함유세포에 관한 전자현미경적 연구 (Ultrastructural Study on the Development of the Small Granule-Containing Cells in Superior Cervical Ganglion of Human Fetus)

  • The development of small granule-containing cell in the superior cervical ganglion was studied by electron microscopic method in human fetuses ranging from 40 mm to 260 mm crown rump length (10 to 30 weeks of gestational age). At 40 mm fetus, the superior cervical ganglion was composed of clusters of undifferentiated cells, primitive neuroblasts, and unmyelinated nerve fibers together with blood vessels. At 90 mm fetus, the superior cervical ganglion consisted of neuroblasts, satellite cell, small granule-containing cells, and unmyelinated nerve fibers. Two morphological types of the small granule-containing cells in the superior cervical ganglion were first indentified at 90 mm fetus, but were rare. Type I granule-containing cell occurred in solitary and had long processes, whereas type II cells tend to appeared in clusters near the blood capillaries. The granule-containing cells were characterized by the presence of dense-cored vesicles ranging from $150{\sim}300nm$ in diameter in both the cell bodies and processes. Other organelles included abundant mitochondria, rough endoplasmic reticulum, neurotubules, and widely distributed ribosomes. The granule-containing cells had long processes similar to those found in principal ganglionic cells. They could be identified by their content in dense-cored vesicles. The small granule-containing cells increased somewhat in size and number with increase of fetal age. Synaptic contacts were first found on the solitary granule-containing cell at 150 mm fetus. Synaptic contacts between the soma and processes of type I granule-containing cells and preganglionic axon terminals were observed. In addition, synaptic junctions between the processes of granule-containing cells and presumed dendrite of postganglionic neuron were also observed from 150 mm onward. On the basis of these features type I granule-containing cells could be considered as interneurons. The clusters of type II granule-containing cells were located in the interstitial or subcapsular portions of the ganglion, and had short processes which ended in close relation to fenestrated capillaries. Therefore it may be infer that clusters of type II granule-containing cells have an endocrine function.
