Nitric oxide(NO) mediating non-adrenergic non-cholinergic(NANC) relaxation in the boar retractor penis muscle I.   • This study was carried out to characterize nonadrenergic, noncholinergic(NANC) relaxation of porcine retractor penis(PRP) muscle induced by electrical field stimulation(EFS) and to investigate the actions of niric oxide(NO) and vasoactive intestinal polypeptide(VIP) as candidates for NANC neurotransmitters. Biphasic relaxations of PRP muscle were induced by EFS to NANC nerve. Rapid-phase relaxation was observed at low frequency(0.5-16Hz) and slow-phase relaxation followed during high frequency(8-60Hz). Both relaxations were frequency-dependent and TTX($1{\times}10^{-6}M$)-sensitive. L-NAME($2{\times}10^{-5}M$) inhibited the rapid-phase relaxation, but not the slow-phase relaxation. The inhibition of the rapid-phase relaxation with L-NAME was reversed by L-arginine ($1{\times}10^{-3}M$) but not by D-arginine($1{\times}10^{-3}M$). Methylene blue($4{\times}10^{-5}M$) reduced the rapid-phase relaxation. Exogenous No(ExoNO, $1{\times}10^{-5}-1{\times}10^{-4}M$) induced dose-dependent relaxations of PRP muscle. Oxyhemoglobin($5{\times}1^{-5}M$) blocked the relaxation induced by ExoNO and inhibited EFS-induced relaxation. Hydroquinone($1{\times}10^{-4}M$) also abolished the relaxation induced by ExoNO, but did not affect EFS-induced relaxation. L-NAME resistant slow-phase relaxation to EFS was inhibited by ${\alpha}$-chymotrypsin(2.5 U/ml). Both methylene blue($4{\times}10^{-5}M$) and Nethylmaleimide($1{\times}10^{-4}M$) reduced the slow-phase relaxation by EFS. [4-Cl-D-$Phe^6$, $Leu^{17}$]-VIP($3{\times}10^{-6}M$) inhibited the slow-phase relaxation by EFS. External applications of VIP ($1{\times}10^{-7}M$) caused relaxations that were simillar to the L-NAME resistant slow-phase relaxations induced by EFS, and relaxant effects of exogenous VIP were blocked by ${\alpha}$-chymotrypsin(2.5 U/ml).

  • Recently, the number of passengers using airplanes is rapidly increasing due to the increase of overseas travelers. Therefore, the probability of exposure to natural radiation due to altitude is increasing due to the increase in flight time. Cosmic-ray penetrates the Earth's magnetic field belt Van Allen, which is located at an altitude of 400 km to 1200 km. Most cosmic rays are blocked at Van Allen belt. However, cosmic-ray could be not completely blocked, and a small amount of cosmic-ray affects the earth. In general, if the altitude was increased by 100m, the natural exposure dose increased by 0.03 mSv on the Earth. In this study, I tried to minimize the exposure to natural radiation in airplanes when boarding airplanes. Especially, I was aimed to minimize radiation exposure by protecting the highly sensitive thyroid gland among human organs. According to the results of the study, the designed shielding handkerchief was able to shield cosmic natural radiation dose by more than 70%. In conclusion, the application of the shielding handkerchief made in this study can be effectively shield natural radiation.

A Study on the Evaluation of Radiation Safety in Opened-Ceiling-Facilities for Radiography Testing (천장 개방형 RT 사용시설의 방사선 안전성 평가 연구)

  • Radiography-Testing that verify the quality of welding structures without destruction are overwhelmingly used in industries, but many safety precautions are required as radiation is used. The workers for Radiography-Testing perform the inspection by moving the Iridium-192 radiation source embedded in the transport container of the gamma-ray irradiator within or outside the facility. The general facility is completely blocked about radiation from the outside with thick concrete, but if it is difficult for worker to handle object of inspection, facilities ceiling can be opened. A general facility may be constructed using a theoretical dose evaluation method because all exterior facilities are blocked, but if the ceiling is open, it is not appropriate to evaluate radiation safety with a simple theoretical calculation method due to the skyshine effect. Therefore, in this study, the radiation safety of the facility was evaluated in the actual field through an ion chamber survey-meter and an accumulated dose-meter called as OSLD, and the actual evaluation environment was modeled and evaluated using the Monte Carlo simulation code as FLUKA. According to the direction of the irradiation, the radiation dose at the facility boundary was difficult to meet the standards set by the regulatory authority, and radiation safety could be secured through additional methods. In addition, it was confirmed that the simulation results using the Iridium-192 source were valid evaluation with the actual measured results.

Calcium Channel Blockers Suppress the Responses of Rat Dorsal Horn Cell to Nociceptive Input (쥐 척수후각세포의 유해자극 반응에 대한 칼슘이온통로 차단제의 억제작용)

  • Calcium ions are implicated in a variety of physiological functions, including enzyme activity, membrane excitability, neurotransmitter release, and synaptic transmission, etc. Calcium antagonists have been known to be effective for the treatment of exertional angina and essential hypertension. Selective and nonselective voltage-dependent calcium channel blockers also have inhibitory action on the acute and tonic pain behaviors resulting from thermal stimulation, subcutaneous formalin injection and nerve injury. This study was undertaken to investigate the effects of iontophoretically applied $Ca^{++}$ and its antagonists on the responses of WDR (wide dynamic range) cells to sensory inputs. The responses of WDR cells to graded electrical stimulation of the afferent nerve and also to thermal stimulation of the receptive field were recorded before and after iontophoretical application of $Ca^{++}$, EGTA, $Mn^{++}$, verapamil, ${\omega}-conotoxin$ GVIA, ${\omega}-conotoxin$ MVIIC and ${\omega}-agatoxin$ IVA. Also studied were the effects of a few calcium antagonists on the C-fiber responses of WDR cells sensitized by subcutaneous injection of mustard oil (10%). Calcium ions and calcium channel antagonists ($Mn^{++}$, verapamil, ${\omega}-conotoxin$ GVIA & ${\omega}-agatoxin$ IVA) current-dependently suppressed the C-fiber responses of WDR cells without any significant effects on the A-fiber responses. But ${\omega}-conotoxin$ MVIIC did not have any inhibitory actions on the responses of WDR cell to A-fiber, C-fiber and thermal stimulation. Iontophoretically applied EGTA augmented the WDR cell responses to C-fiber and thermal stimulations while spinal application of EGTA for about $20{\sim}30\;min$ strongly inhibited the C-fiber responses. The augmenting and the inhibitory actions of EGTA were blocked by calcium ions. The WDR cell responses to thermal stimulation of the receptive field were reduced by iontophoretical application of $Ca^{++}$, verapamil, ${\omega}-agatoxin$ IVA, and ${\omega}-conotoxin$ GVIA but not by ${\omega}-conotoxin$ MVIIC. The responses of WDR cells to C-fiber stimulation were augmented after subcutaneous injection of mustard oil (10%, 0.15 ml) into the receptive field and these sensitized C-fiber responses were strongly suppressed by iontophoretically applied $Ca^{++}$, verapamil, ${\omega}-conotoxin$ GVIA and ${\omega}-agatoxin$ IVA. These experimental findings suggest that in the rat spinal cord, L-, N-, and P-type, but not Q-type, voltage-sensitive calcium channels are implicated in the calcium antagonist-induced inhibition of the normal and the sensitized responses of WDR cells to C-fiber and thermal stimulation, and that the suppressive effect of calcium and augmenting action of EGTA on WDR cell responses are due to changes in excitability of the cell.

MLCK and PKC Involvements via Gi and Rho A Protein in Contraction by the Electrical Field Stimulation in Feline Esophageal Smooth Muscle

  • We have shown that myosin light chain kinase (MLCK) was required for the off-contraction in response to the electrical field stimulation (EFS) of feline esophageal smooth muscle. In this study, we investigated whether protein kinase C (PKC) may require the on-contraction in response to EFS using feline esophageal smooth muscle. The contractions were recorded using an isometric force transducer. On-contraction occurred in the presence of $N^G$-nitro-L-arginine methyl ester (L-NAME), suggesting that nitric oxide acts as an inhibitory mediator in smooth muscle. The excitatory composition of both contractions was cholinergic dependent which was blocked by tetrodotoxin or atropine. The on-contraction was abolished in $Ca^{2+}$-free buffer but reappeared in normal $Ca^{2+}$-containing buffer indicating that the contraction was $Ca^{2+}$ dependent. 4-aminopyridine (4-AP), voltage-dependent $K^+$ channel blocker, significantly enhanced on-contraction. Aluminum fluoride (a G-protein activator) increased on-contraction. Pertussis toxin (a $G_i$ inactivator) and C3 exoenzyme (a rhoA inactivator) significantly decreased on-contraction suggesting that Gi or rhoA protein may be related with $Ca^{2+}$ and $K^+$ channel. ML-9, a MLCK inhibitor, significantly inhibited on-contraction, and chelerythrine (PKC inhibitor) affected on the contraction. These results suggest that endogenous cholinergic contractions activated directly by low-frequency EFS may be mediated by $Ca^{2+}$, and G proteins, such as Gi and rhoA, which resulted in the activation of MLCK, and PKC to produce the contraction in feline distal esophageal smooth muscle.

Effect Analysis of Offshore Wind Farms on VHF band Communications (VHF 대역 통신에 대한 해상풍력 발전단지의 영향성 분석)

  • As the development of renewable energy expands internationally to cope with global warming and climate change, the share of wind power generation has been gradually increasing. Although wind farms can produce electric power for 24 h a day compared to solar power plants, Their interfere with the operation of nearby radars or communication equipment must be analyzed because large-scale wind power turbines are installed. This study analyzed whether a land radio station can receive sufficient signals when a ship sailing outside the offshore wind farm transmits distress signals on the VHF band. Based on the geographic information system digital map around the target area, wind turbine CAD model, and wind farm layout, the area of interest and wind farm were modeled to enable numerical analysis. Among the high frequency analysis techniques suitable for radio wave analysis in a wide area, a dedicated program applying physical optics (PO) and shooting and bouncing ray (SBR) techniques were used. Consequently, the land radio station could receive the electromagnetic field above the threshold of the VHF receiver when a ship outside the offshore wind farm transmitted a distress communication signal. When the line of sight between the ships and the land station are completely blocked, the strength of the received field decreases, but it is still above the threshold. Hence, although a wind farm is a huge complex, a land station can receive the electromagnetic field from the ship's VHF transmitter because the wave length of the VHF band is sufficiently long to have effects such as diffraction or reflection.

Correction of Radar Reflectivity over Beam Blocking Area by Accumulated Radar Reflectivity (레이더 반사도 누적 방법을 이용한 지형에 의한 부분차폐영역의 레이더 반사도 보정)

  • Radar beam blocking which is partially or entirely interrupted by obstacles like a mountain causes underestimation of the rainfall. In this paper, partially blocked radar reflectivity is retrieved using the ARM(Accumulated Reflectivity Map). ARM is made by accumulation of the radar reflectivity and very useful product to analyze the beam blockage. The blockage correcting map could be obtained by assuming the spatially uniform reflectivity field in the ARM. This method is applied to the cases of typhoon and Changma, and we obtain the MFE(Mean Fractional Error) from two radar data, the one is objective radar data which is affected by blockage and the other is comparative radar data which is not affected by blockage. Before blocking correction, MFE is 20-35%. However, after correction, MFE decreases to 7-10%.

Roles of $Na^+\;-Ca^{2+}$ Exchange in the Negative Force-Frequency Relationship

  • Frequency-force relationships (FFR) were studied in electrically field stimulated rat left atria (LA) by reducing the stimulation frequency from resting 3 Hz to test frequencies (0.1-1 Hz) for 5 minutes. The twitch amplitudes of LA elicited the typical negative staircases with 3-phased changes: the initial rapid increase, the second decrease and the following plateau at test frequencies. Verapamil $(3{\times}10^{-5}\;M)$ pretreatment elicited frequency-dependent suppression of the twitch amplitudes, exaggerating the negative staircase. Monensin pretreatment enhanced not the peak but the plateau amplitudes in a concentration-dependent manner. When the $Na^+-Ca^{2+}$ exchange was blocked by $Na^+\;and\;Ca^{2+}$ depletion in the Krebs Hensleit buffer (0 $Na^+-0\;Ca^{2+}$ KHB), the twitch amplitudes increased in a frequency-dependent manner, changing the negtive staircase into the positve one. Meanwhile, the 0 $Na^+-0\;Ca^{2+}$ KHB applicationinduced enhancement was strongly suppressed by caffeine (5 mM) pretreatment. Only dibucaine among the local anesthetics increased the basal tone during frequency reduciton. There were no differences in $^{45}Ca$ uptakes between 0.3 Hz and 3 Hz stimulation except at 1 min when it was significantly low at 0.3 Hz than 3 Hz, illustrating net $Ca^{2+}$ losses. Monensin pretreatment enhanced the rate of this $Ca^{2+}$ loss. Taken together, it is concluded that $Na^+-Ca^{2+}$ exchange extrudes more SR released $Ca^{2+}$ out of the cell in proportion to the frequency, resulting in the negative rate staircase in the rat LA.

Phorbol 12-Myristate 13-Acetate Enhances Long-Term Potentiation in the Hippocampus through Activation of Protein Kinase $C{\delta}$ and ${\varepsilon}$

  • Many intracellular proteins and signaling cascades contribute to the sensitivity of N-methyl-D-aspartate receptors (NMDARs). One such putative contributor is the serine/threonine kinase, protein kinase C (PKC). Activation of PKC by phorbol 12-myristate 13-acetate (PMA) causes activation of extracellular signal-regulated kinase (ERK) and promotes the formation of new spines in cultured hippocampal neurons. The purpose of this study was to examine which PKC isoforms are responsible for the PMA-induced augmentation of long-term potentiation (LTP) in the CA1 stratum radiatum of the hippocampus in vitro and verify that this facilitation requires NMDAR activation. We found that PMA enhanced the induction of LTP by a single episode of theta-burst stimulation in a concentration-dependent manner without affecting to magnitude of baseline field excitatory postsynaptic potentials. Facilitation of LTP by PMA (200 nM) was blocked by the nonspecific PKC inhibitor, Ro 31-8220 ($10{\mu}M$); the selective $PKC{\delta}$ inhibitor, rottlerin ($1{\mu}M$); and the $PKC{\varepsilon}$ inhibitor, TAT-${\varepsilon}V1$-2 peptide (500 nM). Moreover, the NMDAR blocker DL-APV ($50{\mu}M$) prevented enhancement of LTP by PMA. Our results suggest that PMA contributes to synaptic plasticity in the nervous system via activation of $PKC{\delta}$ and/or $PKC{\varepsilon}$, and confirm that NMDAR activity is required for this effect.

Effect of Fluoxetine on the Induction of Long-term Potentiation in Rat Frontal Cortex

  • Serotonin (5-hydroxytroptamine, 5-HT) has been shown to affect the induction of long-term potentiation (LTP) in the cortex such as the hippocampus, the visual cortex and the prefrontal cortex. Fluoxetine, as a selective serotonin reuptake inhibitor, is used in the management of a wide variety of psychological diseases. To study the effect of fluoxetine on the induction of LTP, we recorded the field potential in layer II/III of the frontal cortex from 3-wk-old. LTP was induced in horizontal input by theta burst stimulation (TBS). TBS with two-folds intensity of the test stimulation induced LTP, which was blocked by application of D-AP5 $(50 {\mu}M)$, an NMDA receptor antagonist. Whereas bath application of 5-HT $(10 {\mu}M)$ inhibited the induction of LTP, treatment with the 5-HT depleting agent, para-chloroamphetamine $(PCA,\;10{\mu}M)$, for 2hr did not affect the induction of LTP. Bath application of fluoxetine (1, 3, and $10 {\mu}M)$ suppressed the induction of LTP in concentration-dependent manner, however, fluoxetine did not inhibit the induction of LTP in 5-HT-depleted slices. These results indicate that fluoxetine may inhibit the induction of LTP by modulating serotonergic mechanism in the rat frontal cortex.
