녹차 카테킨류의 항혈전 효과 (The Antithrombotic Effects of Green Tea Catechins)

  • Green tea catechins(GTC) were studied for its inhibitory effect on human platelet aggregation in vitro, for its antithrombotic effect in mice in viro, and bleeding and clotting time in rats. The catechins were isolated and purified from green tea, which were composed of (-)-epigallocatechin gallate, (-)-epigallocatechin, (-)epicatechin gallate and (-)-epicatechin, GTC produced a potent inhibition of human platelet aggregation in a dose-dependent manner against the stimulants such as ADP, collagen, epinephrine and ristocetin n vitro. GTC also prevented death due to the formation of pulmonary thrombosis by platelet aggregates in mice in a dose-de-pendent manner in viro. GTC increased the bleeding time, whole blood clotting time and plasma clotting time in rats, too. These results suggest that GTC is a promising antithrombotic agent.

항혈전작용 및 콜레스테롤 대사에 관한 흰쥐 식이내 삶은 계란 급여 효과 (Effects of the Dietary Boiled Eggs on the Antithrombotic Activity and Cholesterol Metabolism in Rats)

  • The influence of the ditary containing boiled eggs on the plasma cholesterol level and antithrombotic activity in rats was studied. Rats were fed basal diet(0% boiled eggs) as a control group or diets containing 25% and 50% boiled eggs or a mixed diet with 95% boiled eggs plus 5% $\alpha$-cellulose powder as a experimental groups for 30 days. The bleeding time and whole blood clotting time were significantly(P<0.05) increased by feeding diet containing 25% boiled eggs compared to groups of basal diet, 50% or 95% boiled eggs diets. The plasma clotting time was high in group of 25% boiled eggs diet. However, there were no difference in plasma clotting time among rats fed the dietary boiled eggs. The levels of plasma total cholesterol(TC) and low density plus very low density lipoprotein cholesterol(LDL, VLDL-C) were significantly(P<0.05) highest in group 95% boiled eggs diet compared to others. There were no differences in high density lipoprotein cholesterol(HDL-C) among rats fed the dietary boiled eggs. The levels of plasma TC, HDL-C, LDL$.$VLDL-C and the ratios of HDL-C/TC were not significant among the basal diet, 25% and 50% boiled eggs diets. These results suggest that the intakes of the dietary boiled eggs have the antithrombotic activity and plasma cholesterol lowering effect.

Anticoagulant Activity of Ilexoside D, a Triterpenoid Saponin from ilex pubescens

  • The anti-coagulant activity of ilexoside D isolated from the roots of ilex pubescens Hook. et Am. was investigated in in vivo models of blood coagulation in rats. On oral administration, ilexoside D prolonged the bleeding time and the whole blood recalcified clotting time, but not the plasma recalcified clotting time. In vitor, ilexoside D did not affect the recalciffed clotting times of whole blood, platelet-rich plasma(PRP), and platelet-poor plasma(PPP), while in the presence of tissue factor the compound prologed the reduced proth-rombin times of whole blood, PRP and PPP in the dose-dependent manner. These results indicate that ilexoside D has the anit-tissue factor activity as well as the antithromobotic activity.

Hypolipidemic and Antithrombotic Effects of Increasing Intake of Linolenic Acid Derived from Perilla Oil in Rats

  • This study investigated the hypolipidemic and antithrombotic effects of linolenic acid derived from Korean perilla oil. The experimental rats(male, Sprague-Dawley) were divided into 5 groups using a Randomized Complete Block Design and fed one of the five following diets : DO*/O#. D4/O, D4/4, D4/8, or D4/20(D*/# represents the ratio of linoleic to linoenic acid as the percentage of total dietary energy intake) for 4 or 8 months. Bleeding time and whole blood clotting time were determined and the composition of serum and platelet lipids analyzed. Comparisons from the DO/O to the D4/20 group showed that serum lipids (total lipid, triglyceride, total cholesterol, and HDL-cholesterol) gradually decreased with increasing linolenic acid intake - the hypolipidemic effect. The composition of platelet fatty acids[the ratio of eicosapentaenoic acid(EPA)/arachidonci aci(AA)] increased gradually with increasing linolenic acid intake. Higher linolenic acid intake increased bleeding time and whole blood clotting time, and decreased malondialdehyde(MDA) production in the platelets, though no significant differences. These results suggest that linolenic acid derived from perilla oil appears to suppress the conversion of linoleic acid to AA and the EPA transformed from linolenic acid appears to suppress the conversion of AA to TXA2. Since TXA2 is a platelet-aggregating and vasoconstricting agent, the redulction of TXA2 released by platelets with increasing intake of perilla oil containing a lot of linolenic acid confers an antithrombotic effect.

혈전용해효소의 경구투여에 의한 생체 내 작용 (In vivo Biological Function of a Fibrinolytic Enzyme after Oral Adminstration)

  • Bacillus subtilis BB-1 (KFCC l1344P)으로부터 분리된 혈전용해효소 유전자 (BCF-1)를 대량발현 벡터인 pEB 벡터에 크로닝하여 순수분리 된 혈전용해효소를 rat 경구 투여하여 출혈시간, 혈액의 응고, serum의 혈전용해능 등에 대한 in vivo 실험을 실시하였으며, 혈전용해효소의 단회경구투여에 따른 독성을 검사하였다. 효소의 경구투여에 따른 rat의 출혈시간에서는 대조군에 비하여 모든 경구 투여군에서 출혈시간이 유의적으로 약 1.75배 이상 길게 나타남을 확인하였고(P<0.05), 혈액의 출혈시간 또한 활발히 진행됨을 관찰하였다. 혈액으로부터 분리된 serum의 혈전용해작용 있어서는 경구투여 후 1시간부터 채혈한 혈액 내에서 혈전용해효소의 활성이 검출되기 시작하여 3시간째까지 높은 활성을 보였으며 4시간째부터 서서히 활성이 감소하는 것을 확인하였고 혈액의 응고 역시 대조군에 비하여 경구 투여군에서 상당히 지연되는 것을 알 수 있었다. Western blot에 의한 효소 검출에서는 경구 투여군에서 30,000 Da 크기의 단일밴드를 확인하였으며, 혈전용해효소의 rat에 대한 단회경구투여 독성실험에서 중량의 변화, 장기의 이상여부, 사망률 등에서 어떠한 이상이나 병변이 발견되지 않았다. 이상의 결과로 동물실험을 통한 혈전용해 효소의 경구투여에 의한 작용을 혈액 내에서 확인 할 수 있었으며, 본 효소의 단회 경구투여 시의 독성은 전혀 없음을 확인할 수 있었다.

해조 올리고당 음료가 고지혈증 유도 쥐에 있어서 혈액 지질 산화 및 혈액ㆍ혈장 응고에 미치는 영향 (Effect of Seatangle Oligosaccharide Drink on Oxidation of Serum Lipid and Bleeding and Plasma Clotting Time in Rats Fed a Hyperlipidemic Diet)

  • 식이성 고지혈증 model 실험동물을 사용하여 해조 음료를 투여하여 지질 산화와 관련된 효소 변화 및 혈액응고에 미치는 영향을 시험하였다. 혈액의 lipid peroxide 함량과 혈중 hydroxy radical 생성이 식이성 고지혈증의 유도로 증가되었으며, 혈중 superoxide dismutase의 활성은 오히려 감소되던 것이 해조 음료의 투여로 조절되었다. 식이성 고지혈증을 유도한 흰쥐에서 감소되었던 bleeding time과 plasma clotting time이 해조 음료의 투여로 다소 증가하였고, 증가하였던 tissue factor의 활성은 현저히 감소되었다 동물 소화장관에 대한 운동성 시험에서는 해조 음료의 투여로 장관 운동이 증가되는 것을 확인하였다. 이러한 결과는 해조 음료나 해조 음료 소재에 다량 함유되어 있는 해조 올리고당이 기존의 해조 다당과 마찬가지로 혈관계 관련 질병의 예방에 유효한 기능을 할 수 있다는 것을 나타내며 향후로도 다양한 기능성 제품의 소재가 될 수 있을 것으로 여겨진다.

Bleeding Time Prolongation Effect of Methanol Extract of Viscum album var. coloratum

  • The methanol extract of Viscum album var. coloratum, Korean mistletoe, showed potent prolongation effects on the bleeding time in rats in vivo, and whole blood clotting time and plasma recalcification time in rats ex vivo. The prolongation effect on the bleeding time of Korean mistletoe is comparable to that of Viscum album L., European mistletoe, 185.6% and 176.5%, respectively. However, the water extracts of the both plants did not show any prolongation effects. Platelet activating factor (PAF) receptor binding assay was carried out to elucidate the action mechanisms of the extracts, and both of the methanol extracts did not show any inhibitory activity. The $LD_{50}$ of the methanol extracts of both mistletoes are more than 2 g/kg. These results suggest that the mehtanol extract of Korean mistletoe might be a potential candidate to develop new drug to improve microcirculation.

신체냉각이 혈액세포성분 및 응혈기전에 미치는 영향에 관한 실험적 연구 (Hematologic and Coagulation Changes in Hypothermic Dogs)

  • This experiment was carried out to study the responses of cellular component of blood and bone marrow to cold and also the changes of coagulation during cooling. Forty-two mongrel dogs were subjected to hypothermia by ice-water surface cooling technique. Lowest body temperature ranged from 21-23 degree. Dogs were divided into 3 groups,Group I, 12 dogs: pentothal anesthesia for 3 hours, Group II, 20 dogs;hypothermic group and Group III,10 dogs;postsplenectomy hypothermic group. Results were summarized as follows: 1. Hemoglobin, hematocrit and red blood cell count significantly increased when animals were cooled, and increase was noted in similar magnitude among the animals of Group I. 2. White blood cell count extremely decreased after cooling and effect of splenectomy on white blood cell count was not apparent. No significant changes were seen among Group I. 3. Differential count of white blood cell when cooled showed relative increase of polymorphonuclear neutrophil and decrease of lymphocyte. 4. There was marked decrease of platelets when body temperature reached to 21-23degree and essentially. no changes was noted in Group I. 5. Clotting time, bleeding time, plasma prothrombin time, recalcification time, and fibrinolysis showed no significant changes when dogs were cooled. Clot retration and prothrombin consumption during hypothermia appeared to be poor. In Group II, bleeding time decreased after splenctomy and when body temperature was lowered, plasma prothrombin time, clot retraction, and prothrombin consumption decreased. Decreased bleeding time and poor clot retraction were noted in Group I. 6. It was found that megacaryocyte count decreased even though platelet count of peripheral blood markedly diminsished when animals were cooled. There was some tendency of erythroid hyperplasia noted during hypothermia.

도홍음(桃紅飮)이 Collagen 등(等)으로 유발(誘發)한 혈전(血栓) 및 혈액점도(血液粘度)에 미치는 영향(影響)에 관(關)한 연구(硏究)

  • This study was performed to prove the antithrombotic effects of DoHongYeum fluid by way of experimental methods. The thrombosis was induced by injection of collagen the mixture (0.1ml/10g, 2mg/kg B.W)plus serotonin (5mg/kg B.W) into the caudal vein of rat, 2 hours after liquid extract of DoHongYeum was oral administration. The effect of the fluid of DoHongYeum was rxamined by the number of RBC and platelets, bleeding time, blood clotting time, death rate, platelet aggregation, plasmacoagulation factor activity, exvivo and fibrinolytic activity of englobulin fracture in the rats. The results were summerized as followings. 1. The number of RBC and platelets was significantly increased in DoHomeYeum group incomparison with the control group. 2. Bleeding time was significantly shortened in DHY group in comparison with the controlgroup. 3. Blood clotting time was significantly prolonged in DHY group in comparison with the control group. 4. The death rate of mouse was inhibited in DHY group in comparison with the control group. 5. The platelet aggregation was inhibited in DHY group in comparison with the control group. 6. The prothrombin time and activated partial thromboplastin time on the test of plasmacoagulation factor activity was prolonged but was not valuable in DHY group. 7. Fibrinogen lyses time of rat was reduced and lyses area was increased in DHY group incomparison with the control group. 8. Fibrinogen lyses time of rat in vitro assay was reduced in DHY group. From the above results, it was thought that the DoHongYeum could be applied effectively in the thrombosis.

제독처리한 복어 간유의 생리활성 (Bioactive Functions of Detoxified Puffer Liver Oil)

  • 인류는 질병에 대한 예방 및 치료에 주로 합성 의약품을 사용하여 왔으나 근래에 이르러 여러 가지 합성 의약품의 부작용 또는 독성이 밝혀짐에 따라 세계적으로 천연자원으로부터의 의약품 개발이 활발히 추진되고 있다. 한편, 우리나라를 위시한 동양에서는 오랜 시일을 통하여 많은 식물을 치료제로 사용하여 왔으나 약화학적 및 약리학적 검토 자료는 지금에 와서 많이 도출되고 있으며 다양한 연구가 이루어지고 있는 실정이다. 본 연구에서는 해양생물로부터 생리활성을 검색할 목적으로 검은 밀복으로부터 제독처리하여 추출, 정제한 검은 밀복 간유를 대상으로 항피로효과, 간기능에 미치는 영향 및 poloxamer-407로 유도되는 고지혈증에 대한 효과를 검색한 결과, 다음과 같은 결론을 얻었다. 첫째, 식품위생학적 안전성 여부를 조사하기 위하여 검은밀복 간유의 정제품을 mouse assay로 그 잔류 독성을 조사한 결과 모두 무독하였다. 둘째, 생쥐를 유영하게 하여 사망하는 시간을 측정하여 본 결과 정상 생쥐에서는 사망시간보다 복어간유를 용량별로 2주일간 경구투여하였을때 사망 시간은 용량 의존적으로 증가되었다. 셋째, 간장해의 유발 약물로 사염화탄소 및 D-galactosamine(GaIN)을 사용하여 복어로부터 추출한 복어간유를 투여한 결과 사염화탄소 및 GaIN을 투여하였을 때 혈중 AST, ALT 및 SDH의 활성이 정상군에 비하여 현저히 증가되던 것이 복어간유의 전처리로 현저히 억제되었다. 넷째, 혈청 중 중성지방 및 total cholesterol의 함량은 정상군에 비하여 poloxamer-407의 투여로 증가되던 것이 복어간유를 용량별로 투여함으로써 현저히 감소되었다. 한편 poloxamer-407로 유도된 고지혈증의 실험동물의 bleeding time이 복어간유의 투여로 연장되었으며 plasma clotting time도 증가되었다.
