남해안 어류양식장 해수 및 양식어류의 위생지표세균 및 병원성세균 조사 (Survey of Sanitary Indicative Bacteria and Pathogenic Bacteria in Fish Farms on the Southern Coast of Korea)

  • The distributions of sanitary indicative bacteria and pathogenic bacteria in seawater and four species of farmed fishes, including oliver flounder (Paralichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagrus major) and sea bass (Lateolabrax japonicus), collected at fish farms located in the southern coastal area of Korea were investigated from May to October in 2004. The detection rates of fecal coliform and Entirococcus spp. of sanitary indicative bacteria in all samples were $38.9\%$ and $23.8\%$, respectively. The occurrence of fecal coliform was highest of $58.3\%$ in Busan, Geoje and Wando area, followed Yeosu $33.3\%$, Jeju $12.5\%$, Tongyeong $11.1\%$. The occurrence of Enterococcus spp. was highest In Wando area ($45.8\%$), followed by Yeosu ($33.3\%$), Tongyeong ($22.2\%$), Busan ($16.7\%$), Geoje and Jeju ($12.5\%$). The detection rate of fecal coliform was higher than that of Enterococcus spp., except in the Tongyeong area. There was no difference in the detection rate of fecal coliform from May to October, but the detection rate of Enterococcus spp. increased with seasonal warming seawater temperature. Among the pathogenic bacteria, the detection rate of Vibrio alginolyticus ($49.2\%$) in all samples was highest, followed by V. parahaemolyticus ($36.5\%$), Staphylococcus aureus ($6.3\%$), Salmonella sp. ($2.4\%$). However, V cholerae, V. vulnificus and Shigella sp. were not detected in all tested samples. The detection rates of V. parahaemolyticus and V. alginolyticus increased with seasonal warming seawater temperature from May to August.

HPLC를 이용한 어류 중의 Amoxicillin과 Ampicillin 항생제 동시 분석법 (Simultaneous Determination of Amoxicillin and Ampicillin in Fish Meat Using High-Performance Liquid Chromatography)

  • A simultaneous high-performance liquid chromatography assay method for amoxicillin and ampicillin in fish products was developed, evaluated, and validated by monitoring these antibiotics in fish samples obtained from aquaculture and distribution. The recovery rate of this method was higher than those of conventional methods and was 95.3-106.6% for amoxicillin and 81.4-92.4% for ampicillin. Our pretreatment procedure sufficiently removed or reduced materials affecting HPLC analysis, such as low-molecular-weight substances. The performance limit of this method was evaluated as 0.01 ppm of amoxicillin and ampicillin in fish muscle. Finally, 171 fish samples, including olive flounder (Paralichthys olivaceus), common sea bass (Lateolabrax japonicus), and black rock fish (Sebastes schlegeli) collected from fish farms in the coastal area between April and September 2005 were analyzed to evaluate the overall efficiency of the method and to monitor the actual of amoxicillin and ampicillin usage in fish farms. The results indicated that the developed method was suitable for analyzing amoxicillin and ampicillin in fish muscle, and determined that those antibiotics were being used for fish farming but were not detected in fish samples during the shipping and distribution stages.

2000년~2006년 하절기 양식어류의 병원체 감염현황 (Monitoring of Pathogens in Cultured Fish of Korea for the Summer Period from 2000 to 2006)

  • 2000년~2006년 하절기에 국내 동서남해안과 제주의 육상양식장과 가두리양식장에서 사육중인 넙치 (Paralichthys olivaceus), 대하 (Fenneropenaeus chinensis Osbeck), 조피볼락 (Sebastes schlegeli), 돌돔 (Oplegnathus fasciatus), 참돔 (Pagrus major), 감성돔 (Acanthopagrus schlegeli), 농어(Lateolabrax japinicus), 숭어 (Mugil cephalus), 무지재송어 (Onchorhynchus mykiss) 등 25종류 총 3,518마리에 대하여 병원체 감염현황을 조사하였다. 연도별로 검출된 병원체에 대하여 유형별로 감염비율을 살펴보면, 세균이 2000년 22.4%, 2001년 34.5%, 2002년 14.1%, 2003년 15.3%, 2004년 17.7%, 2005년 13.5% 및 2006년 5%로 나타났다. 기생충은 2000년 20%, 2001년 33.8%, 2002년 12.4%, 2003년 14.1%, 2004년 9.2%, 2005년 10.5% 및 2006년 10.7%로 나타났다. 바이러스는 2000년 22.4%, 2001년 13.5%, 2002년 10.3%, 2003년 5.4%, 2004년 9.7%, 2005년 10.2% 및 2006년 15.8%로 나타났다. 혼합감염은 2000년 10.3%, 2001년 0%, 2002년 44.9%, 2003년 50.9%, 2004년 31.9%, 2005년 38.4% 및 2006년 39.6%로 나타났다. 2001년까지 혼합감염은 매우 낮게 검출되었으나, 2002년부터 2006년까지 급격하게 증가하여 높은 검출빈도를 나타내었다. 조사기간을 통해 검출된 병원체 가운데 가장 높은 검출빈도를 나타낸 것은 세균이 비브리오속 (41.2%)과 연쇄구균속 (28.8%), 기생충은 원충류 (85.7%)의 스쿠티카충과 트리코디나충, 바이러스는 VNNV (42.2%)이었다.

  • The antimicrobial resistance of 160 strains of Enterococcus faecalis and 173 strains of E. faecium to 12 antimicrobial agents was investigated. The test strains were isolated from 126 wild seawater and farmed fish, including olive flounder (Paralichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagrus major), and sea bass (Lateolabrax japonicus), in 2005 and 2006. Overall, 91.9% of the E. faecalis isolates and 88.4% of the E. faecium isolates showed antimicrobial resistance to at least one antimicrobial agent. The pattern of antimicrobial resistance of the isolates differed little according to the species of fish. The percentage of E. faecalis and E. faecium with specific antimicrobial resistance differed according to the sample source. For the isolates from farmed fish samples, 66.7% of E. faecalis were tetracycline resistant and 54.5% of E. faecium were erythromycin resistant. By contrast, in the wild fish seawater samples, 92.0% of E. faecalis were rifampin resistant and 88.5% of E. faecium were tetracycline resistant.

남해안 어류양식장에서 분리된 Escherichia coli에 대한 항균제 감수성 (Antimicrobial Susceptibility of Escherichia coli Isolated from Fish Farms on the Southern Coast of Korea)

  • Three-hundred and sixteen Escherichia coli strains from seawater, and a variety of farmed fishes, including oliver flounder (Paralichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagrus major) and sea bass (Lateolabrax japonicus) between May to October in 2004, were tested by agar dilution method to determine their susceptibility patterns to 17 antimicrobial agents. Overall, 92.1% of Escherichia coli isolates from samples showed antimicrobial resistance to at least one antimicrobial agent and the multiple resistance was seen in 173 isolates (54.7%). The resistance of E. coli isolates to tetracycline (74.1%) was highest, followed by cephalothin (69.9%), doxycycline (66.5%), streptomycin (47.2%), ampicillin (46.2%), cefazolin (31.6%), enrofloxacin (31.0%). norfloxacin (28.2%). The most frequent resistance pattern was TE-D-CF-CIP-ENO-NOR-AM-S-C-SXT-AmC-CZ (14.7%), followed by CF (6.2%), TE (5.1%), TE-CF (4.5%) in 177 isolates from fishes and TE-D-CF (7.2%) followed by TE-D-CF-S (5.8%), CF and TE-D-S (3.6%) in 139 isolates from seawater.

  • 무지개 송어, 산천어, 뱀장어, 볼락 및 우럭에 대한 사료 carotenoids의 체내대사와 체색 개선효과를 검토하기 위하여, 사료에 ${\beta}-carotene$, lutein, canthaxanthin, astaxanthin 및 ${\beta}-apo-8’-carotenal$을 각각 첨가하여 4 내지 5주간 사육하여 표피의 carotenoids 성분의 변화를 분석, 비교한 결과는 다음과 같다. 무지개 송어 표피의 carotenoids 조성은, zeaxanthin, ${\beta}-carotene$ 및 canthaxanthin이 주성분이였으며, 그 외 lutein, isocryptoxanthin 및 salmoxanthin을 소량 성분으로 함유하며, 표피의 carotenoids 축적율은 canthaxanthin 첨가구에서 높게 나타나 체색 선명화 효과가 가장 컸으며, astaxanthin, ${\beta}-carotene$ 첨가구의 순으로 나타났다. 무지개 송어 표피에서의 carotenoids 대사경로는, ${\beta}-carotene$이 isocryptoxanthin, echinenone 및 canthaxanthin을 경유하여 astaxanthin으로 lutein은 canthaxanthin으로 산화되고, canthaxanthin은 isozeaxanthin을 경유하여 ${\beta}-carotene$으로 환원되며 astaxanthin은 triol을 경유하여 zeaxanthin으로 환원되는 대사경로를 추정할 수 있었다. 산천어 표피의 carotenoids 조성은, zeaxanthin이 주성분이며, 그 외 triol, lutein, tunaxanthin, ${\beta}-carotene$, ${\beta}-cryptoxanthin$ 및 canthaxanthin을 소량 성분으로 함유하며, 표피의 carotenoids 축적율은 canthaxanthin 첨가구에서 높게 나타나 체색 선명화 효과가 가장 컸으며, lutein, ${\beta}-carotene$ 첨가구의 순으로 나타났다. 산천어 표피에서의 carotenoids 대사경로는, ${\beta}-carotene$이 zeaxanthin으로 산화되고, lutein은 tunaxanthin을 경유하여 zeaxanthin으로 환원되고, canthaxanthin은 ${\beta}-carotene$을 경유하여 zeaxanthin으로 산화되며 astaxanthin은 triol을 경유하여 zeaxanthin으로 환원되는 대사경로를 추정할 수 있었다. 뱀장어 표피의 carotenoids 조성은, ${\beta}-carotene$이 주성분이였으며, 그 외 lutein, zeaxanthin 및${\beta}-cryptoxanthin$등이 소량 성분으로 함유하며, 표피의 carotenoids 축적율은 lutein 첨가구에서 높게 나타나 체색 선명화 효과가 가장 컸었고, canthaxanthin astaxanthin 첨가구의 순으로 나타났다. 뱀장어 표피에서의 carotenoids 대사경로는, ${\beta}-carotene$과 lutein은 그대로 축적되며, canthaxanthin은 ${\beta}-carotene$으로 그리고 astaxanthin은 zeaxanthin으로 환원되는 대사경로를 추정 할 수 있었다. 볼락 표피의 carotenoids 조성은, zeaxanthin, ${\beta}-carotene$, tunaxanthin A, tunaxanthin B, tunaxanthin C 및 lutein이 주성분이였으며, 그 외 ${\beta}-cryptoxanthin$, ${\alpha}-cryptoxanthin$, astaxanthin을 소량 성분으로 함유하며, 표피의 carotenoids 축적율은 lutein 첨가구에서 높게 나타나 체색 선명화 효과가 가장 컸으며, ${\beta}-carotene$, canthaxanthin 첨가구의 순으로 나타났다. 볼락 표피에서의 carotenoids의 대사경로는, ${\beta}-carotene$은 lutein으로 산화되며, lutein, canthaxanthin astaxanthin 및 ${\beta}-apo-8'-carotenal$은 zeaxanthin을 경유하여 tunaxanthin으로 각각 산화 및 환원되는 대사경로를 추정할 수 있었다. 우럭 표피의 carotenoids 조성은, ${\beta}-carotene$, astaxanthin 및 zeaxanthin이 주성분이였으며, 그 외 ${\alpha}-cryptoxanthin$, ${\beta}-cryptoxanthin$, lutein 및 canthaxanthin을 소량 성분으로 함유하며, 표피의 carotenoids 축적율은 lutein 첨가구에서 높게 나타나 체색 선명화 효과가 가장 컸으며, canthaxanthin, ${\beta}-carotene$ 첨가구의 순으로 나타났다. 우럭 표피에서의 carotenoids 대사경로는 ${\beta}-carotene$${\beta}-cryptoxanthin$으로 산화되고, lutein은 ${\alpha}-cryptoxanthin$을 경유하여 ${\beta}-carotene$으로 환원되고, canthaxanthin은 ${\beta}-cryptoxanthin$, zeaxanthin을 경유하여 ${\alpha}-cryptoxanthin$으로 환원되며, astaxanthin은 isocryptoxanthin, zeaxanthin을 경유하여 tunaxanthin으로 대사되며, ${\beta}-apo-8’-carotenal$${\beta}-cryptoxanthin$, zeaxanthin을 경유하여 ${\alpha}-cryptoxanthin$으로 환원되는 대사경로를 추정할 수 있었다.

남해안 어류양식장에서 분리된 Vibrio parahaemolyticus와 Vibrio aiginolyticus의 항균제 감수성 (Antimicrobial Susceptibility of Vibrio parahaemolyticus and Vibrio alginolyticus from Fish Farms on the Southern Coast of Korea)

  • The antimicrobial resistance patterns to the 10 antimicrobial agents of potential pathogenic vibrios isolated from seawater and a variety of farmed fishes, including oliver flounder (Payalichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagyus major) and sea bass (Lateolabyax japonicus), were investigated from May to October, 2004. A total of 314 strains of the genus vibrios were isolated from 126 collected samples, and the number of isolated strains of Vibrio payahaemolyticus and Vibrio alginolyticus were 194 and 120, respectively. Apparently $98.5\%$ of V. parahaemolyticus and $100\%$ of V. alginolyticus isolates demonstrated antimicrobial resistance against at least one antimicrobial agent. The resistance of V. parahaernolyticus isolates to ampicillin ($97.9\%$) was highest, followed by oxolinic acid ($26.8\%$), amikacin ($19.1\%$) and tetracycline and amoxicillin/clavulanic acid ($6.7\%$). V. alginolyticus isolates were resistant to ampicillin ($100\%$), solfamethoxazoleit,imethopenem ($25\%$), amikacln ($21.7\%$), amoxicillin/clavulanic acid ($15.8\%$), ciprofloxacin ($13.3\%$), and tetracycline and doxycycline ($11.7\%$). The rate of multiple antimicrobial resistance to at least four antimicrobials was higher in the V. alginolyticus isolates ($20.8\%$) than in the V. parahaemolyticus ($6.7\%$).
